Stevia rebaudiana, a South American plant normally used as a natural herbal sweetener, has been suggested as exerting beneficial effects on human health, including as an antihypertensive and antihyperglycemic. The present experiment was undertaken to evaluate the renal excretion of steviol, the aglycone of several natural products extracted from the leaves of S. rebaudiana, and to clarify the actual participation of this compound on the renal excretion of glucose in rats, which has been previously suggested as the preferential action of steviol on the Na+-glucose renal tubular transport system. Steviol was obtained by enzymatic hydrolysis of stevioside with pectinase. Thirty normal male Wistar rats weighing 345 g were used. After a control period, steviol was infused iv at three doses (0.5, 1.0 and 3.0 mg.kg-1/h), according to classical clearance techniques. During all the experiments no significant changes in inulin clearance (Cin) and p-aminohipuric acid clearance (C PAH) were observed. Administration of steviol resulted in a statistically significant increase in the fractional sodium excretion (FeNa+), fractional potassium excretion (FeK+), urinary flow as percent of glomerular filtration rate (V/GFR) and glucose clearance (C G) when compared to controls, but these effects were absent with the dose of 0.5 mg.kg-1/h. The steviol clearance (C S) was higher than the Cin and lower than the C PAH at all the doses employed in this study. The data suggest that steviol is secreted by renal tubular epithelium, causing diuresis, natriuresis, kaliuresis and a fall in renal tubular reabsorption of glucose.

Obstructive apnea (OA) can exert significant effects on renal sympathetic nerve activity (RSNA) and hemodynamic parameters. The present study focuses on the modulatory actions of RSNA on OA-induced sodium and water retention. The experiments were performed in renal-denervated rats (D; N = 9), which were compared to sham (S; N = 9) rats. Mean arterial pressure (MAP) and heart rate (HR) were assessed via an intrafemoral catheter. A catheter was inserted into the bladder for urinary measurements. OA episodes were induced via occlusion of the catheter inserted into the trachea. After an equilibration period, OA was induced for 20 s every 2 min and the changes in urine, MAP, HR and RSNA were recorded. Renal denervation did not alter resting MAP (S: 113 ± 4 vs D: 115 ± 4 mmHg) or HR (S: 340 ± 12 vs D: 368 ± 11 bpm). An OA episode resulted in decreased HR and MAP in both groups, but D rats showed exacerbated hypotension and attenuated bradycardia (S: -12 ± 1 mmHg and -16 ± 2 bpm vs D: -16 ± 1 mmHg and 9 ± 2 bpm; P < 0.01). The basal urinary parameters did not change during or after OA in S rats. However, D rats showed significant increases both during and after OA. Renal sympathetic nerve activity in S rats increased (34 ± 9%) during apnea episodes. These results indicate that renal denervation induces elevations of sodium content and urine volume and alters bradycardia and hypotension patterns during total OA in unconscious rats.

-Melanocyte-stimulating hormone (-MSH; 0.6 and 3 nmol) microinjected into the anteroventral region of the third ventricle (AV3V) induced a significant increase in diuresis without modifying natriuresis or kaliuresis. Intraperitoneal (ip) injection of -MSH (3 and 9.6 nmol) induced a significant increase in urinary sodium, potassium and water excretion. Intraperitoneal (3 and 4.8 nmol) or iv (3 and 9.6 nmol) administration of -MSH did not induce any significant changes in plasma atrial natriuretic peptide (ANP), suggesting that the natriuresis, kaliuresis and diuresis induced by the systemic action of -MSH can be dissociated from the increase in plasma ANP. These preliminary results suggest that -MSH may be involved in a -MSHindependent mechanism of regulation of hydromineral metabolism.

A regulação fina do volume e osmolaridade dos líquidos corporais é fundamental para a sobrevivência. Qualquer variação na composição do meio interno ativa mecanismos comportamentais, neurais e hormonais compensatórios que controlam a ingestão e excreção de água e eletrólitos a fim de manter a homeostase hidroeletrolítica. Alterações na faixa de 1-2% na osmolaridade sanguínea estimulam a liberação de arginina vasopressina (AVP) que resulta em antidiurese além de ocitocina (OT) e peptídeo natriurético atrial (ANP) que promovem a natriurese. Trabalhos realizados em nosso laboratório utilizando o modelo experimental de expansão do volume extracelular (EVEC) mostraram ativação de neurônios magnocelulares ocitocinérgicos localizados no núcleo paraventricular (PVN) e núcleo supra-óptico (SON) responsáveis pela secreção de OT e AVP, igualmente alteradas em resposta a este estímulo. A participação do sistema nervoso simpático nestas condições tem sido levantada. Projeções medulares e tronco-encefálicas (simpáticas) para o hipotálamo poderiam atuar de forma seletiva inibindo sinalizações para a ingestão e estimulando sinalizações para excreção de água e eletrólitos. O papel de vias noradrenérgicas tronco-encefálicas nesta regulação ainda precisa ser mais bem estabelecido. Assim sendo, objetivamos neste estudo esclarecer o papel do sistema nervoso simpático (via noradrenérgicas) na regulação das alterações induzidas pelo modelo de EVEC, analisando por cromatografia líquida de alta eficácia o conteúdo de noradrenalina (NA), adrenalina (AD) e serotonina (5-HT) em estruturas do tronco cerebral como núcleo do trato solitário (NTS), bulbo rostro-ventro lateral (RVLM), locus coeruleus (LC) e núcleo dorsal da rafe (NDR) e estruturas hipotalâmicas como SON e PVN. Procuramos ainda, através de estudos imunocitoquímicos determinar alterações no padrão de ativação neuronal pela análise de Fos-TH ou Fos-5HT nas estruturas acima mencionadas em condições experimentais nas quais são induzidas alterações do volume do líquido extracelular.

A área septal medial (ASM), situada no prosencéfalo, está envolvida na regulação cardiovascular e no controle do balanço hidroeletrolítico. Esta área é rica em receptores colinérgicos e a ativação dos mesmos induz ingestão de água, natriurese e anti-diurese. Existem evidências experimentais de que as espécies reativas de oxigênio possam participar do controle de respostas fisiológicas. Resultados recentes de nosso laboratório demonstraram que uma espécie reativa de oxigênio, o peróxido de hidrogênio (H2O2), injetada na ASM reduz a ingestão de água induzida por carbacol (agonista colinérgico) também injetado na ASM. No presente projeto propomos estudar os efeitos do aumento da produção endógena de H2O2 na ASM nos efeitos dipsogênico, natriurético e anti-diurético do carbacol injetado na ASM. O inibidor da catalase, 3 amino 1,2,4 triazole (ATZ) será injetado na ASM para aumentar os níveis endógenos de H2O2

A área septal medial (ASM), situada no prosencéfalo, está envolvida na regulação cardiovascular e no controle do balanço hidroeletrolítico. Esta área é rica em receptores colinérgicos e a ativação dos mesmos induz ingestão de água, natriurese e antidiurese. A ASM também envia projeções aos núcleos paraventricular (NPV) supra-óptico (NSO), os quais contêm os neurônios que secretam vasopressina e ocitocina. Existem evidências experimentais demonstrando que as espécies reativas de oxigênio podem participar do controle de respostas fisiológicas. Resultados recentes de nosso laboratório demonstraram que uma espécie reativa de oxigênio, o peróxido de hidrogênio (H2O2), injetado no ventrículo lateral (VL) reduz a ingestão de água e a resposta pressora induzida por ANG II e carbacol (agonista colinérgico) também injetados no VL. Por isso, o presente estudo teve como objetivo estudar os efeitos da injeção de H2O2 na ASM sobre a ingestão de água, sobre a excreção renal de água e eletrólitos e sobre a expressão da proteína c-Fos no NSO produzidos pela injeção de carbacol também na ASM. Para realizar este trabalho, foram utilizados ratos com cânulas de aço inoxidável implantadas na ASM. A ingestão de água e a excreção renal de água e eletrólitos foram estudadas em ratos que receberam injeções de H2O2 (5 mol/0,5 μl) ou PBS (veículo, 0,5 l) na ASM e, após um minuto, injeção de carbacol (4 nmol/0,5 l) ou salina (NaCl 0,15 M / 0,5 l) também na ASM. A ingestão de água induzida pelo carbacol, através da estimulação colinérgica, foi menor nos ratos que receberam a injeção prévia de peróxido de hidrogênio (8 ± 2,0 ml / 1 h, p<0,05) comparado àqueles que receberam veículo, também na ASM (16,6 ± 1,9 ml / 1 h, p<0,05). Nitric oxide (NO) is a free radical gas, inorganic, which has seven electrons of nitrogen and oxygen eight, possessing an unpaired electron. This radical is produced from L-arginine by a reaction mediated by the enzyme NO synthase. NO it is about a radical of who acts abundant on a variety of biological processes, particularly when produced by endothelial cells plays a significant role in cardiovascular control, as a modulator of peripheral vascular resistance and platelet aggregation. This free radical has also a neurotransmitter and mediator of the immune system. NO kidney function has been considered in many physiological functions such as: (a) regulation of hemodynamics and glomerular function tubuloglomerular, (b) participation in pressure natriuresis (c) maintaining medullar perfusion (d) inhibiting sodium reabsorption tubular, and (e) acting as a modulator of the activity of the sympathetic nervous system. Given these functions, the occurrence of its deficiency is associated with chronic kidney disease (CKD) in vasoconstriction and consequently glomerular hypertension, high blood pressure (HBP), proteinuria and progression of renal dysfunction. This work has the scope to describe the role of NO in renal physiology and pathophysiology of CKD.
