CONTEXT Relationships between mind and body have gradually become accepted. Yogic practices cause modulation of the immune system. Transcendental meditation (TM) is a specific form of mantra meditation. We reported previously different plasma levels of catecholamines and pituitary hormones in TM practitioners comparing with a control group, and patterns of the daytime secretion of these hormones different from those normally described. AIMS The aim of the following study is to evaluate the immune system in these meditation practitioners, by determining leukocytes and lymphocytes subsets. METHODS TM group consisted of 19 subjects who regularly practice either TM or the more advanced Sidhi-TM technique. A control group consisted of 16 healthy subjects who had not previously used any relaxation technique. Total leukocytes, granulocytes, lymphocytes and monocytes were counted by an automated quantitative hematology analyzer, whereas lymphocytes subsets were determined by flow cytometry. Samples were taken from each subject at 0900 h after an overnight fast. RESULTS The results indicated that the TM group had higher values than the control group in CD3+CD4-CD8+ lymphocytes (P < 0.05), B lymphocytes (P < 0.01) and natural killer cells (P < 0.01), whereas CD3+CD4+CD8- lymphocytes showed low levels in meditation practitioners (P < 0.001). No significant differences were observed in total leukocytes, granulocytes, monocytes, total lymphocytes or CD3+ lymphocytes comparing both groups. CONCLUSIONS The technique of meditation studied seems to have a significant effect on immune cells, manifesting in the different circulating levels of lymphocyte subsets analyzed. The significant effect of TM on the neuroendocrine axis and its relationship with the immune system may partly explain our results.

Cardiovascular disease (CVD) is the single greatest cause of death in the United States, accounting for nearly 2400 deaths each day. It is estimated that 79.4 million American adults have some form of the disease, and CVD mortality rates are greater than those of cancer, chronic lower respiratory diseases, accidents and diabetes mellitus combined. Psychosocial stress is a nontraditional risk factor for CVD, and can contribute to the clustering of traditional risk factors as well as to vascular manifestations of the disease. The Transcendental Meditation (TM) technique has been researched as a cost effective intervention aimed at decreasing psychosocial stress. This literature review attempts to analyze randomized controlled clinical trials of TM on cardiovascular disease outcomes. Eleven studies met inclusion criteria and are described below, with statistically significant positive outcomes observed in each study. Studies are grouped by primary outcome reported in the categories of cardiovascular function, blood pressure, and exercise tolerance. The TM intervention significantly decreased insulin resistance, heart rate variability, and carotid intima media thickness and improved exercise tolerance compared to control groups. Seven studies also reported significant decreases in blood pressure among hypertensive and normotensive subjects. Five studies focused solely on African American subjects, a population that has disproportionately higher rates of CVD and hypertension, and found significant improvements in CVD outcomes. Further research is recommended to establish the efficacy of TM on CVD outcomes. Future trials should include larger sample populations, wider ethnic distributions of subjects, and longer follow-up to ascertain the impact of this particular stress reduction technique on cardiovascular disease.^

A C-ficocianina (C-FC), um pigmento comum nas cianobctérias e um dos mais abundantes constituintes da Spirulina platensis, vem sendo estudada por possuir várias propriedades como antioxidante, hepatoprotetora, antiinflamatória e inibidora da enzima COX-2. Alguns autores atribuem também a C-FC um efeito oxidante quando ela é o agente fotossensibilizante utilizado na terapia fotodinâmica (TFD), podendo ser um importante agente no tratamento do câncer. Entretanto ainda pouco se sabe sobre a ação da C-FC, como substância fotosensibilizante, no tratamento de ação fotodinâmica (AFD) em modelos biológicos. A AFD provoca a fotooxidação de substratos biológicos através da geração de espécies reativas de oxigênio produzidas pela associação entre um determinado comprimento de onda, uma substância fotosensível e oxigênio. Existem dois caminhos que levam a morte celular pelo processo de fotooxidação conhecidos como mecanismo do tipo I e tipo II. No mecanismo tipo I são gerados radicais como o radical ânion superóxido e radical hidroxila, enquanto no mecanismo do tipo II a espécie reativa de oxigênio gerada é o oxigênio singlete (1O2). A TFD da C-PC possui muitas vantagens em relação ao uso das hematoporfirinas e seus derivados, como rápida preparação e fácil purificação, ampla faixa de absorção do UV e visível, nenhum efeito local, e significativa redução da fotosensibilidade em tecidos normais por ter uma rápida metabolização em vivo. As pesquisas que avaliam os possíveis efeitos celulares da AFD têm sido também estendidas para as células tumorais que adquirem fenótipo de resistência a múltiplas drogas (MDR). A MDR é um fenômeno no qual células tumorais, selecionadas resistentes a um agente quimioterápico, adquirem resistência a outras drogas, 5 aparentemente não relacionadas. O fenótipo MDR é multifatorial, mas o mecanismo melhor estudado é a super expressão da glicoproteína-P, que é uma proteína de membrana capaz de fazer a extrusão de quimioterápicos para fora de célula. Com isso o objetivo deste estudo é avaliar a sensibilidade das linhagens celulares que expressem (Lucena) ou não (K562) o fenótipo MDR à AFD do pigmento C-FC, extraído da cianobactéria S. platensis, e propor um possível mecanismo de ação. A extração da C-PC foi feita no Laboratorio de Microbiologia e Engenharia de Bioprocesos (FURG). Diferentes concentrações de C-PC (0.025, 0.05, 0.10, 0.20 e 0.40 mg/ml para os testes de PDA da C-PC e 0.05, 0.10, 0.20, 0.40 e 0.60 mg/ml para os testes no escuro) foram usadas. O número de células viáveis foi avaliada imediatamente, 24 h e 48 h após o tratamento com C-PC ou PDA da C-PC através de exclusão por azul de trypan. A concentração de 0.05 mg/ml foi utilizada para determinar o possível papel da Pgp na resposta da linhagem Lucena e a concentração de 0.10 mg/ml foi utilizada nos testes de peroxidação lipídica (LPO), de produção de espécies reativas de oxigênio (ROS) e quantificação de apoptose/necrose. A PDA da CPC causou uma diminuição no número de células viáveis em ambas linhagens K562 (não MDR) e Lucena (MDR), sendo que a linhagem MDR foi menos sensível que a não MDR. Já nos testes realizados no escuro, nenhuma toxicidade foi encontrada para as duas linhagens. Nenhuma alteração na resistência da linhagem Lucena foi encontrada quando o modulador verapamil foi colocado durante o tratamento de APD com C-PC e até às 48h de acompanhamento após o tratamento. Também não foi encontrada diferença significativa de lipoperoxidação (LPO) mas houve uma tendência de aumento na produção de ROS, que foi mais evidente na linhagem K562. Além disso a linhagem Lucena apresentou uma produção basal de ROS significativamente maior que a K562. Nos testes de apoptose/necrose nenhuma diferença foi encontrada entre as células controle e tratadas em ambas linhagens. Os resultados encontrados neste estudo sugerem que a C-PC possa ser um potente agente fotosensibilizante, tanto para linhagens não MDR quanto para linhagens MDR e também que o mecanismo tipo II esteja envolvido em maior parte no efeito observado na PDA da C-PC, mas uma menor participação do mecanismo tipo I não pode ser descartada.

Multitemporal Landsat Thematic Mapper (TM) and Enhanced Thematic Mapper Plus (ETM+) imagery was used to assess coastline morphological changes in southeastern Brazil. A spectral linear mixing approach (SLMA) was used to estimate fraction imagery representing amounts of vegetation, clean water (a proxy for shade) and soil. Fraction abundances were related to erosive and depositional features. Shoreline, sandy banks (including emerged and submerged banks) and sand spits were highlighted mainly by clean water and soil fraction imagery. To evaluate changes in the coastline geomorphic features, the fraction imagery generated for each data set was classified in a contextual approach using a segmentation technique and ISOSEG, an unsupervised classification. Evaluation of the classifications was performed visually and by an error matrix relating ground-truth data to classification results. Comparison of the classification results revealed an intense transformation in the coastline, and that erosive and depositional features are extremely dynamic and subject to change in short periods of time.

Objective To investigate whether standardization of the multiplanar view (SMV) when evaluating the uterus using three-dimensional ultrasonography (3D-US) improves intra-and interobserver reliability and agreement with regard to endometrial measurement. Methods Two-dimensional (2D) and 3D-US was used to measure endometrial thickness by two observers in 30 women undergoing assisted reproduction treatment. Endometrial volume was measured with Virtual Organ Computer-aided AnaLysis (VOCAL (TM)) in the longitudinal (A) and coronal (C) planes using an unmodified multiplanar view (UMV) and a standardized multiplanar view (SMV). Measurement reliability was evaluated by intraclass correlation coefficient (ICC) and agreement was examined using Bland-Altman plots with limits of agreement (LoA). The ease of outlining the endometrial-myometrial interface was compared between the A-and C-planes using subjective assessment. Results Endometrial volume measurements using the SMV and A-plane were more reliable (intra-and interobserver ICCs, 0.979 and 0.975, respectively) than were measurements of endometrial thickness using 2D-US (intra-and interobserver ICCs, 0.742 and 0.702, respectively) or 3D-US (intra-and interobserver ICCs, 0.890 and 0.784, respectively). The LoAs were narrower for SMV than for UMV. Reliability and agreement were not much different between the A- and C-planes. However the observers agreed that delineating the endometrial-myometrial interface using the A-plane was easier (first and second observer, 50.0 and 46.7%, respectively) or `comparable` (50 and 53.3%, respectively), but never more difficult than using the C-plane. Conclusions Endometrial volume measurements are more reliable than endometrial thickness measurements and are best performed using SMV and the A-plane. Copyright (C) 2011 ISUOG. RESUMO:As terapias biológicas revolucionaram o tratamento das doenças autoimunes nos últimos anos. Tipicamente têm como alvos mediadores importantes no mecanismo das doenças. Os antagonistas do fator de necrose tumoral-α (TNF-α) são um grupo de agentes biológicos muito prescrito, pois estão indicados no tratamento de doenças imuno-mediadas comuns, tais como artrite reumatoide, artrite idiopática juvenil, artrite psoriática, espondilite anquilosante, doença de Crohn e colite ulcerosa. Com o uso frequente de inibidores do TNF-α, tem-se tornado evidente que estes agentes têm um potencial imunogénico importante, que pode comprometer o prognóstico a longo prazo dos doentes cronicamente tratados. A produção de anticorpos anti-fármaco parece causar falência terapêutica secundária em muitos doentes. Um dos efeitos dos anticorpos anti-fármaco é o aumento da eliminação do fármaco. A eliminação do fármaco, por sua vez, varia entre indivíduos, refletindo diferentes perfis farmacocinéticos. A determinação dos níveis séricos mínimos do agente anti-TNF-α é assim muito informativa e pode auxiliar nas decisões terapêuticas. Contudo, os testes imunológicos para determinar as concentrações séricas do fármaco não estão facilmente disponíveis na prática clínica. De forma a investigar uma nova técnica potencialmente fidedigna e prática para a deteção e quantificação dos agentes biológicos anti-TNF-α, foi testada a técnica por HTRF (homogeneous time-resolved fluorescence resonance energy transfer) para a determinação de concentrações séricas de infliximab. Apesar de apresentar algumas limitações relacionadas com as condições de leitura da fluorescência, esta técnica provou obter resultados próximos das concentrações obtidas por ELISA (enzyme-linked immunosorbent assay) bridging. Adicionalmente, tem a vantagem de ser de execução muito mais fácil e rápida. Deste modo, a técnica por HTRF poderá ser otimizada e tornar-se uma valiosa ferramenta laboratorial para orientar as decisões terapêuticas em doentes autoimunes com falência da terapêutica anti-TNF-α.--------- ABSTRACT: Biologic therapies revolutionized the treatment of autoimmune diseases in the last years. Typically, they target important disease mediators. Tumor necrosis factor-alpha (TNF-α) antagonists constitute a very prescribed group of biologic agents as they are indicated for the treatment of common immune-mediated diseases, such as rheumatoid arthritis, juvenile idiopathic arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn’s disease and ulcerative colitis. With the increasing use of TNF-α inhibitors it has been noticed that they have an important immunogenic potential that can compromise long-term outcomes in chronically treated patients. The production of anti-drug antibodies seems to cause secondary therapeutic failure in many patients. One of the effects of anti-drug antibodies is the enhancement of drug clearance. Drug clearance, in turn, varies among individuals, reflecting different pharmacokinetic profiles. Determination of serum anti-TNF-α drug trough levels is though very informative and could support treatment decisions. However, immunologic assays to determine drug serum concentrations are not readily available in clinical practice. In order to investigate a potentially reliable and practical new technique for detection and quantification of anti-TNF-α biologic agents, homogeneous time-resolved fluorescence resonance energy transfer (HTRF) technique was tested for determination of serum infliximab concentrations. Although presenting some limitations related with fluorescence reading conditions, this technique proved to give results close to the concentrations obtained by the widely used bridging enzyme-linked immunosorbent assay (ELISA). In addition, it has the advantage of being much easier and faster to perform. Thus, HTRF technique can be optimized and become a valuable laboratorial tool to guide treatment decisions in autoimmune patients with anti-TNF-α therapy failure.

The concern related to environment is growing. Due to this, it is needed to determine chemical elements in a large range of concentration. The neutron activation technique (NAA) determines the elemental composition by the measurement of artificial radioactivity in a sample that was submitted to a neutron flux. NAA is a sensitive and accurate technique with low detection limits. An example of application of NAA was the measurement of concentrations of rare earth elements (REE) in waste samples of phosphogypsum (PG) and cerrado soil samples (clayey and sandy soils). Additionally, a soil reference material of the International Atomic Energy Agency (IAEA) was also analyzed. The REE concentration in PG samples was two times higher than those found in national fertilizers, (total of 4,000 mg kg-1 ), 154 times greater than the values found in the sandy soil (26 mg kg-1 ) and 14 times greater than the in clayey soil (280 mg kg-1 ). The experimental results for the reference material were inside the uncertainty of the certified values pointing out the accuracy of the method (95%). The determination of La, Ce, Pr, Nd, Pm, Sm, Eu, Tb, Dy, Ho, Er, Tm, Yb and Lu in the samples and reference material confirmed the versatility of the technique on REE determination in soil and phosphogypsum samples that are matrices for agricultural interest.

Analytical expressions for the Green’s function of an annular elliptical ring microstrip antenna (AERMA) are developed and reported. The modal, radiation and input impedance characteristics of the TM, modes are determined from these expressions. The resonant frequencies of odd modes are greater than that of the even modes for all TMnl modes (n = 1, 2, 3, ...) udke elliptical microstrip structures. The radiation pattern and input imedance curves of TMI2 mode on comparison with available experimental result shows good agreement whch provides an independent validation to this technique. The performance of the AERMA is then investigated as a function of thickness and substrate dielectric permittivity.

The Phenotype MicroArray (TM) (PM) technology was used to study the metabolic characteristics of 29 Salmonella strains belonging to seven serotypes of S. enterica spp. enterica. Strains of serotypes Typhimurium (six strains among definite phage types DTs 1, 40 and 104) and Agona (two strains) were tested for 949 substrates, Enteritidis (six strains of phage type PT1), Give, Hvittingfoss, Infantis and Newport strains (two of each) were tested for 190 substrates and seven other Agona strains for 95 substrates. The strains represented 18 genotypes in pulsed-field gel electrophoresis (PFGE). Among 949 substrates, 18 were identified that could be used to differentiate between the strains of those seven serotypes or within a single serotype. Unique metabolic differences between the Finnish endemic Typhimurium DT1 and Agona strains were detected, for example, in the metabolism of d-tagatose, d-galactonic acid gamma-lactone and l-proline as a carbon source. Thus, the PM technique is a useful tool for identifying potential differential markers on a metabolic basis that could be used for epidemiological surveillance.

The aim of this study was to analyze, under scanning electron microscopy (SEM), the morphologic characteristics of root surfaces after application of CarisolvTM gel in association with scaling and root planing (SRP). Sixty periodontally compromised extracted human teeth were randomly assigned to 6 groups: 1) SRP alone; 2) passive topical application of CarisolvTM + SRP; 3) active topical application of CarisolvTM + SRP; 4) multiple applications of CarisolvTM + SRP; 5) SRP + 24% EDTA; 6) topical application of CarisolvTM + SRP + 24% EDTA. CarisolvTM gel was applied to root surfaces for 30 s, followed by scaling and root planing, consisting of 50 strokes with Gracey curettes in an apical-coronal direction, parallel to the long axis of the tooth. The only exception was group 4, in which the roots were instrumented until a smooth, hard and glass-like surface was achieved. All specimens were further analyzed by SEM. The results showed that the treatment with CarisolvTM caused significant changes in root surface morphology of periodontally compromised teeth only when the chemical agent was actively applied (burnishing technique). CarisolvTM failed to remove the smear layer completely, especially with a single application, independently of the method of application. Multiple applications of CarisolvTM were necessary to achieve a smear layer reduction comparable to that obtained with 24% EDTA conditioning.

Zusammenfassung Quecksilber kommt in der Natur in seinen vier umweltrelevanten Spezies Hg0, Hg2+, Me2Hg, MeHg+ vor. Die methylierten Spezies wurden dabei in der Vergangenheit aufgrund ihres relativ geringen Anteils am Gesamtquecksilber oft vernachlässigt. Me2Hg und MeHg+ spielen aber eine wichtige Rolle als intermediäre Transportspezies im Quecksilberkreislauf, und eine richtige Analytik dieser Verbindungen ist daher notwendig. Die GC/AFD-Methode (Gaschromatographie/Atomfluoreszenzdetektion) in Verbindung mit einem Purge&Trap-Probenaufgabesystem ermöglicht eine nachweisstarke Analytik dieser Verbindungen in Umweltproben. Dabei müssen die ionischen Quecksilberverbindungen MeHg+ und Hg2+ derivatisiert werden, um sie einer gaschromatographischen Trennung zugänglich zu machen.Das bestehende Verfahren wurde im Rahmen dieser Arbeit mit Hilfe einer alternativen Derivatisierungsmethode verbessert. Das optimierte Verfahren erlaubt auch eine Analyse von MeHg+-Gehalten in Proben mit hoher Chloridbelastung. Solche Proben konnten mit der herkömmlichen Ethylierung als Derivatisierungsmethode nicht analysiert werden, da während der Derivatisierung eine Umwandlung des MeHg+ zu Hg0 auftritt. Bei der Propylierung findet keine solche Transformation statt und sie ist daher eine hervorragende Ergänzung der bestehenden Methode.Die GC/AFD-Methode wurde zur Quecksilberspeziation in der Antarktis als Beispiel für ein anthropogen unbeeinflusstes Gebiet verwendet. Dabei konnten methylierte Quecksilberverbindungen im Meerwasser, in der Atmosphäre und erstmals im antarktischen Schnee nachgewiesen werden und somit die bedeutende Rolle der methylierten Quecksilberspezies am biogeochemischen Stoffkreislauf des Quecksilbers in dieser Region gezeigt werden. Vergleiche mit einer kontinentalen Region zeigten, dass dort MeHg+ und Me2Hg wesentlich weniger produziert bzw. schneller abgebaut werden.

Hallux rígidus (HR) affects the first metatarsophalangeal joint (MTPJ) between 35% and 60% of the population over 65 years and there are multiple ways of treatment. Depending on the radiological stage where you find the deformity determines the procedure to be performed; in the early stages cheilectomy techniques and corrective osteotomy is performed while the more advanced ratings, the surgeon chooses destructive techniques considered as arthrodesis and arthroplasty. This final of degree project aims to focus on 1 MTPJ destructive techniques to clarify which of the procedures generates better results by a number of parameters; outcomes of the American Orthopaedic Foot scale and Ankle Society Hallux metatarsophalangeal Interphalangeal-scale (AOFAS), range of motion (ROM) of the 1ºAMTF, radiological classification. As for the implant arthroplasty technique, this article offers information on material and design that generates better relating to patient characteristics such as age, inflammatory joint diseases, viability and durability of the implant results. The conclusion from this review is that the values obtained in the arthrodesis according AOFAS decrease due to loss of mobility, but both techniques have similar values of effectiveness and concludes with the decision that the technique used is determined taking into account various factors and patient characteristics. To evaluate the outcomes in patients treated for humerus distal third fractures with MIPO technique and visualization of the radial nerve by an accessory approach, in those without radial palsy before surgery. The patients were treated with MIPO technique. The visualization and isolation of the radial nerve was done by an approach between the brachialis and the brachiorradialis, with an oblique incision, in the lateral side of the arm. MEPS was used to evaluate the elbow function. Seven patients were evaluated with a mean age of 29.8 years old. The average follow up was 29.85 months. The radial neuropraxis after surgery occurred in three patients. The sensorial recovery occurred after 3.16 months on average and also of the motor function, after 5.33 months on average, in all patients. We achieved fracture consolidation in all patients (M=4.22 months). The averages for flexion-extension and prono-supination were 112.85° and 145°, respectively. The MEPS average score was 86.42. There was no case of infection. This approach allowed excluding a radial nerve interposition on site of the fracture and/or under the plate, showing a high level of consolidation of the fracture and a good evolution of the range of movement of the elbow.
