As rações comerciais devem ser um alimento balanceado que supra todas as exigências nutricionais nas diferentes fases da vida do animal. Sua formulação deve conter ingredientes de qualidade, em proporções adequadas. É de fundamental importância o conhecimento do valor nutricional das rações, para assegurar que o cão esteja ingerindo diariamente quantidades corretas dos nutrientes. O desbalanço de elementos essenciais e a presença de elementos tóxicos podem causar desequilíbrios nutricionais, doenças e, até mesmo, consequências fatais aos cães. O cobre é um dos vários elementos de importância a ser estudado quanto aos defeitos metabólicos nos cães. O acúmulo de cobre no fígado pode ocasionar lesões progressivas nas organelas dos hepatócitos, resultando em hepatite crônica e cirrose. Diante disso, este trabalho teve como objetivos (I) quantificação dos elementos químicos com função nutricional e com potencial tóxico presentes nas rações para cães adultos e filhotes, (II) avaliação da composição centesimal das rações amostradas, (III) avaliação da variação dos elementos químicos entre as amostras de ração de um mesmo lote de produção (IV), estudo da representatividade de pequenas porções-teste, (V) avaliação da bioacessibilidade de cobre em rações, com experimento in vitro. Os elementos químicos Al, As, Br, Ca, Cl, Co, Cr, Cs, Cu, Fe, I, K, La, Mg, Mn, Na, P, Rb, Sb, Sc, Se, Ti, U e Zn foram determinados pela análise por ativação neutrônica instrumental (INAA). A composição centesimal foi avaliada de acordo com os métodos recomendados da AOAC. A homogeneidade de distribuição dos elementos químicos nas rações foi avaliada pela análise de amostras grandes (LS-NAA). A bioacessibilidade de cobre nas rações foi estimada por meio da simulação da digestão gastrointestinal in vitro. Foi possível determinar por INAA todos os nutrientes minerais, isto é, Ca, P, K, Na, Cl, Mg, Fe, Cu, Mn, Zn, I e Se, com limites estabelecidos pela Association of American Feed Control Officials. Foram notadas em algumas rações altas concentrações de Al, Sb e U, elementos com grande potencial tóxico. Cerca de 16 % das amostras de ração apresentaram, pelo menos, um parâmetro não conforme quanto à composição centesimal. Os resultados obtidos por LS-NAA e NAA convencional mostraram variação na composição entre os sacos de ração para Br, Ca, Na e Zn, com boa concordância entre ambos os métodos. O emprego da LS-NAA combinada com NAA convencional permitiu observar que pequenas porções-teste (350 mg) de ração são representativas comparadas com aquelas de 1 kg para Br, Ca, K, Na e Zn. Em todas as rações para cães, 50 % do cobre presente estava sob a forma bioacessível

The effects of refrigeration, freezing and substitution of milk fat by inulin and whey protein concentrate (WPC) on Lactobacillus acidophilus La-5 viability and resistance to gastric and enteric simulated conditions in synbiotic guava mousses effects were investigated. Refrigerated mousses supplemented with WPC presented the best probiotic viability. ranging from 7.77 to 6.24 log cfu/g during 28 days of storage. The highest probiotic populations, above 7.45 log cfu/g, were observed for all frozen mousses during 112 days of storage. Decreased L acidophilus survival during the in vitro gastrointestinal simulation was observed both for refrigerated and frozen mousses. Nonetheless, for the refrigerated mousses, the addition of inulin enhanced the probiotic survival during the in vitro assays in the first week of storage. L acidophilus survival in simulated gastrointestinal fluids was also improved through freezing. The frozen storage may be used to provide increased shelf-life for synbiotic guava mousses. Even though the protective effect of inulin and WPC on the probiotic microorganism tested was shown to be more specific for the refrigerated products, the partial replacement of milk fat by these ingredients may also help, as it improves the nutritional value of mousses in both storage conditions. This study evaluated the influence of gastrointestinal environmental factors (pH, digestive enzymes, food components, medicaments) on the survival of Lactobacillus casei Shirota and Lactobacillus casei LC01, using a semi-dynamic in vitro model that simulates the transit of microorganisms through the human GIT. The strains were first exposed to different simulated gastric juices for different periods of time (0, 30, 60 and 120 min), and then to simulated intestinal fluids for zero, 120, 180 and 240 min, in a step-wise format. The number of viable cells was determined after each step. The influence of food residues (skim milk) in the fluids and resistance to medicaments commonly used for varied therapeutic purposes (analgesics, antiarrhythmics, antibiotics, antihistaminics, proton pump inhibitors, etc.) were also evaluated. Results indicated that survival of both cultures was pH and time dependent, and digestive enzymes had little influence. Milk components presented a protective effect, and medicaments, especially anti-inflammatory drugs, influenced markedly the viability of the probiotic cultures, indicating that the beneficial effects of the two probiotic cultures to health are dependent of environmental factors encountered in the human gastrointestinal tract.

Probiotic properties of Lactobacillus amylovorus DSM 16698 were previously demonstrated in piglets. Here, its potential as a human probiotic was studied in vitro, using the TIM-1 system, which is fully validated to simulate the human upper gastrointestinal tract. To evaluate the effect of the food matrix composition on the survival of L amylovorus DSM 16698 in TIM-1, the microorganism was inoculated alone or with prebiotic galactooligosaccharides (GOS), partially skimmed milk (PSM) and/or commercial probiotic Bifidobacterium animalis subsp. lactis Bb-12 (Bb-12). Samples were collected from TIM-1 for six hours, at one-hour intervals and L amylovorus populations were enumerated on MRS agar plates with confirmation of identity of selected isolates by randomly amplified polymorphic DNA (RAPD) fingerprinting. The cumulative survival for L amylovorus alone (control) was 30% at the end of the experiment (t = 6 h). Co-administration of L amylovorus with GOS. PSM and/or Bb-12 increased its survival in comparison with the control significantly from the 4th hour after ingestion onwards (P<0.05). Furthermore, by the use of High Performance Anion Exchange Chromatography, both L amylovorus and Bb-12 were observed to promptly degrade GOS compounds in samples collected from TIM-1, as assessed at t = 2 h. Hence, food matrix composition interfered with survival and growth of L. amylovorus during passage through TIM-1, providing leads towards optimization of probiotic properties in vivo. PNU-87407 and PrNU-88509, beta-ketoamide anthelmintics that are structurally related to each other and to the salicylanilide anthelmintic closantel, exhibit different anthelmintic spectra and apparent toxicity in mammals, The basis for this differential pharmacology was examined in experiments that measured motility and adenosine triphosphate (ATP) levels in larval and adult stages of the gastrointestinal nematode, Haemonchus contortus, and in a vertebrate liver cell line and mitochondria, PNU-87407 and PNU-88509 both exhibited functional cross-resistance with closantel in larval migration assays using closantel-resistant and -sensitive isolates of H, contortus. Each compound reduced motility and,ATP levels in cultured adult H. contortus in a concentration- and time-dependent manner: however, motility was reduced more rapidly by PNU-88509, and ATP levels were reduced by lower concentrations of closantel than the beta-ketoamides. Tension recordings from segments of adult H, contortus showed that PNU-88509 induces spastic paralysis, while PNU-87407 and closantel induce flaccid paralysis of the somatic musculature. Marked differences in the actions of these compounds were also observed in the mammalian preparations. In Chang liver cells, ATP levels were reduced after 3 h exposures to greater than or equal to 0.25 mu M PNU-87407 1 mu M closantel or 10 mu M PNU-88509, Reductions in ATP caused by PNU-88509 were completely reversible, while the effects of closantel and PNU-87407; were irreversible. PNU-87407, closantel and PNU-88509 uncoupled oxidative phosphorylation in isolated rat liver mitochondria, inhibiting the respiratory control index (with glutamate or succinate as substrate) by 50% at concentrations of 0.14, 0.9 and 7.6 mu M respectively.

An in vitro colon extended physiologically based extraction test (CEPBET) which incorporates human gastrointestinal tract (GIT) parameters (including pH and chemistry, solid-to-fluid ratio, mixing and emptying rates) was applied for the first time to study the bioaccessibility of brominated flame retardants (BFRs) from the 3 main GIT compartments (stomach, small intestine and colon) following ingestion of indoor dust. Results revealed the bioaccessibility of γ-HBCD (72%) was less than that for α- and β-isomers (92% and 80% respectively) which may be attributed to the lower aqueous solubility of the γ-isomer (2 μg L−1) compared to the α- and β-isomers (45 and 15 μg L−1 respectively). No significant change in the enantiomeric fractions of HBCDs was observed in any of the studied samples. However, this does not completely exclude the possibility of in vivo enantioselective absorption of HBCDs, as the GIT cell lining and bacterial flora – which may act enantioselectively – are not included in the current CE-PBET model. While TBBP-A was almost completely (94%) bioaccessible, BDE-209 was the least (14%) bioaccessible of the studied BFRs. Bioaccessibility of tri-hepta BDEs ranged from 32–58%. No decrease in the bioaccessibility with increasing level of bromination was observed in the studied PBDEs.

Scope Epidemiological and clinical studies have demonstrated that the consumption of red haem-rich meat may contribute to the risk of colorectal cancer. Two hypotheses have been put forward to explain this causal relationship, i.e. N-nitroso compound (NOC) formation and lipid peroxidation (LPO). Methods and Results In this study, the NOC-derived DNA adduct O6-carboxymethylguanine (O6-CMG) and the LPO product malondialdehyde (MDA) were measured in individual in vitro gastrointestinal digestions of meat types varying in haem content (beef, pork, chicken). While MDA formation peaked during the in vitro small intestinal digestion, alkylation and concomitant DNA adduct formation was observed in seven (out of 15) individual colonic digestions using separate faecal inocula. From those, two haem-rich meat digestions demonstrated a significantly higher O6-CMG formation (p < 0.05). MDA concentrations proved to be positively correlated (p < 0.0004) with haem content of digested meat. The addition of myoglobin, a haem-containing protein, to the digestive simulation showed a dose–response association with O6-CMG (p = 0.004) and MDA (p = 0.008) formation. Conclusion The results suggest the haem-iron involvement for both the LPO and NOC pathway during meat digestion. Moreover, results unambiguously demonstrate that DNA adduct formation is very prone to inter-individual variation, suggesting a person-dependent susceptibility to colorectal cancer development following haem-rich meat consumption.

Foi avaliada a digestibilidade in vitro/gás da matéria seca (MS) de quatro volumosos, exclusivos e suas combinações, na proporção de 50% na MS. Foram incubados 100 mg de amostra por tratamento, em triplicatas, em 48 h de digestão. As digestibilidades da MS e a produção de gás foram: cana-de-açúcar = 60,6%, 17,3 mL; cana-de-açúcar + silagem de milho = 63,9%, 19,6 mL; cana-de-açúcar + capim-elefante com 60 dias = 60,5%, 16,9 mL; cana-de-açúcar + capim-elefante com 180 dias = 48,6%, 14,1 mL; silagem de milho = 66,3%; silagem de milho + capim-elefante com 60 dias = 62,1%, 16,7 mL; silagem de milho + capim-elefante com 180 dias = 52,7%, 15,8 mL; capim-elefante com 60 dias = 61,5%, 16,9 mL; capim-elefante com 180 dias = 34,6%, 11,7 mL. Constatou-se que a combinação de volumosos, em alguns casos, pode diminuir a digestibilidade da MS dos volumosos de melhor qualidade.

The aim of this study was to develop multiparticulate therapeutic systems of alginate (AL) and chitosan (CS) containing triamcinolone (TC) to colonic drug delivery. Multiparticulate systems of AL-CS, prepared by a complex coacervation/ionotropic gelation method, were characterized for morphological and size aspects, swelling degree, encapsulation content and efficiency, in vitro release profile in different environments simulating the gastrointestinal tract (GIT) and in vivo gastrointestinal transit. The systems showed suitable morphological characteristics with particle diameters of approximately 1.6 mm. In simulated gastric environment, at pH 1.2, the capsules presented low degree of swelling and in vitro release of drug. A higher swelling degree was observed in simulated enteric environment, pH 7.5, followed by erosion. Practically all the drug was released after 6 h of in vitro assay. The in vivo analysis of gastrointestinal transit, carried out in rats, showed that the systems passed practically intact through the stomach and did not show the same profile of swelling observed in the in vitro tests. It was possible to verify the presence of capsules in the colonic region of GIT. The results indicate that AL-CS multiparticulate systems can be used as an adjuvant for the preparation of therapeutic systems to colonic delivery of drugs. (C) 2010 Elsevier Ltd. O objetivo deste trabalho foi testar o sistema de monitoramento computadorizado da produção de gás in vitro. Essa técnica facilita estudos da degradação das frações solúveis e insolúveis das forragens, quantificadas pela produção de gás (CO2 e CH4) oriunda do metabolismo microbiano e medida por sensor de pressão. Diversas quantidades de amostras (50 a 110 mg de feno de alfafa) e outros alimentos foram testadas. Também, a influência da quantidade de líquido ruminal (2,0 ou 3,0 mL), com ou sem barras magnéticas agitadoras nos frascos incubatórios, na digestibilidade de 100 mg de feno de alfafa, foi estudada. A quantidade de 100 mg de amostra, 2,0 mL de líquido ruminal e sem barra magnética agitadora, proporcionou os menores coeficientes de variação na produção de gás. em conclusão, 2,0 mL de líquido ruminal, sem barra e com 100 mg de amostra, apresentaram maior precisão na curva de digestão.
