Using CD and 2D H-1 NMR spectroscopy, we have identified potential initiation sites for the folding of T4 lysozyme by examining the conformational preferences of peptide fragments corresponding to regions of secondary structure. CD spectropolarimetry showed most peptides were unstructured in water, but adopted partial helical conformations in TFE and SDS solution. This was also consistent with the H-1 NMR data which showed that the peptides were predominantly disordered in water, although in some cases, nascent or small populations of partially folded conformations could be detected. NOE patterns, coupling constants, and deviations from random coil Her chemical shift values complemented the CD data and confirmed that many of the peptides were helical in TFE and SDS micelles. In particular, the peptide corresponding to helix E in the native enzyme formed a well-defined helix in both TFE and SDS, indicating that helix E potentially forms an initiation site for T4 lysozyme folding. The data for the other peptides indicated that helices D, F, G, and H are dependent on tertiary interactions for their folding and/or stability. Overall, the results from this study, and those of our earlier studies, are in agreement with modeling and IID-deuterium exchange experiments, and support an hierarchical model of folding for T4 lysozyme.

Conantokin-G and conantokin-T are two paralytic polypeptide toxins originally isolated from the venom of the fish-hunting cone snails of the genus Conus. Conantokin-G and conantokin-T are the only naturally occurring peptidic compounds which possess N-methyl-D-aspartate receptor antagonist activity, produced by a selective non-competitive antagonism of polyamine responses, They are also structurally unusual in that they contain a disproportionately large number of acid labile post-translational gamma-carboxyglutamic acid (Gla) residues, Although no precise structural information has previously been published for these peptides, early spectroscopic measurements have indicated that both conantokin-G and conantokin-T form alpha-helical structures, although there is some debate whether the presence of calcium ions is required for these peptides to adopt this fold, We now report a detailed structural study of synthetic conantokin-G and conantokin-T in a range of solution conditions using CD and H-1 NMR spec troscopy. The three-dimensional structures of conantokin-T and conantokin-G were calculated from H-1 NMR-derived distance and dihedral restraints. Both conantokins were found to contain a mixture of alpha- and 3(10) helix, that give rise to curved and straight helical conformers. Conantokin-G requires the presence of divalent cations (Zn2+, Ca2+, Cu2+, Or Mg2+) to form a stable iv-helix, while conantokin-T adopts a stable alpha-helical structure in aqueous conditions, in the presence or absence of divalent cations (Zn2+, Ca2+, Cu2+, Or Mg2+).

The free H(2)xspa ligands [xspa = pspa, Clpspa, tspa or fspa where p = 3-(phenyl), Clp = 3-(2-chlorophenyl), t = 3-(2-thienyl), f = 3-(2-furyl) and spa = 2-sulfanylpropenoato], their Zn(II) complexes of formula [HQ](2)[Zn(xspa)(2)] (HQ=diisopropylammonium) and the Cd(II) equivalents were prepared and characterized by elemental analysis and by IR, Raman and NMR ((1)H, (13)C) spectroscopy. X-Ray studies of the crystal structures of [HQ](2)[Zn(pspa)(2)], [HQ](2)[Zn(Clpspa)2], [HQ](2)[Zn(tspa)(2)] and [HQ](2)[Zn(fspa)(2)] show that the zinc atom is coordinated to two O atoms and two S atoms of the ligands in a distorted tetrahedral ZnO(2)S(2) environment. In the structures of [HQ](2)[Cd(pspa)(2)] and [HQ](2)[Cd(Clpspa)(2)] the cadmium atom is coordinated to three S atoms and two carboxylato O atoms of the ligands in a distorted trigonal bipyramidal environment. The interchange of ligands between Zn( II) and Cd( II) was studied by (113)Cd NMR spectroscopy. The in vitro protective effect of H(2)xspa and their Zn( II) complexes against Cd toxicity was investigated using the human hepatocarcinoma HepG2 cell line and the pig renal proximal tubule LLC-PK1 cell line. The incorporation of Zn( II) was found to be relevant in the case of H(2)pspa, with an increase observed in the cell viability of the LCC-PK1 cells with respect to the value for the free ligand.

Characteriza of the inclusion complex ropivacaine: beta-cyclodextrin. Ropivacaine (RVC) is a widely used local anesthetic. The complexation of RVC with beta-cyclodextrin (beta-CD) is of great interest for the development of more efficient local anesthetic formulations. The present work focuses on the characterization of the RVC:beta-CD complex by nuclear magnetic resonance (NMR). The stoichiometry of the complex is 1:2 RVC:beta-CD. DOSY-NMR shows that the association constant is 55.5 M-1. Longitudinal relaxation time results show that RVC changes its mobility in the presence of beta-CD. This study is focused on the physicochemical characterization of inclusion complexes that are potentials options for pain treatment.

The solution structure of A beta(1-40)Met(O), the methionine-oxidized form of amyloid beta-peptide A beta(1-40), has been investigated by CD and NMR spectroscopy. Oxidation of Met35 may have implications in the aetiology of Alzheimer's disease. Circular dichroism experiments showed that whereas A beta(1-40) and A beta(1-40)Met(O) both adopt essentially random coil structures in water (pH 4) at micromolar concentrations, the former aggregates within several days while the latter is stable for at least 7 days under these conditions. This remarkable difference led us to determine the solution structure of A beta(1-40)Met(O) using H-1 NMR spectroscopy. In a water-SDS micelle medium needed to solubilize both peptides at the millimolar concentrations required to measure NMR spectra, chemical shift and NOE data for A beta(1-40)Met(O) strongly suggest the presence of a helical region between residues 16 and 24. This is supported by slow H-D exchange of amide protons in this region and by structure calculations using simulated annealing with the program XPLOR. The remainder of the structure is relatively disordered. Our previously reported NMR data for A beta(1-40) in the same solvent shows that helices are present over residues 15-24 (helix 1) and 28-36 (helix 2), Oxidation of Met35 thus causes a local and selective disruption of helix 2. In addition to this helix-coil rearrangement in aqueous micelles, the CD data show that oxidation inhibits a coil-to-beta-sheet transition in water. These significant structural rearrangements in the C-terminal region of A beta may be important clues to the chemistry and biology of A beta(1-40) and A beta(1-42).

The solution conformation of a peptide LYS(11-36), which corresponds to the beta-sheet region in T4 lysozyme, has been examined in aqueous solution, TFE, and SDS micelles by CD and H-1 NMR spectroscopy. Secondary structure predictions suggest some beta-sheet and turn character in aqueous solution but predict a helical conformation in a more hydrophobic environment. The predictions were supported by the CD and NMR studies which showed the peptide to be relatively unstructured in aqueous solution, although there was some evidence of a beta-turn conformer which was maintained in 200 mM SDS and, to a lesser extent, in 50% TFE. The peptide was significantly helical in the presence of either 50% TFE or 200 mM SDS. TFE and SDS titrations showed that the peptide could form helical, sheet, or extended structure depending on the TFE or SDS concentration. The studies indicate that peptide environment is the determining factor in secondary structure adopted by LYS(11-36).

O cultivo de trigo de sequeiro constitui ótima opção para o Estado de Minas Gerais em regiões com altitude superior a 800 m. Neste estudo objetivou-se estimar a divergência genética entre genótipos de trigo com o auxílio de métodos de análise multivariada, objetivando identificar potenciais genótipos para obtenção de combinações híbridas promissoras. Foi utilizado o delineamento experimental em blocos ao acaso, com quatro repetições. Avaliaram-se seis caracteres morfoagronômicos em 11 genótipos de trigo cultivados em condições de sequeiro no município de Patos de Minas-MG, nos anos de 2007 e 2008. Houve grande variabilidade fenotípica entre os genótipos de trigo avaliados em cultivo de sequeiro. Os genótipos IAC 289-L22 e IAC 289-L4 foram os mais similares, enquanto os CD 113 e EP 011106, os maisdivergentes, promissores nos cruzamentos para obtenção de materiais adaptados e com bons atributos fenotípicos em cultivo de sequeiro.

O objetivo deste trabalho foi identificar as melhores épocas de semeadura e avaliar a adaptabilidade e a estabilidade de cultivares de trigo, em duas regiões tritícolas do Paraná. Avaliou-se a produtividade de grãos de sete cultivares, em Guarapuava, e de nove, em Palotina, em quatro épocas de semeadura, em 2006, 2007 e 2008. Utilizou-se o delineamento experimental de blocos ao acaso, com quatro repetições em Guarapuava, e três em Palotina. Foram utilizadas a metodologia REML/BLUP e a dos efeitos principais dos genótipos e da interação genótipo x ambiente (GGE biplot) para a avaliação da adaptabilidade e da estabilidade das cultivares, e o métodoAMMI para a identificação das melhores épocas de semeadura. Semeaduras em julho, em Guarapuava, e em abril, em Palotina, maximizam a produtividade de grãos. As cultivares Safira, em Guarapuava, e CD 113, em Palotina, são estáveis, amplamente adaptadas e apresentam alta produtividade de grãos.

Complexation between acyclovir (ACV), an antiviral drug used for the treatment of herpes simplex virus infection, and beta-cyclodextrin (beta-CD) was studied in solution and in solid states. Complexation in solution was evaluated using solubility studies and nuclear magnetic resonance spectroscopy (¹H-NMR). In the solid state, X-ray diffraction, differential scanning calorimetry (DSC), thermal gravimetric analysis (TGA) and dissolution studies were used. Solubility studies suggested the existence of a 1:1 complex between ACV and beta-CD. ¹H-NMR spectroscopy studies showed that the complex formed occurs with a stoichiometry ratio of 1:1. Powder X-ray diffraction indicated that ACV exists in a semicrystalline state in the complexed form with beta-CD. DSC studies showed the existence of a complex of ACV with beta-CD. The TGA studies confirmed the DSC results of the complex. Solubility of ACV in solid complexes was studied by the dissolution method and it was found to be much more soluble than the uncomplexed drug.

This paper compares and contrasts, for the first time, one- and two-component gelation systems that are direct structural analogues and draws conclusions about the molecular recognition pathways that underpin fibrillar self-assembly. The new one-component systems comprise L-lysine-based dendritic headgroups covalently connected to an aliphatic diamine spacer chain via an amide bond, One-component gelators with different generations of headgroup (from first to third generation) and different length spacer chains are reported. The self-assembly of these dendrimers in toluene was elucidated using thermal measurements, circular dichroism (CD) and NMR spectroscopies, scanning electron microscopy (SEM), and small-angle X-ray scattering (SAXS). The observations are compared with previous results for the analogous two-component gelation system in which the dendritic headgroups are bound to the aliphatic spacer chain noncovalently via acid-amine interactions. The one-component system is inherently a more effective gelator, partly as a consequence of the additional covalent amide groups that provide a new hydrogen bonding molecular recognition pathway, whereas the two-component analogue relies solely on intermolecular hydrogen bond interactions between the chiral dendritic headgroups. Furthermore, because these amide groups are important in the assembly process for the one-component system, the chiral information preset in the dendritic headgroups is not always transcribed into the nanoscale assembly, whereas for the two-component system, fiber formation is always accompanied by chiral ordering because the molecular recognition pathway is completely dependent on hydrogen bond interactions between well-organized chiral dendritic headgroups.

The total synthesis of 8,9-licarinediols was selectively carried out from licarin A, previously obtained by oxidative coupling of (E)-isoeugenol. The corresponding enantiomerically pure (+)- and(-)-licarin A ester derivatives were subjected to Sharpless oxidation to yield the asymmetric C-8, C-9 dihydroxylation products, whose absolute configurations were established by means of the CD and NMR spectroscopic analyses of their Mosher ester derivatives. Im Rahmen dieser Arbeit wurde die Bindung von Koffein und verwandten Oxopurinen in C¬3-symmetrischen Rezeptoren auf der Basis von Triphenylenketalen untersucht. Dabei stand vor allem die Evaluierung für eine spätere Anwendung im Vordergrund. Für die Anwendung als Chemosensor wurden mehrere optische Verfahren getestet. Die Verwendung von UV/Vis-Spektroskopie gelingt nur unter Einsatz eines elektronenarmen Konkurrenzgastes, welcher durch das stärker bindende Koffein unter Entfärbung verdrängt wird. Obwohl dieser Effekt sogar mit bloßem Auge zu erkennen ist und somit eine einfache Untersuchung ermöglichen würde, machen die besondere Reaktivität des Konkurrenzgastes und dessen geringe Affinität zum Rezeptor eine weitere Anwendung als Chemosensor für Koffein unwahrscheinlich. Den entscheidenden Durchbruch lieferte der Wechsel auf Fluoreszenzspektroskopie. Die Bindung von Gästen lässt sich mit dieser Methode direkt beobachten und für quantitative Studien nutzen. Die Signalzunahme bei Zugabe von Koffein liegt bei maximal 30%. Durch Verwendung eines vom Koffein abgeleiteten Konkurrenzgastes können weitere Verbesserungen erzielt werden. So konnte eine maximale Signaldynamik von fast 400% erzielt werden. Durch die Entwicklung eines geeigneten Probenvorbereitungsprotokolls war es möglich, mit dem fluoreszenzbasierten System einen Nachweis von Koffein an kommerziell verfügbaren Getränkeproben durchzuführen. Die Ergebnisse waren in guter Übereinstimmung mit HPLC-Kontrollexperimenten. Die Eignung von Rezeptoren auf Triphenylenketalbasis für die enantiofaciale Differenzierung an Heteroaromaten wurde durch Untersuchung verschiedener Wirt-Gast-Komplexe mittels CD-Spektroskopie und Tieftemperatur-NMR systematisch demonstriert. Rezeptoren mit Menthyl-Substituenten liefern laut NMR die stärkste Seitendifferenzierung. Anhand des CD wird ein vollständiges und schlüssiges Bild über den Zusammenhang zwischen dem Raumbedarf am Gast, der Ausrichtung der chiralen Gruppen am Wirt und dem erhaltenen CD hergestellt. Durch umfangreiche molekulardynamische Simulationen und nachfolgende semiempirische Berechnungen wurden Referenzspektren berechnet, welche die Zuordnung der Stereochemie anhand des CD eindeutig belegen. Die Ergebnisse sind zudem in guter Übereinstimmung mit den Ergebnissen aus röntgenkristallographischen Untersuchungen. (Diese Methode ließ sich erfolgreich auf die helicale Faltung von Alkanen in Kapseln von Rebek, jr. umsetzen.) Obwohl die Energieunterschiede zwischen den diastereomeren Komplexen klein sind, konnte anhand der CD-Spektroskopie somit erstmalig die enantiofaciale Differenzierung an einem heterocyclischen System bei Raumtemperatur beobachtet werden. Die beste enantiofaciale Differenzierung erzielen die Menthyl-abgeleiteten Rezeptoren. Diese sind hinsichtlich einer möglichen Anwendung als chirales „Auxiliar“ ungeeignet, da sie mit den sperrigen Cyclohexylgruppen auch den Raum oberhalb des gebundenen Gastes blockieren. Daher wird für die weitere Entwicklung auf die praktische Einführung chiraler Information in Form des Isocyanats verzichten werden müssen. Stattdessen zielen aktuelle Bemühungen auf den Aufbau chiraler Rückgrate, welche den Raum in der unteren Peripherie des Gastes beeinflussen.

The synthesis and incorporation into oligonucleotides of C-nucleosides containing the two aromatic, non-hydrogen-bonding nucleobase substitutes biphenyl (I) and bipyridyl (Y) are described. Their homo- and hetero-recognition properties in different sequential arrangements were then investigated via UV-melting curve analysis, gel mobility assays, CD- and NMR spectroscopy. An NMR analysis of a dodecamer duplex containing one biphenyl pair in the center, as well as CD data on duplexes with multiple insertions provide further evidence for the zipper-like interstrand stacking motif that we proposed earlier based on molecular modeling. UV-thermal melting experiments with duplexes containing one to up to seven I- or Y base pairs revealed a constant increase in T(m) in the case of I and a constant decrease for Y. Mixed I/Y base pairs lead to stabilities in between the homoseries. Insertion of alternating I/abasic site- or Y/abasic site pairs strongly decreases the thermal stability of duplexes. Asymmetric distribution of I- or Y residues on either strand of the duplex were also investigated in this context. Duplexes with three natural base pairs at both ends and 50 % of I pairs in the center are still readily formed, while duplexes with blunt ended I pairs tend to aggregate unspecifically. Duplexes with one natural overhang at the end of a I-I base pair tract can both aggregate or form ordered duplexes, depending on the nature of the natural bases in the overhang
