This work evaluates a spline-based smoothing method applied to the output of a glucose predictor. Methods:Our on-line prediction algorithm is based on a neural network model (NNM). We trained/validated the NNM with a prediction horizon of 30 minutes using 39/54 profiles of patients monitored with the Guardian® Real-Time continuous glucose monitoring system The NNM output is smoothed by fitting a causal cubic spline. The assessment parameters are the error (RMSE), mean delay (MD) and the high-frequency noise (HFCrms). The HFCrms is the root-mean-square values of the high-frequency components isolated with a zero-delay non-causal filter. HFCrms is 2.90±1.37 (mg/dl) for the original profiles.

Drimys angustifolia Miers. (Winteraceae) is a Brazilian medicinal plant used as analgesic, antiulcer and anti-inflammatory without studies to assure its efficacy and safety Leaf and stem bark extracts were evaluated to determine the antiulcer, analgesic, antiinflammatory and antioxidant activities. Preliminary toxic effects and qualitative phytochemical profile were also performed. The antiulcer activity was detected in both extracts. Administration of the leaf extract at 250 mg/kg inhibited total lesion area by 76.50% (p < 0.01 in ethanol/HCl method), while carbenoxolone at 250 mg/kg reduced lesions by 69.48%. Stem bark extract (250 mg/kg) inhibited lesion by 81.42%, while carbenoxolone by 74.10%. Similar effects were observed in the ethanol-induced ulcer method, but no activity was observed in piroxican model. The effects involve nitric oxide in gastric protection, since the L-NAME treatment reversed the protection given by the extracts. Antioxidant effects suggest an involvement against oxidative stress. In the pain (writhing, tail-flick and hot-plate tests) and inflammation (carrageenan-induced paw edema) models, the extracts did not present any effect. The phytochemical studies demonstrated that both extracts contain flavonoids, saponins, glycosilated triterpenoids, fixed acids, cyanogenic glycosides, quinones, tannins, xanthone and steroidal aglycones. Toxicological studies showed that the extracts are safe at the effective antiulcer doses. (c) 2006 Elsevier B.V.. All rights reserved.

O método de polarização induzida, consagrado como ferramenta de prospecção de sulfetos metálicos e indiretamente, do ouro em associações polimetálicas, vem crescendo em outras aplicações como prospecção de águas subterrâneas, investigações de interesse à Engenharia Civil e ao uso de ocupação do solo. Neste trabalho, inicialmente, faz-se uma visualização geral do método IP, suas aplicabilidades, metodologia de trabalhos, arranjos de campo, fundamentos teóricos, vantagens e desvantagens etc. O objetivo principal, entretanto, é o modelamento analógico em laboratório, no sentido de simular-se as condições encontradas na natureza. Para tanto, utilizou-se de modelos geométricos simples, cilindros e placas. Na confecção destes modelos, utilizou-se cimento, areia quartzitica e grafita. Como a grafita apresenta um intenso fenômeno IP, variou-se o teor de grafita contido nos modelos. Assim, para diferentes formas e medidas dos modelos, variou-se também o conteúdo de partículas polarizáveis. As medidas foram feitas no sentido de verificar-se o comportamento da resposta IP quanto a forma, conteúdo de grafita, profundidade e direção do modelo estudado.

A geofísica aplicada em estudos ambientais é uma alternativa viável e promissora diante da demanda crescente por novas técnicas de investigação, essencialmente na caracterização e monitoramento de áreas contaminadas. Este trabalho apresenta os resultados de aplicação do método Polarização Induzida (IP), por meio da técnica de caminhamento elétrico, para determinação da área de percolação de material lixiviado proveniente de aterro de resíduos sólidos domiciliares do tipo vala. Os resultados sugerem uma associação entre anomalias de alta cargabilidade e prováveis acumulações de minerais férricos na base das valas de resíduos recentemente fechadas, em contraste com valas mais antigas, onde a cargabilidade tende aos valores naturais para a área. Investigações geofísicas, pelos métodos geoelétricos de eletrorresistividade e polarização induzida (IP), através das técnicas de sondagem elétrica vertical e caminhamento elétrico (configurações Schlumberger e dipolo-dipolo), foram executadas na área do aterro controlado da cidade de Piracicaba (SP), com o objetivo de avaliar as potencialidades da integração dos métodos na caracterização geoelétrica da área. A área sobre a qual está assentado o aterro de Piracicaba é constituída por sedimentos da Formação Corumbataí (sedimentos argilosos e/ou silto-argilosos, com intercalações de arenitos). A interpretação conjunta da resistividade e da polarizabilidade permitiu mapear zonas de percolação de chorume e identificar os diferentes litotipos das formação, identificando materiais arenosos e siltosos. Ficou evidente que a polarizabilidade é sensível à presença de resíduos urbanos e que o efeito de IP é relacionado a materiais polarizáveis dispostos na cava, como latas, papéis, restos eletrônicos e materiais de empréstimo utilizados para a cobertura dos resíduos. Além disso, o método de IP pode ser mais indicado para o mapeamento de zonas de resíduos onde não exista contraste de resistividade entre o meio natural e os resíduos. In the present study, thin functional conducting polyaniline (PANI) films, either doped or undoped, patterned or unpatterned, were prepared by different approaches. The properties of the obtained PANI films were investigated in detail by a combination of electrochemistry with several other techniques, such as SPR, QCM, SPFS, diffraction, etc. The sensing applications (especially biosensing applications) of the prepared PANI films were explored. Firstly, the pure PANI films were prepared by the electropolymerisation method and their doping/dedoping properties in acidic conditions were investigated in detail by a combination of electrochemistry with SPR and QCM. Dielectric constants of PANI at different oxidation states were obtained quantitatively. The results obtained here laid a good foundation for the following investigations of PANI films in neutral pH conditions. Next, PANI multilayer films doped by a variety of materials were prepared by the layer-by-layer method in order to explore their biosensing applications, because of the loss of redox activity of pure PANI in neutral pH conditions. The dopants used include not only the traditionally used linear polyelectrolytes, but also, for the first tim, some other novel materials, like modified gold nanoparticles or modified carbon nanotubes. Our results showed that all the used dopants could form stable multilayer films with PANI. All the obtained PANI multilayer films showed good redox activity in a neutral pH environment, which makes them feasible for bioassays. We found that all the prepared PANI multilayer films can electrocatalyze the oxidation of NADH in neutral conditions at a low potential, although their catalytic efficiencies are different. Among them, PANI/carbon nanotube system showed the highest catalytic efficiency toward the oxidation of NADH, which makes it a good candidate as a NADH sensor. Besides, because some of the prepared PANI multilayer systems were end-terminated with –COOH groups (like PANI/Au nanoparticles system), which can be utilized to easily link biomolecules for biosensing applications. Here, we demonstrated, for the first time, to use the prepared PANI multilayer films for the DNA hybridisation detection. The detection event was monitored either by direct electrochemical method, or by enzyme-amplified electrochemical method, or by surface plasmon enhanced fluorescence spectroscopic method. All the methods can effectively differentiate non-complementary DNA from the complementary ones, even at the single-base mismatch level. It should also be noted that, our success in fabricating PANI multilayer films with modified Au nanoparticles or carbon nanotubes also offered another novel method for incorporating such novel materials into (conducting) polymers. Because of the unique electrochemical and optical properties of each component of the obtained PANI multilayer films, they should also find potential applications in many other fields such as microelectronics, or for electrochromic and photovoltaic devices. Finally, patterned PANI films were fabricated by the combination of several patterning techniques, such as the combination of electrocopolymerization with micromolding in capillaries (EP-MIMIC), the combination of microcontact printing with the layer-by-layer technique (µCP-LBL), and the polystyrene (PS) template induced electropolymerisation method. Using the obtained stripe-shaped PANI/PSS film, a redox-switchable polymer grating based on the surface-plasmon-enhanced mode was constructed and its application in the field of biosensing was explored. It was found that the diffraction efficiency (DE) of the grating was very sensitive to the applied potential (i.e. redox state of the film) as well as the pH environment of the dielectric medium. Moreover, the DE could also be effectively tuned by an electrocatalytic event, such as the electrocatalytic oxidation of NADH by the grating film. By using PS colloidal crystal assemblies as templates, well-ordered 3D interconnected macroporous PANI arrays (PANI inverse opals) were fabricated via electropolymerisation method. The quality of the obtained inverse opals was much higher than those reported by chemical synthesis method. By electrochemical method, the structures of the prepared inverse opals can be easily controlled. To explore the possible biosensing applications of PANI inverse opals, efforts were also done toward the fabrication of PANI composite inverse opals. By selecting proper dopants, high quality inverse opals of PANI composites were fabricated for the first time. And the obtained opaline films remained redox-active in neutral pH conditions, pointing to their possible applications for electrobioassays.

Es wurde untersucht, wie sich das Substitutionsmuster organischer Peroxyradikale (RO2) auf die Ratenkonstante k1 und die Verzweigungsverhältnisse α, β und γ der Reaktionen von RO2 mit HO2 auswirkt. Die Effekte der Deuterierung von HO2 wurden ebenfalls studiert. Für zwei RO2 wurde zusätzlich das UV-Absorptionsspektrum bestimmt.rnrn αrnRO2 + HO2 → RO + OH + O2 R1arnrn βrn → RO2H + O2 R1brnrn γrn → ROH + O3 R1crnrnIn dieser Arbeit wurde ein neues Experiment aufgebaut. Für die direkte und zeitaufgelöste Messung der OH-Konzentration wurde das Verfahren der Laser-induzierten Fluoreszenz angewendet. Die Radikalerzeugung erfolgte mittels gepulster Laserphotolyse, wodurch unerwünschte Nebenreaktionen weitgehend unterdrückt werden konnten. Mittels transienter Absorptionsspektroskopie konnten die Menge der photolytisch erzeugten Radikale bestimmt und die Ozonbildung über R1c quantifiziert werden. Für die Auswertung wurden kinetische Modelle numerisch an die Messdaten angepasst. Um die experimentellen Unsicherheiten abzuschätzen, wurde ein Monte-Carlo-Ansatz gewählt.rnrnk1 und α reagieren sehr empfindlich auf Veränderungen des RO2-Substitutionsmusters. Während sich eine OH-Bildung für das unsubstituierte C2H5O2 (EtP) mit α EtP ≤ 5 % nicht nachweisen lässt, stellt R1a bei den α-Oxo-substituierten H3CC(O)O2 (AcP) und HOCH2C(O)O2 (HAP) mit α AcP = (63 ± 11) % bzw. α HAP = (69 ± 12) % den Hauptkanal dar. Wie die mit α HEP = (10 ± 4) % geringfügige OH-Bildung bei HOC2H4O2 (HEP) zeigt, nimmt die OH-Gruppe in β-Stellung weniger Einfluss auf den Wert von α als die Oxogruppe in α-Stellung. Bei der Erzeugung α-Oxo-substituierter RO2 kann ebenfalls OH entstehen (R+O2→RO2/OH). Die Druckabhängigkeit dieser OH-Quelle wurde mit einem innovativen Ansatz bestimmt. Mit γ AcP = (15+5-6) % bzw. γ HAP = (10+2-3) % lässt sich für die Reaktionen der α-Oxo-substituierten RO2 eine erhebliche Ozonbildung nachweisen. Durch die Einführung der α-Oxogruppe steigt k1 jeweils um 1,3 • 10-11 cm3s-1 an, der Effekt der β-Hydroxygruppe ist halb so groß (k1 AcP = (2,0 ± 0,4) • 10-11 cm3s-1, k1 HAP = (2,6 ± 0,4) • 10-11 cm3s-1). Das Verzweigungsverhältnis α steigt weiter, wenn das HO2 deuteriert wird (α AcP,iso = (80 ± 14) %, k1 AcP,iso = (2,1 ± 0,4) • 10-11 cm3s-1). Vergleiche mit älteren Studien zeigen, dass die OH-Bildung über R1a bislang deutlich unterschätzt worden ist. Die möglichen Ursachen für die Unterschiede zwischen den Studien werden ebenso diskutiert wie die Hintergründe der beobachteten Substituenteneffekte.

Statistical shape models (SSMs) have been used widely as a basis for segmenting and interpreting complex anatomical structures. The robustness of these models are sensitive to the registration procedures, i.e., establishment of a dense correspondence across a training data set. In this work, two SSMs based on the same training data set of scoliotic vertebrae, and registration procedures were compared. The first model was constructed based on the original binary masks without applying any image pre- and post-processing, and the second was obtained by means of a feature preserving smoothing method applied to the original training data set, followed by a standard rasterization algorithm. The accuracies of the correspondences were assessed quantitatively by means of the maximum of the mean minimum distance (MMMD) and Hausdorf distance (H(D)). Anatomical validity of the models were quantified by means of three different criteria, i.e., compactness, specificity, and model generalization ability. The objective of this study was to compare quasi-identical models based on standard metrics. Preliminary results suggest that the MMMD distance and eigenvalues are not sensitive metrics for evaluating the performance and robustness of SSMs.
