DNA is the first SINE isolated from zebrafish (Danio rerio) exhibiting all the hallmarks of these tRNA-derived elements. DANA is unique in its clearly defined substructure of distinct cassettes. In contrast to generic SINE elements, DANA appears to have been assembled by insertions of short sequences into a progenitor, tRNA-derived element. Once associated with each other, these subunits were amplified as a new transposable element with such a remarkable success that DANA-related sequences comprise approximately 10% of the modern zebrafish genome. At least some of the sequences comprised by the full-length element were capable of movement, forming a new group of mobile, composite transposons, one of which caused an insertional mutation in the zebrafish no tail gene. Being present only in the genus Danio, and estimated to be as old as the genus itself, DANA may have played a role in Danio speciation by massive amplification and genome-wide dispersion. There are extensive DNA polymorphisms between zebrafish populations and strains detected by PCR amplification using primers specific to DANA, suggesting that the DANA element will be useful as a molecular tool for genetic and phylogenetic analyses.

Cyanobacteria ("blue-green algae") are known to produce a diverse repertoire of biologically active secondary metabolites. When associated with so-called "harmful algal blooms", particularly in freshwater systems, a number of these metabolites have been associated—as "toxins", or commonly "cyanotoxins"—with human and animal health concerns. In addition to the known water-soluble toxins from these genera (i.e. microcystins, cylindrospermopsin, and saxitoxins), our studies have shown that there are metabolites within the lipophilic extracts of these strains that inhibit vertebrate development in zebrafish embryos. Following these studies, the zebrafish embryo model was implemented in the bioassay-guided purification of four isolates of cyanobacterial harmful algal blooms, namely Aphanizomenon, two isolates of Cylindrospermopsis, and Microcystis, in order to identify and chemically characterize the bioactive lipophilic metabolites in these isolates. ^ We have recently isolated a group of polymethoxy-1-alkenes (PMAs), as potential toxins, based on the bioactivity observed in the zebrafish embryos. Although PMAs have been previously isolated from diverse cyanobacteria, they have not previously been associated with relevant toxicity. These compounds seem to be widespread across the different genera of cyanobacteria, and, according to our studies, suggested to be derived from the polyketide biosynthetic pathway which is a common synthetic route for cyanobacterial and other algal toxins. Thus, it can be argued that these metabolites are perhaps important contributors to the toxicity of cyanobacterial blooms. In addition to the PMAs, a set of bioactive glycosidic carotenoids were also isolated because of their inhibition of zebrafish embryonic development. These pigmented organic molecules are found in many photosynthetic organisms, including cyanobacteria, and they have been largely associated with the prevention of photooxidative damage. This is the first indication of these compounds as toxic metabolites and the hypothesized mode of action is via their biotransformation to retinoids, some of which are known to be teratogenic. Additional fractions within all four isolates have been shown to contain other uncharacterized lipophilic toxic metabolites. This apparent repertoire of lipophilic compounds may contribute to the toxicity of these cyanobacterial harmful algal blooms, which were previously attributed primarily to the presence of the known water-soluble toxins.^

The industrial effluents are one of the main sources of water pollution. For an appropriate characterization and control of their discharges, the most efficient strategy is the integrated use of chemical, physical and ecotoxicological analyses. The aims of this study were to asses the efficiency of the treatment plant of a textile industry performing acute toxicity tests and physical-chemical analyses of the effluents before and after the treatment, besides evaluate the toxicity of the effluents of the Treatment System of Liquids Effluents (Sistema de Tratamento de Efluentes Líquidos - SITEL) of Distrito Industrial de Natal (DIN) and some of their physical-chemical variables. The species used in the ecotoxicological tests was the fish Danio rerio. The results showed that the treatment plant reduced significantly (around 50%) the toxicity of the raw textile effluent in only three of the seven tests but, in general, it promoted the reduction of the physical-chemical parameters analyzed. The toxicity and the physical-chemical factors of the effluents of SITEL of DIN varied among the tests and show the importance of monitoring their discharges in the Potengi river, one of the most important rivers of the Rio Grande do Norte state

The objectives of this research were to investigate the agrotoxic most used in the Gramorezinho region in the green belt of Natal, and to evaluate the acute toxicity of these, based on the LC50-48h values estimated in tests for Danio rerio, internationally used as test organism. The acute toxicity tests were performed under laboratory conditions, according to standardized methods (ABNT/NBR/15088/04) for this species. The LC50-48h estimated to Tamaron BR was 352.89 mg.L-1, which characterizes that as practically non-toxic, according to toxicological classes cited by Zucker. For Decis 25EC, the LC50-48h estimated was 0.0004156 mg.L-1 (4.156 X 10-4 mg.L-1), which classifies it as highly toxic to this species

Methomyl (Lannate®) is an insecticide from the carbamate group, frequently used in pest control in various types of crops. This compound works inhibiting the activity of the enzyme acetylcholinesterase. The use of physicochemical and ecotoxicological analysis is the most efficient strategy for the correct characterization and control of residues of metomil. The main objectives of this study were to evaluate the acute toxicity of methomyl in 96 hours of exposure and, through a sublethal assay of 5 hours, to assess its effect on the activity of acetylcholinesterase present in brain and squeletic dorsal muscle of the Danio rerio fish. The results showed that the LC50-96 found to D. rerio was 3.4 mg/L and it was found through the average of four definitive tests. In vitro assays were used to test the inhibitory action of methomyl directly over soluble AChE, extracted from the squeletic dorsal muscle, with maximum inhibition of 68.57% to the insecticide concentrations of 0.2 mg/L. In sublethal tests with D. rerio, inhibitory effect of methomyl was found over the soluble form of AChE in the squeletic dorsal muscle, both in one and five hours of fish exposure to the insecticide. In both period, the average values of inhibition were around 61%. In the same condition, no significant inhibitory effect of methomyl soluble and membrane AChE of the D. rerio was observed in the 0.42, 0.85, 1.70 and 2.50 mg/L concentrations and in both times of fish exposure

O desenvolvimento de organismos transgênicos para o hormônio do crescimento (GH) tem sido considerado uma importante alternativa para o aumento nas taxas de crescimento animal. Entretanto, os efeitos do excesso do GH não se limitam aos processos do crescimento. Sistemas fisiológicos como o sistema imunológico, já demonstraram ser prejudicados pelo desbalanço dos níveis do GH. Tendo em vista a importância da geração de organismos transgênicos para o GH no âmbito da aquicultura, esforços se fazem necessários na elaboração de estratégias com o intuito de reduzir ou compensar os efeitos adicionais do excesso de GH. Nos hipotetizamos que a geração de peixes duplo transgênicos os quais superexpressem GH e GHR apenas no músculo esquelético, poderia ser uma possível alternativa para compensar os efeitos prejudiciais ocasionados pelo excesso de GH sobre os sistemas fisiológicos mantendo, ao mesmo tempo, as altas taxas de crescimento. Afim de testar esta hipótese, nós avaliamos a morfometria de órgãos imunes, como o timo, o rim cefálico e o baço; os conteúdos de células T CD3+ e CD4+ no timo e no rim anterior e a expressão de genes relacionados à imunidade. Contrariando as expectativas iniciais, os resultados revelaram que a superexpressão do GHR no músculo esquelético não é capaz de diminuir os efeitos danosos causados pelo GH no tamanho do timo e rim anterior e no conteúdo de células T CD3+ e CD4+ nestes órgãos. Inesperadamente, zebrafish transgênicos somente para o GHR revelaram prejuízos nos aspectos imunes similares aos observados aos transgênicos para GH. De forma geral, estes resultados indicam que a dupla transgenia para o GH/GHR não é capaz de atenuar os efeitos negativos causados pelo excess de GH sobre o sistema imunológico de zebrafish transgênicos. Além disso, a transgenia para genes componentes do eixo somatotrófico pode ainda reforçar os danos as funções imunes em transgênicos, não recuperando os danos causados pelo excesso de GH.

Agroquímicos são amplamente utilizados na atividade agrícola com o objetivo de aumentar a produção e melhorar a qualidade dos alimentos, no entanto podem vir a gerar danos ao meio ambiente e a organismos não-alvo. Dentre esses pesticidas encontra-se o herbicida glifosato, o qual vem sendo mais utilizado mundialmente. Seu mecanismo de ação se dá através da inibição da enzima 5-enolpiruvilshikimato-3- fosfatosintase, intermediária da síntese de aminoácidos aromáticos essenciais em plantas. Pouco se sabe sobre os efeitos da substância glifosato em animais, pois os estudos realizados visam principalmente os efeitos da formulação comercial, a qual contém surfactantes e outras substâncias inertes. Tendo isso em vista, esse estudo avaliou o efeito do glifosato no teleósteo Danio rerio considerando parâmetros de estresse oxidativo, atividade e expressão da acetilcolinesterase e parâmetros reprodutivos. Foram feitas exposições a 5 mg/L e 10 mg/L de glifosato, mais um grupo controle por 24 e 96 horas, somente com peixes machos. Para análise bioquímica foram retirados cérebro, brânquias e músculo; para análise molecular, cérebro e músculo; e para análise na qualidade espermática dos peixes, os testículos. Quanto às análises bioquímicas houve um aumento na capacidade antioxidante contra radicais peroxil nas brânquias na concentração de 5 mg/L após 24 horas de exposição; uma redução na peroxidação lipídica no cérebro na maior concentração (10 mg/L) após 24h e um aumento da mesma em músculo, também em 10 mg/L, após 96 horas. Não foi observada alteração na geração de espécies reativas de oxigênio decorrente da exposição ao glifosato, assim como na atividade da enzima acetilcolinesterase; já na expressão gênica desta enzima houve uma diminuição no cérebro após 24 horas de exposição e um aumento no cérebro e no músculo após 96 horas. Quanto à qualidade espermática dos peixes, houve uma redução na motilidade e período de motilidade dos espermatozóides nas concentrações de 5 mg/L e 10 mg/L em ambos tempos de exposição; na concentração de 10 mg/L ainda houve uma redução da funcionalidade mitocondrial, integridade de membrana do espermatozóide e integridade de DNA após 24 e 96 horas. Sendo assim, o glifosato se mostrou capaz de alterar o balanço oxidativo dos tecidos do peixe Danio rerio bem como alterar significativamente a expressão gênica da enzima acetilcolinesterase. Além disso, nossos resultados demonstram que o glifosato pode interferir na reprodução deste animal, através da redução de sua qualidade espermática.

As Microcistinas são heptapeptídios cíclicos produzidos como metabólitos secundários por diferentes espécies de cianobactérias, sendo relevantes pelo seu potencial hepatotóxico. Peixes apresentam estratégias bioquímicas para detoxificar contaminantes ambientais, incluindo a ativação de enzimas de fase II de biotransformação, que incluem as isoformas de glutationa S-transferase (GST). As GST catalizam a conjugação de glutationa reduzida (GSH) com uma variedade de xenobióticos, incluindo as microcistinas. O presente estudo avaliou os níveis transcricionais de quinze isoformas de GST a fim de identificar isoformas possivelmente envolvidas na detoxificação de contaminantes ambientais como a microcistina-LR (MC-LR) em Danio rerio. A técnica de PCR em tempo real (RT-qPCR) foi utilizada para avaliação dos níveis transcricionais, permitindo análise das GST em diferentes órgãos, abundância e a ativação/repressão das isoformas de GST pela exposição à MC-LR. Foram avaliados os possíveis efeitos causados em brânquia e fígado após exposição por 24 hs às concentrações de 5 µg.L-1 e 50 µg.L-1 de MC-LR. Baseado nos scores de estabilidade para oito genes normalizadores, foram selecionados glicose-6-fosfato desidrogenase (g6pdh), β-actina1 e beta-2-microglobulina (b2m); b2m, alfa-tubulina 1 (tuba) e β- actin1; e tuba, b2m e g6pdh, para normalização dos níveis trancricionais de GST para distribuição órgão-específica, abundância e efeito da MC-LR em brânquia e fígado, respectivamente. A avaliação transcricional da distribuição órgão-específica revelou níveis significativos de gstal e gstk1.1 no fígado; gstp1 e gstp2 em brânquia; mgst3a, gstr1, gstm2, gstm33, gstp1, gstp2 e gstk1.1 no intestino; gstm2, gstm3 e gstal no olho e gstt1a e gsta2.1 no cérebro. Considerando os níveis de transcritos para um dado órgão, gstk1.1, gstal, gstp1 e gstt2 foram mais abundantes nos órgãos de detoxificação, tais como o fígado, brânquias e intestino, enquanto gstt1a e gsta2.1 foram mais abundantes no rim. Em brânquia, gsta2.1 e gstt1b foram reprimidas por 5 µg.L-1 de MC-LR e mgst1.1 foi reprimida em 50 µg.L-1 de MC-LR. No fígado, as isoformas gst2.2 e gstp2 foram reprimidas em ambas as concentrações, gstal foi reprimida em 5 µg.L-1, e gstt1a e gstk1.1 foram reprimidas em 50 µg.L-1 de MC-LR. As isoformas gstal, gstr1, gstp1, mgst3a, gstm1, gstm2 e gstm3 não foram alteradas pela exposição a MC-LR. Os resultados obtidos fornecem informações para a escolha de isoformas específicas de GST possivelmente envolvidas na detoxificação/toxicidade de MC-LR, a serem melhores caracterizadas ao nível protéico e também contribui para a escolha de genes normalizadores a serem utilizados em outros estudos da mesma natureza

Ornamental fish may be severely affected by a stressful environment. Stressors impair the immune response, reproduction and growth rate; thus, the identification of possible stressors will aid to improve the overall quality of ornamental fish. The aim of this study was to determine whole-body cortisol of adult zebrafish, Danio rerio, following visual or direct contact with a predator species. Zebrafish were distributed in three groups: the first group, which consisted of zebrafish reared completely isolated of the predator, was considered the negative control; the second group, in which the predator, Parachromis managuensis was stocked together with zebrafish, was considered the positive control; the third group consisted of zebrafish stocked in a glass aquarium, with direct visual contact with the predator. The mean whole-body cortisol concentration in zebrafish from the negative control was 6.78 +/- 1.12 ng g(-1), a concentration statistically lower than that found in zebrafish having visual contact with the predator (9.26 +/- 0.88 ng g(-1)) which, in turn, was statistically lower than the mean whole-body cortisol of the positive control group (12.35 +/- 1.59 ng g(-1)). The higher whole-body cortisol concentration found in fish from the positive control can be attributed to the detection, by the zebrafish, of relevant risk situations that may involve a combination of chemical, olfactory and visual cues. One of the functions of elevated cortisol is to mobilize energy from body resources to cope with stress. The elevation of whole-body cortisol in fish subjected to visual contact with the predator involves only the visual cue in the recognition of predation risk. We hypothesized that the zebrafish could recognize predator characteristics in P managuensis, such as length, shape, color and behavior. Nonetheless, the elevation of whole-body cortisol in zebrafish suggested that the visual contact of the predator may elicit a stress response in prey fish. This assertion has a strong practical application concerning the species distribution in ornamental fish markets in which prey species should not be allowed to see predator species. Minimizing visual contact between prey and predator fish may improve the quality, viability and welfare of small fish in ornamental fish markets. (c) 2007 Elsevier B.V. All rights reserved.

Acid-sensing ion channels (ASICs) are epithelial Na+ channels gated by external H+. Recently, it has been demonstrated that ASICs play a role in Na+ uptake in freshwater rainbow trout. The current paper investigated the potential involvement of ASICs in Na+ transport in another freshwater fish species, the zebrafish (Danio rerio). Using molecular and histological techniques we found that asic genes and the ASIC4.2 protein are expressed in the gill of adult zebrafish. Immunohistochemistry revealed that mitochondrion-rich cells positive for ASIC4.2 do not co-localize with Na+/K+-ATPase (NKA)-rich cells, but co-localize with cells expressing vacuolar-type H+-ATPase (VHA). Furthermore, pharmacological inhibitors of ASIC and Na+/H+-exchanger (NHEs) significantly reduced uptake of Na+ in adult zebrafish exposed to low Na+ media, but did not cause the same response in individuals exposed to ultra-low Na+ water. Our results suggest that in adult zebrafish ASICs play a role in branchial Na+ uptake in media with low Na+ concentrations and that mechanisms used for Na+ uptake by zebrafish may depend on the Na+ concentration in the acclimation media.
