L'ototoxicitat per cisplatí es presenta fins en un 70% dels pacients i pogués estar relacionada amb els haplogrups de l'ADN mitocondrial de cada individu. Material i Mètode: es va determinar el haplogrup de 26 pacients tractats amb cisplatí i es va realitzar un seguiment audiomètric durant el tractament. Resultats: 61,5% va presentar pèrdua auditiva significativa. Haplogrups: 16 pertanyents al grup HV, 6 al UK, 3 al WIX i 1 al JT. No es va trobar relació entre la pèrdua auditiva i els haplogrups de l'ADN mitocondrial. Conclusió: L'ototoxicitat per cisplastí no guarda relació amb els haplogrups de l'ADN mitocondrial.

INTRODUCCIÓ L’angiogènesi és el procés que condueix a la formació de nous vasos sanguinis a partir de la vascularització preexistent. Disposem en l’actualitat de diversos fàrmacs que inhibeixen un factor proangiogènic, el factor de creixement de l’endoteli vascular (VEGF). L’objectiu del nostre estudi és determinar l’efectivitat de la mol·lècula bevacizumab (Avastin ®) en el tractament de diverses patologies amb neovascularització i/o edema macular. MATERIAL I MÈTODES Estudi prospectiu obert no randomitzat. Àmbit d’estudi: pacients del Servei d’Oftalmologia de l’Hospital Municipal de Badalona. Població a estudi: pacients amb Oclusió venosa de l’artèria central de la retina i oclusió de branca venosa / Edema macular diabètic / Edema macular uveític / Retinopatia diabètica proliferativa / Glaucoma neovascular / Metàstasi a iris de tumor oat cell de pulmó Metodologia: comparació de l’espessor macular central o de la neovascularització, abans i després del tractament RESULTATS En tots els subgrups a estudi s’observa millora del paràmetre estudiat globalment. Pacients amb edema macular diabètic: milloren agudesa visual 0’22 de mitjana, 169 micres de disminució promig de l’edema. Pacients amb edema macular secundari a oclusió venosa: milloren agudesa visual 0’23 de mitjana, 143 micres de disminució promig de l’edema. Pacients amb edema macular uveític: milloren agudesa visual 0’22 de mitjana, amb una disminució promig de l’edema macular de 263 micres. Pacients amb retinopatia diabètica, glaucoma neovascular o metàstasi iridiana: regressió parcial/completa de la neovascularització. CONCLUSIONS El nostre estudi suggereix que el bevacizumab intravitri és una teràpia eficaç i segura en casos d’edema macular o neovascularització de diversa etiologia diferent de DMAE, encara que d’efecte temporal. Cal tenir en compte els riscos locals i sistèmics, així com el seu ús compassiu. Serien necessaris estudis prospectius aleatoris més extensos i amb major temps de seguiment per reforçar els resultats favorables actuals d’aquest tractament.

Objectiu: comparar la eficàcia entre Ranibizumab y Bevacizumab en el tractament de la DMAE de tipus exudatiu. Material i mètode Es tracta d’un estudi retrospectiu en el que es comparen a curt termini dos fàrmacs antiangiogènics utilitzats en el tractament de DMAE exudativa per via intravitrea: el Ranibizumab iel Bevacizumab. Hem seleccionat pacients amb DMAE exudativa tractats amb teràpia antiangiogènica i s’han distribuït en dos grups segons el tractament realitzat. Els pacients van ser tractats en el període comprés entre Gener del 2007 i Novembre del 2007, deprés d’uns mesos de l’aprovació del Ranibizumab intravitri per tractament de DMAE per la FDA. Resultats: La diferència entre l’AV mitja abans i després del tractament no era estadísticament significativa en cap dels dos grups tractats, mentre que la diferència entre el gruix macular antes i després del tractament en ambdós grups ha millorat de manera estadísticament significativa. Comparem a continuació el grau de millora per OCT en ambdós grups. Procedim a l’aplicació del test de Mann-Whitney per comparar si existeix diferència en la disminució del gruix de la màcula entre els dos grups. Evidenciem que la diferència no era clínicament significativa (p = 0.705) Conclusió: Els resultats obtinguts en el nostre estudi no mostren diferència entre els dos fàrmacs en el tractament de la DMAE exudativa a curt termini però reforcen la necessitat de futurs estudis prospectius i randomitzats

C75 is a synthetic compound described as having antitumoral properties. It produces hypophagia and weight loss in rodents, limiting its use in cancer therapy but identify- ing it as a potential anti-obesity drug. C75 is a fatty acid synthase (FAS) inhibitor and, through its coenzyme A (CoA) derivative, it acts as a carnitine palmitoyltransferase (CPT) 1 inhibitor. Racemic mixtures of C75 have been used in all the previous studies; however, the potential dif- ferent biological activities of C75 enantiomers have not been examined yet. To address this question we synthesized the two C75 enantiomers separately. Our results showed that ( )- C75 inhibits FAS activity in vitro and has a cytotoxic effect on tumor cell lines, without affecting food consumption. (+)-C75 inhibits CPT1 and its administration produces anorexia, suggesting that central inhibition of CPT1 is essential for the anorectic effect of C75. The differential activity of C75 enantiomers may lead to the development of potential new specific drugs for cancer and obesity.

Pd-catalysed reactions of 2-Cl, 2-Br and 2-I derivatives of a 6-chloropurine nucleoside with benzamide have been compared, using Pd2dba3, Xantphos and Cs2CO3 in toluene, between 20 and 80 °C. The reactivity order was 2-I > 2-Br > 6-Cl ≫ 2-Cl. The 2-I substituent could be replaced even at 0 °C, under conditions disclosed here for the first time. On the other hand, the replacement of the chlorine atom at position 2 (2-Cl) required 110 °C.

Lamellarins are a large family of marine alkaloids with potential anticancer activity that have been isolated from diverse marine organisms, mainly ascidians and sponges. All lamellarins feature a 3,4-diarylpyrrole system. Pentacyclic lamellarins, whose polyheterocyclic system has a pyrrole core, are the most active compounds. Some of these alkaloids are potently cytotoxic to various tumor cell lines. To date, Lam-D and Lam-H have been identified as lead compounds for the inhibition of topoisomerase I and HIV-1 integrase, respectively nuclear enzymes which are over-expressed in deregulation disorders. Moreover,these compounds have been reported for their efficacy in treatment of multi-drug resistant (MDR) tumors cells without mediated drug efflux, as well as their immunomodulatory activity and selectivity towards melanoma cell lines. This article is an overview of recent literature on lamellarins, encompassing their isolation, recent synthetic strategies for their total synthesis, the preparation of their analogs, studies on their mechanisms of action, and their structure-activity relationships (SAR).

Kahalalide compounds are peptides that are isolated from a Hawaiian herbivorous marine species of mollusc, Elysia rufescens, and its diet, the green alga Bryopsis sp. Kahalalide F and its synthetic analogues are the most promising compounds of the Kahalalide family because they show anti-tumoral activity. Linear solid-phase syntheses of Kahalalide F have been reported. Here we describe several new improved synthetic routes based on convergent approaches with distinct orthogonal protection schemes for the preparation of Kahaladide analogues. These strategies allow a better control and characterization of the intermediates because more reactions are performed in solution. Five derivatives of Kahalalide F were synthesized using several convergent approaches.

Molecular characterization of radical prostatectomy specimens after systemic therapy may identify a gene expression profile for resistance to therapy. This study assessed tumor cells from patients with prostate cancer participating in a phase II neoadjuvant docetaxel and androgen deprivation trial to identify mediators of resistance. Transcriptional level of 93 genes from a docetaxel-resistant prostate cancer cell lines microarray study was analyzed by TaqMan low-density arrays in tumors from patients with high-risk localized prostate cancer (36 surgically treated, 28 with neoadjuvant docetaxel þ androgen deprivation). Gene expression was compared between groups and correlated with clinical outcome. VIM, AR and RELA were validated by immunohistochemistry. CD44 and ZEB1 expression was tested by immunofluorescence in cells and tumor samples. Parental and docetaxel-resistant castration-resistant prostate cancer cell lines were tested for epithelial-to-mesenchymal transition (EMT) markers before and after docetaxel exposure. Reversion of EMT phenotype was investigated as a docetaxel resistance reversion strategy. Expression of 63 (67.7%) genes differed between groups (P < 0.05), including genes related to androgen receptor, NF-k B transcription factor, and EMT. Increased expression of EMT markers correlated with radiologic relapse. Docetaxel-resistant cells had increased EMT and stem-like cell markers expression. ZEB1 siRNA transfection reverted docetaxel resistance and reduced CD44 expression in DU-145R and PC-3R. Before docetaxel exposure, a selected CD44 þ subpopulation of PC-3 cells exhibited EMT phenotype and intrinsic docetaxel resistance; ZEB1/CD44 þ subpopulations were found in tumor cell lines and primary tumors; this correlated with aggressive clinical behavior. This study identifies genes potentially related to chemotherapy resistance and supports evi-dence of the EMT role in docetaxel resistance and adverse clinical behavior in early prostate cancer.

Barmumycin was isolated from an extract of the marine actinomycete Streptomyces sp. BOSC-022A and found to be cytotoxic against various human tumor cell lines. Based on preliminary one- and two-dimensional 1H- and 13C-NMR spectra, the natural compound was initially assigned the structure of macrolactone-type compound 1, which was later prepared by two different routes. However, major spectroscopic differences between isolated barmumycin and 1 led to revision of the proposed structure as E-16. Based on synthesis of this new compound, and subsequent spectroscopic comparison of it to an authentic sample of barmumycin, the structure of the natural compound was indeed confirmed as that of E-16.

Somatostatin analogues (SAs) are potential anticancer agents. This study was designed to investigate the expression of somatostatin receptors (SSTRs) in melanoma cells and the effect of two SAs on cell proliferation and viability. Eighteen primary and metastatic human cutaneous melanoma cell lines were treated with octreotide and SOM230. Expression of SSTR1, SSTR2, SSTR3 and SSTR5 was assessed by real-time polymerase chain reaction. Proliferation, viability and cell death were assessed using standard assays. Inhibition was modelled by mixed-effect regression. Melanoma cells expressed one or more SSTR. Both SAs inhibited proliferation of most melanoma cell lines, but inhibition was less than 50%. Neither SA affected cell viability or induced cell death. The results suggest that melanoma cell lines express SSTRs. The SAs investigated, under the conditions used in this study, did not, however, significantly inhibit melanoma growth or induce cell death. Novel SAs, combination therapy with SAs and their anti-angiogenic properties should be further investigated.

Four new metal complexes {M = Pd(II) or Pt(II)} containing the ligand 9-aminoacridine (9AA) were prepared. The compounds were characterized by FT-IR and 1H, 13C, and 195Pt NMR spectroscopies. Crystal structure of the palladium complex of formulae [Pd(9AA)(μ-Cl)]2 · 2DMF was determined by X-ray diffraction. Two 9-acridine molecules in the imine form bind symmetrically to the metal ions in a bidentate fashion through the imine nitrogen atom and the C(1) atom of the aminoacridine closing a new five-membered ring. By reaction with phosphine or pyridine, the Cl bridges broke and compounds with general formulae [Pd(9AA)Cl(L)] (where L = PPh3 or py) were formed. A mononuclear complex of platinum of formulae [Pt(9AA)Cl(DMSO)] was also obtained by direct reaction of 9-aminoacridine and the complex [PtCl2(DMSO)2]. The capacity of the compounds to modify the secondary and tertiary structures of DNA was evaluated by means of circular dichroism and electrophoretic mobility. Both palladium and platinum compounds proved active in the modification of both the secondary and tertiary DNA structures. AFM images showed noticeable modifications of the morphology of the plasmid pBR322 DNA by the compounds probably due to the intercalation of the complexes between base pairs of the DNA molecule. Finally, the palladium complex was tested for antiproliferative activity against three different human tumor cell lines. The results suggest that the palladium complex of formula [Pd(9AA)(μ-Cl)]2 has significant antiproliferative activity, although it is less active than cisplatin.

Chemoprevention is a pragmatic approach to reduce the risk of colorectal cancer, one of the leading causes of cancerrelated death in western countries. In this regard, maslinic acid (MA), a pentacyclic triterpene extracted from wax-like coatings of olives, is known to inhibit proliferation and induce apoptosis in colon cancer cell lines without affecting normal intestinal cells. The present study evaluated the chemopreventive efficacy and associated mechanisms of maslinic acid treatment on spontaneous intestinal tumorigenesis in ApcMin/+ mice. Twenty-two mice were randomized into 2 groups: control group and MA group, fed with a maslinic acid-supplemented diet for six weeks. MA treatment reduced total intestinal polyp formation by 45% (P,0.01). Putative molecular mechanisms associated with suppressing intestinal polyposis in ApcMin/+ mice were investigated by comparing microarray expression profiles of MA-treated and control mice and by analyzing the serum metabolic profile using NMR techniques. The different expression phenotype induced by MA suggested that it exerts its chemopreventive action mainly by inhibiting cell-survival signaling and inflammation. These changes eventually induce G1-phase cell cycle arrest and apoptosis. Moreover, the metabolic changes induced by MA treatment were associated with a protective profile against intestinal tumorigenesis. These results show the efficacy and underlying mechanisms of MA against intestinal tumor development in the ApcMin/+ mice model, suggesting its chemopreventive potential against colorectal cancer.
