We evaluated the efficiency of callus induction and plantlet regeneration from hypocotyl explants of broccoli (Brassica oleracea var. italica). The cultivars were ‘Marathon’, ‘Greenbelt’, and ‘Shogun’. Transformation success was not affected by the presence of tobacco feeder-cell layers on the culture media. The frequency of shoot regeneration was greater from 10-d-old hypocotyls than from 14-d-old hypocotyls. Both ‘Marathon’ and ‘Greenbelt’ had higher potentials for tissue regeneration than did ‘Shogun’. We found that for transformation selection, the optimum concentration was either 50 mg/L kanamycin or 100 mg/L genetkin.

Broccoli is a vegetable of important economic value as well as a source of vitamin C and anticarcinogenic property substances. Although it can be minimally processed, this vegetable presents fast senescence, characterized by yellowing, loss of turgescence, development of off-odors, increase in enzymatic activity and reduction of nutritional value. Among the frequently used techniques that can extend the shelf-life of minimally processed broccoli, we have the use of cool storage and modified atmosphere packaging (MAP). Several factors interfere in the efficiency of MAP, influencing the speed and intensity of the changes on atmosphere composition. This work aimed to study the influence of the amount packaged broccoli on its postharvest quality, when stored at 1ºC. Results demonstrated strong influence of the use of MAP compared to control treatment. It was observed that for all packaged treatments there was better vitamin C retention, lower loss of weight, lower peroxidase activity and better turgidity score of broccoli. This study demonstrated that the treatments with higher amounts of product showed the worst performance on vitamin C and aroma evaluation. The peroxidase activity was also influenced, and the best results were found in the treatment with higher amounts, which presented lowest values. However there is a tendency of increase in the activity along the time, especially at the end of the experiment. It was not detected the influence of the fill weight about color evaluation.

Sulforaphane, a naturally occurring cancer chemopreventive, is the hydrolysis product of glucoraphanin, the main glucosinolate in broccoli. The hydrolysis requires myrosinase isoenzyme to be present in sufficient activity; however processing leads to its denaturation and hence reduced hydrolysis. In this study, the effect of adding mustard seeds, which has a more resilient isoform of myrosinase, to processed broccoli was investigated with a view to intensify the formation of sulforaphane. Thermal inactivation of myrosinase from both broccoli and mustard seeds was studied. Thermal degradation of broccoli glucoraphanin was investigated in addition to the effects of thermal processing on the formation of sulforaphane and sulforaphane nitrile. Limited thermal degradation of glucoraphanin (less than 12 %) was observed when broccoli was placed in vacuum sealed bag (sous vide) and cooked in a water bath at 100 ºC for 8 and 12 min. Boiling broccoli in water prevented the formation of any significant levels of sulforaphane due to inactivated myrosinase. However, addition of powdered mustard seeds to the heat processed broccoli significantly increased the formation of sulforaphane.

We investigated the gene expression profiles of different members of the 1-aminocyclopropane-1-carboxilic acid (ACC) synthase (EC 4.4.1.14) gene family in broccoli (Brassica oleracea L. var. italica) during the post-harvest-induced senescence process. Using RT-PCR, three different cDNAs coding for ACC synthase (BROCACS1, BROCACS2 and BROCACS3) were amplified from floret tissue at the start of the senescence process. The three genes share relatively little homology, but have highly homologous sequences in Arabidopsis thaliana, and could be functionally related to these counterparts. Southern analyses suggest that BROCACS1 and BROCACS3 are present as single copy genes, while there are probably two copies of BROCACS2. All three genes showed different expression patterns: BROCACS1 is likely to be either wound - or mechanical stress-induced showing high transcript levels after harvesting, but no detectable expression afterwards. BROCACS2 shows steady expression throughout senescence, increasing at the latest stages, and BROCACS3 is almost undetectable until the final stages. Our results suggest that BROCACS1 could be required to initiate the senescence process, while BROCACS2 would be the main ACC synthase gene involved throughout the post-harvest-induced senescence. BROCACS3's expression pattern indicates that it is not directly involved in the initial stages of senescence, but in the final remobilization of cellular resources.

Os testes de vigor e o teste de germinação são componentes essenciais no controle de qualidade das empresas de produção de sementes. Com o objetivo de verificar a eficiência de diferentes testes de vigor e de variações de suas metodologias na avaliação da qualidade de sementes de couve-brócolos visando diferenciação de lotes e previsão de emergência em bandeja, cinco lotes de sementes do híbrido Flórida foram submetidos aos seguintes testes: germinação; primeira contagem de germinação; emissão de raiz primária (após 48, 56, 72, 80 e 96 h após a instalação do teste de germinação); emergência de plântulas em substrato; envelhecimento acelerado com água (1g de sementes mantidas a 41ºC por 48 e 72 h a 100%UR); envelhecimento acelerado com solução saturada de sal (mesmo procedimento do item anterior, porém usando solução de NaCl, 40% e 76%UR); condutividade elétrica (50 sementes em 25 mL de água destilada a 25ºC e leituras após 2, 4, 6, 8 e 24 h). Todos os testes apresentaram correlação significativa com a porcentagem de emergência de plântulas em substrato, a 1% de probabilidade. Os testes de envelhecimento acelerado com solução saturada de sal por 48 h e de condutividade elétrica após 8 e 24 h de embebição foram eficientes e tiveram resultados semelhantes aos da emergência em substrato. Os testes da primeira contagem de germinação, emissão da raiz primária após 56 h e envelhecimento acelerado com solução saturada de sal por 72 h, apresentaram-se mais eficientes que a emergência de plântulas em substrato na diferenciação do vigor dos lotes.

A eficiência dos testes de vigor na avaliação da qualidade de sementes, depende da sua padronização para as diferentes espécies. Neste contexto, os objetivos deste trabalho foram elaborar as curvas de absorção de água para determinar o período necessário ao ajuste do teor de água das sementes e aprimorar o teste de deterioração controlada para sementes de brócolis. Foram utilizadas sementes de três variedades: dois lotes de Piracicaba precoce, dois lotes de Flórida e quatro lotes de Ramoso. Os lotes foram caracterizados mediante a determinação do teor de água e germinação; o vigor foi determinado pelos testes de primeira contagem, comprimento de plântula, emergência e índice de velocidade de emergência. Foram determinadas as curvas de absorção de água por 84 horas. Para averiguar a eficiência do teste de deterioração controlada para sementes de brócolis, foi feito o ajuste do conteúdo de água das sementes para: 18, 20, 21, 23 e 24%, sendo estas expostas a 45ºC em banho-maria, por 24 horas. O delineamento utilizado foi o inteiramente casualizado. As médias foram comparadas pelo teste de Tukey a 5% de probabilidade. As curvas de absorção permitiram estimar os períodos de 15, 20, 22, 32 e 48 horas para elevação do teor de água a 18, 20, 21, 23 e 24% respectivamente. O teste de deterioração controlada, para sementes de brócolis, propiciou resultados promissores, tendo sido eficiente na separação de lotes, em todos os teores de água estudados, tanto quanto os testes de emergência de plântula e índice de velocidade de emergência.

The first topic area of this thesis involved studies on the accumulation and translocation of glucosinolates (GSs), bioactive secondary plant compounds, in broccoli plants. Changes in GS accumulation and gene expression levels in response to exogeneous methyl jasmonate (MeJA) treatment were analyzed in different tissue types at different developmental stages of broccoli. Greater accumulation of GSs with MeJA treatment was observed in apical leaves of broccoli seedlings and florets of plants at harvest maturity. Increases in indolyl GS in apical leaves of seedlings and florets were coupled with the up-regulation of indolyl GS biosynthesis genes. The accumulation of indolyl GSs appears to be modulated by MeJA treatment in an organ-specific manner for optimal distribution of defense substances in the plant. Metabolic profiling of hydrophilic metabolites using GC-MS demonstrated increased accumulation of various phenolics, ascorbates and amino acids in broccoli tissues after MeJA treatment. Distinct changes in carbohydrate levels observed between different tissues (vegetative leaves and floret tissues) of broccoli plants after treatment suggest that carbon metabolism is differentially modulated by MeJA treatment in different tissue types depending on sink-source relationships. Reduced levels of hexose sugars and tricarboxylic acid intermediates after MeJA treatment may reflect the increased requirement for carbon and energy needed to drive secondary product biosynthesis to accumulate metabolites for defense against insects and other herbivores. Substantial increases of indolyl and aromatic GSs after exogenous treatment with MeJA in stem and petioles of seedlings and the existence of intact indolyl-GS forms in phloem exudates suggest enhanced de novo synthesis in combination with active transport. Indoly GSs share structural similarities with the auxin, IAA, and may interact with components of the auxin transport system for intra- and extra-cellular transport or translocation. Application of the auxin efflux inhibitor, 1-naphthylphthalamic acid (NPA) reduced MeJA-mediated accumulation of indolyl GSs in broccoli florets and seedling tissues. NPA did not inhibit expression of indolyl GS biosynthesis genes shown to be upregulated by MeJA treatment or the accumulation of tryptophan, the amino acid precursor of indolyl GSs. Exogenous application of benzyl GS to Arabidopsis roots induced ectopic expression of the PIN1 protein associated with the auxin transport system similar to treatment with NPA, again suggesting GS interaction with the auxin efflux carrier system. The inhibitory effect of NPA on MeJA-mediated accumulation of GS may be due to competitive binding of NPA to auxin efflux carrier components and that GS transport is mediated by the auxin transport system. The inhibitory effect of NPA on indolyl and aromatic GS accumulation and the bioactivity of exogenous treatment of these GS compounds in PIN1 localization, Arabidopsis root growth, and gravitrophic response suggest that indolyl and aromatic GSs may be antagonistic to IAA transport and biosynthesis. Indolyl and aromatic GSs can also be potentially converted into IAA by hydrolysis. This intrinsic feature of GSs may be the part of a sophisticated regulatory process where the metabolic pathways in the plant shift from active growth to a reversible defense posture in response to biotic or abiotic stress. It seems likely that indolyl and aromatic GSs are important compounds that provide connections between jasmonate and auxin signaling. Further studies are required to reveal the regulatory mechanism for crosstalk between the two hormones. The third part of this research was to investigate effect of selenium fertilization and MeJA treatment on accumulation of GSs in broccoli florets. Increasing dietary intake of the element selenium (Se) has been shown to reduce the risk of cancer. Simultaneous enhancement of both Se and GS concentrations in broccoli floret tissue were conducted through the combined treatment of MeJA with Se fertilization. A low level of Se fertilization (concentration) with MeJA treatment displayed no significant changes in total aliphatic GS concentrations with 90% and 50% increases in indolyl and total GSs concentrations, respectively. This result suggests that Se- and GS-enriched broccoli with improved health-promoting properties can be generated by this combined treatment. The second topic of this thesis was conducted to provide basic information required to improve biomass quality and productivity and develop tools for gene transformation in Miscanthus x giganteus. The perennial rhizomatous grass, Miscanthus x giganteus is an ideal biomass crop due to its rapid vegetative growth and high biomass yield potential. As a naturally occurring sterile hybrid, M. x giganteus must be propagated vegetatively by mechanicalling divided rhizomes or from micropropagated plantlets. The effect of callus type, age and culture methods on regeneration competence was studied to improve regeneration efficiency and shorten the period of tissue culture in M. x giganteus propagation. Seven lignin biosynthesis genes and one putative flowering gene were isolated from M. x giganteus by PCR reactions using maize othologous sequences. Southern hybridization and nuclear DNA content analysis indicated that the genes isolated from M. x giganteus exist in the genome of other Miscanthus species as multiple copies. Analysis of lignin content and histological staining of lignin deposition indicated that higher lignin content is found in mature stem node tissues compared to young leaves and apical stem nodal tissues. Cell wall lignification is associated with increasing tissue maturity in Miscanthus species. RNAi and antisense constructs harboring sequences of these genes were developed to generate Miscanthus transgenic plants with suppressed of lignin biosynthesis and delayed flowering.

Literature has documented beneficial effects of seed priming on speed, synchronization and uniformity of germination. often leading to improved stand establishment. However. doubts still persist about the possible reversal effects, after drying and during storage of primed seeds that could overcome, partial or totally, the improved performance. The objectives of this research were to identify drying and storage procedures that would maintain the physiological performance achieved after seed priming, without negative effects on storability. First. hydroprimed cauliflower Seeds cv. Sharon and cv. Teresopolis Gigante, each represented by three seed lots were submitted to fast drying, slow drying, and treatments of pre-drying incubation (exposure to 35 degrees C, to a polyethylene glycol 6000 solution or a heat shock) followed by fast drying. In the second phase of this study, hydroprimed seed samples were submitted to fast drying (30-35 degrees C and 40-50% R.H.) and stored under laboratory conditions or in a chamber at 20 degrees C and 50% relative humidity for six months. Seed physiological potential was evaluated after 60-day intervals for germination (speed and percentage), Seedling emergence and saturated salt accelerated aging tests. All drying treatments efficiently preserved the favourable priming effects, except for the incubation at 35 degrees C for 96-144 hours. The beneficial priming effects followed by fast drying persisted for four months under controlled conditions (20 degrees C and 50% relative humidity).

Amostras de repolho (Brassica oleracea var. capitata) de densidade de 20.833, 25.641, 37.037, 55.555 e 111.111 plantas/ha foram analisadas quanto aos teores de ácido ascórbico e carboidratos solúveis. Não foram observadas diferenças significativas entre estes teores nas densidades de população utilizadas. O teor médio de ácido ascórbico foi 25,8 mg/100 g peso fresco. Os principais constituintes da fração carboidratos solúveis foram sacarose, glucose e frutose, perfazendo acima de 80% do total. Os teores médios de carboidratos solúveis, expressos em g/100 g de peso fresco foram os seguintes carboidratos solúveis totais (4,60), sacarose (0,45), glucose (1,94) e frutose (1,83).

Os teores de carboidratos solúveis, glucose, frutose, sacarose e ácido ascórbico foram determinados em amostras de couve-flor (Brassica oleracea var. botrytis), obtidas de tratamentos apresentando diversas densidades de plantais. O efeito do espaçamento não foi significativo para os dviersos constituintes analisados, embora o tratamento com 20.833 plantas/ha apresentasse maior teor de ácido ascórbico. O peso seco das amostras variou de 8,24 a 10,13% e ácido ascórbico de 93,1 a 113,5 mg/100 g de peso fresco. Os teores de carboidratos solúveis, expressos em g/100 g de peso fresco foram: totais (3,17-4,54) sacarose (0,26-0,66), glucose (0,92-1,50) e frutose (0,91-145).

Foi conduzido na Área Experimental do Setor de Horticultura, DAH-ESALQ, Piracicaba, um experimento sobre o uso de alguns herbicidas na cultura do repolho (Brassica oleracea var. capitata). Os herbicidas testados foram Dacthal (DCPA), tenoran (cloroxuron), Afalon (linuron) e Ramrod (propacloro). O Dacthal e o Ramrod não foram fitotóxicos ao repolho; o Tenoran foi levemente fitotóxico e o Afalon, muito tóxico. Uma única aplicação de herbicida não foi suficiente para o controle das plantas daninhas até o final da cultura.

Foi conduzido na Área Experimental do Setor de Horticultura, DAH-ESALQ, Piracicaba, um experimento sobre o uso de alguns herbicidas na cultura da couve-flor (Brassica oleracea var. botrytis). Os herbicidas foram Dacthal (DCPA), Tenoran (cloroxuron), Afalon (linuron) e Ramrod (propacloro). O Dacthal, Ramrod, e o Tenoran não foram fitotóxicos à couve-flor, enquanto que o Afalon foi muito fitotóxico. O herbicida que melhor resultado apresentou para a couve-flor foi o Ramrod.

Amostras de couve-flor (Brassica oleracea var. botrytis) foram armazenadas à temperatura ambiente, a 5°C (geladeira) e a - 20°C (congelador) e foram feitas as análises para ácido ascórbico. O teor de ácido ascórbico não variou durante o período de resfriamento e as amostras de couve-flor congelada mostrou uma redução de 50% durante 12 dias de armazenamento. Amostras colocadas à temperatura ambiente apresentaram um pequeno aumento até os 3 dias. Não foram observadas diferenças significativas entre os tipos de embalagens usadas (garrafa de vidro, sacos plásticos e folhas de alumínio) quanto ao teor de ácido ascórbico.
