The biological characteristics of Aedes aegypti (Diptera, Culicidae), which is a vector of dengue and yellow fever, make this organism a good model for studying population structure and the events that may influence it under the effect of human activity. We assessed the genetic variability of five A. aegypti populations using RAPD-PCR technique and six primers. Four populations were from Brazil and one was from the USA. A total of 165 polymorphic DNA loci were generated. Considering the six primers and the five populations, the mean value of inter-population genetic diversity (Gst) was 0.277, which is considered high according to the Wright classification. However, pairwise comparisons of the populations gave variable Gst values ranging from 0.044 to 0.289. This variation followed the population's geographic distance to some extent but was also influenced by human activity. The lowest Gst values were obtained in the comparison of populations from cities with intensive commercial and medical contacts. These mosquito populations were previously classified as insecticide resistant, susceptible, or with decreased susceptibility; this parameter apparently had an effect on the Gst values obtained in the pairwise comparisons. ©FUNPEC-RP.

INTRODUCTION: Despite years of efforts towards the elimination of Triatoma infestans in Brazil, residual foci still persist in some areas of the States of Bahia and Rio Grande do Sul. The persistence of these T. infestans populations in the country has two different origins of equal concern: operational failures or insecticide resistance. Thus, the objective of this study was to characterize the susceptibility profile of the residual Brazilian populations of T. infestans to deltamethrin. METHODS: The susceptibility reference lineage was derived from Cipein/Argentina. The populations studied were manually collected using a dislodging agent in peridomiciles in the States of Bahia (Novo Horizonte) and of Rio Grande do Sul (Santa Rosa and Doutor Maurício Cardoso). Serial dilutions of deltamethrin were prepared and applied at the dorsal abdomen of first instar nymphs. The control group received only pure acetone. Mortality was evaluated after 72h. Qualitative tests assessed the mortality of a diagnostic dose of 1xLD99 (2.76ng a.i./nymph) determined for the susceptibility reference lineage. RESULTS: The susceptibility profile characterization of the T. infestans populations revealed an RR50 ranging from 1.73 to 3.26. The mortality percentage in response to a diagnostic dose was 100%. The results obtained in the quantitative and qualitative assays corresponded for all populations. CONCLUSIONS: The results of this study indicate that the persistence of residual foci of T. infestans in Bahia and Rio Grande do Sul is not related to insecticide resistance but may be associated with operational failures. In Rio Grande do Sul, we must consider the possibility of continuous reinfestation by Argentinian individuals, which justifies active and efficient epidemiological surveillance.

The residual potential of an aqueous solution of Deltamethrin (FW 25 mg i.a./m2) was evaluated on raffia curtains. These are sheets of synthetic material used in the construction of huts to house miners. Experiments were conducted during 420 days and the curtains were always rolled up in the daytime and unrolled in late afternoon. Data analyzed by logarithmic regression indicated that raffia treated with Deltamethrin had higher mortality indices than that covered with DDT. The residual capacity of Deltamethrin on raffia was high. The mortality percentage was above 85% after 360 days and dropped to about 50% at 420 days. The effect of DDT was reduced after 180 days and reached zero by the end of the experiment. Based on the results of these experiments, it is recommended that Deltamethrin be used to spray raffia curtains in mining regions and other areas that are endemic for malaria.

Saint Louis encephalitis virus caused an outbreak of febrile illness and encephalitis cases in Córdoba, Argentina, in 2005. During this outbreak, the strain CbaAr-4005 was isolated from Culex quinquefasciatus mosquitoes. We hypothesised that this epidemic variant would be more virulent in a mouse model than two other non-epidemic strains (78V-6507 and CorAn-9275) isolated under different epidemiological conditions. To test this hypothesis, we performed a biological characterisation in a murine model, including mortality, morbidity and infection percentages and lethal infection indices using the three strains. Mice were separated into age groups (7, 10 and 21-day-old mice) and analysed after infection. The strain CbaAr-4005 was the most infective and lethal of the three variants, whereas the other two strains exhibited a decreasing mortality percentage with increasing animal age. The strain CbaAr-4005 produced the highest morbidity percentages and no significant differences among age groups were observed. The epidemic strain caused signs of illness in all inoculated animals and showed narrower ranges from the onset of symptoms than the other strains. CbaAr-4005 was the most virulent for Swiss albino mice. Our results highlight the importance of performing biological characterisations of arbovirus strains likely to be responsible for emerging or reemerging human diseases.

The organophosphate temephos has been the main insecticide used against larvae of the dengue and yellow fever mosquito (Aedes aegypti) in Brazil since the mid-1980s. Reports of resistance date back to 1995; however, no systematic reports of widespread temephos resistance have occurred to date. As resistance investigation is paramount for strategic decision-making by health officials, our objective here was to investigate the spatial and temporal spread of temephos resistance in Ae. aegypti in Brazil for the last 12 years using discriminating temephos concentrations and the bioassay protocols of the World Health Organization. The mortality results obtained were subjected to spatial analysis for distance interpolation using semi-variance models to generate maps that depict the spread of temephos resistance in Brazil since 1999. The problem has been expanding. Since 2002-2003, approximately half the country has exhibited mosquito populations resistant to temephos. The frequency of temephos resistance and, likely, control failures, which start when the insecticide mortality level drops below 80%, has increased even further since 2004. Few parts of Brazil are able to achieve the target 80% efficacy threshold by 2010/2011, resulting in a significant risk of control failure by temephos in most of the country. The widespread resistance to temephos in Brazilian Ae. aegypti populations greatly compromise effective mosquito control efforts using this insecticide and indicates the urgent need to identify alternative insecticides aided by the preventive elimination of potential mosquito breeding sites.

Entomopathogenic nematodes, Steinernema carpocapsae, S. feltiae (Steinernematids) Heterorhabditis indica and H. bacteriophora (Heterorhabditids) were studied to control nymphs of desert locust Schistocerca gregaria. Results of all experiments showed a significant difference in mortality percentage among all isolates. All nematodes were found more effective when exposure time was increased up to 10 days. On the other hand, both Heterorhabditids caused maximum mortality as compared to Steinernematids at 30 degree C. When different moisture levels were tested in the sand arena, a medium level of moisture (1%) caused maximum insect mortality in all isolates. However, highest concentration of each isolate (200 IJs per ml) proved to be most appropriate for maximum insect death. Similarly, both Heterorhabditis nematodes when orally applied to insects killed maximum nymphs as compared to other two Steinernematids. A similar response was observed in infectivity test when maximum percentage of IJs of both isolates of Heterorhabditis successfully penetrated into the body of locust nymphs. This research suggests some useful basic findings in developing biocides with suitable virulent of entomopathogenic nematode for controlling nymphs of desert locust.

This work was developed with the objetive to evaluate the insecticidal effect of the Neem (Azadirachta indica A. Juss) seeds against grass bug nymphs Collaria scenica Stal. For that, an extract was prepared from green fruits of Neem by rotaevaporacion. The extract was diluted in three ppm concentrations corresponding to treatments. Using thin-layer chromatography we identified the presence of Azaridactina. DCA essay was carried out with 4 treatments and 5 repetitions that included the three extract concentrations and a control. In each repetition were used 15 nymphs, randomly placed in plastic boxes with food and the corresponding concentration. Every day an extract application was made and recorded mortality percentage, number of exuviae and number of individuals who came to adulthood in each treatment. The results were analyzed using Kruskal-Wallis and Games-Howell for each variable. The three concentrations of Neem seed extract had a negative effect on the development of the bugs. The more concentrated treatment (250 ppm) was the most effective, showing a 97% mortality, fewer exuvias and and fewer adults in the end of the experiment.

O objetivo desta pesquisa foi obter informações que possam favorecer o entendimento da atuação dos compostos alelopáticos sobre o banco de sementes do solo e sobre a seleção de plantas invasoras e indicar espécies que favoreçam a renovação dos pastos. Assim, soluções de solo de uma área de pastagem de Brachiaria brizantha cv. Marandu formada há mais de cinco anos foram extraídas para avaliar os efeitos alelopáticos dessa espécie sobre a germinação, a dormência de sementes e o vigor de plântulas de B. brizantha cv. Marandu, Panicum maximum cv. Tanzânia, Sida rhombifolia e Peschiera fuchsiaefolia. O experimento foi instalado e conduzido no Laboratório de Matologia do Departamento de Produção Vegetal da FCA/UNESP - campus de Botucatu-SP. O substrato de germinação foi umedecido com 12 mL dos seguintes tratamentos: solução do solo de uma área cultivada com B. brizantha; solução do solo de uma área sem B. brizantha (mata nativa); água destilada; solução de polietilenoglicol com potencial osmótico idêntico ao da solução do solo sob B. brizantha; e solução de polietilenoglicol com potencial osmótico idêntico ao da solução de solo de uma mata nativa. A porcentagem de sementes normais, mortas, anormais e dormentes de B. brizantha não foi influenciada por nenhuma das soluções testadas, o que evidencia a não-ocorrência de efeito auto-alelopático. Foram verificados possíveis efeitos alelopáticos negativos sobre a porcentagem e velocidade de germinação de P. maximum cv. Tanzânia e sobre o crescimento radicular de S. rhombifolia. Esta última também se mostrou sensível ao efeito alelopático da solução de solo de mata, que promoveu redução no crescimento radicular. P. fuchsiaefolia não foi afetada por nenhuma das soluções testadas.

O trabalho objetivou avaliar os efeitos de diferentes rações comerciais com alto nível energético na incidência de ascite em frangos de corte. Foram utilizadas 1.200 aves de uma mesma linhagem comercial (Hubbard), distribuídas em 12 boxes, segundo um delineamento inteiramente ao acaso, com quatro tratamentos e três repetições de 100 aves cada. Os tratamentos foram constituídos por três diferentes rações comerciais trituradas (B, C e D) comparadas com o controle, uma ração farelada inicial (A), do primeiro ao 39º dia de idade. Não houve diferenças entre os tratamentos quanto ao consumo, peso e ganho de peso das aves. em relação à conversão alimentar, o tratamento C apresentou resultado significativamente melhor; entretanto, foi observada neste mesmo tratamento, a maior taxa de mortalidade. O maior motivo dos óbitos registrados foi a síndrome ascítica. Conclui-se que existe um favorecimento de surto de ascite pelas rações com melhor conversão alimentar nas aves.

El objetivo del presente trabajo fue estudiar la actividad tóxica del aceite esencial de laurel (Laurus nobilis L.) y del cineol, monoterpeno cíclico considerado un aleloquímico puro, sobre Brevicoryne brassicae L. en repollo (Brassica oleracea var. capitata L.). Las concentraciones de aceite esencial utilizadas fueron: 1; 1,5; 2 y 3%, y para cineol: 0,5; 1,5 y 2,5%, ambas formuladas en solución acuosa con 2% de oleato de polietilenglicol como emulsionante. Se utilizaron dos técnicas de aplicación: papeles impregnados y pulverización directa. A las 24 horas del tratamiento se evaluó el porcentaje de mortalidad. Los resultados se analizaron por ANOVA de dos vías y Test de Tukey. Se encontraron diferencias significativas para los productos evaluados en todas las concentraciones ensayadas. Las técnicas de aplicación no dieron diferencias significativas. La mayor mortalidad correspondió a 52% para el aceite esencial de laurel y 27,5% para cineol a las mayores concentraciones en ambos casos. Se concluye que los productos ensayados podrían ser una herramienta para el Manejo Integrado de Plagas para control de áfidos en cultivos hortícolas.

El objetivo del presente trabajo fue comparar la eficiencia de 4 pesticidas: dimetoato, metidation, etion y clorpirifos, en el control de la cochinilla H [Saissetia oleae Bernard (Homoptera, Coccoidea, Lecaniidae)] también llamada cochinilla negra del olivo. Estos productos, conocidos en Mendoza (Argentina), se utilizan localmente. El ensayo se realizó en Vistalba (Luján de Cuyo). Para la evaluación se tuvo en cuenta el porcentaje de mortalidad. El análisis estadístico se efectuó transformando los datos según arcsen ( x / 1 0 0 ) .

La actividad biológica de Lonchocarpus guaricensis Pittier fue evaluada utilizando dos dosis de Tecnona® en el control de larvas de Tuta absoluta (Meyrick), Valle de Azapa, Chile, mediante una pulverización sobre plantas de tomate cv. Naomi en macetas, ubicadas aleatoriamente en un invernáculo dentro de un vivero. Semanas previas a la pulverización, las macetas se infestaron artificialmente con adultos del fitófago para obtener larvas en los foliolos. Los tratamientos evaluados fueron los siguientes: T1 (0,21 g de IA de L. guaricensis·L-1), T2 (0,43 g de IA de L. guaricensis·L-1), T3 (control positivo a base de spinosad 0,048 g de IA·L-1) y T0 (control negativo a base de agua de pozo). De acuerdo con el porcentaje de mortalidad acumulada de larvas contabilizadas a las 24, 48, 120 horas y 9no día post aplicación, no hay diferencias estadísticas entre los tratamientos T0 y T1, a su vez, T2 alcanza una media de 53,05% de mortalidad, no diferenciándose de T3 que logra un 73,9%. Se concluye que la dosis experimental L. guaricensis de 0,43 g de IA∙ L-1 puede constituir una alternativa interesante de utilizar en el Manejo Integrado de Plagas del cultivo de tomate en el Valle de Azapa.

As rodovias podem contribuir para a diminuição de populações de anfíbios de várias formas, sendo a mais visível a mortalidade por atropelamento. Neste trabalho procurou-se observar a forma como o tráfego pode afectar a mortalidade de anfíbios. A recolha de dados foi efectuada em duas estradas rurais próximas onde foram registadas as espécies observadas e se estas estavam vivas ou mortas, entre outros. Os resultados comprovaram não haver diferenças significativas em relação ao número de indivíduos ou à proporção das espécies que atravessam as vias e confirmaram que uma das estradas possui uma percentagem de mortalidade muito superior à outra. Os testes estatísticos também revelaram existir uma diferença significativa entre o tráfego nas duas estradas. Desta forma, na estrada com mais trânsito foi observado um número muito superior de mortos. Foi, portanto, possível relacionar os resultados e concluir que os anfíbios são negativamente afectados pela intensidade do trânsito rodoviário. ABSTRACT; The roads can contribute to the amphibian populations decline in many ways, being the most visible the roadkills. The purpose of this study was to observe how the traffic can affect the amphibian mortality. The data collection was conducted in two rural roads, near to each other, where the species observed were recorded, and if they were alive or dead, among other information. The results showed no significant differences in the number of individual or in the proportion of species that cross the roads and confirmed that one of the road has a much higher mortality percentage than the other. Statistical tests also revealed a significant difference between the traffic of the two roads. Thus, on the road with more traffic was observed a much higher number of deaths. It was therefore possible to relate the results and conclude that the amphibians are negatively affected by the traffic intensity.
