ABSTRACT The analysis of Fernando Higueras's creative thought as an architect begins with a study of his main creative themes and their chronological evolution, divided into nine directions of research. Here we see the importance of his UNBUILT work as the germ of the creative themes of his thought, part of which we study, analyse and reconstruct graphically in 3-D. Fernando Higueras drew on his earlier architectural works and ideas and reached an ultimate constructive and formal maturity that was infused with beauty through composition and the amazingly natural combination of these keynote themes. In order to gain a greater understanding of the key themes of the architect's creative thought it is also worth noting the element of artists' intuition, their way of seeing and observing the world as a source of inspiration. The architect's "way of seeing" reveals his sensitivity to the popular and traditional human habitat. Thanks to Antonio Miró, in the period between 1963 and 1970 Fernando Higueras was able to provide a real framework for a large part of his constructive ideas. The object of study of the present work is to determine the architect's “invariables” as a means of understanding his creative thought. The aims of the present study are to explore his thoughts in greater depth, to examine his work from the chronological-evolutionary point of view for an insight into the relationships and the creative connections between them, to observe and apprehend the evolution of his main creative themes and compositional keynotes, to analyse graphically part of his unbuilt work in order to improve our understanding of these underlying themes through their reconstruction using the latest graphic digital resources. The 3-D reconstruction of part of his unbuilt work highlights the magnitude of his creative thought. The methodology used to collect all the necessary information includes an examination of his writings and articles in journals and newspapers, criticisms, observations, plans, projects, building specifications, period photographs, models, the opinions of his contemporaries, family and friends, and visits to some of his built work. Nine directions of research were then established for each main theme of his creative thought. Each direction includes an analysis from two different points of view: the first, theoretical or chronological-evolutionary aspect, based on a selection of some of his built and unbuilt works; and a second graphic aspect based on the study and reconstruction of part of his unbuilt work. The result is a survey of the evolution of his work with a view to establishing the connections and their interrelationships, through to the ultimate maturity of his creative themes in both his built and unbuilt work. Fernando Higueras is on a constant quest for excellence, a perpetual desire to do things a little better, until a point where he gains mastery over his own recurrent themes, in the fullness of maturity and of his formal and constructive expression. The creation of three-dimensional models in 13 projects of his unbuilt work offers a new look at the capacity of his creative thought, in addition to the analysis of the evidence in some of these projects of the constructive and compositional germs of many of his ideas and themes. Finally, this work has contributed to the understanding of Fernando Higueras in a broader dimension, from the freshness of his unbuilt work through to the capacity of his creative thought, the evolution of his main themes, the study of his final unbuilt works, his sensitivity to the arts as a whole and to everything beautiful. The 360 plates of illustrations provided form the basis of the study and analysis of his extensive unbuilt work.

This paper addresses the theme of retrotif applied to buildings that belong to the modernist architectural production of historical interest located in the urban area of Natal . The overall objective is to identify and harmonize procedures for retrofit and architectural heritage preservation using elements of constructive analysis of expression and Benefit Cost Ratio ( BCR ) parameters established by the National Electric Energy Agency - ANEEL . The hypothesis put forward is that by stimulating the projetual year analysis , retrofit interventions , it is possible to obtain better results with projects RCB addressing the issues of preservation of architectural heritage . For both flow analysis of process solutions and proposals for action of elements and systems that seek to improve the energy performance of the building , restoring or preserving the architectural elements were developed . The proposed interventions undergo performance through computer simulations of systems such as DesignBuilder, Solar and Sun Tool. The energy results were converted to the analysis of RCB parameter and compared to the constructive expression of the project because the prenatal and intervention. From the results , a plot was constructed which results in a comparison between the RCB and the constructive expression of the simulated interventions

Abstract Background Transcript enumeration methods such as SAGE, MPSS, and sequencing-by-synthesis EST "digital northern", are important high-throughput techniques for digital gene expression measurement. As other counting or voting processes, these measurements constitute compositional data exhibiting properties particular to the simplex space where the summation of the components is constrained. These properties are not present on regular Euclidean spaces, on which hybridization-based microarray data is often modeled. Therefore, pattern recognition methods commonly used for microarray data analysis may be non-informative for the data generated by transcript enumeration techniques since they ignore certain fundamental properties of this space. Results Here we present a software tool, Simcluster, designed to perform clustering analysis for data on the simplex space. We present Simcluster as a stand-alone command-line C package and as a user-friendly on-line tool. Both versions are available at: http://xerad.systemsbiology.net/simcluster. Conclusion Simcluster is designed in accordance with a well-established mathematical framework for compositional data analysis, which provides principled procedures for dealing with the simplex space, and is thus applicable in a number of contexts, including enumeration-based gene expression data.

The tissue kallikreins are serine proteases encoded by highly conserved multigene families. The rodent kallikrein (KLK) families are particularly large, consisting of 13 26 genes clustered in one chromosomal locus. It has been recently recognised that the human KLK gene family is of a similar size (15 genes) with the identification of another 12 related genes (KLK4-KLK15) within and adjacent to the original human KLK locus (KLK1-3) on chromosome 19q13.4. The structural organisation and size of these new genes is similar to that of other KLK genes except for additional exons encoding 5 or 3 untranslated regions. Moreover, many of these genes have multiple mRNA transcripts, a trait not observed with rodent genes. Unlike all other kallikreins, the KLK4-KLK15 encoded proteases are less related (25–44%) and do not contain a conventional kallikrein loop. Clusters of genes exhibit high prostatic (KLK2-4, KLK15) or pancreatic (KLK6-13) expression, suggesting evolutionary conservation of elements conferring tissue specificity. These genes are also expressed, to varying degrees, in a wider range of tissues suggesting a functional involvement of these newer human kallikrein proteases in a diverse range of physiological processes.

Banana bunchy top is regarded as the most important viral disease of banana, causing significant yield losses worldwide. The disease is caused by Banana bunchy top virus (BBTV), which is a circular ssDNA virus belonging to the genus Babuvirus in the family Nanoviridae. There are currently few effective control strategies for this and other ssDNA viruses. “In Plant Activation” (InPAct) is a novel technology being developed at QUT for ssDNA virus-activated suicide gene expression. The technology exploits the rolling circle replication mechanism of ssDNA viruses and is based on a unique “split” gene design such that suicide gene expression is only activated in the presence of the viral Rep. This PhD project aimed to develop a BBTV-based InPAct system as a suicide gene strategy to control BBTV. The BBTV-based InPAct vector design requires a BBTV intergenic region (IR) to be embedded within an intron in the gene expression cassette. To ensure that the BBTV IR would not interfere with intron splicing, a TEST vector was initially generated that contained the entire BBTV IR embedded within an intron in a β-glucuronidase (GUS) expression vector. Transient GUS assays in banana embryogenic cell suspensions indicated that cryptic intron splice sites were present within the IR. Transcript analysis revealed two cryptic intron splice sites in the Domain III sequence of the CR-M within the IR. Removal of the CR-M from the TEST vector resulted in an enhancement of GUS expression suggesting that the cryptic intron splice sites had been removed. An InPAct GUS vector was subsequently generated that contained the modified BBTV IR, with the CR-M (minus Domain III) repositioned within the InPAct cassette. Using transient histochemical and fluorometric GUS assays in banana embryogenic cells, the InPAct GUS vector was shown to be activated in the presence of the BBTV Rep. However, the presence of both BBTV Rep and Clink was shown to have a deleterious effect on GUS expression suggesting that these proteins were cytotoxic at the levels expressed. Analysis of replication of the InPAct vectors by Southern hybridisation revealed low levels of InPAct cassette-based episomal DNA released from the vector through the nicking/ligation activity of BBTV Rep. However, Rep-mediated episomal replicons, indicative of rolling circle replication of the released circularised cassettes, were not observed. The inability of the InPAct cassette to be replicated was further investigated. To examine whether the absence of Domain III of the CR-M was responsible, a suite of modified BBTV-based InPAct GUS vectors was constructed that contained the CR-M with the inclusion of Domain III, the CR-M with the inclusion of Domain III and additional upstream IR sequence, or no CR-M. Analysis of replication by Southern hybridisation revealed that neither the presence of Domain III, nor the entire CR-M, had an effect on replication levels. Since the InPAct cassette was significantly larger than the native BBTV genomic components (approximately 1 kb), the effect of InPAct cassette size on replication was also investigated. A suite of size variant BBTV-based vectors was constructed that increased the size of a replication competent cassette to 1.1 kbp through to 2.1 kbp.. Analysis of replication by Southern hybridisation revealed that an increase in vector size above approximately 1.5 - 1.7 kbp resulted in a decrease in replication. Following the demonstration of Rep-mediated release, circularisation and expression from the InPAct GUS vector, an InPAct vector was generated in which the uidA reporter gene was replaced with the ribonuclease-encoding suicide gene, barnase. Initially, a TEST vector was generated to assess the cytotoxicity of Barnase on banana cells. Although transient assays revealed a Barnase-induced cytotoxic effect in banana cells, the expression levels were sub-optimal. An InPAct BARNASE vector was generated and tested for BBTV Rep-activated Barnase expression using transient assays in banana embryogenic cells. High levels of background expression from the InPAct BARNASE vector made it difficult to accurately assess Rep-activated Barnase expression. Analysis of replication by Southern hybridisation revealed low levels of InPAct cassette-based episomal DNA released from the vector but no Rep-mediated episomal replicons indicative of rolling circle replication of the released circularised cassettes were again observed. Despite the inability of the InPAct vectors to replicate to enable high level gene expression, the InPAct BARNASE vector was assessed in planta for BBTV Rep-mediated activation of Barnase expression. Eleven lines of transgenic InPAct BARNASE banana plants were generated by Agrobacterium-mediated transformation and were challenged with viruliferous Pentalonia nigronervosa. At least one clonal plant in each line developed bunchy top symptoms and infection was confirmed by PCR. No localised lesions were observed on any plants, nor was there any localised GUS expression in the one InPAct GUS line challenged with viruliferous aphids. The results presented in this thesis are the first study towards the development of a BBTV-based InPAct system as a Rep-activatable suicide gene expression system to control BBTV. Although further optimisation of the vectors is necessary, the preliminary results suggest that this approach has the potential to be an effective control strategy for BBTV. The use of iterons within the InPAct vectors that are recognised by Reps from different ssDNA plant viruses may provide a broad-spectrum resistance strategy against multiple ssDNA plant viruses. Further, this technology holds great promise as a platform technology for the molecular farming of high-value proteins in vitro or in vivo through expression of the ssDNA virus Rep protein.
