Três experimentos de competição entre genótipos de feijão-fradinho (Vigna unguiculata (L.) Walp.) foram instalados em Coimbra (17 de novembro de 1993), Ponte Nova (19 de novembro de 1993) e Viçosa (1º de dezembro de 1994), municípios da Zona da Mata de Minas Gerais, com o objetivo de avaliar-lhes o comportamento na primavera-verão, nessa região. Foram testados 18 genótipos em 1993 e 10 em 1994. Foi utilizado o delineamento de blocos ao acaso, com três (1993) e quatro (1994) repetições. Os rendimentos variaram de 106 a 2.562 kg/ha. Em 1993, quando houve veranico nas três primeiras semanas de fevereiro, os rendimentos máximos foram de 1.587 (Coimbra) e 1.438 kg/ha (Ponte Nova), obtidos com a linhagem precoce e de porte intermediário IT 85F-2687. Em 1994, quando choveu regularmente até o mês de março, o rendimento máximo (2.562 kg/ha) foi alcançado com a linhagem de ciclo tardio e porte semi-ereto IT 85F-899. A mancha-café (Colletotrichum falcatum f. truncata) foi a única doença observada e as linhagens IT 85F-2687, IT 85D-34284, IT 83S-818, IT 83S-899 e IT 86D-716 foram consideradas resistentes. A primeira colheita foi realizada entre 69 e 79 dias após a emergência, e o período de colheita variou de 16 a 62 dias, dependendo do genótipo e do regime de chuvas.

O objetivo deste trabalho foi avaliar o efeito da fertirrigação nitrogenada sobre os danos causados pelo complexo broca-podridão à produtividade e à qualidade da cana-de-açúcar. O experimento foi realizado em campo, em delineamento de blocos ao acaso, com quatro repetições e cinco doses de N (0, 50, 100, 150 e 200 kg ha-1), aplicadas por irrigação por gotejamento. Quantificaram-se as seguintes variáveis: número de orifícios causados por Diatraea saccharalis (Lepidoptera: Crambidae); número de internódios infectados com a podridão-vermelha; percentagem de incidência de podridão-vermelha; produtividade de colmos; e teor de açúcar. Em laboratório, foram avaliados a atratividade e o consumo de fragmentos de colmos por lagartas de quarto instar, em testes com ou sem chance de escolha. A irrigação nitrogenada aumentou a incidência de D. saccharalis e de podridão-vermelha. O número de orifícios causados pelas lagartas correlacionou-se à ocorrência de podridão-vermelha. A percentagem de açúcar diminuiu com o aumento do número de internódios danificados pela podridão-vermelha. As doses de N não afetaram a atratividade dos fragmentos de colmos de cana-de-açúcar; no entanto, maiores doses de N fertilizante aumentaram o consumo destes fragmentos por lagartas de D. saccharalis. Apesar dos danos à qualidade, a adubação nitrogenada aumentou expressivamente a produtividade de colmos e de açúcar.

Background: Sugarcane is one of the most important crops in Brazil, mainly because of its use in biofuel production. Recent studies have sought to determine the role of sugarcane endophytic microbial diversity in microorganism-plant interactions, and their biotechnological potential. Epicoccum nigrum is an important sugarcane endophytic fungus that has been associated with the biological control of phytopathogens, and the production of secondary metabolites. In spite of several studies carried out to define the better conditions to use E. nigrum in different crops, little is known about the establishment of an endophytic interaction, and its potential effects on plant physiology. Methodology/Principal Findings: We report an approach based on inoculation followed by re-isolation, molecular monitoring, microscopic analysis, plant growth responses to fungal colonization, and antimicrobial activity tests to study the basic aspects of the E. nigrum endophytic interaction with sugarcane, and the effects of colonization on plant physiology. The results indicate that E. nigrum was capable of increasing the root system biomass and producing compounds that inhibit the in vitro growth of sugarcane pathogens Fusarium verticillioides, Colletotrichum falcatum, Ceratocystis paradoxa, and Xanthomomas albilineans. In addition, E. nigrum preferentially colonizes the sugarcane surface and, occasionally, the endophytic environment. Conclusions/Significance: Our work demonstrates that E. nigrum has great potential for sugarcane crop application because it is capable of increasing the root system biomass and controlling pathogens. The study of the basic aspects of the interaction of E. nigrum with sugarcane demonstrated the facultative endophytism of E. nigrum and its preference for the phylloplane environment, which should be considered in future studies of biocontrol using this species. In addition, this work contributes to the knowledge of the interaction of this ubiquitous endophyte with the host plant, and also to a better use of microbial endophytes in agriculture.

Os fungos do gênero Metarhizium são entomopatogênicos e ainda apresentam relações endofíticas e podem viver saprofiticamente no solo. A associação desses microrganismos com insetos é bem conhecida, mas as interações diretas com as plantas ainda são incipientes. Objetivou-se com este estudo, determinar a capacidade de colonização endofítica em raízes de cana-de-açúcar (Saccharum spp.) de M. robertsii, M. anisopliae e três linhagens brasileiras recém-descobertas, bem como revelar o potencial destes fungos como antagonistas a dois fungos fitopatogênicos responsáveis pela podridão-vermelha (Fusarium moniliforme e Colletotrichum falcatum) e no controle de duas importantes pragas, a broca da cana-de-açúcar, Diatraea saccharalis e o nematóide-das-galhas, Meloidogyne javanica. Foram utilizados 24 isolados de Metarhizium spp. nos experimentos de promoção de crescimento de plantas após a inoculação em gemas de cana-de-açúcar em condições de casa-de-vegetação e campo. O efeito da inoculação do fungo em mudas de cana-de-açúcar foi investigado quanto a mortalidade e redução no desenvolvimento de D. saccharalis e redução nas populações de M. javanica. Experimentos de inibição in vitro em cultivo pareado foram conduzidos com os fungos F. moniliforme e C. falcatum e Trichoderma harzianum. Além disto, foram realizados testes qualitativos \"in vitro\" para avaliar a capacidade desses fungos em produzir β-1,3-Glucanase e Sideróforos. A inoculação de Metarhizium de todas as espécies testadas promoveu o crescimento de parte aérea, peso úmido e seco de raízes em relação ao controle (água). Enquanto o tratamento de gemas rotineiramente usado na usina onde se realizou a produção de mudas pré-brotadas com aplicação do fungicida Comet® + o fertilizante Biozyme TF resultou em pegamento de 32% e 59,6%, apenas com o uso de isolados de Metarhizium spp. estes valores foram de até 50% e 86,4%, nos dois experimentos em campo, respectivamente. As mortalidades observadas em lagartas de D. saccharalis que se alimentaram das plantas inoculadas com isolados de Metarhizium spp. atingiram valores de até 80%, sendo a mortalidade confirmada pela esporulação do fungo de até 55%. As lagartas sobreviventes apresentaram menor peso do que aquelas do controle não tratado. A inoculação de Metarhizium spp. em mudas de cana-de-açúcar conferiu proteção a M. javanica, resultando em uma densidade de massa de ovos por grama de raiz nas mudas inoculadas com o fungo até 59,8% menor no primeiro experimento e 64,1% menor no segundo em relação as mudas sem inoculação. Os ensaios de antagonismo mostraram que, todos os isolados de Metarhizium spp. apresentaram alguma forma de inibição tanto para F. moniliforme como para C. falcatum. Dos 24 isolados testados, 20,8% produziram a enzima hidrolítica, β-1,3-glucanase, o que pode estar associado a capacidade de inibição dos fungos fitopatogênicos. Constatou-se que 8 (33,3%) dos isolados produziram sideróforos e sugere que esses fungos apresentam mecanismos de disponibilização do ferro. O conhecimento gerado com este estudo poderá subsidiar novas estratégias de utilização de Metarhizium spp. em campo na produção de MPB de cana-de-açúcar.

The effect of methyl jasmonate treatment on gene expression in sugarcane roots signalling between roots and shoots was studied. A collection of 829 ESTs were obtained from sugarcane roots treated with the defence-regulator methyl jasmonate (MJ) treatment. A subset of 747 of these were combined with 4793 sugarcane ESTs obtained from stem tissues in a cDNA microarray and experiments undertaken to identify genes that were induced in roots 24-120 h following treatment with MJ. Two data analysis systems (t-statistic and tRMA) were used to analyse the microarray results and these methods identified a common set of 21 ESTs corresponding to transcripts significantly induced by MJ in roots and 23 that were reduced in expression following MJ treatment. The induction of six transcripts identified in the microarray analysis was tested and confirmed using northern blotting. Homologues of genes encoding lipoxygenase and PR-10 proteins were induced 824 It after MJ treatment while the other four selected transcripts were induced at later time points. Following treatment of roots with MJ, the lipoxygenase homologue, but not the PR-10 homologue, was induced in untreated stem and leaf tissues. The PR-10 homologue and a PR-1 homologue, but not the lipoxygenase homologue, were induced in untreated tissues after the application of SA to roots. Repeated foliar application of MJ had no apparent effects on plant growth and was demonstrated to increase lipoxygenase transcripts in roots, but did not increase transcript levels-of other genes tested. Introduction. This protocol aims at detecting and quantifying quiescent infections of Colletotrichum musae on bananas. The principle, key advantages, starting plant material, time required and expected results are presented. Materials and methods. The materials required and details of the three steps of the protocol (fruit sampling, fruit ripening and anthracnose lesion quantification) are described. Possible troubleshooting is discussed. Results. The protocol results in the quantification of anthracnose lesions on the fruits, which makes it possible to predict postharvest losses due to anthracnose (peel rot), and also to propose a better management of postharvest fungicide applications.

From a genomic enriched library, we developed 27 primer pairs from microsatellite flanking sequences for Colletotrichum acutatum, associated to postbloom fruit drop disease on citrus. Loci were characterized using 40 monosporic C. acutatum isolates. Nine primer pairs successfully amplified polymorphic microsatellite regions, with 3-6 alleles per locus, and mean heterozygosities ranging 0.093-0.590 across loci. The suitability of these primers was investigated in four Colletotrichum species as well. These microsatellite markers will be useful for genetic analyses and epidemiological studies of C. acutatum.

Endophytic microorganisms reside asymptomatically within plants and are a source of new bioactive products for use in medicine, agriculture, and industry. Colletotrichum (teleomorph Glomerella) is a fungus widely cited in the literature as a producer of antimicrobial substances. Identification at the species level, however, has been a problem in this type of study. Several authors have reported the presence of endophytic fungi from the medicinal plant Maytenus ilicifolia (espinheira-santa) in Brazil that has antimicrobial activity against various pathogens. Therefore, Colletotrichum strains were isolated from M. ilicifolia and identified based on morphology, RAPD markers, sequence data of the internal transcribed spacer regions (ITS-1 and ITS-2), the 5.8S gene, and species-specific PCR. The analyses suggested the presence of 2 species, Colletotrichum gloeosporioides and Colletotrichum boninense. Two morphological markers were characterized to allow C. gloeosporioides and C. boninense to be distinguished quickly and accurately. The molecular diagnosis of C. boninense was confirmed by using Coll and ITS4 primers. This species of Colletotrichum is reported for the first time in M. ilicifolia.

Citrus post-bloom fruit drop (caused by Colletotrichum acutatum) frequently occurs in the southwestern region of So Paulo State, Brazil. A survey of Colletotrichum isolates associated with symptoms of post-bloom fruit drop in So Paulo State showed C. gloeosporioides in addition to C. acutatum. The objectives of this study were to confirm the identification of C. gloeosporioides isolated from symptomatic citrus flowers, to test the pathogenicity of C. gloeosporioides isolates, to compare the development of disease caused by C. gloeosporioides and C. acutatum, and to determine the frequency of C. gloeosporioides in a sample of isolates obtained from symptomatic flowers in different regions of So Paulo State. Through the use of species-specific primers by PCR, 17.3% of 139 isolates were C. gloeosporioides, and the remaining 82.7% were C. acutatum. The pathogenicity tests, carried out in 3-year old potted plants of sweet oranges indicated that both species caused typical symptoms of the disease including blossom blight and persistent calyces. Incubation periods (3.5 and 3.9 days, respectively, for C. acutatum and C. gloeosporioides) and fruit sets (6.7 and 8.5%, respectively for C. acutatum and C. gloeosporioides) were similar for both species. The incidences of blossom blight and persistent calyces were higher on plants inoculated with C. acutatum than in those inoculated with C. gloeosporioides. Conidial germination was similar for both species under different temperatures and wetness periods. Under optimal conditions, appressorium formation and melanisation were higher for C. gloeosporioides than for C. acutatum. These results indicated that Colletotrichum gloeosporioides is a new causal agent of post-bloom fruit drop.

Anthracnose, caused by Colletotrichum gloeosporioides and C acutatum, is one of file main post-harvest diseases in guavas. This study aimed to determine the influence of environmental variables oil germination and appressorium formation of Colletotrichum gloeosporioides and C acutatum and infection of Kumagai guavas by these pathogens. The germination rate and the apressorium formation rate in vitro were determined under temperatures of 10, 15, 20, 25, 30, 35 and 40 degrees C, with wetting periods of 6, 12 and 24 hours, The infection of guavas was determined under temperatures of 15, 20, 25 and 30 degrees C and wetting period of 24 hours. There was no germination at 40 degrees C for either species. The germination and apressorium formation rate were rather high in the range of 15 to 30 degrees C for C. gloeosporioides, with a maximum at 25 degrees C. For the species C. acutatum, germination and apressorium formation rates were more sensitive to variations in temperature, with a maximum at 20 degrees C. The wetting periods tested somewhat influenced the germination of C. gloeosporioides, whereas in C acutatum the germination was significantly lower with 6 hours of wetting than 12 and 24 hours. The infection of guavas, for both fungal species, increased with the temperature, unlike conidium germination and apressorium formation. Incidences of 100% occurred with 30 degrees C, at 10 days after the inoculation.

Phytophthora root rot (Phytophthora medicaginis) and colletotrichum crown rot (Colletotrichum trifoli) are the 2 most serious pathogens of lucerne in eastern Australia. Work reported in this paper shows that in glasshouse tests of the 11 most commonly grown Australian lucerne cultivars, the proportion of individual plants with resistance to both pathogens ranges from 0 (Hunter River and Aurora) through to a maximum of 19.8% (Sequel HR). Within 9 of the cultivars, the proportion of individual plants resistant to the 2 pathogens was <7%. Since these 2 diseases are known to cause serious losses in eastern Australia, the results indicate further improvement in lucerne production can be obtained by increasing the proportion of individual plants in a cultivar resistant to both pathogens. This would be best achieved by identifying dominant sources of resistance and incorporating this into on-going lucerne breeding programs.

Genetic markers that distinguish fungal genotypes are important tools for genetic analysis of heterokaryosis and parasexual recombination in fungi. Random amplified polymorphic DNA (RAPD) markers that distinguish two races of biotype B of Colletotrichum gloeosporioides infecting the legume Stylosanthes guianensis were sought. Eighty-five arbitrary oligonucleotide primers were used to generate 895 RAPD bands but only two bands were found to be specifically amplified from DNA of the race 3 isolate. These two RAPD bands were used as DNA probes and hybridised only to DNA of the race 3 isolate. Both RAPD bands hybridised to a dispensable 1.2 Mb chromosome of the race 3 isolate. No other genotype-specific chromosomes or DNA sequences were identified in either the race 2 or race 3 isolates. The RAPD markers hybridised to a 2 Mb chromosome in all races of the genetically distinct biotype A pathogen which infects other species of Stylosanthes as well as S. guianensis. The experiments indicate that RAPD analysis is a potentially useful tool for obtaining genotype-and chromosome-specific DNA probes in closely related isolates of one biotype of this fungal pathogen.

The Australian-bred lucerne cultivars, Trifecta and Sequel, were found to possess useful levels of resistance to both Colletotrichum trifolii races 1 and 2. Race 2 has only been previously observed in the United States and surveys did not reveal its presence in Australia. Multilocus fingerprinting using random amplified polymorphic DNA (RAPDs) analysis revealed low diversity (<10% dissimilarity) within Australian C. trifolii collections, and between the Australian race 1 isolates and a US race 2 isolate. Studies on the inheritance of resistance to C. trifolii race 1 in individual clones from Trifecta and Sequel revealed the presence of 2 different genetic mechanisms. One inheritance was for resistance as a recessive trait, and the other indicated that resistance was dominant. The recessive system has never been previously reported, whereas in the US, 2 completely dominant and independent tetrasomic genes Anl and Ant have been reported to condition C. trifolii resistance. It was not possible to fit the observed segregations from our studies to a single-gene model. In contrast to US studies, clones of cv. Sequel exhibiting the recessive resistance reacted differently to spray and stem injection with C. trifolii inoculum, being resistant to the former and susceptible to the latter, providing additional evidence for the presence of a different genetic mechanism conditioning resistance to those previously reported in the US. As C. trifolii is one of the most serious diseases of lucerne worldwide, the future development of molecular markers closely linked to the dominant and recessive resistances identified in these studies, and the relationships between these resistances and Anl and Ans as determined by genetic mapping, appear to be useful areas of future study.

Two biotypes (A and B) of Colletotrichum gloeosporioides infect the tropical legumes Stylosanthes spp. in Australia. These biotypes are asexual and vegetatively incompatible. However, field isolates of biotype B carrying a supernumerary 2-Mb chromosome, thought to originate from biotype A, have been reported previously. We tested the hypothesis that the 2-Mb chromosome could be transferred from biotype A to biotype B under laboratory conditions. Selectable marker genes conferring resistance to hygromycin and phleomycin were introduced into isolates of biotypes A and B, respectively. A transformant of biotype A, with the hygromycin resistance gene integrated on the 2-Mb chromosome, was cocultivated with phleomycin-resistant transformants of biotype B. Double antibiotic-resistant colonies were obtained from conidia of these mixed cultures at a frequency of approximately 10(-7). Molecular analysis using RFLPs, RAPDs, and electrophoretic karyotypes showed that these colonies contained the 2-Mb chromosome in a biotype B genetic background. In contrast, no double antibiotic colonies developed from conidia obtained from mixed cultures of phleomycin-resistant transformants of biotype B with biotype A transformants carrying the hygromycin resistance gene integrated in chromosomes >2 Mb in size. The results demonstrated that the 2-Mb chromosome was selectively transferred from biotype A to biotype B. The horizontal transfer of specific chromosomes across vegetative incompatibility barriers may explain the origin of supernumerary chromosomes in fungi.

Glasshouse experiments determined effects of a moth, Helicoverpa armigera (Lepidoptera: Noctuidae), and the anthracnose pathogen, Colletotrichum gloeosporioides (Penz.) Penz. and Sacc., on each other when attacking the same host plant, Stylosanthes scabra (Vog.) (Leguminosae) cv. Fitzroy. The host was treated with both organisms in 2 ways of succession and at 2 different life stages each. Larvae of the moth preferred to feed on healthy plants rather than plants recently infected with C. gloeosporioides, and preferred such newly infected plants to severely diseased ones. Adult female moths laid more eggs on healthy and recently infected plants than on diseased plants, when given a choice of all 3 plant types. Severity of anthracnose disease was neither promoted nor retarded by damage to leaves caused by larvae of the moth.
