O objetivo deste trabalho foi avaliar o efeito do choque térmico e mecânico na produtividade de Lentinula edodes em 140 toras de Eucalyptus saligna, completamente colonizadas pelo fungo, em diferentes tempos de imersão em água e no primeiro fluxo de produção. As toras foram imersas em água resfriada (16ºC) ou à temperatura ambiente (22ºC); os períodos de imersão corresponderam a 6, 10, 14, 18, 22, 26 e 38 horas; o choque mecânico foi acompanhado por três quedas consecutivas da tora, em posição vertical, no chão. A temperatura da água e o tempo de imersão afetaram a produção de L. edodes, resultando em aumentos significativos (2 a 4 vezes) nos tratamentos em que as toras foram submetidas à água resfriada e nos tempos de imersão mais curtos (6 e 10 horas). O choque mecânico não resultou em aumento na produção de basidiomas.

This study was performed to evaluate the efficiency of four different lineages (95/01, L1, 96/22 and JABK) of Lentinula edodes (BERK.) Pegler mushroom (shiitake) for inhibiting the N-ethyl-N-nitrosourea (ENU) clastogenicity in vivo. Male Swiss mice (10 animals/group) were treated during 15 consecutive days with dried mushroom added to basal diet under three different concentrations (1, 5 and 10%). At day 15, mice were intraperitoneally injected with ENU (50 mg/kg body weight) and sacrificed 24 h later for evaluation of micronucleated bone marrow polychromatic erythrocytes (MNPCE). Negative and positive controls (10 animals each), receiving basal diet and saline or ENU ip injection, respectively, were also evaluated. Results showed that pretreatments with diets containing the lineages 95/01, L1 and 96/22 reduce the frequencies of MNPCE induced by ENU. The absence of an antimutagenic activity for the lineage JABK might be related to intrinsic differences among the lineages such as biochemical composition. Taken together, our data show that the differences in protective activities of the mushrooms need to be clarified in further studies and the mechanisms for such activities need to be investigated. The radial mycelial growth of Lentinula edodes (Berk) Pegler, strain LE-96/13, was studied in culture media prepared with organic residues extract, by using substrates prepared with pineapple (Ananas comosus (L.) Merril) crown, Astrocaryum aculeatum Meyer peel, Theobroma grandiflorum Schum shell, Musa sp. (genomic group AAB, subgroup Pacovan) peel, and Musa sp. (genomic group AAB, subgroup Praia) peel, with three supplementation levels with wheat bran (0, 10 and 20%), and incubated at 25 degrees C. The experimental design was totally randomized, in a 5x3 factorial scheme, adding up 15 treatments with 4 repetitions, and each repetition corresponding to a Petri dish. The diameter of the colony was evaluated daily during nine days of incubation. After that period, it was verified that the highest mycelial growth averages of strain LE-96/13 of L. edodes were found in culture media prepared with T. grandiflorum Schum shell (whose supplementation with wheat bran was favorable for Mushroom development) and A. aculeatum Meyer peel (whose supplementation did not favor the mycelial growth of L. edodes in relation to the medium not supplemented).

This study was carried out in the towns of Dracena, Junquciropolis, Mirandopolis, Aliancas, Ilha Solteira, Castilho, Aracatuba, Birigui and Guararapes, São Paulo State, by surveying 17 Shiitake growers through a questionnaire. Data pertaining to the stages of log-Shiitake growing, recording and characterization of growers and growing systems were entered into Microsoft Excel for Windows. The results showed that Shiitake cultivation is recent and increasing in this region, and that growers have a high education level. Shiitake cultivation is mainly located in rural areas, with both Brazilians of Japanese descent and native Brazilians growing it. The most commonly used trees are eucalyptus and mango. The high level of log contamination is perhaps due to growing without temperature or moisture control and to the inappropriate growing system.. In 2004, there were 45,000 Shiitake-inoculated logs in this region, and the yield stood around 200 g of fresh mushroom/log. The mushrooms are picked in boxes of 200 g, and are sold mainly to Ceasa in open markets.

This work aimed to study the stationary and periodically mixed culture of L. edodes to the production of lignocellulolitic enzymes activity. LE 95/17, LE 96/22 and Leax strains were incubated in 25 g of eucalyptus sawdust substrate in Erlenmeyer flasks in stationary culture at 25 degrees C and in a bioreactor with four complete rotations daily at 25 degrees C and 3% CO2. The samples were collected at 8, 11, 14, 17 and 20 days after the incubation. Oxidative and hydrolytic enzymes analyses were performed. Lignin peroxidase enzyme was not found in the lignolytic systernfor LE 95/17, LE 96/22 and Leax strains in the different incubation methods. The use of bioreactor could be a practicable system to induce the laccase activity for L22 and Leax and MnP activity for L17 and L22. The activity of the hydrolytic enzymes was higher in the stationary system in comparison to periodically mixed system in the bioreactor.

Basidyomycete Lentinula edodes has its related enzymatic activity mainly on the agribusiness waste kind used as a substrate. The objective of this work was to verify the activity of oxidative enzymes lacase (Lac), lignina peroxidase (LiP) and manganese peroxidase (MnP) of three L. edodes strains, in stationary system, cultivated the 25 degrees C, in the absence of light, in substrates wtih 20% of bran of rice, 1% of CaCO(3) and 79% of rice husk (CA), eucalyptus sawdust (SE), cassava bagasse (BM) and sugarcane bagasse (BC), adjusted to 60% of humidity. The Lac and MnP activities were bigger in eucalyptus sawdust (SE) and sugarcane bagasse (BC). The UP activity was not induced for tested substrates. The rice husk (CA) and cassava bagasse (BM) Substrates, although are not adequate to produce Lac or MnP, can be used as additives to increase the porosity, air availability and easy metabolism polysaccharides.

O Lentinula edodes é um alimento de qualidade elevada, sendo rico em proteínas, vitaminas e sais minerais e pobre em calorias e gorduras. No entanto, seu valor nutricional varia em função da linhagem cultivada, do processamento após a colheita, do estágio de desenvolvimento do basidioma e do substrato utilizado. Assim, este trabalho teve como objetivo caracterizar bromatologicamente os basidiomas de oito linhagens de L. edodes (LE-95/01, LE-95/02, LE-95/07, LE-96/17, LE-96/18, LE-98/47, LE-98/55 e LE-96/13), inteiros ou em partes (estípete e píleo), produzidos em toras de E. grandis. Verificou-se que tanto as linhagens de L. edodes como as partes dos basidiomas analisados influenciaram nas respostas nutricionais das amostras. O estípete apresentou as maiores médias de fibra bruta e as menores médias de proteína bruta. O píleo, por sua vez, apresentou as maiores médias de cinzas. Nas avaliações de basidiomas inteiros, as maiores médias, quanto ao teor de proteína bruta, foram obtidas nas linhagens LE-95/07, LE-96/17 e LE-96/18. Já a maior média de fibra bruta foi obtida pela linhagem LE-96/13 de L. edodes. O conteúdo de lipídios do píleo e do estípete não variaram entre as linhagens de L. edodes.

O valor nutricional do Lentinula edodes (Berk.) Pegler varia em função da linhagem cultivada, do processamento após colheita, do estágio de desenvolvimento do basidioma e do tipo de substrato utilizado. Este trabalho teve como objetivo avaliar nutricionalmente basidiomas de L. edodes em função da linhagem e do tipo de eucalipto cultivado. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 2 x 10 (linhagens de L. edodes x tipo de eucalipto), totalizando 20 tratamentos com 2 repetições, sendo que cada repetição correspondeu a uma amostra de basidiomas desidratados e moídos. de acordo com os resultados obtidos, verificou-se que as propriedades nutricionais do L. edodes (proteína bruta, extrato etéreo, cinzas e fibra bruta) demonstraram sofrer influência da interação eucalipto x fungo. Assim, os melhores resultados foram: Proteína bruta: Linhagem LE-96/18 cultivada em toras de E. urophylla, cuja média foi de 24,3%; Extrato etéreo: Linhagem LE-96/18 cultivada em toras do clone 23, cuja média foi de 3,0%; Cinzas: Linhagem LE-96/18 cultivada em toras de E. paniculata e E. camaldulensis e Linhagem LE-95/01 cultivada em toras de E. citriodora, cujas médias foram de 5,0%; Fibra bruta: Linhagem LE-95/01 cultivada em toras de E. paniculata, cuja média foi de 20,5%.

In Brazil there was little research related to Shiitake axenic culture. The aim of this research was to understand the substratum effects in the kinetics of the Shiitake mycelium growth. It was used two Shiitake strains and two different base substrate (eucalyptus sawdust and sugar cane bagasse) varying in three proportions of the supplements. The supplements, a blend of rice and wheat brans, were added in the proportion of 0, 10 and 20% of the base substrate. The experiment was composed of six treatments. The mycelium growth kinetics in volume had no effect relation to the strains and substrate and it followed a mathematical model represented by logarithmic equation. Beta, gamma and delta parameters didn't show any correlation with the growth velocity in volume. The strain L55 was better adapted than L17.
