The new flavonoid glycoside kaempferol-3-O-α-L-rhamnopyranosyl(1→2)-O-[α-L- rhamnopyranosyl(1→6)]-O-β-D-galactopyranoside-7-O-α-L- rhamnopyranoside was isolated together with (S)-zierin from the leaves of Zollernia ilicifolia (Fabaceae), a medicinal plant used as analgesic and antiulcerogenic effects in Brazilian Tropical Atlantic Rain Forest. The structures were established on the basis of 1H, 13C NMR and 2D NMR (COSY, HMBC, HMQC), UV, MS and IV spectra. The infusion of Zollernia ilicifolia was qualitatively compared to the infusion of the espinheiras-santas (Maytenus aquifolium and Maytenus ilicifolia) by HPLC-DAD.

Reactive oxygen species (ROS) and free radical species have been implicated in initiating, accompanying or causing many diseases in living organisms; there is thus, a continual need for antioxidants molecules to inactivate ROS/free radicals. Many studies of plants crude extracts have demonstrated free-radical scavenging and antioxidant action. Maytenus species have long been used, in several countries, as traditional medicines against gastric ulcers, dyspepsia and others gastric problems and for their anti-inflammatory properties. In this study, Maytenus aquifolium (Celastraceae) root bark ethanol extract was assessed for its ability to scavenge free radicals and reactive oxygen species. The results were expressed as percentage inhibition of the active species. The extract was efficient against studied reactive species: DPPH radical (obtained inhibition = 35.5 ± 1.3 %), ABTS.+ (IC50 = 0.0036 ± 0.0003 mg/mL), HOCl (IC50 = 0.002 ± 0.0001 mg/mL), O2 .- (obtained inhibition = 36.0 ± 2.1 %), and NO. (obtained inhibition = 18.3 ± 0.4 %). Uniterms Oxidant species Free radicals Maytenus aquifolium Oxidative damage.

Maytenus ilicifolia and Maytenus aquifolia (Celastraceae) both designated espinheira-santa have proven anti-ulcer activity. Morphologic similarities between leaves of espinheira-santa and mata-olho (Sorocea bonplandii), has motivated fakes in the market of phytotherapy. The present work consisted of the anatomical study, including stem and leaf, of the species M. ilicifolia, M. aquifolia and S. bomplandii. Samples of adult leaves and stem of plants located in the cities of Maringá and Marialva were collected. Both are located in the northwest region of Paraná State. The botanical material was prepared with using usual techniques of anatomy. The leaves of both Maytenus species presented great similarities, characterizing itself for the presence of epidermal cells with straight walls, biseriate palisade tissue, petiole vascular system represented by unique amphicribal bundle and sclereids, which were present in the stems of these two species. S. bomplandii leaves differed of Maytenus species for presenting epidermal cells with undulated walls, uniserite palisade tissue, petiole vascular system represented by many collateral bundles and gelatinous fibres. Non-glandular trichomes, glandular trichomes, and laticifer only occur in S. bomplandii.

Maytenus aquifolium (Celastraceae), Sorocea bomplandii (Moraceae) and Zolernia ilicifolia (Fabaceae) are native plants from the Tropical Atlantic Forest (Mata Atlantica, Brazil) known a 'espinheira-santa'. These plants are traditionally used as analgesic and antiulcerogenic medicine, with the same traditional uses of the true 'espinheira-santa' (Maytenus ilicifolia, Celastraceae), an efficient antiulcerogenic agent. Pharmacological and toxicological studies with these plants have not been carried out. The purpose in this study was to evaluate the efficacy (analgesic and antiulcerogenic activities), safety (acute toxicity) and quality (phytochemical profile) of these three plants. The analgesic effect was analyzed by writhing and tail flick tests, while anti Ulcerogenic effect was performed through ulcer induction by ethanol and indomethacin/bethanecol assays. LD., and acute toxic effects, as well as phytochemical profiles of all plants also were carried. Surprisingly, the three plants showed analgesic and antiulcerogenic effects at dose of 1000 mg/kg, v.o. Maytenus aquifolium lowering all ulcerogenic parameters (ethanol test), but increased the ulcerogenic effects in the indoniethacin/bethanecol test. Sorocea bomplandii produced antiulcerogenic effects in both experimental models used, while Zolernia ilicifolia showed significant effects only in indomethacin/bethanecol-induced gastric lesions. Pre-treatment with Zolernia ilicifolia induced someone toxic effects, A phytochemical profile for each plant species was determined and its main chemical classes of compounds were described. Essa nota registra a recoleta e a primeira informação sobre a distribuição de Maytenus comocladiaeformis Reissek. A espécie era conhecida por apenas duas coletas pouco informativas, a última delas de 1950, e era considerada extinta.

Maytenus s. l. (including Gymnosporia) is a morphologically diverse genus of about 300 species that is widely distributed in the tropics and subtropics of both the Old and New Worlds. Its delimitation has been extensively debated and despite the segregation of Gymnosporia, Maytenus s. s. remains a heterogeneous, polyphyletic group. To delimit natural segregate genera we increased taxon sampling and generated sequences from two nuclear gene regions (ITS and 26S rDNA) and two plastid loci (matK and trnL-F) to analyze together with morphological characters. Both Moya and Tricerma were found to be nested within the New World Maytenus and are recognized as synonyms of Maytenus s. s.. In contrast, the three New World species of Gymnosporia are recognized as a new genus that is closely related to Gyminda. Haydenia is erected for these three species: H. gentryi, H. haberiana, and H. urbaniana. One or more previously proposed or novel genera are required to accommodate the systematically difficult African Maytenus. Putterlickia, and most likely Gloveria, are nested within Gymnosporia and should be synonymized with that genus. New binomials are required for four Chinese and one Rapan species of Gymnosporia that have been previously treated only as Maytenus: Gymnosporia austroyunnanensis, G. confertiflora, G. dongfangensis, G. guangxiensis, and G. pertinax. Austral-Pacific Maytenus are transferred to Denhamia, requiring eight new binomials: Denhamia bilocularis, D. cunninghamii, D. cupularis, D. disperma, D. fasciculiflora, D. ferdinandii, D. fournieri, and D. silvestris. Existing intrageneric classifications of Gymnosporia and Maytenus s. s. were not supported in their entirety. Gymnosporia is inferred to have had an African origin followed by dispersals to Madagascar, southeast Asia and the Austral-Pacific.

Maytenus aquifolium (Celastraceae) and Salacia campestris (Hippocrateaceae) species accumulate friedelane and quinonemethide triterpenoids in their leaves and root bark, respectively. Enzymatic extracts obtained from leaves displayed cyclase activity with conversion of the substrate oxidosqualene to the triterpenes, 3 beta -friedelanol and friedelin. In addition, administration of (+/-)5-H-3 mevalonolactone in leaves of M. aquifolium seedlings produced radio labelled friedelin in the leaves, twigs and stems, while the root bark accumulated labelled maytenin and pristimerin. These experiments indicated that the triterpenes once biosynthesized in the leaves are translocated to the root bark and further transformed to the antitumoral quinonemethide triterpenoids. Bioactivity-guided fractionation of several bioactive extracts obtained from Cerrado and Atlantic Forest plant species led to the isolation of potent DNA-damaging piperidine 1-5 and guanidine alkaloids 6-9 from Cassia leptophylla and Pterogyne nitens respectively, two common Leguminosae from Atlantic Forest. By means of biotechnological approach on Maytenus aquifolium, a species from Cerrado, moderate DNA-damaging sesquiterpene pyridine alkaloid 10-11 was isolated. Bioassay-guided fractionation on Casearia sylvestris, a medicinal plant species found in Cerrado and Atlantic Forest, led to the isolation of clerodane diterpenes 12-13 which showed effect on DNA. In addition, we have reported several interesting potent antifungal iridoids: 1β-hydroxy-dihydrocornin (14), 1α-hydroxy-dihydrocornin (15), α-gardiol (16), β-gardiol (17), plumericin (18), isoplumericin (19), 11-O-trans-caffeoylteucrein (20); ester derivative: 2-methyl-4-hydroxy-butyl-caffeoate (21), amide N-[7-(3'.4'-methylenedioxyphenyl)-2Z, 4Z-heptadienoyl] pyrrolidine (22) and triterpene viburgenin (23).

Due to the confirmation of its medicinal properties, the demand and the area planted with 'espinheira-santa' have been increasing. However, there are no sources of propagation material with known characteristics. Progenies of Maytenus ilicifolia Martius ex Reiss (espinheira-santa), originating from Southern Brazil have been analyzed, and currently comprise a germplasm inventory for the species. Studies included the assessment of in natura growth features (height and production of biomass canopy) and plant chemistry (lignin, phenols and mineral element contents), and targeted the creation of a data base, characterizing populations/progenies to be used in future selection and breeding programs of the species. Among the features assessed, were considered desirable the ones related to productivity (high foliar mass to branch mass ratio) and also the absence of thorns on the leaves. High total phenol and tannin content were observed in all the progenies assessed and these features is desirable as long the absence of toxic effects and preservation of the therapeutic quality of the plant medicinal elements produced can be proved.
