[ES]El presente trabajo se centra en el RD 1254/1999, decreto que obliga a aquellos establecimientos que trabajan con sustancias peligrosas y pueden ocasionar accidentes graves con consecuencias en el exterior (nube tóxica, explosión, incendio, radiación, etc.) a hacer un análisis de riesgo de los mencionados accidentes. En base a este análisis se programan los planes de autoprotección así como los planes de emergencia exterior y se establecen los sistemas de gestión de la seguridad. Se estudia un caso práctico en el que se elabora el Análisis de Riesgo de una planta en la que se almacena metanol, y se calculan las distancias a las que llegarían las consecuencias dañinas en caso de accidentes con el metanol.

When designing a new passenger ship or modifying an existing design, how do we ensure that the proposed design and crew emergency procedures are safe from an evacuation resulting from fire or other incident? In the wake of major maritime disasters such as the Scandinavian Star, Herald of Free Enterprise, Estonia and in light of the growth in the numbers of high density, high-speed ferries and large capacity cruise ships, issues concerning the evacuation of passengers and crew at sea are receiving renewed interest. Fire and evacuation models with features such as the ability to realistically simulate the spread of fire and fire suppression systems and the human response to fire as well as the capability to model human performance in heeled orientations linked to a virtual reality environment that produces realistic visualisations of the modelled scenarios are now available and can be used to aid the engineer in assessing ship design and procedures. This paper describes the maritimeEXODUS ship evacuation and the SMARTFIRE fire simulation model and provides an example application demonstrating the use of the models in performing fire and evacuation analysis for a large passenger ship partially based on the requirements of MSC circular 1033. The fire simulations include the action of a water mist system.

Burkholderia cenocepacia is an opportunistic pathogen that survives intracellularly in macrophages and causes serious respiratory infections in patients with cystic fibrosis. We have previously shown that bacterial survival occurs in bacteria-containing membrane vacuoles (BcCVs) resembling arrested autophagosomes. Intracellular bacteria stimulate IL-1ß secretion in a caspase-1-dependent manner and induce dramatic changes to the actin cytoskeleton and the assembly of the NADPH oxidase complex onto the BcCV membrane. A Type 6 secretion system (T6SS) is required for these phenotypes but surprisingly it is not required for the maturation arrest of the BcCV. Here, we show that macrophages infected with B. cenocepacia employ the NLRP3 inflammasome to induce IL-1ß secretion and pyroptosis. Moreover, IL-1ß secretion by B. cenocepacia-infected macrophages is suppressed in deletion mutants unable to produce functional Type VI, Type IV, and Type 2 secretion systems (SS). We provide evidence that the T6SS mediates the disruption of the BcCV membrane, which allows the escape of proteins secreted by the T2SS into the macrophage cytoplasm. This was demonstrated by the activity of fusion derivatives of the T2SS-secreted metalloproteases ZmpA and ZmpB with adenylcyclase. Supporting this notion, ZmpA and ZmpB are required for efficient IL-1ß secretion in a T6SS dependent manner. ZmpA and ZmpB are also required for the maturation arrest of the BcCVs and bacterial intra-macrophage survival in a T6SS-independent fashion. Our results uncover a novel mechanism for inflammasome activation that involves cooperation between two bacterial secretory pathways, and an unanticipated role for T2SS-secreted proteins in intracellular bacterial survival.

This paper discusses the work of Claude Parent and The Serving Library, considering the critiques generated by their intersecting of architecture, art and editorial design. Through focus on the ways in which hosting environment, architecture and forms of expanded publishing can serve to dissolve disciplinary boundaries and activities of production, spectatorship and reception, it draws on the lineage of 1960s/70s Conceptual Art in considering these practices as a means through which to escape medium specificity and spatial confinement. Relationships between actual and virtual space are then read against this broadening of aesthetic ideas and the theory of critical modernity.

In Tumoren und Onkogen-transformierten Zellen finden sich häufig Defizienzen in der Expression von Komponenten der MHC Klasse I-Antigenprozessierung, die mit einer verminderten MHC Klasse I-Oberflächenexpression und einer reduzierten Sensitivität der Zellen gegenüber einer ZTL-vermittelten Lyse gekoppelt sein können. Da in den meisten Fällen die reduzierten Expressionsmuster über Zytokine revertiert werden können, werden verschiedene Regulationsmechanismen als Ursache für die Defizienzen postuliert. Auch in Zellen, die den „human epidermal growth factor receptor 2“ (HER-2/neu) überexprimieren, wurden derartige „Immune escape“-Mechanismen identifiziert. Aufgrund der Amplifikation und/oder Überexpression dieses Onkogens in Tumoren, die mit einer schnellen Progression der Erkrankung und einer schlechten Heilungsprognose assoziiert ist, wurden zahlreiche Therapien entwickelt, die auf einer Mobilisierung des Immunsystems gegenüber HER-2/neu oder dessen Blockade durch spezifische Antikörper abzielen. Die bisher jedoch nur unzureichenden Erfolge dieser Therapien könnten ihre Ursache in einer verminderten Immunogenität der HER-2/neu+-Zellen aufgrund von Defizienzen in der MHC Klasse I-Antigenprozessierung haben, weshalb die Untersuchung der molekularen Ursachen dieser Suppression für die Therapie von HER-2/neu+-Tumoren von besonderer Bedeutung ist. In dieser Arbeit wurde anhand eines in vitro-Systems ein HER-2/neu-vermittelter „Immune escape“-Phänotyp charakterisiert und die zugrunde liegenden molekularen Mechanismen untersucht. Hierzu wurden murine, HER-2/neu--NIH3T3-Zellen mit HER-2/neu-transfizierten NIH3T3-Zellen verglichen. Die Untersuchung zeigte, dass die Oberflächenexpression von MHC Klasse I-Antigenen bei einer HER-2/neu-Überexpression vermindert ist. Dies ist assoziiert mit reduzierten Expressionen von LMP2, LMP10, PA28a, PA28b, ERAAP, TAP1, TAP2, und Tapasin, einem blockiertem TAP-Transport und einer fehlenden Sensitivität gegenüber einer ZTL-vermittelten Lyse. Da die analysierten Defekte durch eine Stimulation mit IFN‑g wieder revertiert werden können, wird eine transkriptionelle oder translationelle Regulation der betroffenen Gene durch HER-2/neu postuliert. Aufgrund dieser Ergebnisse ist eine T-Zell-vermittelte Therapie von HER-2/neu+-Tumoren als kritisch anzusehen. Die Untersuchung der Promotoren von TAP1/LMP2, TAP2 und Tapasin ergab geringere und durch IFN‑g-induzierbare Promotoraktivitäten in den HER-2/neu+-Zellen im Vergleich zu den HER-2/neu—-Zellen. Mittels Mutagenese-PCR und Gelretardationsanalysen konnte die Bindung eines Komplexes an zwei E2F- und einer P300-Bindungsstelle im Tapasin-Promotor identifiziert werden, die für die HER-2/neu-vermittelte Hemmung der Tapasin-Promotor­aktivität essentiell ist. Eine Inaktivierung der E2F- und P300-Motve in den TAP1/LMP2- und TAP2-Promotoren hatte dagegen keinen Einfluss auf die HER-2/neu-vermittelte Blockade der Promotoraktivität. Ein Vergleich der Promotoraktivitäten der HER-2/neu+- mit Ras-transformierten Zellen ergab, dass die TAP1/LMP2- und TAP2-Promotoren in beiden Zellen supprimiert werden, während der Tapasin-Promotor bei Ras-Transformation nicht beein­trächtigt ist. Der Einsatz von Inhibitoren zeigte, dass die Suppression des Tapasin-Promotors vermutlich über die PLC-g-PKC-Kaskade erfolgt. Dagegen konnte mit Inhibitoren gegen MAPK und PI3Kinase kein vergleichbarer Effekt erzielt werden. Aufgrund dieser Daten wird postuliert, dass HER-2/neu über die Signalkaskade PLC-g–PKC–E2F/P300 die Tapasin-Promotoraktivität supprimiert, wohingegen noch bisher unbekannte Signalkaskaden von HER-2/neu und Ras zu einer Hemmung der TAP1/LMP2- und TAP2-Promotoraktivität führen. Da die Komplexbildung von E2F und P300 auch im Zellzyklus eine Rolle spielt, wird eine negative Korrelation zwischen Zell-Proliferation und MHC Klasse I-Antigenpräsentation postuliert, die Gegenstand künftiger Studien sein wird.

This work comprises three different types of unconventional correlated systems.rnChapters 3-5 of this work are about the open shell compounds Rb4O6 and Cs4O6. These mixed valent compounds contain oxygen in two different modifications: the closed-shell peroxide anion is nonmagnetic, whereas the hyperoxide anion contains an unpaired electrons in an antibonding pi*-orbital. Due to this electron magnetic ordering is rendered possible. In contrast to theoretical predictions, which suggested half-metallic ferromagnetism for Rb4O6,rndominating antiferromagnetic interactions were found in the experiment. Besidesrna symmetry reduction due to the mixed valency, strong electronic correlations of this highly molecular system determine its properties; it is a magnetically frustrated insulator. The corresponding Cs4O6 was found to show similar properties.rnChapters 6-9 of this work are about intermetallic Heusler superconductors. rnAll of these superconductors were rationally designed using the van Hove scenario as a working recipe. A saddle point in the energy dispersion curve of a solid leads to a van Hove singularity in the density of states. In the Ni-based and Pd-based Heusler superconductors presented in this work this sort of a valence instability occurs at the high-symmetry L point and coincides or nearly coincides with the Fermi level. The compounds escape the high density of states at the Fermi energy through a transition into the correlated superconducting state.rnChapter 10 of this work is about the tetragonally distorted ferrimagnetic DO22 phase of Mn3Ga. This hard-magnetic modification is technologically useful for spin torque transfer applications. The phase exhibits two different crystallographic sites that are occupied by Mn atoms and can thus be written as Mn2MnGa. The competition between the mainly itinerant moments of the Mn atoms at the Wyckoff position 4d and the localized moments of the Mn atoms at the Wyckoff position 2b leads to magnetic correlations. The antiferromagnetic orientation of these moments determines the compound to exhibit a resulting magnetic moment of approximately 1 µB per formula unit in a partially compensated ferrimagnetic configuration.

Introduction Gene expression is an important process whereby the genotype controls an individual cell’s phenotype. However, even genetically identical cells display a variety of phenotypes, which may be attributed to differences in their environment. Yet, even after controlling for these two factors, individual phenotypes still diverge due to noisy gene expression. Synthetic gene expression systems allow investigators to isolate, control, and measure the effects of noise on cell phenotypes. I used mathematical and computational methods to design, study, and predict the behavior of synthetic gene expression systems in S. cerevisiae, which were affected by noise. Methods I created probabilistic biochemical reaction models from known behaviors of the tetR and rtTA genes, gene products, and their gene architectures. I then simplified these models to account for essential behaviors of gene expression systems. Finally, I used these models to predict behaviors of modified gene expression systems, which were experimentally verified. Results Cell growth, which is often ignored when formulating chemical kinetics models, was essential for understanding gene expression behavior. Models incorporating growth effects were used to explain unexpected reductions in gene expression noise, design a set of gene expression systems with “linear” dose-responses, and quantify the speed with which cells explored their fitness landscapes due to noisy gene expression. Conclusions Models incorporating noisy gene expression and cell division were necessary to design, understand, and predict the behaviors of synthetic gene expression systems. The methods and models developed here will allow investigators to more efficiently design new gene expression systems, and infer gene expression properties of TetR based systems.
