OBJECTIVE To evaluate the effects of medetomidine, midazolam and ketamine (MMK) in captive gorillas after premedication with oral zuclopenthixol. STUDY DESIGN Case series. ANIMALS Six gorillas, two males and four females, aged 9-52 years and weighing 63-155 kg. METHODS The gorillas were given zuclopenthixol dihydrochloride 0.2 ± 0.05 mg kg(-1) per os twice daily for 3 days for premedication. On the day of anaesthesia the dose of zuclopenthixol was increased to 0.27 mg kg(-1) and given once early in the morning. Anaesthesia was induced with medetomidine 0.04 ± 0.004 mg kg(-1) , midazolam 0.048 ± 0.003 mg kg(-1) and ketamine 4.9 ± 0.4 mg kg(-1) intramuscularly (IM). Upon recumbency, the trachea was intubated and anaesthesia was maintained on 1-2% isoflurane in oxygen. Physiological parameters were monitored every 10 minutes and arterial blood gas analysis was performed once 30-50 minutes after initial darting. At the end of the procedure, 42-115 minutes after initial darting, immobilisation was antagonized with atipamezole 0.21 ± 0.03 mg kg(-1) and sarmazenil 5 ± 0.4 μg kg(-1) IM. RESULTS Recumbency was reached within 10 minutes in five out of six animals. One animal required two additional darts before intubation was feasible. Heart rate ranged from 60 to 85 beats minute(-1) , respiratory rate from 17 to 46 breaths minute(-1) and temperature from 36.9 to 38.3 °C. No spontaneous recoveries were observed and anaesthetic level was stable. Blood gas analyses revealed mild respiratory acidosis, and mean PaO(2) was 24.87 ± 17.16 kPa (187 ± 129 mmHg) with all values being above 13.4 kPa (101 mmHg). Recovery was smooth and gorillas were sitting within 25 minutes. CONCLUSION AND CLINICAL RELEVANCE The drug combination proved to be effective in anaesthetizing captive gorillas of various ages and both sexes, with minimal cardio-respiratory changes.

BACKGROUND: The only known albino gorilla, named Snowflake, was a male wild born individual from Equatorial Guinea who lived at the Barcelona Zoo for almost 40 years. He was diagnosed with non-syndromic oculocutaneous albinism, i.e. white hair, light eyes, pink skin, photophobia and reduced visual acuity. Despite previous efforts to explain the genetic cause, this is still unknown. Here, we study the genetic cause of his albinism and making use of whole genome sequencing data we find a higher inbreeding coefficient compared to other gorillas.RESULTS: We successfully identified the causal genetic variant for Snowflake's albinism, a non-synonymous single nucleotide variant located in a transmembrane region of SLC45A2. This transporter is known to be involved in oculocutaneous albinism type 4 (OCA4) in humans. We provide experimental evidence that shows that this amino acid replacement alters the membrane spanning capability of this transmembrane region. Finally, we provide a comprehensive study of genome-wide patterns of autozygogosity revealing that Snowflake's parents were related, being this the first report of inbreeding in a wild born Western lowland gorilla.CONCLUSIONS: In this study we demonstrate how the use of whole genome sequencing can be extended to link genotype and phenotype in non-model organisms and it can be a powerful tool in conservation genetics (e.g., inbreeding and genetic diversity) with the expected decrease in sequencing cost.

Le degré de rétention de l’arboricolisme dans le répertoire locomoteur des hominines fossiles du Pliocène est toujours matière à débat, les études ayant principalement porté sur la courbure des phalanges et la proportion des membres. Vu la récente découverte de DIK-1-1 (A. afarensis) et de la scapula qui lui est associée, l’étude de cet os d’un point de vue fonctionnel est intéressante, puisqu’il est directement impliqué dans la locomotion de presque tous les hominoïdes. Le but de cette étude est de tenter d’établir un lien entre l’orientation supéro-inférieure (SI) et antéro-postérieure (AP) de la cavité glénoïde de la scapula et les comportements locomoteurs chez les grands singes et l’humain moderne. Des analyses comparatives sur les adultes ont été réalisées pour 1) voir s’il existe des différences dans la morphologie étudiée entre les espèces et 2) voir si ces différences peuvent être expliquées par la taille corporelle. Des analyses ontogéniques ont aussi été réalisées pour voir si un accroissement de la taille corporelle pendant le développement et les changements locomoteurs qui y sont associés correspondent à un changement d’orientation de la cavité glénoïde. Les résultats montrent que les humains ont une cavité glénoïde qui est orientée moins supérieurement que les grands singes, mais que Pongo, bien qu’étant le plus arboricole, n’a pas l’orientation la plus supérieure. Les « knuckle-walkers » (Pan et Gorilla) se distinguent des autres hominoïdes avec une orientation de la surface glénoïde relative à l’épine plus inférieure. La taille corporelle ne semble pas influencer la morphologie étudiée, sauf parfois chez le gorille. Seuls l’humain et les mâles Pongo montrent un changement ontogénique dans l’orientation de la cavité glénoïde relativement à l’épine. Sur la base de ces résultats, l’orientation de la cavité glénoïde semble refléter partiellement la fonction du membre supérieur dans la locomotion, mais des recherches plus poussées sont nécessaires. Comprendre l'évolution de la bipédie est un élément essentiel à la recherche en paléoanthropologie, car ce comportement est le trait le plus important utilisé pour identifier les fossiles comme appartenant à la lignée des hominines. La topographie de la surface infradiaphysaire du fémur et du tibia pourrait donner un aperçu du comportement locomoteur des espèces fossiles, mais n'a pas été étudiée de façon approfondie. Ce trait reflète directement les différences dans la locomotion, puisque la surface change de topographie pour mieux résister aux charges encourues par les mouvements réguliers. Le plan infradiaphysaire du fémur chez les humain est relativement plat, tandis que la surface est plus irrégulière chez les grands singes. Dans ce projet, les métaphyses du genou ont été étudiées d’une manière quantifiée afin de percevoir les différences entre espèces et mieux comprendre le développement ontogénique de ces traits. Les angles formés par les protrusions et les creux de ces surfaces ont été mesurés à partir de points de repère enregistrés en trois-dimensions sur les métaphyses du genou chez les humains, chimpanzés, gorilles, et orangs-outans, et chez trois fossiles Australopithecus afarensis, afin d’observer de l’effet de facteurs tel le stade de croissance et l’appartenance à une espèce sur la topographie des plaques de croissance du genou. Les angles d’obliquité du fémur et du tibia ont aussi été mesurés et analysés. Les résultats ont révélé que le stade développemental et l’appartenance à une espèce et, par association, le mode de locomotion, ont un effet significatif sur les métaphyses du genou. Il a également été constaté que les mesures d'Australopithecus afarensis chevauchent les valeurs trouvées chez les humains et chez les grands singes, ce qui suggère que cette espèce avait possiblement conservé une composante arboricole dans son comportement locomoteur habituel.

Hepatitis B viruses (HBV) and related viruses, classified in the Hepadnaviridae family, are found in a wide variety of mammals and birds. Although the chimpanzee has been the primary experimental model of HBV infection, this species has not been considered a natural host for the virus. Retrospective analysis of 13 predominantly wild-caught chimpanzees with chronic HBV infection identified a unique chimpanzee HBV strain in 11 animals. Nucleotide and derived amino acid analysis of the complete HBV genome and the gene coding for the hepatitis B surface antigen (S gene) identified sequence patterns that could be used to reliably identify chimpanzee HBV. This analysis indicated that chimpanzee HBV is distinct from known human HBV genotypes and is closely related to HBVs previously isolated from a chimpanzee, gibbons, gorillas, and orangutans.

Self-recognition has been explored in nonlinguistic organisms by recording whether individuals touch a dye-marked area on visually inaccessible parts of their face while looking in a mirror or inspect parts of their body while using the mirror's reflection. Only chimpanzees, gorillas, orangutans, and humans over the age of approximately 2 years consistently evidence self-directed mirror-guided behavior without experimenter training. To evaluate the inferred phylogenetic gap between hominoids and other animals, a modified dye-mark test was conducted with cotton-top tamarins (Saguinus oedipus), a New World monkey species. The white hair on the tamarins' head was color-dyed, thereby significantly altering a visually distinctive species-typical feature. Only individuals with dyed hair and prior mirror exposure touched their head while looking in the mirror. They looked longer in the mirror than controls, and some individuals used the mirror to observe visually inaccessible body parts. Prior failures to pass the mirror test may have been due to methodological problems, rather than to phylogenetic differences in the capacity for self-recognition. Specifically, an individual's sensitivity to experimentally modified parts of its body may depend crucially on the relative saliency of the modified part (e.g., face versus hair). Moreover, and in contrast to previous claims, we suggest that the mirror test may not be sufficient for assessing the concept of self or mental state attribution in nonlinguistic organisms.

We introduce a new genetic distance for microsatellite loci, incorporating features of the stepwise mutation model, and test its performance on microsatellite polymorphisms in humans, chimpanzees, and gorillas. We find that it performs well in determining the relations among the primates, but less well than other distance measures (not based on the stepwise mutation model) in determining the relations among closely related human populations. However, the deepest split in the human phylogeny seems to be accurately reconstructed by the new distance and separates African and non-African populations. The new distance is independent of population size and therefore allows direct estimation of divergence times if the mutation rate is known. Based on 30 microsatellite polymorphisms and a recently reported average mutation rate of 5.6 x 10(-4) at 15 dinucleotide microsatellites, we estimate that the deepest split in the human phylogeny occurred about 156,000 years ago. Unlike most previous estimates, ours requires no external calibration of the rate of molecular evolution. We can use such calibrations, however, to test our estimate.

The aim in this study was to investigate the association between infants' developing interest in their self-image and the onset of mirror self-recognition (MSR). A longitudinal study was conducted with 98 infants who were seen at intervals of 3 months from 9-24 months of age. At each session, the infants were administered a preferential-looking test whereby they were presented with a video image of themselves alongside a video image of a same-aged peer in two conditions, unmarked and marked. From the 12-month session onwards, the infants were also administered a version of the standard mark test of MSR. The infants showed a significant preference for looking at images of themselves in both conditions coincident with the onset of MSR. This result indicates that developing an interest in the self-image is an important component in the development of MSR. (C) 2003 Elsevier Science Inc. All rights reserved.

Psychologists have studied self-recognition in human infants as an indication of self-knowledge (Amsterdam, 1972) and the development of abstract thought processes. Gallup (1970) modified the mark test used in human infant work to examine if nonhuman primates showed similar evidence of mirror self-recognition. Chimpanzees (Pan troglodytes) and orangutans (Pongo pygmnaeus) pass the mirror self-recognition test with limited mirror training or exposure. Other species of primates, such as gorillas and monkeys, have not passed the mirror test, despite extensive mirror exposure and training (Gallup, 1979). This project examined a gorilla (G. gorilla gorilla) named Otto in the traditional mark test. Using the modified mark-test, there were more incidents of touching the marked area while Otto was in front of the mirror than when he was not in front of the mirror. These results indicated that Otto was able to show some evidence of selfawareness.
