In the Mediterranean region the fruits of the strawberry tree (Arbutus unedo L.) may be fermented and distilled to produce a traditional beverage very much appreciated in Southern Europe. The aim of the present work was to study the diversity of the yeast population and the killer activity of the isolates identified as Saccharomyces cerevisiae, obtained during solid state industrial fermentations of the arbutus berries. The identification of the isolates was performed by the 5.8S rRNA-ITS region restriction analysis and by sequencing the D1/D2 region of the large subunit of the rRNA gene. At the start of the fermentations, various non-Saccharomyces species were detected including Aureobasidium pullulans, Dothichiza pithyophila, Dioszegia zsoltii, Hanseniaspora uvarum and yeasts belonging to the genera Metschnikowia, Cryptococcus and Rhodotorula. However, as the biological processes progressed the number of different species decreased with S. cerevisiae and Pichia membranaefaciens becoming dominant at advanced stages of the must fermentation that is characterized by high concentrations of ethanol. Forty three isolates identified as S. cerevisiae were tested for killer activity against two sensitive reference strains and Zygosaccharomyces bailii. Their killer sensitivity in relation to five killer referenced toxins (K2, K5, K8, K9 and K10) was also studied. Out of the isolates analyzed, 95.3% were sensitive and 4.7% were tolerant against the killer toxins tested. Only three isolates revealed killer activity against one sensitive strain and two of them against the spoiler yeast Z. bailii. The microbiota obtained revealed an interesting potential to be used as starter cultures to overcome unpredictable uncontrolled fermentations of the arbutus fruits as well as in other applications of biotechnological interest. An unstructured mathematical model is proposed to describe the fermentation kinetics of growth, lactic acid production, pH and sugar consumption by Lactobacillus plantarum as a function of the buffering capacity and initial glucose concentration of the culture media. Initially the experimental data of L plantarum fermentations in synthetic media with different buffering capacity and glucose were fitted to a set of primary models. Later the parameters obtained from these models were used to establish mathematical relationships with the independent variables tested. The models were validated with 6 fermentations of L. plantarum in different cereal-based media. In most cases the proposed models adequately describe the biochemical changes taking place during fermentation and are a promising approach for the formulation of cereal-based probiotic foods. Previously, using an in vitro static batch culture system, it was found that rice bran (RB), inulin, fibersol, mannanoligosaccharides (MOS), larch arabinogalactan and citrus pectin elicited prebiotic effects (in terms of increased numbers of bifidobacteria and lactic acid bacteria) on the faecal microbiota of a dog. The aim of the present study was to confirm the prebiotic potential of each individual substrate using multiple faecal donors, as well as assessing the prebiotic potential of 15 substrate blends made from them. Anaerobic static and stirred, pH-controlled batch culture systems inoculated with faecal samples from healthy dogs were used for this purpose. Fluorescence in situ hybridization (FISH) analysis using seven oligonucleotide probes targeting selected bacterial groups and DAPI (total bacteria) was used to monitor bacterial populations during fermentation runs. High-performance liquid chromatography was used to measure butyrate produced as a result of bacterial fermentation of the substrates. RB and a MOS/RB blend (1:1, w/w) were shown to elicit prebiotic and butyrogenic effects on the canine microbiota in static batch culture fermentations. Further testing of these substrates in stirred, pH-controlled batch culture fermentation systems confirmed the prebiotic and butyrogenic effects of MOS/RB, with no enhancement of Clostridium clusters I and II and Escherichia coli populations.

Cyclodextrin glycosyltransferase (CGTase) is an enzyme that produces cyclodextrins from starch by an intramolecular transglycosylation reaction. Cyclodextrins have been shown to have a number of applications in the food, cosmetic, pharmaceutical, and chemical industries. In the current study, the production of CGTase by Paenibacillus campinasensis strain H69-3 was examined in submerged and solid-state fermentations. P. campinasensis strain H69-3 was isolated from the soil, which grows at 45 C, and is a Gram-variable bacterium. Different substrate sources such as wheat bran, soybean bran, soybean extract, cassava solid residue, cassava starch, corn starch, and other combinations were used in the enzyme production. CGTase activity was highest in submerged fermentations with the greatest production observed at 48-72 h. The physical and chemical properties of CGTase were determined from the crude enzyme produced from submerged fermentations. The optimum temperature was found to be 70-75 degrees C, and the activity was stable at 55 degrees C for 1 h. The enzyme displayed two optimum pH values, 5.5 and 9.0 and was found to be stable between a pH of 4.5 and 11.0.

The selected yeast strains were examined for their ability lo grow, to retain cell viability and to ferment diluted sugar cane juice (15% total sugar, w/v) to ethanol at 40-degrees-C. The degree of agitation (aeration) affects the thermotolerance while the method used for isolation of the strains appears to have no significant effect. The yeast isolated are aerobically fermentative with increased levels of fermentation and growth resulting from agitation (aeration), the exact level of these increases being dependent on the strain used.

The growth kinetics, sporulation, and toxicity of Bacillus thuringiensis var. israelensis were evaluated through the analysis of batch cultures with different dissolved oxygen (DO) profiles. Firstly, DO was maintained constant at 5%, 20%, or 50% throughout fermentation in order to identify the most suitable one to improve the main process parameters. Higher biomass concentration, cell productivity, and cell yield based on glucose were obtained with 50% DO. The higher aeration level also resulted in higher spore counts and markedly improved the toxic activity of the fermentation broth, which was 9-fold greater than that obtained with 5% DO (LC50 of 39 and 329 mg/L, respectively). Subsequently, using a two-stage oxygen supply strategy, DO was kept at 50% during the vegetative and transition phases until the maximum cell concentration was achieved. Then, DO was changed to 0%, 5%, 20%, or 100% throughout sporulation and cell lysis phases. The interruption of oxygen supply strongly reduced the spore production and thoroughly repressed the toxin synthesis. On the contrary, when DO was raised to 100% of saturation, toxic activity increased approximately four times (LC50 of 8.2 mg/L) in comparison with the mean values reached with lower DO levels, even though spore counts were lower than that from the 50% DO assay. When pure oxygen was used instead of normal air, it was possible to obtain 70% of the total biomass concentration achieved in the air assays; however, cultures did not sporulate and the toxin synthesis was consequently suppressed.

Das Ziel dieser Arbeit war es, mehr Informationen über unkonventionelle Gründe für Gärstockungen zu gewinnen und neue Wege zu finden, diese zu überwinden. Mikrobielle Sukzession und die chemische Zusammensetzung bei der Gärung wurden in zwei aufeinander folgenden Jahren in einem Weingut von der oberen Mosel in Deutschland studiert. Es gab keinen Hinweis darauf, dass die isolierten Bakterienspezies oder chemischen Komponenten von Most und Jungwein an schleppenden oder stockenden Gärungen beteiligt waren. Ferner konnte während dieser Arbeit gezeigt werden, dass Saccharomyces bayanus die dominierende Weinhefe in diesem Weingut war statt der klassischen und bekannten Weinhefe Saccharomyces cerevisiae. Während der Gärstockung konnte ein Dreifach-Hybrid Saccharomyces cerevisiae x Saccharomyces kudriavzevii x Saccharomyces bayanus wachsen, Saccharomyces bayanus ersetzen und die Gärung beenden. Beide isolierten Hefestämme Saccharomyces bayanus Stamm HL 77 und der Dreifach-Hybrid Saccharomyces cerevisiae x Saccharomyces kudriavzevii x Saccharomyces bayanus Stamm HL 78 konnten Glucose und Fructose von Anfang an verwerten und konnten bei niedrigen Temperaturen von 15 °C und in der Abwesenheit von Hefe-verwertbarem Stickstoff in Form von Ammonium wachsen, solange Aminosäuren im Medium vorhanden waren, im Gegensatz zu einer kommerziellen Saccharomyces cerevisiae-Starterkultur. Chemische Untersuchungen ergaben, dass Hefe-verwertbarer Stickstoff in dem kooperierenden Weingut mit einem Maximum von 160 mg/l zu Beginn der Gärung vorhanden war und auf 40 mg/L verringert war nach zwei Wochen. Aus diesem Grund sind beide isolierten Hefestämme interessant als Starterkulturen in diesem Weingut und dies kann neben der niedrigen Temperatur im Keller auch ein Grund sein, warum Saccharomyces cerevisiae nicht die dominierende Weinhefe in diesem Fall ist. Der Dreifach-Hybrid Saccharomyces cerevisiae x Saccharomyces kudriavzevii x Saccharomyces bayanus Stamm HL 78 ist in der Lage, Fructose noch effizienter zu nutzen als Saccharomyces bayanus Stamm HL 77 und ist weniger abhängig von der Aminosäurekonzentration. Dieser Stamm wurde bereits erfolgreich bei diesem Projekt eingesetzt, um eine Gärstockung in dem kooperierenden Weingut zu beheben. Es ist bekannt, dass Saccharomyces-Hybride in der Weinherstellung vorkommen aber ihre Rolle bei der Überwindung von Gärstockungen wurde bisher noch nicht beschrieben. Diese Ergebnisse sind nützlich, um Gärstockungen zu vermeiden oder zu überwinden mit der selektiven Verwendung dieser Hefestämme in verschiedenen Stadien der Gärung. Das kooperierende Weingut, welches im oberen Qualitätssegment platziert ist, hatte jedes Jahr Probleme mit Gärstockungen. Daher ist die Anwendung der Dreifach-Hybriden Saccharomyces cerevisiae x Saccharomyces kudriavzevii x Saccharomyces bayanus Stamm HL 78 eine große Chance, Gärstockungen und finanzielle Verluste ohne kommerzielle Starterkulturen oder andere übliche Praktiken, die zu einer Veränderung des Aromaprofils führen können, zu vermeiden. Die beschriebenen Untersuchungen stellen ein Modell dar, um Gärstockungen auch in anderen Weingütern, die Spontangärungen anwenden, zu überwinden.

Torulaspora delbrueckii is a non-Saccharomyces yeast with interesting metabolic and physiological properties of potential use in oenology. This work examines the fermentative behaviour of five strains of T. delbrueckii in sequential fermentations with Saccharomyces cerevisiae, analysing the formation of aromatic compounds, polyalcohols and pigments. The fermentative power of these five strains ranged between 7.6 and 9.0% v/v ethanol; the associated volatile acidity was 0.2e0.7 g/l acetic acid. The production of glycerol was inferior to that of S. cerevisiae alone. The mean 2,3-butanediol concentration reached in single-culture S. cerevisiae fermentations was 73% higher than in the five sequential T. delbrueckii/S. cerevisiae fermentations. However, these fermentations produced larger quantities of diacetyl, ethyl lactate and 2-phenylethyl acetate than single-culture S. cerevisiae fermentation. 3-ethoxy propanol was produced only in the sequential fermentations. The five sequential fermentations produced smaller quantities of vitisin A and B than single-culture S. cerevisiae fermentation. In tests performed prior to the addition of the S. cerevisiae in the sequential fermentations, none of the T. delbrueckii strains showed any extracellular hydroxycinnamate decarboxylase activity. They therefore produced no vinyl phenolic pyranoanthocyanins.

This work examines the physiology of a new commercial strain of Torulaspora delbrueckii in the production of red wine following different combined fermentation strategies. For a detailed comparison, several yeast metabolites and the strains implantation were measured over the entire fermentation period. In all fermentations in which T. delbrueckii was involved, the ethanol concentration was reduced; some malic acid was consumed; more pyruvic acid was released, and fewer amounts of higher alcohols were produced. The sensorial properties of final wines varied widely, emphasising the structure of wine in sequential fermentations with T. delbrueckii. These wines presented the maximum overall impression and were preferred by tasters. Semi-industrial assays were carried out confirming these differences at a higher scale. No important differences were observed in volatile aroma composition between fermentations. However, differences in mouthfeel properties were observed in semi-industrial fermentations, which were correlated with an increase in the mannoprotein content of red wines fermented sequentially with T. delbrueckii.
