Structural Health Monitoring (SHM) schemes are useful for proper management of the performance of structures and for preventing their catastrophic failures. Vibration based SHM schemes has gained popularity during the past two decades resulting in significant research. It is hence evitable that future SHM schemes will include robust and automated vibration based damage assessment techniques (VBDAT) to detect, localize and quantify damage. In this context, the Damage Index (DI) method which is classified as non-model or output based VBDAT, has the ability to automate the damage assessment process without using a computer or numerical model along with actual measurements. Although damage assessment using DI methods have been able to achieve reasonable success for structures made of homogeneous materials such as steel, the same success level has not been reported with respect to Reinforced Concrete (RC) structures. The complexity of flexural cracks is claimed to be the main reason to hinder the applicability of existing DI methods in RC structures. Past research also indicates that use of a constant baseline throughout the damage assessment process undermines the potential of the Modal Strain Energy based Damage Index (MSEDI). To address this situation, this paper presents a novel method that has been developed as part of a comprehensive research project carried out at Queensland University of Technology, Brisbane, Australia. This novel process, referred to as the baseline updating method, continuously updates the baseline and systematically tracks both crack formation and propagation with the ability to automate the damage assessment process using output only data. The proposed method is illustrated through examples and the results demonstrate the capability of the method to achieve the desired outcomes.

The Commission has released a Draft Report on Business Set-Up, Transfer and Closure for public consultation and input. It is pleasing to note that three chapters of the Draft Report address aspects of personal and corporate insolvency. Nevertheless, we continue to make the submission to national policy inquiries and discussions that a comprehensive review should be undertaken of the regulation of insolvency and restructuring in Australia. The last comprehensive review of the insolvency system was by the Australian Law Reform Commission (the Harmer Report) and was handed down in 1988. Whilst there have been aspects of our insolvency laws that have been reviewed since that time, none has been able to provide the clear and comprehensive analysis that is able to come from a more considered review. Such a review ought to be conducted by the Australian Law Reform Commission or similar independent panel set up for the task. We also suggest that there is a lack of data available to assist with addressing questions raised by the Draft Report. There is a need to invest in finding out, in a rigorous and informed way, how the current law operates. Until there is a willingness to make a public investment in such research with less reliance upon the anecdotal (often from well-meaning but ultimately inadequately informed participants and others) the government cannot be sure that the insolvency regime we have provides the most effective regime to underpin Australia’s commercial and financial dealings, nor that any change is justified. We also make the submission that there are benefits in a serious investigation into a merged regulatory architecture of personal and corporate insolvency and a combined personal and corporate insolvency regulator.

Multi-step electron tunneling, or “hopping,” has become a fast-developing research field with studies ranging from theoretical modeling systems, inorganic complexes, to biological systems. In particular, the field is exploring hopping mechanisms in new proteins and protein complexes, as well as further understanding the classical biological hopping systems such as ribonuclease reductase, DNA photolyases, and photosystem II. Despite the plethora of natural systems, only a few biologically engineered systems exist. Engineered hopping systems can provide valuable information on key structural and electronic features, just like other kinds of biological model systems. Also, engineered systems can harness common biologic processes and utilize them for alternative reactions. In this thesis, two new hopping systems are engineered and characterized.

The protein Pseudomonas aeruginosa azurin is used as a building block to create the two new hopping systems. Besides being well studied and amenable to mutation, azurin already has been used to successfully engineer a hopping system. The two hopping systems presented in this thesis have a histidine-attached high potential rhenium 4,7-dimethyl-1,10-phenanthroline tricarbonyl [Re(dmp)(CO)3] + label which, when excited, acts as the initial electron acceptor. The metal donor is the type I copper of the azurin protein. The hopping intermediates are all tryptophan, an amino acid mutated into the azurin at select sites between the photoactive metal label and the protein metal site. One system exhibits an inter-molecular hopping through a protein dimer interface; the other system undergoes intra-molecular multi-hopping utilizing a tryptophan “wire.” The electron transfer reactions are triggered by excitation of the rhenium label and monitored by UV-Visible transient absorption, luminescence decays measurements, and time-resolved Infrared spectroscopy (TRIR). Both systems were structurally characterized by protein X-ray crystallography.

Nicotinic acetylcholine receptors (nAChRs) are pentameric, ligand-gated, cation channels found throughout the central and peripheral nervous system, whose endogenous ligand is acetylcholine, but which can also be acted upon by nicotine. The subunit compositions of nAChR determine their physiological and pharmacological properties, with different subunits expressed in different combinations or areas throughout the brain. The behavioral and physiological effects of nicotine are elicited by its agonistic and desensitizing actions selectively on neuronal nAChRs. The midbrain is of particular interest due to its population of nAChRs expressed on dopaminergic neurons, which are important for reward and reinforcement, and possibly contribute to nicotine dependence. The α6-subunit is found on dopaminergic neurons but very few other regions of the brain, making it an interesting drug target. We assayed a novel nicotinic agonist, called TI-299423 or TC299, for its possible selectivity for α6-containing nAChRs. Our goal was to isolate the role of α6-containing nAChRs in nicotine reward and reinforcement, and provide insight into the search for more effective smoking cessation compounds. This was done using a variety of in vitro and behavioral assays, aimed dually at understanding TI-299423’s exact mechanism of action and its downstream effects. Additionally, we looked at the effects of another compound, menthol, on nicotine reward. Understanding how reward is generated in the cholinergic system and how that is modulated by other compounds contributes to a better understand of our complex neural circuitry and provides insight for the future development of therapeutics.

EXTRACT (SEE PDF FOR FULL ABSTRACT): The annual cycle and non-seasonal variability of streamflow over a network of stations in western North America and Hawaii is studied in terms of atmospheric forcing elements. The phase lag between the annual cycle of streamflow and precipitation varies considerably over this network, as does the persistence of monthly streamflow anomalies. This lag effect appears to be largely a function of the relative amount of snow laid down in a particular basin. In addition to the rather strong annual cycle that exists in mean streamflow and its variance at most of the stations, there is also a distinct annual cycle in the autocorrelation of streamflow anomalies that is related to the interplay between the temperature and precipitation annual cycles; of particular importance is the existence of stored water in the form of a snow pack.

Linear regression models are constructed to predict seasonal runoff by fitting streamflow to temperature, precipitation, and snow water content across a range of elevations. The models are quite successful in capturing the differences in discharge between different elevation watersheds and their interannual variations. This exercise thus provides insight into seasonal changes in streamflow at different elevation watersheds that might occur under a changed climate.

A realistic alternative to traditional technology development and transfer has been utilized by the International Center for Living Aquatic Resources Management (ICLARM) to integrate pond fish culture into low-input farming systems in Malawi. Resource mapping was used to assess farm resources and constraints and introduce the concept of integrated resource management (IRM), the synergistic movement of resources between and among farm and household enterprises. Farmer-led IRM research projects are conducted on-farm and monitored by researchers through direct observation and on-station simulation of constraints and management practices. Technology-adoption rates by farmers involved in a pilot activity was 65% compared to 0% by farmers exposed only to top-down extension approaches. Within two years of adoption, every participating farmer had transferred the technology to an average of four other farmers without the involvement of the extension services.

Precipitation is a difficult variable to understand and predict. In this study, monthly precipitation in California is divided into two classes according to the monthly temperature to better diagnose the atmospheric circulation that causes precipitation, and to illustrate how temperature compounds the precipitation to runoff process.

This study examines the seasonal variability of an extensive history of snow observations over the western United States and Canada. It links variations in snowpack to variations in atmospheric circulation, surface temperature, and precipitation.

First, ENSO-associated circulation patterns over the North Pacific/North America sector were identified using an REOF (rotated empirical orthogonal function) analysis of the 700-mb height field and compositing these for extreme phases of the Southern Oscillation Index. ... EXTRACT (SEE PDF FOR FULL ABSTRACT): The characterization of inter-decadal climate variability in the Southern Hemisphere is severely constrained by the shortness of the instrumental climate records. To help relieve this constraint, we have developed and analyzed a reconstruction of warm-season (November-April) temperatures from Tasmanian tree rings that now extends back to 800 BC. A detailed analysis of this reconstruction in the time and frequency domains indicates that much of the inter-decadal variability is principally confined to four frequency bands with mean periods of 31, 57, 77, and 200 years. ... In so doing, we show how a future greenhouse warming signal over Tasmania could be masked by these natural oscillations unless they are taken into account.

EXTRACT (SEE PDF FOR FULL ABSTRACT): Bidecadal radiocarbon measurements on tree rings provide a detailed series of carbon-14 activities at isotopic equilibrium with atmospheric carbon dioxide. ... Most marine environments do not permit development of a comparable series of carbon-14 ages with which to compare the terrestrial tree ring series. However, we have recently begun work on such a series using material from the varved sediments of the Santa Barbara Basin off southern California. ... We now have a nearly continuous record of carbon-14 dates representing the age of the water over the upper 100 meters. ... The ocean reservoir ages show an increase prior to 1450 and a progressive decrease with time after 1450. Although there may be other explanations, we believe this trend is principally the result of changes in large-scale upwelling of water from below 500 meters. These changes were probably also associated with changes in the intensity of the California Current.

This report continues to cover the period of the financial year. Although, as before, for the sake of continuity and accuracy many of the statistics refer to the calendar year. The year has been notable in particular for the launching of extensive research projects on wild-life conservation problems, and much progress has already been made. This has been rendered possible by the greatly appreciated help and interest of the Fulbright Commission, who arranged for three experienced American wild-life scientists to work on these problems in Uganda. The latter have been working in conjunction with The Game and Fisheries Department's Biologist, as a team under the general Direction of the recently created Fauna Research Committee.
