The basis for the neuroprotectant effect of D-mannitol in reducing the sensory neurological disturbances seen in ciguatera poisoning, is unclear. Pacific ciguatoxin-1 (P-CTX-1), at a concentration 10 nM, caused a statistically significant swelling of rat sensory dorsal root ganglia (DRG) neurons that was reversed by hyperosmolar 50 MM D-mannitol. However, using electron paramagnetic resonance (EPR) spectroscopy, it was found that P-CTX-1 failed to generate hydroxyl free radicals at concentrations of toxin that caused profound effects on neuronal excitability. Whole-cell patch-clamp recordings from DRG neurons revealed that both hyper- and iso-osmolar 50 MM D-mannitol prevented the membrane depolarisation and repetitive firing of action potentials induced by P-CTX-1. In addition, both hyper- and iso-osmolar 50 MM D-mannitol prevented the hyperpolarising shift in steady-state inactivation and the rise in leakage current through tetrodotoxin (TTX)-sensitive Na-v channels, as well as the increased rate of recovery from inactivation of TTX-resistant Nav channels induced by P-CTX-1. D-Mannitol also reduced, but did not prevent, the inhibition of peak TTX-sensitive and TTX-resistant I-Na amplitude by P-CTX-1. Additional experiments using hyper- and isoosmolar D-sorbitol, hyperosmolar sucrose and the free radical scavenging agents Trolox (R) and L-ascorbic acid showed that these agents, unlike D-mannitol, failed to prevent the effects of P-CTX-1 on spike electrogenesis and Na-v channel gating. These selective actions of D-mannitol indicate that it does not act purely as an osmotic agent to reduce swelling of nerves, but involves a more complex action dependent on the Nav channel subtype, possibly to alter or reduce toxin association. 天花粉蛋白(Trichosanthin,TCS)是一种由247 个氨基酸组成的Ⅰ型核糖 体失活蛋白(Ribosome Inactivating Protein,RIP),从葫芦科植物栝楼 (Trichosanthes Kirilowii)球根中提纯获得。它具有广谱的生物学和药学活性, 包括抗肿瘤、免疫抑制、中期引产以及抗病毒活性。上世纪八十年代末, McGrath 等发现TCS 可以抑制HIV-1 在急性感染的T 淋巴细胞和慢性感染的 巨噬细胞中的复制,从而引起了研究者们极大的兴趣。但迄今为止,其抗HIV-1 的作用机制仍不清楚。 我们实验室对TCS 的免疫毒理作用和抗HIV-1 作用进行了多年的研究, 前期工作显示TCS 对HIV-1 的直接作用并不明显,但对于HIV-1 感染细胞却 具有很强的毒性作用。提示TCS 可能通过作用于宿主细胞来发挥其抗HIV-1 活性。在此基础上,我们从细胞方面着手,对TCS 选择性杀伤HIV-1 感染细 胞的作用及机制进行了探讨。首先,通过MTT 法检测发现,相同条件下,TCS 对于H9/HIV-1IIIB 细胞的毒性远远大于其对正常H9 细胞的毒性。其次,流式 细胞仪检测亚二倍体小峰和琼脂糖凝胶电泳检测片断化DNA 的实验证实了 TCS 对H9/HIV-1IIIB 的细胞杀伤作用是通过诱导细胞凋亡实现的。流式细胞仪 的结果显示TCS 以剂量依赖的方式诱导较多的H9/HIV-1IIIB 细胞凋亡,25μ g/mlTCS 作用24h 时,有8.4%的H9 细胞凋亡,而H9/HIV-1IIIB 细胞的凋亡率 则达到24.5%;随着作用浓度的降低,这种差异也在缩小。阳性对照D-Sorbitol 对两种细胞的凋亡率没有明显差别,约为25%。琼脂糖凝胶电泳的结果进一 步证实了这种推测,相同条件下,TCS 诱导H9/HIV-1IIIB 细胞出现更多的DNA 片断化。 TCS 可以选择性的诱导H9/HIV-1IIIB 细胞凋亡,为了进行更深入的机制研 究,我们建立了另外一株HIV-1 慢性感染细胞系,HIV-1 慢性感染的Jurkat 细胞系(Jurkat/HIV-1ⅢB)来验证TCS的作用。发现相同条件下,TCS可以诱 导同等程度的Jurkat/HIV-1ⅢB和Jurkat 细胞凋亡,25μg/mlTCS作用24h 时, 分别有21.08%的Jurkat 细胞和27.27%的Jurkat/HIV-1IIIB 细胞凋亡。以上的结 果说明HIV-1 感染H9 细胞后增强了感染细胞对TCS 的敏感性,而HIV-1 感 染Jurkat 细胞后并不影响其对TCS 的敏感性。根据细胞凋亡途径中是否依赖 线粒体的参与可以将细胞分成TypeⅠ和TypeⅡ两种类型,H9 细胞采取的是 线粒体非依赖性的TypeⅠ型凋亡途径,而Jurkat 细胞则采取线粒体依赖性的 TypeⅡ型凋亡途径。由于Jurkat 细胞对TCS 诱导的凋亡更加敏感,我们推测 HIV-1 感染H9 细胞后,诱导了细胞凋亡途径由TypeⅠ向TypeⅡ型转变。为 此,我们采用流式细胞仪检测了TCS 对凋亡细胞内的线粒体膜电位及 caspase-8 蛋白酶活性的影响,结果显示,H9/HIV-1IIIB、Jurkat 和Jurkat/HIV-1IIIB 细胞对TCS 诱导的凋亡具有相同程度的敏感性,并且伴随着细胞线粒体膜电 位的耗散和caspase-8 蛋白酶的活化;而相同情况下,TCS 对H9 细胞的影响 则很微弱。由此,进一步证实了我们的推测,即HIV-1 感染H9 细胞后,通过 改变细胞内的某些信号,使H9/HIV-1ⅢB细胞的性状更加倾向于Type Ⅱ型细 胞,从而增强其对于TCS 的敏感性.

Heating 2,5-di-O-methanesulfonyl-1,4:3,6-dianhydro-D-sorbitol (1) in a range of solvents led to the formation of a gel state at low concentrations. 1 was found to gel aromatics, alcohols and water. The structure of 1 in the solid state was solved by single crystal X-ray crystallography; no strong hydrogen bonds or associated solvents were found in the crystal. Electron micrographs revealed the morphology of the gels to be predominantly rod-like. The ethanol alcogel was used to template silica by sol-gel transcription.

The synthesis of Y(0.9)Er(0.1)Al(3)(BO(3))(4) crystalline powders and vitreous thin films were studied. Precursor solutions were obtained using a modified polymeric precursor method using D-sorbitol as complexant agent. The chemical reactions were described. Y(0.)9Er(0.1)Al(3)(BO(3))(4) composition presents good thermal stability with regard to crystallization. The Y(0.9)Er(0.1)Al(3)(BO(3))(4) crystallized phase can be obtained at 1,150 degrees C, in agreement with other authors. Crack- and porosity-free films were obtained with very small grain size and low RMS roughness. The films thickness revealed to be linearly dependent on precursor solution viscosity, being the value of 25 mPa s useful to prepare high-quality amorphous multi-layers (up to similar to 800 nm) at 740 degrees C during 2 h onto silica substrates by spin coating with a gyrset technology.

Lokale sekundäre Prozesse in glasbildenden Materialien sind wegen ihrer Wechselwirkung mit der primären Relaxation von besonderem Interesse. Für D-Sorbitol wurde die beta-Relaxation mit drei verschiedenen dielektrischen Meßtechniken untersucht und die Ergebnisse miteinander verglichen. Im Gegensatz zu konventionellen Messungen der dielektrischen Retardation, detektiert eine echte dielektrische Relaxationsmessung den Zerfall des elektrischen Feldes unter der Bedingung einer konstanten dielektrischen Verschiebung. Eine weitere dielektrische Relaxationsmethode ist die Solvatationsdynamik. Sie detektiert die dielektrische Relaxation in der direkten Umgebung eines Farbstoffmoleküls. Die Übereinstimmung der lokal ermittelten und der makroskopisch gemittelten Ergebnisse für den Glaszustand weisen darauf hin, daß die Sekundärrelaxation in D-Sorbitol eine räumlich homogene Eigenschaft ist. Im Gegensatz zu beta-Prozessen anderer Materialien, zeigt nur der abgeschreckte Glaszustand von ortho-Terphenyl einen sekundären Prozeß. Es wurde beobachtet, daß die beta-Amplitude beim Ausheilen langsam abnimmt und im Gleichgewichtszustand der Flüssigkeit oberhalb der Glasübergangstemperatur völlig verschwindet. Viele glasbildende Materialien, wie z. B. Salol zeigen keinen dielektrischen beta-Prozeß. Im Gegensatz zu Messungen mit Standardkühlraten, zeigt eine stark abgeschreckte Salol-Probe eine symmetrische dielektrische Sekundärrelaxation. Diese neuartige Eigenschaft von Salol verschwindet irreversibel, wenn sich die Temperatur der Glasübergangstemperatur nähert.

With the aim of analysing the relative importance of sugar supply and nitrogen nutrition for the regulation of sulphate assimilation, the regulation of adenosine 5′‐phosphosulphate reductase (APR), a key enzyme of sulphate reduction in plants, was studied. Glucose feeding experiments with Arabidopsis thaliana cultivated with and without a nitrogen source were performed. After a 38 h dark period, APR mRNA, protein, and enzymatic activity levels decreased dramatically in roots. The addition of 0.5% (w/v) glucose to the culture medium resulted in an increase of APR levels in roots (mRNA, protein and activity), comparable to those of plants kept under normal light conditions. Treatment of roots with D‐sorbitol or D‐mannitol did not increase APR activity, indicating that osmotic stress was not involved in APR regulation. The addition of O‐acetyl‐L‐serine (OAS) also quickly and transiently increased APR levels (mRNA, protein, and activity). Feeding plants with a combination of glucose and OAS resulted in a more than additive induction of APR activity. Contrary to nitrate reductase, APR was also increased by glucose in N‐deficient plants, indicating that this effect was independent of nitrate assimilation. [35S]‐sulphate feeding experiments showed that the addition of glucose to dark‐treated roots resulted in an increased incorporation of [35S] into thiols and proteins, which corresponded to the increased levels of APR activity. Under N‐deficient conditions, glucose also increased thiol labelling, but did not increase the incorporation of label into proteins. These results demonstrate that (i) exogenously supplied glucose can replace the function of photoassimilates in roots; (ii) APR is subject to co‐ordinated metabolic control by carbon metabolism; (iii) positive sugar signalling overrides negative signalling from nitrate assimilation in APR regulation. Furthermore, signals originating from nitrogen and carbon metabolism regulate APR synergistically.

The optimal formulation for the preparation of amaranth flour films plasticized with glycerol and sorbitol was obtained by a multi-response analysis. The optimization aimed to achieve films with higher resistance to break, moderate elongation and lower solubility in water. The influence of plasticizer concentration (Cg, glycerol or Cs, sorbitol) and process temperature (Tp) on the mechanical properties and solubility of the amaranth flour films was initially studied by response surface methodology (RSM). The optimized conditions obtained were Cg 20.02 g glycerol/100 g flour and Tp 75 degrees C, and Cs 29.6 g sorbitol/100 g flour and Tp 75 degrees C. Characterization of the films prepared with these formulations revealed that the optimization methodology employed in this work was satisfactory. Sorbitol was the most suitable plasticizer. It furnished amaranth flour films that were more resistant to break and less permeable to oxygen, due to its greater miscibility with the biopolymers present in the flour and its lower affinity for water. This study assesses the Vitamin D status of 126 healthy free-living adults aged 18–87 years, in southeast Queensland, Australia (27°S) at the end of the 2006 winter. Participants provided blood samples for analysis of 25(OH)D (the measure of an individual’s Vitamin D status), PTH, Calcium, Phosphate, and Albumin, completed a questionnaire on sun-protective/sun-exposure behaviours, and were assessed for phenotypic characteristics such as skin/hair/eye colour and BMI. We found that 10.2% of the participants had serum 25(OH)D levels below 25 nmol/l (considered deficient) and a further 32.3% had levels between 25 nmol/l and 50 nmol/l (considered insufficient). Our results show that low levels of 25(OH)D can occur in a substantial proportion of the population at the end of winter, even in a sunny climate. 25(OH)D levels were higher amongst those who spent more time in the sun and lower among obese participants (BMI > 30) than those who were not obese (BMI < 30). 25(OH)D levels were also lower in participants who had black hair, dark/olive skin, or brown eyes, when compared with participants who had brown or fair hair, fair skin, or blue/green eyes. No associations were found between 25(OH)D status and age, gender, smoking status, or the use of sunscreen.

Vitamin D deficiency and insufficiency are now seen as a contemporary health problem in Australia with possible widespread health effects not limited to bone health1. Despite this, the Vitamin D status (measured as serum 25-hydroxyvitamin D (25(OH)D)) of ambulatory adults has been overlooked in this country. Serum 25(OH)D status is especially important among this group as studies have shown a link between Vitamin D and fall risk in older adults2. Limited data also exists on the contributions of sun exposure via ultraviolet radiation and dietary intake to serum 25(OH)D status in this population. The aims of this project were to assess the serum 25(OH)D status of a group of older ambulatory adults in South East Queensland, to assess the association between their serum 25(OH)D status and functional measures as possible indicators of fall risk, obtain data on the sources of Vitamin D in this population and assess whether this intake was related to serum 25(OH)D status and describe sun protection and exposure behaviors in this group and investigate whether a relationship existed between these and serum 25(OH)D status. The collection of this data assists in addressing key gaps identified in the literature with regard to this population group and their Vitamin D status in Australia. A representative convenience sample of participants (N=47) over 55 years of age was recruited for this cross-sectional, exploratory study which was undertaken in December 2007 in south-east Queensland (Brisbane and Sunshine coast). Participants were required to complete a sun exposure questionnaire in addition to a Calcium and Vitamin D food frequency questionnaire. Timed up and go and handgrip dynamometry tests were used to examine functional capacity. Serum 25(OH)D status and blood measures of Calcium, Phosphorus and Albumin were determined through blood tests. The Mean and Median serum 25-Hydroxyvitamin D (25(OH)D) for all participants in this study was 85.8nmol/L (Standard Deviation 29.7nmol/L) and 81.0nmol/L (Range 22-158nmol/L), respectively. Analysis at the bivariate level revealed a statistically significant relationship between serum 25(OH)D status and location, with participants living on the Sunshine Coast having a mean serum 25(OH)D status 21.3nmol/L higher than participants living in Brisbane (p=0.014). While at the descriptive level there was an apparent trend towards higher outdoor exposure and increasing levels of serum 25(OH)D, no statistically significant associations between the sun measures of outdoor exposure, sun protection behaviors and phenotypic characteristics and serum 25(OH)D status were observed. Intake of both Calcium and Vitamin D was low in this sample with sixty-eight (68%) of participants not meeting the Estimated Average Requirements (EAR) for Calcium (Median=771.0mg; Range=218.0-2616.0mg), while eighty-seven (87%) did not meet the Adequate Intake for Vitamin D (Median=4.46ug; Range=0.13-30.0ug). This raises the question of how realistic meeting the new Adequate Intakes for Vitamin D is, when there is such a low level of Vitamin D fortification in this country. However, participants meeting the Adequate Intake (AI) for Vitamin D were observed to have a significantly higher serum 25(OH)D status compared to those not meeting the AI for Vitamin D (p=0.036), showing that meeting the AI for Vitamin D may play a significant role in determining Vitamin D status in this population. By stratifying our data by categories of outdoor exposure time, a trend was observed between increased importance of Vitamin D dietary intake as a possible determinant of serum 25(OH)D status in participants with lower outdoor exposures. While a trend towards higher Timed Up and Go scores in participants with higher 25(OH) D status was seen, this was only significant for females (p=0.014). Handgrip strength showed statistically significant association with serum 25(OH)D status. The high serum 25(OH)D status in our sample almost certainly explains the limited relationship between functional measures and serum 25(OH)D. However, the observation of an association between slower Time Up and Go speeds, and lower serum 25(OH)D levels, even with a small sample size, is significant as slower Timed Up and Go speeds have been associated with increased fall risk in older adults3. Multivariable regression analysis revealed Location as the only significant determinant of serum 25(OH)D status at p=0.014, with trends (p=>0.1) for higher serum 25(OH)D being shown for participants that met the AI for Vitamin D and rated themselves as having a higher health status. The results of this exploratory study show that 93.6% of participants had adequate 25(OH)D status-possibly due to measurement being taken in the summer season and the convenience nature of the sample. However, many participants do not meet their dietary Calcium and Vitamin D requirements, which may indicate inadequate intake of these nutrients in older Australians and a higher risk of osteoporosis. The relationship between serum 25(OH)D and functional measures in this population also requires further study, especially in older adults displaying Vitamin D insufficiency or deficiency.
