O experimento foi conduzido com o objetivo de desenvolver técnicas de manejo durante a larvicultura de trairão, realizando cultivos em água doce ou ligeiramente salinizada, utilizando náuplios de Artemia como alimento vivo. No início da alimentação exógena, oito dias após a eclosão, as larvas foram contadas e estocadas em 12 recipientes com volume útil de 1,5 L cada, dotados de sistema de aeração, numa densidade de 10 larvas/L, distribuídos dentro de três tanques de 130 L, contendo água com temperatura controlada (29,5ºC). Os tanques foram totalmente cobertos com lona plástica preta, mantendo o ambiente interno escuro, e descobertos somente para os manejos diários. As larvas foram submetidas a três tratamentos: cultivo em água doce; em água a 2 de salinidade; e em água a 4 de salinidade. Cada tratamento teve quatro repetições. A alimentação foi fornecida nas proporções diárias de 300 náuplios de Artemia/larva, do primeiro ao quinto dia, de 600 náuplios de Artemia/larva, do sexto ao décimo dia e de 900 náuplios de Artemia/larva, do décimo primeiro ao décimo quinto dia, divididos em três refeições. Ao final do experimento, foram avaliadas a sobrevivência, o crescimento (comprimento e peso), a taxa de crescimento específico e a taxa de resistência ao estresse. Após 15 dias de tratamento, não foram verificadas diferenças significativas entre os resultados das variáveis analisadas. As condições de cultivo mostraram-se eficientes, possibilitando altas taxas de sobrevivência (valores médios superiores a 91,6%), sendo a Artemia um alimento atrativo. Os níveis de salinidade utilizados não afetaram o desenvolvimento das larvas e alevinos.

Com esta pesquisa buscou-se avaliar diferentes níveis de arraçoamento, em porcentagem de peso corporal (PC), para juvenis de trairão em início do período de engorda. Peixes com comprimento de 15,0 ± 1,0 cm e peso de 38,5 ± 8,2 g foram distribuídos em seis aquários com 250 litros de água cada, em densidade de cinco juvenis/aquário. Utilizou-se delineamento inteiramente casualisado com três tratamentos (níveis de arraçoamento diário: 2, 4 e 6% PC) e duas repetições. Os juvenis foram alimentados com ração comercial extrusada (42% PB), às 8 e 13 h. Ao final de 45 dias, avaliaram-se a sobrevivência, a conversão alimentar aparente e os ganhos em biomassa, peso diário e comprimento. Não houve diferença para sobrevivência e ganhos em biomassa, peso diário e comprimento. Houve diferença para a conversão alimentar aparente em função do manejo adotado. Os melhores resultados de desempenho foram encontrados nos peixes alimentados com níveis de 2 e 4% PC de ração por dia. Conclui-se que a melhor taxa de arraçoamento para juvenis de trairão é de 4% PC.

This experiment was carried out to evaluate the effects of the stocking density on trairao larvae rearing. Seven days old larvae were submitted to four different stocking densities: 10, 30, 60 and 90 larvae/L. Sixteen vessels with 5 L each supplied with constant aeration were kept in a thermostatic bath. Each treatment had four replications. In order to keep the internal environment dark, all the experimental units were covered with a black plastic and it was uncovered only during the daily management. At the end of the experiment, after 15 days of feeding with Artemia nauplii, no significant differences were found (P>0.05) on growth among the evaluated densities. The same results were also registered for the survival, mortality and cannibalism rates. Higher stocking density resulted in higher production of trairao larvae rearing, allowing intensive rearing of this species in this phase.

Sixty-six trairão (Hoplias lacerdae) fingerlings (average weight of 2.0±0.5 g and total length of 5.8±0.2 cm), trained to accept dry rations, were allotted to six 15-L aquariums, with aeration and controlled temperature (24.0±0.5°C), in a density of 0.7 juveniles/L, aiming to evaluate the effects of darkness on fish productive performance. The treatments consisted of two photoperiods: 12 hours light: 12 hours dark (12L:12D) and 0 hour light: 24 hours dark (OL:24D), with three replicates. Fingerlings were fed ad libitum a commercial extruded diet (42% CP), twice a day. The aquariums were cleaned daily for excrement withdrawal through siphoning, exchanging 1/4 total volume. At the end of the experiment (30 days), weight gain, feed:gain ratio and survival and cannibalism rates were evaluated. The results showed that darkness did not affect the productive performance of trairão juveniles.

The aim of this work was to investigate the s of daily prey concentration during the first 15 days of active feeding of Hoplias lacerdae larvae, and the juvenile size on the feed training. In the first phase, the larvae received five Artemia nauplii concentrations (P). In the second phase, the juveniles from each treatment were trained to accept formulated diet. Superior growth was related to higher initial daily prey concentrations (900 and 1100 nauplii larvae-1). During feed training, the growth tendency was similar to that verified in the first phase. The lowest values of specific growth rate (SGR) were registered after the introduction of the semi-moist diet used in the feed training. However, the values of SGR recovered along the experiment and similar rates were found among the treatments. Survival, mortality and cannibalism were similar in the different treatments at the end of both phases. It can be concluded that: the prey concentration affects growth of H. lacerdae during the first 15 days of active feeding, and feed training can be initialized with juveniles of about 16 mm of total length.

Alevinos de trairão (Hoplias cf. lacerdae), com peso e comprimento total médios de 12,52 g e 10,5 cm, respectivamente, previamente condicionados ao aceite de dietas artificiais, foram distribuídos em seis tanques de alvenaria (5m²) nas densidades de: 1 e 4 alevinos/m². Os peixes dos diferentes tratamentos foram alimentados ad libitum nos horários de 8 e 14 h com ração comercial extrusada contendo 42% de proteína bruta. A cada trinta dias realizou-se a troca do volume total de água de todos os tanques. Ao final do experimento (120 dias) foi avaliado o desempenho produtivo dos peixes, por meio da taxa de sobrevivência, conversão alimentar, ganhos em comprimento e em peso diário. Foram observadas taxas médias de sobrevivência de 86,7 e 96,7% para as densidades de 1 e 4 peixes/m², respectivamente. A conversão alimentar foi de 1,6 e 1,2; o ganho em peso diário e em comprimento de 0,70 e 0,38 g e 8,01 e 7,27 cm, respectivamente. Estes valores não diferiram entre si. Com os resultados obtidos pode-se concluir que as densidades de estocagem de 1 e 4 peixes/m² podem ser utilizadas no cultivo de alevinos de trairão sem que haja comprometimento no seu desempenho produtivo.

Chromosomal localization of 5S rDNA and 5SHindIII repetitive sequences was carried out in several representatives of the Erythrinidae family, namely in karyomorphs A, D, and F of Hoplias malabaricus, and in H. lacerdae, Hoplerythrinusunitaeniatus and Erythrinus erythrinus. The 5S rDNA mapped interstitially in two chromosome pairs in karyomorph A and in one chromosome pair in karyomorphs D and F and in H. lacerdae. The 5SHindIII repetitive DNA mapped to the centromeric region of several chromosomes (18 to 22 chromosomes) with variations related to the different karyomorphs of H. malabaricus. on the other hand, no signal was detected in the chromosomes of H. lacerdae, H. unitaeniatus and E. erythrinus, suggesting that the 5SHindIII-DNA sequences have originated or were lost after the divergence of H. malabaricus from the other erythrinid species. The chromosome distribution of 5S rDNA and 5SHindIII-DNA sequences contributes to a better understanding of the mechanisms of karyotype differentiation among the Erythrinidae members.Copyright (c) 2007 S. Methyl mercury (MeHg) is highly neurotoxic, affecting visual function in addition to other central nervous system functions. The effect of mercury intoxication on the amplitude of horizontal cell responses to light was studied in the retina of the fish Hoplias malabaricus. Intracellular responses were recorded from horizontal cells of fish previously intoxicated with MeHg by intraperitoneal injection (IP group) or by trophic exposure (T group). Only one retina per fish was used. The doses of MeHg chloride administered to the IP group were 0.01, 0.05, 0.1, 1.0, 2.0, and 6.0 mg/kg. The amplitudes of the horizontal cell responses were lower than control in individuals exposed to 0.01 (N = 4 retinas), 0.05 (N = 2 retinas) and 0.1 mg/kg (N = 1 retina), whereas no responses were recorded in the 1.0, 2.0, and 6.0 mg/kg groups. T group individuals were fed young specimens of Astyanax sp previously injected with MeHg corresponding to 0.75 (N = 1 retina), 0.075 (N = 8 retinas) or 0.0075 (N = 4 retinas) mg/kg fish body weight. After 14 doses, one every 5 days, the amplitude of the horizontal cell response was higher than control in individuals exposed to 0.075 and 0.0075 mg/kg, and lower in individuals exposed to 0.75 mg/kg. We conclude that intoxication with MeHg affects the electrophysiological response of the horizontal cells in the retina, either reducing or increasing its amplitude compared to control, and that these effects are related to the dose and/or to the mode of administration.

A substantial fraction of the eukaryotic genome consists of repetitive DNA sequences that include satellites, minisatellites, microsatellites, and transposable elements. Although extensively studied for the past three decades, the molecular forces that generate, propagate and maintain repetitive DNAs in the genomes are still discussed. To further understand the dynamics and the mechanisms of evolution of repetitive DNAs in vertebrate genome, we searched for repetitive sequences in the genome of the fish species Hoplias malabaricus. A satellite sequence, named 5SHindIII-DNA, which has a conspicuous similarity with 5S rRNA genes and spacers was identified. FISH experiments showed that the 5S rRNA bona fide gene repeats were clustered in the interstitial position of two chromosome pairs of H. malabaricus, while the satellite 5SHindIII-DNA sequences were clustered in the centromeric position in nine chromosome pairs of the species. The presence of the 5SHindIII-DNA sequences in the centromeres of several chromosomes indicates that this satellite family probably escaped from the selective pressure that maintains the structure and organization of the 5S rDNA repeats and become disperse into the genome. Although it is not feasible to explain how this sequence has been maintained in the centromeric regions, it is possible to hypothesize that it may be involved in some structural or functional role of the centromere organization.

The differentiation of spermatids in Hoplias malabaricus is characterized by chromatin compaction, flagellum development, nuclear rotation, nuclear fossa formation, and excess cytoplasm elimination. In the resulting spermatozoon, the head is round and the nucleus contains chromatin compacted in thick filaments, peripherically arranged, to a central electron-lucent area. The acrosome is absent. The nuclear fossa is eccentric but not pronounced. The proximal centriole penetrates it and is oblique to the flagellum. The long midpiece has several converging elongate vesicles, forming membranous hoops in the initial segment of the flagellum, but has no cytoplasmic channel. The mitochondria are elongate and branched or C-shaped and located around the initial segment of the axoneme. The lateral flagellum does not show lateral projections. The ultrastructural characteristics of H. malabaricus spermatozoa are similar to the Cypriniformes. (C) 2001 the Fisheries Society of the British Isles.

The Hoplias malabaricus primary spermatogonium shows a large nucleus, central nucleolus, and low electron-dense cytoplasm containing nuages. In cysts, they undergo several mitotic divisions with incomplete cytokinesis, giving rise to secondary spermatogonia. These are smaller than the primary spermatogonia and their nuclei have one or two eccentric nucleoli. Spermatocytes I can be identified by the presence of synaptonemal complexes. Spermatocytes II are smaller than spermatocytes 1, displaying roughly compacted chromatin. All these cell types remain interconnected by thick-walled intercellular bridges, which have membranous reinforcements during mitosis and meiosis. These cell types show a well-developed endomembranous system, one of the centrioles anchored to the plasma membrane and small nuages. Their mitochondria are large and circular, with few cristae. In the last generations of spermatogonia, the mitochondria are smaller, elongate and have more cristae. In the spermatocytes, the mitochondria are small and round. Similarities found in relation to germ cells of other teleosts are discussed.
