Frequently there is a disconnectedness, either perceived or actual, between theoretical principles and laboratory practice in science education and this holds true for clinical microbiology where traditionally knowledge is delivered in ‘chunks’ in a lecture format with the misguided belief that students have to know ‘everything about everything’. This preoccupation with content delivery often leaves no time for active class discussion or reflection. Moreover, laboratory classes are treated as add-ons to the process, rather than an integrated part of the whole learning experience. In redesigning our units (subjects) we have bridged the gap between the theory and practice of clinical bacteriology. In doing so, we have seen a transformation in the learning experiences of our students and in the way we teach.

Microbiological investigation of fresh and frozen fishes such as pomfret, surmai and mackerel was carried out under various conditions of preservation. Glazing, block-freezing and preservation in gunny bag were affected. Determination of bacterial load and isolation, identification and classification of the resistant bacteria were made. Spore-formers of Subtilis mesentericus group were found to be resistant to freezing as well as glazing by ascorbic acid, citric acid and sodium nitrite except a mixture of sodium chloride and glucose. Bacterial load was reduced to a good extent and maintained low till the end of frozen storage period.

Except for the preliminary studies at Torry Research Station in Scotland, no results have been reported on the succession of the bacterial flora during the storage of fish in chilled water. The present work was undertaken to elucidate the dynamics of bacterial population changes in chilled fresh water under comparable conditions of storage in melting ice (+1° to +3°C.) which has been earlier studied by de Silva in 1960.

Spoilage characteristics of different types of bacteria isolated from bacteriologically defective cans and processing factory environs were studied by inoculating pure cultures into sterile prawn meat. The pattern of spoilage, namely, production of off odour, bulging of the cans and disintegration of meat were observed. Data on spoilage under aerobic as well as anaerobic conditions are presented. Most of the cultures produced some kind of spoilage, though differences were observed in the extent of spoilage produced by different types of bacteria. Gram positive spore formers were found to be the major spoilers and the extent of spoilage was more with mixed cultures.

The total aerobic viable plate counts (TPCs) of skin, gills and intestine of newly caught oil sardine (Sardinella longiceps) and Indian mackerel ( Rastrelliger kanagurta) at four different temperatures, namely 36 ± 1°C, 28 ± 2°C (RT), 8 ± 1°C and 1 ±1°C, are reported. The total plate count at RT of the skin of oil sardine and Indian mackerel were in the range of l0 super(3) to 10 super(7) and 10 super(4) to 10 super(6) per cm², that of gills in the range of 10 super(5) to 10 super(9) and 10 super(4) to 10 super(8) per g and that intestine in the range of 10 super(5) to 10 sueper(9) and 10 super(5) to 10 super(8) per g respectively. The TPCs were markedly affected by the incubation temperature. Incubation at 28 ± 2°C gave the highest count; at 36 ± 1°C and 8 ± 1°C, the counts decreased by nearly 1-2 log cycles from that at RT. Incubation at 1 ± 1°C registered the lowest count. The peak values for bacterial counts of these fishes occurred at different periods of the year.

80% of the flora of skin, gills and intestines of oil sardine and mackerel at isolation temperature 28 ± 2°C consisted of Gram negative asporogenous rods or cocci, belonging to the genera Vibrio, Pseudomonas, Moraxella, Acinetobacter and Flavobacteria/Cytophaga. Nearly 10% of the flora was constituted by Gram positives, Micrococcus and Arthrobacter. Incubation temperature of 36 ± 1°C recovered more Vibrio spp. and Gram positives, while at lower temperatures of 8 ± 1°C and 1 ± 1°C, more Pseudomonas, Acinetobacter and Moraxella spp. were recovered. Significant changes with respect to season were observed in the relative distribution of different genera.

The thesis is mainly concerned with the study of the bacteriology of freezing of mackerel(Rastrelliger kanaggrta)and prawn (Metgpggaeus ddbsoni) The thesis in four sections describes the salient features of the observations and inferences on the bacteriology of freezing of fishes and prawns. This includes the evaluation of methodology for the optimum recovery of bacteria, bacteriology of the newly caught fish and prawn, the effect of freezing temperatures on the survival of selected species of isolates from fish and prawn and the bacteriology of freezing, frozen storage and thawing of prawn/fish emphasizing the effect contributed by each.
