We characterized 28 new isolates of Trypanosoma cruzi IIc (TCIIc) of mammals and triatomines from Northern to Southern Brazil, confirming the widespread distribution of this lineage. Phylogenetic analyses using cytochrome b and SSU rDNA sequences clearly separated TCIIc from TCIIa according to terrestrial and arboreal ecotopes of their preferential mammalian hosts and vectors. TCIIc was more closely related to TCIId/e, followed by TCIIa, and separated by large distances from TCIIb and TCI. Despite being indistinguishable by traditional genotyping and generally being assigned to Z3, we provide evidence that TCIIa from South America and TCIIa from North America correspond to independent lineages that circulate in distinct hosts and ecological niches. Armadillos, terrestrial didelphids and rodents, and domestic dogs were found infected by TCIIc in Brazil. We believe that, in Brazil, this is the first description of TCIIc from rodents and domestic dogs. Terrestrial triatomines of genera Panstrongylus and Triatoma were confirmed as vectors of TCIIc. Together, habitat, mammalian host and vector association corroborated the link between TCIIc and terrestrial transmission cycles/ecological niches. Analysis of ITS1 rDNA sequences disclosed clusters of TCIIc isolates in accordance with their geographic origin, independent of their host species. (C) 2009 Elsevier B.V. All rights reserved.

Analysis of the phylogenetic relationships among trypanosomes from vertebrates and invertebrates disclosed a new lineage of trypanosomes circulating among anurans and sand flies that share the same ecotopes in Brazilian Amazonia. This assemblage of closely related trypanosomes was determined by comparing whole SSU rDNA sequences of anuran trypanosomes from the Brazilian biomes of Amazonia, the Pantanal, and the Atlantic Forest and from Europe, North America, and Africa, and from trypanosomes of sand flies from Amazonia. Phylogenetic trees based on maximum likelihood and parsimony corroborated the positioning of all new anuran trypanosomes in the aquatic clade but did not support the monophyly of anuran trypanosomes. However, all analyses always supported four major clades (An01-04) of anuran trypanosomes. Clade An04 is composed of trypanosomes from exotic anurans. Isolates in clades An01 and An02 were from Brazilian frogs and toads captured in the three biomes studied, Amazonia, the Pantanal and the Atlantic Forest. Clade An01 contains mostly isolates from Hylidae whereas clade An02 comprises mostly isolates from Bufonidae; and clade An03 contains trypanosomes from sand flies and anurans of Bufonidae, Leptodactylidae, and Leiuperidae exclusively from Amazonia. To our knowledge, this is the first study describing morphological and growth features, and molecular phylogenetic affiliation of trypanosomes from anurans and phlebotomines, incriminating these flies as invertebrate hosts and probably also as important vectors of Amazonian terrestrial anuran trypanosomes.

The seasonal distribution of Lutzomyia longipalpis was studied in two forested and five domiciliary areas of the urban area of Campo Grande; MS, from December 2003 to November 2005. Weekly captures were carried out with CDC light traps positioned on ground and in the canopy inside a residual forest and on the edge (ground) of a woodland and in at least one of the following ecotopes in peridomiciles-a cultivated area, a chicken coop, a pigsty, a kennel, a goat and sheep shelter and an intradomicile. A total of 9519 sand flies were collected, 2666 during the first year and 6853 during the second. L. longipalpis was found throughout the 2-year period, presenting smaller peaks at intervals of 2-3 months and two greater peaks, the first in February and the second in April 2005, soon after periods of heavy rain. Only In one of the woodlands was a significant negative correlation (p < 0.05) between the number of insects and temperature during the first year and the climatic factors (temperature, RHA and rain) was observed. In the domiciliary areas in four domiciles some positive correlations (p < 0.05) occurred in relation to one or more climatic factors; however, the species shows a clear tendency to greater frequency (72%) in the rainy season than in the dry (28%). Thus, we recommend an intensification of the VL control measures applied in Campo Grande, MS, during the rainy season with a view to reducing the risk of the transmission of the disease. The scarcity of occurrence records of culicid species, transmitting insects of etiologic agents for arboviruses, protozoan diseases and philarioses, and lack of information about their interactions with the forest environment make the Caatinga one of the most unknown biomes in Brazil. The aim of this study was to identify culicid species in a conservation unit located in a preserved area of the Caatinga and the possible associations with local abiotic factors, as well as characterize the natural and artificial ecotopes present in the Caatinga of Rio Grande do Norte state. The Ecological Station of Serido (ESEC-Seridó) is a Caatinga Conservation Unit located in a rural area of Rio Grande do Norte. The hot and dry climate of the Caatinga, along with a short rainy season, favor the adaptation and resistance of plants, resulting in completely distinct landscapes over the course of the year. The study took place over the course of one year, with monthly collections between 10h-12h, 14h-16h and 18h-20h. Adult mosquitoes were collected in a forest area using a manual suction trap with human bait as attractant at the three times and a Shannon trap for night collections. During the rainy season ovitraps were randomly distributed at different sites. The different capture sites were observed and photographed. Immature and adult insects captured were taken to the laboratory for identification. Of the 5081 insects collected in human-bait and Shannon traps, 75% were Mansonia wilsoni. In addition, Aedeomyia, Aedes, Anopheles, Coquillettidia, Culex, Haemagogus, Mansonia and Psorophora were identified. Of the 92 Haemagogus eggs found, 71 were collected on the banks of a reservoir. Mosquito larvae were found and photographed in an artificial breeding site of a neighboring rural area. Humidity had the greatest influence on culicid density in the Caatinga. Rain was important in the appearance of Haemagogus, Anopheles, Aedes and Psorophora. Temperature had a negative influence on Anopheles triannulatus. The occurence of important vector species of diseases in a little known preserved area of the Caatinga as well as environmental and sociocultural questions are factors that favor the emergence of transmitting insects, making these areas potential subject to the resurgence of diseases

Results are given of a phytosociological study of a forest located in the Morro do Diabo State Park. in the county of Teodoro Sampaio (22 degrees 31'S, 52 degrees 10'W), Pontal do Paranapanema region, state of São Paulo. The point-centered quarter method, using 300 sampling points, was used establishing 3 trails of 100 sampling points each, 10 m interdistant. All woody vegetation with a breast height diameter of 5 cm or more were samplied. Three distinct ecotopes could be identified in the forest: the lower, thc slope and the peak, which were analised separately.The families that presented the greatest numbers of genera, species and individuals were Leguminosae, Euphorbiaceae and Myrtaceae, followed by Rutaceae, Lauraceae, Meliaceae and Moracea. A total of 104 species, 85 genera and 37 families were sampled by the phytosociological survey. The relative sociological position of each plant family or species varied according to ecotope.

A doença de Chagas aguda (DCA) é endêmica na Amazônia Brasileira sendo a via oral a principal forma de transmissão com surtos familiares ou multifamiliares. Esta via independe da colonização de triatomíneos no domicílio e a ocorrência é regular com média de 100 casos/ano e 5% de óbitos. Apresenta distribuição espaço-temporal bem definida, colocando a enfermidade como emergência de importância em saúde pública nos estados do Pará, Amapá e Amazonas. A presença de mamíferos e triatomíneos silvestres, infectados naturalmente com o c e habitando distintos ecótopos terrestres e arbóreos, mantém um intenso ciclo enzoótico em toda a Amazônia. Perfis moleculares de linhagens de T. cruzi na região estão associados a hospedeiros mamíferos (incluindo o homem), triatomíneos, ecótopos e manifestações clínicas. Foram estudados quatro surtos de DCA ocorridos nos Municípios de Barcarena, Belém e Cachoeira do Arari no Estado do Pará e em Santana, no Estado do Amapá e abordados os aspectos epidemiológicos (parasitológico e sorológico manifestações clínicas, reservatórios e triatomíneos silvestres associados aos surtos). Foi investigado também em São Luís, Estado do Maranhão, o ciclo domiciliar e silvestre do T. cruzi, porém sem a ocorrência de casos de DCA. O estudo incluiu também a genotipagem molecular de T. cruzi pelo gene de mini-exon dos isolados (homem, mamíferos e triatomíneos silvestres) associados aos diferentes ciclos de transmissão. O diagnóstico parasitológico foi confirmado em 63 pacientes com a seguinte sensibilidade nos testes aplicados: 41,3% (26/63) pela gota espessa; 58,7% (37/63) no QBC; 79,4% (50/63) no xenodiagnóstico e 61,9% (39/63) na hemocultura. A sorologia de 2648 pessoas por hemaglutinação indireta (HAI) foi de 3,05% (81/2648) e imunofluorescência indireta IFI apresentaram respectivamente resultados de e 2,49% (66/2648) para IgG e 2,37 (63/2648) para IgM. Os resultados em São Luís foram todos negativos. Foram capturados 24 mamíferos, 13 Didelphis marsupialis, 1 Marmosa cinerea, 5 Philander opossum, 3 Metachirus nudicaudatus, 1 Oryzomys macconnelli, 1 Oecomys bicolor e 433 R. rattus. A taxa de infecção para T. cruzi foi de 7,14% (29/404). Um total de 3279 triatomíneos foi capturado sendo: Triatoma rubrofasciata (n=3008), com taxa de infecção (TI) de 30.46%, (39/128), Rhodnius robustus (n=137), com TI de 76% (79/104), R. pictipes (n=94), TI de 56,9% (49/86%), E. mucronatus (n=6) e P. geniculatus (n=12) com TI de 50% e as demais espécies sem infecção R. neglectus (n=5) e P. lignarius (n=6). As palmeiras foram os principais ecótopos dos triatomíneos silvestres. O urucurizeiro (S. martiana) apresentava infestação de 47,41% (101/213) dos triatomíneos; o “inajazeiro” (Maximiliana regia) 35,21% (75/213); o “babaçueiro” (Orbgnya. speciosa) 5,16% (11/213); o “dendezeiro” (Eleas melanoccoca) 1,87% (4/213) e a “bacabeira” (Oenocarpus bacaba) 10,32% (22/213). Para a genotipagem foram obtidos 46 isolados de tripanossomas de origem humana, 31 isolamentos de mamíferos silvestres e 74 amostras de triatomíneos. Todos os isolados foram caracterizados como da linhagem TcI de T. cruzi. Todos os casos humanos no Pará foram caracterizados como positivos por exame parasitológico. Nem todos os casos de Santana, Amapá, apresentaram casos parasitológicos positivos, pela demora do diagnóstico, mesmo assim estes foram definidos como DCA. Exames como xenodiagnóstico, hemocultura e o QBC® foram mais sensíveis do que a gota espessa. A sorologia por HAI e IFI (IgG e IgM) tiveram excelente sensibilidade para detectar os casos agudos em tempos distintos de infecção. O achado de mamíferos (D. marsupilais) e triatomíneos silvestres (R. pictipes e P. geniculatus) infectados com consideráveis taxas de infecção para T. cruzi no entorno das residências dos pacientes sustentam a importância destes hospedeiros associados à transmissão da DCA. Apesar de na Amazônia circularem vários genótipos de T. cruzi nos diferentes hospedeiros, neste trabalho foi identificada somente a linhagem TCI de T. cruzi, a mais predominante na Região. Em São Luís, Maranhão, embora sem registro de casos de DCA apresenta um ciclo domiciliar associados ao rato doméstico e o triatomíneo da espécie T. rubrofasciata, e um ciclo silvestre mantido por didelfídeos. Nos dois ciclos circulam a linhagem TCI de T. cruzi. Estudos com marcadores de maior resolução com isolados de T. cruzi regionais podem ajudar a esclarecer os ciclos de transmissão, as rotas de contaminação e os hospedeiros envolvidos em casos de DCA na Amazônia.

Introduction: This study analyzed the occurrence and the contamination of triatomines by trypanosomatids in Orbignya speciosa (babassu) specimens in the State of Rondonia, Brazil, in two different environments (pasture and woods). Methods: Capture of triatomines on babassus and microscopic search for trypanosomatids in their digestive tube were carried out. Results: Four hundred ninety-four (494) specimens were captured (Rhodnius prolixus and R.robustus), of which 35.6% of the triatomines were positive for trypanosomatids. Conclusions: The high index of natural infection along with the abundance of triatomines points out to the necessity to create an epidemiological surveillance system to monitor vector-borne transmission and deepen the studies on the ecology of such vectors in the Amazon.

We determined the lipid distributions (n-alkanes, n-alkan-2-ones, n-alkanoic acids), total organic carbon (TOC), total nitrogen (TN), Ca/Mg and ash content in Las Conchas mire, a 3.2 m deep bryophyte-dominated mire in Northern Spain covering 8000 cal yr BP. Bog conditions developed in the bottom 20 cm of the profile, and good preservation of organic matter (OM) was inferred from n-alkanoic acid distribution, with the exception of the uppermost 20 cm (last ca. 200 yr). Microbial synthesis of long chain saturated fatty acids from primary OM likely produced a dominance of short chain n-alkanoic acids with a bimodal distribution, as well as the lack of correspondence between the n-alkane and n-alkanoic acid profiles in the upper 20 cm. This was accompanied by an increase in ash content, a decrease in TOC and variation in n-alkane ratios, thereby suggesting significant changes in the mire, namely drainage and transformation to a meadow, in the last ca. 200 yr. The distribution of n-alkan-2-ones indicated an increase in bacterial source from the bottom of the record to 94 cm, whereas their distribution in the upper part could be attributed mainly to plant input and/or the microbial oxidation of n-alkanes. The different n-alkane proxies showed variations, which we interpreted in terms of changes in vegetation (Sphagnum vs. non-Sphagnum dominated phases) during the last 8000 cal yr BP. C23 was the most abundant homolog throughout most of the record, thereby suggesting dominant humid conditions alternating with short drier phases. However, such humid conditions were not linked to paleoclimatic variation but rather to geomorphological characteristics: Las Conchas mire, at the base of the Cuera Range, receives continuous runoff—even during drier periods—which is not necessarily accompanied by additional mineral input to peat, producing the development of Sphagnum moss typical of waterlogged ecotopes and damp habitats. Thus, although geochemical proxies indicated an ombrotrophic regime in the mire, geomorphological characteristics may make a considerable contribution to environmental conditions.

Rhodnius neglectus Lent, 1954 {Rn}e Psammolestes tertius Lent & Jurberg, 1965 {Pt} são triatomíneos que ocorrem em ninhos de aves, principalmente da família Furnariidae. O ciclo biológico dessas espécies é conhecido em condições de laboratório, sendo poucos estudos em ecótopos silvestres. Para analisar a infestação e estrutura de populações de Rn e Pt em ninhos de aves presentes na palmeira Mauritia flexuosa Linnaeus, em duas estações climáticas do Brasil Central, foram amostradas 41 palmeiras com evidências de nidificação dePhacellodomus ruber Vieillot, 1817 (22 na estação chuvosa e 19 na estação seca) em quatro áreas do Distrito Federal. Os insetos foram capturados usando-se coleta manual na copa da palmeira, identificados morfologicamente, separados por sexo e estádio ninfal. Fezes e glândulas salivares de Rn foram examinadas para verificar infecção por Trypanosoma cruzi Chagas, 1909 e/ou T. rangeli Tejera, 1920. Trinta e cinco palmeiras com ninhos de P. ruber estavam infestadas por Rn (85%) e 22 por Pt (53%). 442 indivíduos foram coletados na estação seca (200 Rn e 242 Pt) e 267 na estação chuvosa (136 Rn e 131 Pt). O único fator relacionado significativamente com a densidade de triatomíneos nas palmeiras foi a área. A estrutura etária das populações mostrou: a) maior abundância de adultos nas populações de Pt, b) maior abundância de machos em ambas as espécies e c) presença de fêmeas ovipondo em ambas as estações. Nenhum dos 177 triatomíneos examinados estava infectado por T. cruzi ou T. rangeli. A estrutura etária das populações de Rn e Pt não diferiu significativamente entre as estações amostradas, indicando ausência de marcada sazonalidade para essas espécies. _______________________________________________________________________________________ ABSTRACT
