Under de senaste åren har intresset för utnyttjandet av förnybara resurser kraftigt ökat. I samband med detta utgör kolhydrater en viktig del av den tillgängliga förnybara biomassan och den har därefter blivit föremål för ett stort intresse inom hållbar kemi. Sockeralkoholer är en särskilt viktig grupp av molekyler som vanligtvis erhålls ur kolhydrater och som har mångsidiga tillämpningar som t.ex. lågkalorihaltiga sötningsmedel. Forskningen i doktorsarbetet omfattar hydreringen av naturligt förekommande sockerarter L-arabinos, D-galaktos, D-maltos och L-ramnos till respektive sockeralkoholer. Dessa sockeralkoholer kan användas bl.a. som hälsosamma sötningsmedel på samma sätt som xylitol. Grunden för detta arbete består av hydreringsexperiment som utfördes på en dispergerad ruteniumkatalysator i syfte att studera bildningskinetiken av de motsvarande sockeralkoholerna. Reaktionerna genomfördes vid temperaturer mellan 90 och 130 °C och vätetryck mellan 40 och 60 bar. Under dessa betingelser var det möjligt att åstadkomma sockeromvandlingar upp till 100 %. Reaktionshastigheterna modellerades matematiskt. Konkurrerande kinetiska modeller som baserades på Langmuir-Hinshelwood-konceptet föreslogs för att beskriva reaktionerna. Parametrar i hastighetsekvationerna bestämdes därefter genom icke-linjär regression. Dessa modeller kunde väl förutsäga hydreringsreaktionernas förlopp och de kan följaktligen användas för design av industriella anläggningar. Ytterligare hydreringsexperiment med sockerblandningar genomfördes för att fördjupa kunskaper i kinetik och reaktionsmekanismer av sockerhydreringen. Studierna genomfördes med syntetiska sockerblandningar av L-arabinos och D-galaktos (de viktigaste komponenterna i hemicellulosan arabinogalaktan). Fullständig omsättning uppnåddes med utmärkta selektiviteter som överskred 95 % och dessutom inverkade varken temperatur eller vätetryck på reaktionens förlopp på något oväntat sätt. Antagandet av konkurrerande adsorption för en samtidig reduktion av båda sockermolekylerna gav en kinetisk modell som noggrant beskrev de experimentella resultaten. Idén om att utforska potentiella sätt att påskynda bildningen av sockeralkoholer ledde till utföringen av hydreringsexperiment med L-arabinos och D-galaktos i närvaro av ultraljud. Det visade sig att ultraljudets inverkan var oberoende av sockerhalten och vätetrycket och att bestrålningen gynnade hydreringen av D-galaktos trots att den inte förhindrade Ru/C-katalysatorns deaktivering överhuvudtaget. En kinetisk modell som beaktade deaktiveringen utvecklades. Kontinuerlig hydrering av L-arabinos genomfördes med tre olika Ru-katalysatorer på tre olika bärare: tyg av aktivt kol, kolnanorör på svampliknande metalliska strukturer samt krossade partiklar av en kommersiell Ru/C-katalysator. Det visade sig att det var möjligt att omvandla L-arabinos till arabitol med höga selektiviteter med hjälp av Ru/koltyg-katalysatorn. Dessa experiment demonstrerade att hydreringen av de valda sockerarterna är en helt genomförbar rutt till framställning av fin- och specialkemikalier, som kan förverkligas i större skala.

L-arabinose is widely used in food, medicine, chemistry, and biology fields; however, solubility and seeded metastable zone width (MSZW) of L-arabinose have not been reported in the literature. In this paper, solubility and MSZW of L-arabinose in aqueous solution were determined. Solubility of L-arabinose was measured in the range of 20-68 °C by a conventional equilibrium solubility method and quantitation was determined using the ion chromatography technique. Seeded MSZW was determined in the range of 51-73% by the calorimetric method. The effect of two salts (potassium chloride and calcium chloride) on the solubility and MSZW of L-arabinose were also evaluated. Results showed that both potassium chloride and calcium chloride increased the solubility of L-arabinose, and this increase was intensified with temperature rise. The MSZW of L-arabinose was not constant but a spread. Potassium chloride increased the MSZW of L-arabinose. However, the effect of calcium chloride on MSZW of L-arabinose was concentration dependent. Conclusion: the L-arabinose solubility increased with the increase in temperature, and both potassium chloride and calcium chloride increased the solubility of L-arabinose in aqueous solution. The seeded MSZW of L-arabinose is not a constant; it increases in the presence of potassium chloride and varies with the change in calcium chloride concentration.

La diarrhée post-sevrage est une maladie d’importance dans l’industrie porcine et est principalement causée Escherichia coli O149. Le traitement habituellement utilisé est la néomycine. Cependant, en raison de l’antibiorésistance, les vétérinaires se tournent vers la colistine sulfate (CS). La CS lie les lipopolysaccharides (LPS) et provoque un déplacement des cations divalents causant la formation de pores entrainant la mort cellulaire. Le système à deux composantes PmrA/PmrB est le plus incriminé dans la résistance à la colistine en ajoutant un groupement 4-amino-4-déoxy-L-arabinose (L-Ara4N) au lipide A des LPS, augmentant ainsi la charge du LPS et diminuant son affinité pour la CS. L’objectif principal est d’évaluer l’acquisition de la résistance à la CS d’E. coli in vitro et dans un modèle in vivo. Nous avons utilisé des souches associées à des cas cliniques d’E. coli O149 et avons créé 22 mutants résistants à la CS. La concentration minimale inhibitrice (CMI) a été mesurée par une méthode de double dilution et comparée au seuil de résistance. Suite au séquençage des gènes pmrA/pmrB, nous avons identifié sept nouveaux polymorphismes, trois dans PmrA : A80V, N128I, S144G et quatre dans PmrB : V87E, D148Y, D148V et T156M. Pour l’essai in vivo, nous avons suivi une souche expérimentale ETEC:F4 (E. coli O149) et isolé des E. coli de la flore commensale. Le séquençage des gènes pmrA et pmrB de ces isolats a montré un polymorphisme spécifique, G15R et T156M respectivement. Cependant, plusieurs souches récoltées possédaient une résistance à la CS, mais sans polymorphisme de PmrA/PmrB, suggérant d’autre(s) mécanisme(s) de résistance.

Sugarcane bagasse hemicellulose was isolated in a one-step chemical extraction using hydrogen peroxide in alkaline media. The polysaccharide containing 80.9% xylose and small amounts of L-arabinose, 4-O-methyl-D-glucuronic acid and glucose, was hydrolyzed by crude enzymatic extracts from Thermoascus aurantiacus at 50 degrees C. Conditions of enzymatic hydrolysis leading to the best yields of xylose and xylooligosaccharides (DP 2-5) were investigated using substrate concentration in the range 0.5-3.5% (w/v), enzyme load 40-80 U/g of the substrate, and reaction time from 3 to 96 h, applying a 22 factorial design. The maximum conversion to xylooligosaccharides (37.1%) was obtained with 2.6% of substrate and xylanase load of 60 U/g. The predicted maximum yield of xylobiose by a polynomial model was 41.6%. Crude enzymatic extract of T. aurantiacus generate from sugarcane bagasse hemicellulose 39% of xylose, 59% of xylobiose, and 2% of other xylooligosaccharides.

Bacillus subtilis tem a capacidade de utilizar arabinoligossacáridos presentes na parede das células vegetais, através de um consórcios de enzimas envolvidas na hidrólise enzimática dos mesmos. A captação destes carbohidratos por parte do Bacillus subtilis depende de dois transportadores membranares AraE e AraNPQ-MsmX. Após a captação de L-arabinose e arabinoligossacáridos, estes substratos serão metabolizados pelos produtos enzimáticos codificados por genes pertencentes ao operão responsável pela metabolização de arabinose araABDLMNPQabfA. Neste operão, para além dos genes responsáveis pelo transportador AraNPQ, o gene araL determina a síntese de uma enzima com atividade de fosfatase, AraL, que tem putativamente como principal função a destoxificação de intermediários metabólicos fosforilados, em situações particulares. O objectivo deste trabalho consistiu em descobrir quais os determinantes moleculares envolvidos em ambos os processos de reconhecimento molecular, ora o reconhecimento de arabinoligossacáridos por parte da AraN, do ponto de vista dos carbohidratos ora de carbohidratos fosforilados do ponto de vista da AraL. Para elucidar estes processos de reconhecimento molecular proteína-carbohidrato foi utilizada a técnica de RMN, modelação computacional e mutagénese dirigida. No primeiro capitulo são introduzidos conceitos fundamentais para a percepção da ação de ambas as proteínas (AraN e AraL), no organismo Bacillus subtilis. O segundo capitulo, refere-se ao estudo dos mecanismos envolvidos no reconhecimento proteína-carbohidrato através da técnica de STD-RMN para estudar interações do ponto de vista do carbohidrato, bem como abordagens bioinformáticas tais como alinhamentos de sequencia primária e dockings moleculares, para identificar resíduos do ponto de vista da proteína passiveis de se encontrarem envolvidos no reconhecimento molecular, que numa última instância são mutados para se confirmar a sua relevância no processo de reconhecimento molecular do ponto de vista da proteína. Neste capitulo é demonstrado que a AraN é responsável pelo reconhecimento de arabinoligossacáridos e celoligossacáridos. No que diz respeito aos primeiros, a AraN reconhece preferencialmente arabinoligossacáridos com três subunidades, verificando-se a perda de saturação na subunidade não redutora da arabinotetraose. O processo de reconhecimento molecular apresenta uma constante de dissociação com uma ordem de grandeza na ordem dos μM. Do ponto de vista da proteína são identificados resíduos (W253, Y254 e Y54) putativos de se encontrarem também eles envolvidos no reconhecimento molecular do ponto de vista da AraN através de interações CHstacking. Finalmente no terceiro capitulo, é apresentada a optimização da sobrexpressão da AraL, dado que numa primeira fase a sua produção ocorria sob a forma de corpos de inclusão. Para tal são descritas metodologias de solubilização e renaturação da mesmas, com recurso a agentes caotrópicos (ureia e GmdCl) e detergentes (SDS).

Experiments for the investigation of dehydrogenase activity of washed cells of a strains of Br. abortus and another of Br. suis in presence of different single added substrates are reported. The activity was measured as the amount of formazan produced by the reduction of 2, 3, 5-triphenyltetrazolum chloride acting as a hydrogen ions acceptor, at pH 7.0. In a general manner the dehydrogenase activity of Br. suis was much more intense than that of Br. abortus (fig. 5). In the conditions of the experiments Br. abortus oxidized L-arabinose, D-galactose, D-glucose, glycerol, D-xylose, DL-alanine, D-fructose, and D-sorbitol. Brucella suis oxidized D-xylose, L-arabinose, D-glucose, D-galactose, DL-alanine, sodium acetate, maltose, glycine, D-fructose, and D-sorbitol. Glycerol was oxidized by Br. abortus but its oxidation by Br. suir was very slight. Sodium acetate and maltose were intensely oxidized by Br. suir but not by Br. abortus. The sites of more intense enzymatic acitivity were seen as small red colored round granules located in one pole of the cells.

La synthèse énantiosélective de la (+)-ambruticine S, un produit naturel antifongique a été effectuée au sein de notre groupe. Trois approches ont été développées pour la synthèse du fragment lactone (cycle A). Ces trois voies d’accès au cycle A ont pour intermédiaire commun le methyl α-D-glycopyranoside déjà porteur du diol requis et disponible commercialement à bon prix. Une désoxygénation de l’hydroxyle en C-4 et l’homologation d’un carbone de la chaine latérale en C-6 ont permis l’obtention du cycle lactonique A. Le deuxième projet est une collaboration entre le groupe Hanessian et ISIS Pharmaceuticals afin de développer de nouveaux oligonucléosides antisens. Les nucléosides antisens [4.3.0]-bicycliques cis et trans ont été synthétisés avec succès à partir d’un monosaccharide naturel commun, L-arabinose, porteur des stéréocentres requis. Un réaction clé d’allylation de Sakurai a permis d’obtenir les diastéréoisomères cis et trans dans des conditions de contrôle de type Felkin-Ahn et de contrôle par chélation respectivement. Les composés bicycliques finaux cibles ont été obtenus par une réaction d’aldol intramoléculaire catalyzéé par la proline, par métathèse de fermeture de cycle et par l’application de la méthode de Vorbrüggen pour la synthèse de nucléosides.

Sugar esters are substances which possess surfactant, antifungical and bactericidal actions and can be obtained through two renewable sources of raw materials: sugars and vegetable oils. Their excellent biodegradability, allied to lhe fact that they are non toxic, insipid, inodorous, biocompatible, no-ionic, digestible and because they can resist to adverse conditions of temperature, pH and salinity, explain lhe crescent use of these substances in several sections of lhe industry. The objective of this thesis was to synthesize and characterize surfactants and polymers containing sugar branched in their structures, through enzymatic transesterification of vinyl esters and sugars, using alkaline protease from Bacillus subtilis as catalyst, in organic medium (DMF).Three types of sugars were used: L-arabinose, D-glucose and sucrose and two types of vinyl esters: vinyl laurate and vinyl adipate. Aiming to reach high conversions from substrates to products for a possible future large scale industrial production, a serie of variables was optimized, through Design of Experiments (DOE), using Response Surface Methodology (RSM).The investigated variables were: (1) enzyme concentration; (2) molar reason of substrates; (3) water/solvent rale; (4) temperature and (5) time. We obtained six distinct sugar esters: 5-0-lauroyl L-arabinose, 6-0-lauroyl D-glucose, 1'-O-lauroyl sucrose, 5-0-vinyladipoyl L-arabinose, 6-0-vinyladipoyl D-glucose and 1 '-O-vinyladipoyl sucrose, being lhe last three polymerizable. The progress of lhe reaction was monitored by HPLC analysis, through lhe decrease of sugar concentration in comparison to lhe blank. Qualitative analysis by TLC confirmed lhe formation of lhe products. In lhe purification step, two methodologies were adopted: (1) chromatographic column and (2) extraction with hot acetone. The acylation position and lhe chemical structure were determined by 13C-RMN. The polymerization of lhe three vinyl sugar esters was possible, through chemical catalysis, using H2O2 and K2S2O8 as initiators, at 60°C, for 24 hours. IR spectra of lhe monomers and respective polymers were compared revealing lhe disappearance of lhe vinyl group in lhe polymer spectra. The molar weights of lhe polymers were determined by GPC and presented lhe following results: poly (5-0-vinyladipoyl L-arabinose): Mw = 7.2 X 104; PD = 2.48; poly (6-0-vinyladipoyl D-glucose): Mw = 2.7 X 103; PD = 1.75 and poly (1'-O-vinyladipoyl sucrose): Mw = 4.2 X 104; PD = 6.57. The six sugar esters were submitted to superficial tension tests for determination of the critical micelle concentrations (CMC), which varied from 122 to 167 ppm. Finally, a study of applicability of these sugar esters, as lubricants for completion fluids of petroleum wells was' accomplished through comparative analysis of lhe efficiency of these sugar esters, in relation to three commercial lubricants. The products synthesized in this thesis presented equivalent or superior action to lhe tested commercial products

Biotechnological conversion of biomass into fuels and chemicals requires hydrolysis of the polysaccharide fraction into monomeric sugars. Hydrolysis can be performed enzymatically and with dilute or concentrate mineral acids. The present study used dilute sulfuric acid as a catalyst for hydrolysis of Eucalyptus grandis residue. The purpose of this paper was to optimize the hydrolysis process in a 1.41 pilot-scale reactor and investigate the effects of the acid concentration, temperature and residue/acid solution ratio on the hemicellulose removal and consequently on the production of sugars (xylose, glucose and arabinose) as well as on the formation of by-products (furfural, 5-hydroxymethylfurfural and acetic acid). This study was based on a model composition corresponding to a 2 3 orthogonal factorial design and employed the response surface methodology (RSM) to optimize the hydrolysis conditions, aiming to attain maximum xylose extraction from hemicellulose of residue. The considered optimum conditions were: H2SO4 concentration of 0.65%, temperature of 157 degrees C and residue/acid solution ratio of 1/8.6 with a reaction time of 20 min. Under these conditions, 79.6% of the total xylose was removed and the hydrolysate contained 1.65 g/l glucose, 13.65 g/l xylose, 1.55 g/l arabinose, 3.10 g/l acetic acid, 1.23 g/l furfural and 0.20 g/l 5-hydroxymethylfurfural.
