Reports show that cold atmospheric-pressure plasmas can induce death of cancer cells in several minutes. However, very little is presently known about the mechanism of the plasma-induced death of cancer cells. In this paper, an atmospheric-pressure plasma plume is used to treat HepG2 cells. The experimental results show that the plasma can effectively control the intracellular concentrations of ROS, NO and lipid peroxide. It is shown that these concentrations are directly related to the mechanism of the HepG2 death, which involves several stages. First, the plasma generates NO species, which increases the NO concentration in the extracellular medium. Second, the intracellular NO concentration is increased due to the NO diffusion from the medium. Third, an increase in the intracellular NO concentration leads to the increase of the intracellular ROS concentration. Fourth, the increased oxidative stress results in more effective lipid peroxidation and consequently, cell injury. The combined action of NO, ROS and lipid peroxide species eventually results in the HepG2 cell death. The mechanism of death of human hepatocellular carcinoma cells (HepG2) induced by atmospheric-pressure room-temperature plasma, related to the plasma-controlled intracellular concentrations of reactive oxygen species (ROS), nitric oxide (NO) and lipid peroxide is revealed. Only 34.75 s are required to reduce the number of the viable HepG2 cells by 50%.

This study was designed to determine cytotoxic effects of PBDE-47 and HBCDs individually or with a mixture of both compounds exposure to Hep G2 cells. The results showed PBDE-47 and HBCDs induced increase of nitric oxide synthase (NOS) activity, release of NO. dissipation of mitochondria membrane potential and cell apoptosis. Exposure to HBCDs induced ROS formation. Moreover, preincubation with PTIO (NO scavanger) and N-acetylcysteine (ROS scavanger) partially reversed cytotoxic effects of these compounds. The possible mechanism is that PBDE-47 and HBCDs could boost generation of NO and/or ROS, impact mitochondria, and result in start-ups of apoptosis program. Cells exposed to mixture of both compounds and each of them showed non-apoptotic rate significant difference, but the combination of them caused more adverse effects on cells. These results Suggest that PBDE-47 and HBCDs in single and complex exposure have the cytotoxic activity of anti-proliferation and induction of apoptosis in tumor cells in vitro. 分别制备经1 Gy C离子辐照和未经辐照人肝癌HepG2细胞的总蛋白样品,采用固相pH梯度双向凝胶电泳技术进行蛋白分离,用ImageMaster 2D双向电泳凝胶图像分析软件分析数字化图谱。结果显示,差异表达的蛋白质点为17个,其中1个仅在未经C离子辐照的HepG2细胞中表达,8个蛋白质点在辐照后的细胞中表达上调,8个蛋白质点在辐照后的细胞中表达下调。建立了重复性较好和分辨率较高的分离HepG2细胞总蛋白的双向电泳技术。实验结果表明,C离子辐照HepG2细胞后其蛋白质组发生了改变。

本工作研究不同LET射线辐照对HepG2肝癌细胞辐射敏感性、周期进程和凋亡的影响,为重离子治疗癌症的临床应用积累基础数据。以0、0.5、1、2、4、8Gy剂量的12C6+离子及X射线分别照射处于指数生长期的HepG2细胞,用克隆形成率测定细胞辐射敏感性,通过流式细胞术测定细胞DNA含量以确定各时相细胞的比例及细胞凋亡情况。实验结果显示,12C6+离子辐照所致的HepG2细胞存活率明显低于X射线。随着吸收剂量的增加和修复时间的延长,12C6+离子能导致更显著的细胞S期阻滞、G2/M期阻滞延迟和细胞凋亡。说明与X射线相比,12C6+离子辐照能更有效地杀伤HepG2肝癌细胞并诱导其凋亡。

以人肝癌细胞 hepG2 为研究对象,用流式细胞术测定并分析了低剂量 γ 射线(5cGy)预照射对高剂量照射(3Gy)诱导的细胞周期阻滞的影响。结果显示:(1)低剂量照射可引起 hepG2 细胞在 G2/M 期短暂累积(至照射后 4h);(2)低剂量照射促进 hepG2 细胞的生长;(3)高剂量照射后,hepG2 细胞的 G2期发生阻滞,而 S 期只发生短暂延迟;(4)与单纯高剂量照射相比,低剂量照射预处理后 4h 给予高剂量照射可进一步促进hepG2 细胞在 G2/M 期累积,但预处理后 8h 给予高剂量可促进细胞通过 G2/M 期。实验结果表明,低剂量照射预处理对高剂量照射诱导的细胞周期阻滞的影响,取决于两次照射的时间间隔。

Survivin is a member of the inhibitors of apoptosis (IAP) protein family that interferes with post-mitochondrial events including activation of caspases. To examine the regulation of survivin expression in response to irradiation with different linear energy transfer (LET), human hepatoma HepG2 cells were irradiated in vitro with X-rays and carbon ions. Cellular sensitivities to low- and high-LET radiation were determined by colony formation. Survivin expression at mRNA and protein level were measured with RT-PCR and Western blot analyses, respectively. Radiation-induced cell cycle arrest and apoptosis were investigated with flow cytometry. We found that low-LET X-rays induced dose-dependent increases in survivin expression. After exposure to high-LET carbon ions, survivin expression gradually increased from 0 to 4 Gy, and then declined at 6 Gy. More pronounced survivin expression, stronger G(2)/M phase arrest was observed after exposure to carbon ions in comparison with X-rays at doses from 0 to 4 Gy. These observations indicate that there is a differential survivin expression in response to different LET radiations and the cycle arrest mechanism may be associated with it. In addition, our data on induction of apoptosis are compatible with the assumption that survivin expression induced by low-LET X-rays radiation may play a critical role in inhibiting apoptosis. However, after irradiation with ions, an anti-apoptotic function of survivin is not evident, possibly because of the serious damage produced by densely ionizing radiation.

探讨了肿瘤细胞中survivin的表达对高线性能量转移(LET)射线辐射敏感性的影响.根据Gen Bank提供的survivin序列,合成特异性survivin-siRNA寡核苷酸,转染人肝癌HepG2细胞,抑制survivin的表达.发现siRNA转染后诱导了HepG2细胞G2/M期阻滞,增加了自发性和辐射诱导的细胞凋亡.在高线性能量转移(LET)碳离子辐照后,siRNA转染细胞的克隆存活率明显下降.这些结果表明survivin表达是HepG2细胞产生对高LET射线辐射抗性的关键因素.

The influence of survivin expression on the radiosensitivity of tumor cells to high linear energy transfer (LET) radiation is investigated. Survivin-specific short-interfering RNA (siRNA) oligonucleotides were synthesized based on the survivin sequence provided by GenBank. Human hepatoma HepG2 cells were transfected with survivin-specific siRNA to inhibit its expressions. It was found that the transfection with surviving-specific siRNA increased the levels of G2/M arrest and the apoptotic rates induced by radiation in HepG2 cells. After exposure to high-LET carbon ions, a reduced clonogenic survival effect was observed in the cells treated with siRNA. These results show that survivin plays a key role in mediating the radioresistance of cells to high-LET radiation.

放射治疗是肿瘤三大治疗手段之一(手术治疗、放疗、化疗),如何提高肿瘤细胞的放射敏感性一直是科研人员关注的研究方向。电离辐射导致细胞死亡的主要方式是细胞凋亡,然而肿瘤细胞内往往细胞凋亡信号通路异常,降低了治疗效果。其中细胞内高水平表达的细胞凋亡抑制蛋白(Inhibitor of Apoptosis Protein,IAP)抑制了caspase分子的活性,而caspase分子正是细胞凋亡的执行分子。因此科学家们通过各种手段尤其是RNA干涉的方法以抑制肿瘤细胞内细胞凋亡抑制蛋白的表达及蛋白活性来达到提高肿瘤治疗效果的目的。 Survivin是凋亡抑制蛋白家族的一员,该蛋白在大多数恶性肿瘤中高表达,而在正常组织中检测不到,因此具有组织特异性。Survivin参与肿瘤细胞分化并抑制肿瘤细胞凋亡,它的高表达被证明与很多恶性肿瘤对放射治疗中产生的辐射抗性相关。本文主要研究了不同LET射线辐照下人肝癌HepG2细胞 survivin的表达及其表达对重离子诱导的生物学效应的影响。首先,我们使用不同LET的碳离子束和X射线辐照HepG2细胞,采用标准克隆形成法确定其辐射敏感性,利用流式细胞技术(FCM)检测辐射后细胞周期分布,RT-PCR和western blotting检测survivin的表达。结果显示,人肝HepG2癌细胞经不同LET射线照射,survivin表达是不同的。与低LET的X射线相比,高LET碳离子诱导的细胞损伤和周期阻滞更明显,从而诱导了更强烈的survivin表达。 接着,根据Genbank提供的survivin序列,合成特异性survivin-siRNA寡核苷酸,转染HepG2细胞,抑制survivin的表达。我们发现siRNA转染后诱导了细胞G2/M期阻滞,增加了自发性和辐射诱导的细胞凋亡。在碳离子辐照后,siRNA细胞克隆存活率明显下降。这些结果显示survivin表达是细胞产生高LET辐射抗性的关键因素。最后,我们初步探讨了在细胞凋亡过程中,survivin基因的作用机制。发现抑制survivin表达,对离子束辐射诱导的Bcl-2和Bax表达没有明显的影响。Survivin表达直接抑制了caspase-3和-9的活性,从而抑制了细胞凋亡。以上的实验结果表明:不同LET射线辐照细胞后survivin出现差异表达,与X射线相比,高LET重离子诱导的HepG2细胞中survivin表达更明显,以survivin为靶基因的siRNA技术应用于HepG2细胞,可以极大提高该细胞对重离子辐射的敏感性。本论文研究为临床应用重离子束治疗癌症提供了非常有用的基础数据,同时也为重离子束放射治疗联合基因治疗提供了新的思路

以控制LDLR基因表达的调控元件SRE为靶位点 ,使用含有荧光素酶报告基因的转染人肝细胞系HepG2筛选模型高通量筛选微生物来源的能够增加低密度脂蛋白受体 (LDLR)基因表达的具有潜在的降胆固醇作用的生物活性物质。在筛选过程中 ,通过对一株真菌产生的化合物ascofuranone的研究发现该化合物对测活用细胞株的LDLR荧光素酶报告基因的表达有中等强度的激活作用 ,其SC150 为 40 .4μmol/L。Ascofuranone激活LDLR基因表达这一新的生物活性的发现为深入研究该化合物的降血脂作用提供了新的重要启示

Persistent organic pollutants (POPs) are toxic substances, highly resistant to environmental degradation, which can bio-accumulate and have long-range atmospheric transport potential. Most studies focus on single compound effects, however as humans are exposed to several POPs simultaneously, investigating exposure effects of real life POP mixtures on human health is necessary. A defined mixture of POPs was used, where the compound concentration reflected its contribution to the levels seen in Scandinavian human serum (total mix). Several sub mixtures representing different classes of POP were also constructed. The perfluorinated (PFC) mixture contained six perfluorinated compounds, brominated (Br) mixture contained seven brominated compounds, chlorinated (Cl) mixture contained polychlorinated biphenyls and also p,p'-dichlorodiphenyldichloroethylene, hexachlorobenzene, three chlordanes, three hexachlorocyclohexanes and dieldrin. Human hepatocarcinoma (HepG2) cells were used for 2h and 48h exposures to the seven mixtures and analysis on a CellInsight™ NXT High Content Screening platform. Multiple cytotoxic endpoints were investigated: cell number, nuclear intensity and area, mitochondrial mass and membrane potential (MMP) and reactive oxygen species (ROS). Both the Br and Cl mixtures induced ROS production but did not lead to apoptosis. The PFC mixture induced the ROS production and likely induced cell apoptosis accompanied by the dissipation of MMP. Synergistic effects were evident for ROS induction when cells were exposed to the PFC+Br mixture. No significant effects were detected in the Br+Cl, PFC+Cl or total mixtures, which contain the same concentrations of chlorinated compounds as the Cl mixture plus additional compounds; highlighting the need for further exploration of POP mixtures in risk assessment.

The ability of human postprandial triacylglycerol-rich lipoproteins (TRLs), isolated after meals enriched in saturated fatty acids (SFAs), n-6 PUFAs, and MUFAs, to inhibit the uptake of I-125-labeled LDL by the LDL receptor was investigated in HepG2 cells. Addition of TRLs resulted in a dose-dependent inhibition of heparin-releasable binding, cell-associated radioactivity, and degradation products of I-125-labeled LDL (P < 0.001). SFA-rich Svedberg flotation rate (S-f) 60-400 resulted in significantly greater inhibition of cell-associated radioactivity than PUFA-rich particles (P = 0.016) and total uptake of I-125-labeled LDL compared with PUFA- and MUFA-rich particles (P = 0.02). Normalization of the apolipoprotein (apo)E but not apoC-III content of the TRLs removed the effect of meal fatty acid composition, and addition of an anti-apoE antibody reversed the inhibitory effect of TRLs on the total uptake of I-125-labeled LDL. Real time RT-PCR showed that the SFA-rich Sf 60-400 increased the expression of genes involved in hepatic lipid synthesis (P < 0.05) and decreased the expression of the LDL receptor-related protein 1 compared with MUFAs (P = 0.008). In conclusion, these findings suggest an alternative or additional mechanism whereby acute fat ingestion can influence LDL clearance via competitive apoE-dependent effects of TRL on the LDL receptor.-Jackson, K. Aims: The ATP-binding cassette transporters, ABCA1 and ABCG1, are LXR-target genes that play an important role in reverse cholesterol transport. We examined the effects of inhibitors of the cholesterol absorption (ezetimibe) and synthesis (statins) on expression of these transporters in HepG2 cells and peripheral blood mononuclear cells (PBMCs) of individuals with primary (and nonfamilial) hypercholesterolemia (HC). Materials & methods: A total of 48 HC individuals were treated with atorvastatin (10 mg/day/4 weeks) and 23 were treated with ezetimibe (10 mg/day/4 weeks), followed by simvastatin (10 mg/day/8 weeks) and simvastatin plus ezetimibe (10 mg of each/day/4 weeks). Gene expression was examined in statin- or ezetimibe-treated and control HepG2 cells as well as PBMCs using real-time PCR. Results: In PBMCs, statins and ezetimibe downregulated ABCA1 and ABCG1 mRNA expression but did not modulate NR1H2 (LxR-beta) and NR1H3 (LXR-alpha) levels. Positive correlations of ABCA1 with ABCG1 and of NR1H2 with NR1H3 expressions were found in all phases of the treatments. In HepG2 cells, ABCA1 mRNA levels remained unaltered while ABCG1 expression was increased by statin (1.0-10.0 mu M) or ezetimibe (5.0 mu M) treatments. Atorvastatin upregulated NR1H2 and NR1H3 only at 10.0 mu M, meanwhile ezetimibe (1.0-5.0 mu M) downregulated NR1H2 but did not change NR1H3 expression. Conclusion: Our findings reveal that lipid-lowering drugs downregulate ABCA1 and ABCG1 mRNA expression in PBMCs of HC individuals and exhibit differential effects on HepG2 cells. Moreover, they indicate that the ABCA1 and ABCG1 transcript levels were not correlated directly to LXR mRNA expression in both cell models treated with lipid-lowering drugs.
