Vuoden 2004 iskukykytutkimus esittää lentotiedustelujärjestelmän hankkimista tukemaan HN-kalustoon perustuvaa ilmasta maahan -järjestelmää. Tässä tutkimustyössä on tutkittu kolmen erilaisen miehittämättömän ilma-aluksen käyttömahdollisuuksia ilmasta maahan -aseiden tarvitseman maalitiedon hankkimisessa. Tutkimuksessa mukana olleet ilmasta maahan -aseet olivat JDAM (Joint Direct Attack Munition), AGM-154 JSOW (Joint Standoff Weapon) ja toistaiseksi epävarman hankintansa vuoksi vähäisemmässä määrin AGM-158 JASSM (Joint Air to Surface Standoff Weapon). Miehittämättömiksi ilmaaluksiksi valittiin Predator-koneen kolme eri kehitysversiota, MQ-1 Predator A, MQ-9 Reaper (Predator B) ja Avenger (Predator C). Tutkimuksen pääasiallinen tutkimusmenetelmä on ollut laadullinen kirjallisuusselvitys julkisia lähteitä käyttäen. Suoritusarvoista johdettuihin päätelmiin on käytetty soveltuvin osin määrällisiä tutkimusmenetelmiä. Lähestymistapa tutkimukseen on ollut hermeneuttinen. Alakysymysten osalta kyseessä on ollut deskriptiivinen tutkimus ja tavoitteena on ollut löytää avainasiat miehittämättömien ilma-alusten käytön kannalta. Tutkimusraportin ensimmäinen luku on johdanto. Toisessa luvussa on selvitetty toimintaympäristön yleiset piirteet ja kolmannessa luvussa ilmasta maahan -aseiden ominaisuudet. Neljäs luku käsittelee tutkimuksessa olleiden miehittämättömien ilma-alusten ominaisuuksia maalitiedon hankkimismahdollisuuksien kannalta. Tutkimustyön johtopäätökset on kirjattu viidenteen lukuun. Tulosten perusteella perinteisten, ilman häiveominaisuuksia rakennettujen miehittämättömien ilma-alusten ei ole mahdollista hankkia kaikkea ilmasta maahan -aseiden tarvitsemaa maalitietoa. Lähialueen ilmatorjunnan vaikutuksen väistämiseksi häiveominaisuudet ja jonkin asteinen tunkeutumiskyky ovat välttämättömät erityisesti pitkän kantaman ilmasta maahan -aseiden tarvitseman maalitiedon hankkimisessa.

Recently we have described an HPMA copolymer conjugate carrying both the aromatase inhibitor aminoglutethimide (AGM) and doxorubicin (Dox) as combination therapy. This showed markedly enhanced in vitro cytotoxicity compared to the HPMA copolymer-Dox (FCE28068), a conjugate that demonstrated activity in chemotherapy refractory breast cancer patients during early clinical trials. To better understand the superior activity of HPMA copolymer-Dox-AGM, here experiments were undertaken using MCF-7 and MCF-7ca (aromatase-transfected) breast cancer cell lines to: further probe the synergistic cytotoxic effects of AGM and Dox in free and conjugated form; to compare the endocytic properties of HPMA copolymer-Dox-AGM and HPMA copolymer-Dox (binding, rate and mechanism of cellular uptake); the rate of drug liberation by lysosomal thiol-dependant proteases (i.e. conjugate activation), and also, using immunocytochemistry, to compare their molecular mechanism of action. It was clearly shown that attachment of both drugs to the same polymer backbone was a requirement for enhanced cytotoxicity. FACS studies indicated both conjugates have a similar pattern of cell binding and endocytic uptake (at least partially via a cholesterol-dependent pathway), however, the pattern of enzyme-mediated drug liberation was distinctly different. Dox release from PK1 was linear with time, whereas the release of both Dox and AGM from HPMA copolymer-Dox-AGM was not, and the initial rate of AGM release was much faster than that seen for the anthracycline. Immunocytochemistry showed that both conjugates decreased the expression of ki67. However, this effect was more marked for HPMA copolymer-Dox-AGM and, moreover, only this conjugate decreased the expression of the anti-apoptotic protein bcl-2. In conclusion, the superior in vitro activity of HPMA copolymer-Dox-AGM cannot be attributed to differences in endocytic uptake, and it seems likely that the synergistic effect of Dox and AGM is due to the kinetics of intracellular drug liberation which leads to enhanced activity. The AGM theory of belief revision provides a formal framework to represent the dynamics of epistemic states. In this framework, the beliefs of the agent are usually represented as logical formulas while the change operations are constrained by rationality postulates. In the original proposal, the logic underlying the reasoning was supposed to be supraclassical, among other properties. In this paper, we present some of the existing work in adapting the AGM theory for non-classical logics and discuss their interconnections and what is still missing for each approach.

In this dissertation we present a model for iteration of Katsuno and Mendelzon’s Update, inspired in the developments for iteration in AGM belief revision. We adapt Darwiche and Pearls’ postulates of iterated belief revision to update (as well as the independence postulate proposed in [BM06, JT07]) and show two families of such operators, based in natural [Bou96] and lexicographic revision [Nay94a, NPP03]. In all cases, we provide a possible worlds semantics of the models.

Forty-six allogeneic hematopoietic stem cell transplantation (HSCT) patients were monitored for the presence of CMV antibodies, CMV-DNA and CMV antigens after transplantation. Immunoenzymatic serological tests were used to detect IgM and the increase in CMV IgG antibodies (increase IgG), a nested polymerase chain reaction (N-PCR) was used to detect CMV-DNA, and an antigenemia assay (AGM) was used to detect CMV antigens. The presence of CMV-IgM and/or CMV-increase IgG antibodies was detected in 12/46 (26.1%) patients, with a median time between HSCT and the detection of positive serology of 81.5 days. A positive AGM was detected in 24/46 (52.2%) patients, with a median time between HSCT and antigen detection of 62 days. Two or more consecutive positive N-PCR results were detected in 32/46 (69.5%) patients, with a median time between HSCT and the first positive PCR of 50.5 days. These results confirmed that AGM and mainly PCR are superior to serology for the early diagnosis of CMV infection. Six patients had CMV-IgM and/or CMV-increase IgG with a negative AGM (five cases) or N-PCR assay (one case). In five of these cases the serological markers were detected during the first 100 days after HSCT, the period of highest risk. These findings support the idea that serology may be useful for monitoring CMV infections in HSCT patients, especially when PCR is unavailable.
