The present study aimed to evaluate the acaricidal efficacy of fluazuron (2.5 mg/kg), administered as a pour-on, in comparison to an injectable formulation containing fluazuron (1.6 mg/kg) + ivermectin (0.63 mg/kg), against Rhipicephalus (Boophilus) microplus in naturally and experimentally infested cattle. Two studies were conducted with different tick strains, one with artificial infestations (Stall Test, using leight animals per group) and one with natural infestations (utilizing ten animals per group): In both studies, the animals were randomized, according to average tick counts performed on days -3, -2 and -1, into four groups: T01, negative control (saline solution); T02, pour-on fluazuron (2.5 mg/kg); T03: subcutaneous fluazuron (1.6 mg/kg) + ivermectin (0.63 mg/kg); and T04 subcutaneous ivermectin (0.63 mg/kg). Based on obtained results, and considering the utilized tick strains, it was possible to conclude that the pour-on fluazuron (2.5 mg/kg) formulation demonstrated high acaricidal efficacy, with protection periods ranging from 49 to 77 days against Rhipicephalus (Boophilus) microplus. On the other hand, for the injectable fluazuron (1.6 mg/kg) + ivermectin (0.63 mg/kg) formulation, it was not possible to observe elevated anti-R. (B.) microplus effect on both artificial and experimental infestation studies. Results observed for this combination were similar or inferior to those obtained by subcutaneous ivermectin (0.63 mg/kg). Future studies with this formulation containing fluazuron (1.6 mg/kg) + ivermectin (0.63 mg/kg), regarding pharmacokinetic and/or bioavailability profiles, or even studies analyzing both this active principles separately, are needed, seeking to better understand the effects of such combination against Rhipicephalus (Boophilus) microplus parasitizing cattle. O objetivo deste trabalho foi estudar os efeitos do ácido giberélico (GA3), do ethephon e da interação de ambos os reguladores vegetais no processo germinativo de sementes de atemoia (Annona cherimola Mill. x A. squamosa L. ), cultivar 'Gefner'. Empregou-se delineamento experimental inteiramente casualizado, em esquema fatorial 5², com os tratamentos constituídos pela combinação de cinco concentrações de GA3 (ácido giberélico) e cinco concentrações de ethephon, resultando em 25 tratamentos, com quatro repetições de 25 sementes por parcela. As concentrações de GA3 empregadas foram: 0; 250; 500; 750 e 1.000 mg L-1 i.a.e de ethephon: 0; 25; 50; 75 e 100 mg L-1 i.a.. Os tratamentos com os reguladores vegetais foram aplicados na semente por imersão das mesmas nas soluções de GA3 e ethephon por período de 36 horas. As sementes foram semeadas em rolo de papel germitest e levadas à câmara de germinação onde permaneceram no escuro, com temperatura alternada entre 20ºC por 8 horas e 30ºC por 16 horas. As variáveis avaliadas foram: percentagem, tempo e índice de velocidade de germinação, percentagem de plântulas normais e percentagem de sementes dormentes. Existe interação da ação dos reguladores vegetais estudados no processo germinativo de sementes de atemoia, o que permite concluir que a percentagem de germinação de sementes de atemoia (Annona cherimola Mill. x A. squamosa L. ) cv 'Gefner' é aumentada com o emprego de 778 mg L-1 de GA3, enquanto a associação entre elevadas concentrações de GA3 e 75 a 100 mg L-1 de ethephon incrementam o índice de velocidade de germinação e a percentagem de plântulas normais.

Most atypical antipsychotic drugs (APDs), e. g. risperidone (RIS), produce more extensive blockade of brain serotonin (5-HT)(2A) than dopamine (DA) D-2 receptors. This distinguishes them from typical APDs, e.g. haloperidol (HAL). Our objective was to test the hypothesis that augmentation of low doses of RIS or HAL (2 mg/day) with pimavanserin (PIM), a selective 5-HT2A inverse agonist, to enhance 5-HT2A receptor blockade, can achieve efficacy comparable to RIS, 6 mg/day, but with lesser side effects. In a multi-center, randomized, double-blind, 6 week trial, 423 patients with chronic schizophrenia experiencing a recent exacerbation of psychotic symptoms were randomized to RIS2mg + placebo (RIS2PBO), RIS2mg + PIM20mg (RIS2PIM), RIS6mg + PBO (RIS6PBO), HAL2mg + PBO (HAL2PBO), or HAL2mg + PIM20mg (HAL2PIM). Improvement in psychopathology was measured by the PANSS and CGI-S. The reduction in PANSS Total Score with RIS2PIM at endpoint was significantly greater than RIS2PBO: -23.0 vs. -16.3 (p = 0.007), and not significantly different from the RIS6PBO group: -23.2 points. The percentage of patients with >= 20% improvement at day 15 in the RIS2PIM group was 62.3%, significantly greater than the RIS6PBO (42.1%; p = 0.01) and the RIS2PBO groups (37.7%; p = 0.002). Weight gain and hyperprolactinemia were greater in the RIS6PBO group than the RIS2PIM group but there was no difference in extrapyramidal side effects (EPS). HAL2PBO and HAL2PIM were not significantly different from each other in efficacy but HAL2PIM had less EPS at end point. Both HAL groups and RIS6PBO showed equal improvement in psychopathology at endpoint, indicating HAL 2 mg/day is effective to treat an acute exacerbation in chronic schizophrenia patients. In conclusion, a sub-effective RIS dose combined with PIM to enhance 5-HT2A receptor blockade provided faster onset of action, and at endpoint, equal efficacy and better safety, compared to standard dose RIS. These results support the conclusion that 5-HT2A receptor blockade is a key component of the action of some atypical APDs and can reduce EPS due to a typical APD. (C) 2012 Elsevier B.V. OBJECTIVE: To evaluate safety of same-day administration of verteporfin and ranibizumab. METHODS: Prospective, open-label, multicentre study; patients with predominantly classic (n = 13) or occult (n = 19) choroidal neovascularisation secondary to age-related macular degeneration received standard-fluence verteporfin at baseline and months 3, 6 and 9, based on fluorescein angiography (FA). Ranibizumab 0.5 mg was administered at baseline and months 1, 2 and 3. MAIN OUTCOME MEASURE: The incidence of severe vision loss (best-corrected visual acuity (BCVA) loss > or = 30 letters; primary safety assessment). RESULTS: No severe vision loss due to ocular inflammation or uveitis occurred. One patient had moderate vision loss (BCVA loss > or = 15 letters). Three patients had mild/moderate uveitis. Two serious ocular adverse events occurred (retinal pigment epithelial tear and moderate BCVA decrease). No systemic adverse events occurred. At 9 months, all lesions were inactive with no recurrent leakage on FA and optical coherence tomography; macular oedema and subretinal fluid resolved. The mean BCVA measured at 2 m improved by 6.9 letters at 4 months and 2.4 letters at 9 months. CONCLUSIONS/APPLICATION TO CLINICAL PRACTICE: Same-day verteporfin and ranibizumab was safe and not associated with severe vision loss or severe ocular inflammation. Lesions stabilized, with minimal treatment required after month 3.

Context: In the Health Outcomes and Reduced Incidence with Zoledronic Acid Once Yearly - Pivotal Fracture Trial (HORIZON-PFT), zoledronic acid (ZOL) 5 mg significantly reduced fracture risk. Objective: To identify factors associated with greater efficacy during ZOL 5 mg treatment. Design, Setting and Patients: Subgroup analysis (preplanned and post hoc) of a multicenter, double-blind, placebo-controlled, 36-month trial in 7765 women with postmenopausal osteoporosis. Intervention: Single infusion of ZOL 5 mg or placebo at baseline, 12 and 24 months. Main Outcome Measures: Primary endpoints: new vertebral fracture and hip fracture. Secondary endpoints: non-vertebral fracture, change in femoral neck bone mineral density (BMD). Baseline risk factor subgroups: age, BMD T-score and vertebral fracture status, total hip BMD, race, weight, geographical region, smoking, height loss, history of falls, physical activity, prior bisphosphonates, creatinine clearance, body mass index (BMI), concomitant osteoporosis medications. Results: Greater ZOL induced effects on vertebral fracture risk with younger age (treatment-by-subgroup interaction P=0.05), normal creatinine clearance (P=0.04), and BMI >/=25 kg/m(2) (P=0.02). There were no significant treatment-factor interactions for hip or non-vertebral fracture or for change in BMD. Conclusions: ZOL appeared more effective in preventing vertebral fracture in younger women, overweight/obese women and women with normal renal function. ZOL had similar effects irrespective of fracture risk factors or femoral neck BMD.

Entre las técnicas moleculares, RAPD-PCR es una de las más rápidas y operativamente simple para la caracterización de cultivares. Sin embargo, la confiabilidad de sus resultados radica, en gran parte, en la optimización previa de variables que pueden afectar los patrones de amplificación. Se investigó la repetibilidad de patrones RAPD de olivo bajo diferentes condiciones experimentales. Entre ellas, la pureza del ADN, el tipo de Taq ADN-polimerasa, las concentraciones de Mg+2 y dNTPs, el uso de tejidos atacados por patógenos y el uso de diferentes termocicladores, modificaron los patrones de bandas. Por el contrario, pequeñas modificaciones de la temperatura de apareamiento de cebadores, la concentración del ADN molde y la edad de los tejidos vegetales de los cuales se aisló ADN, no afectaron los patrones amplificados.

En una plantación de kiwi situada en Balcarce (provincia de Buenos Aires, Argentina), se evaluó el efecto de la fertilización nitrogenada y potásica sobre el crecimiento de la planta durante la etapa de implantación, en la temporada 2008 - 2009. Los tratamientos fueron T: control sin fertilizar, N: fertilizado con nitrógeno (48 kg/ha N), K: fertilizado con potasio (60 kg/ha K) y NK: fertilizado con nitrógeno y potasio (48 kg/ha N - 60 kg/ha K). Se realizaron muestreos periódicos de: longitud de tallo, longitud del último entrenudo y área foliar. Se determinó el diámetro del tallo a fin de temporada y la distribución de raíces en el perfil de suelo en plena dormición. Se efectuaron análisis foliares de macronutrientes, así como N mineral, N anaeróbico (NAN) y carbono orgánico (CO) en el suelo. El suelo correspondió a un Argiudol, con niveles de NAN entre 85 - 100 mg/kg y de CO de 45 - 50 g/kg en los primeros 30 cm. Los resultados obtenidos, en cuanto al crecimiento de la parte aérea de la planta, no constituyen evidencia suficiente para justificar la fertilización con N y K bajo las condiciones experimentales del estudio. Aun en el T, los niveles foliares de N y K se mantuvieron dentro de los estándares normales. El sistema radical, observado durante el reposo, se habría visto beneficiado por la fertilización nitrogenada. En el estrato de suelo comprendido entre 10 y 20 cm se determinó un promedio de 61 raíces/100 cm2, en el tratamiento N, 40 en el tratamiento combinado NK, 28 en el tratamiento K, y 19 en el testigo.

In order to investigate a possible connection between tropical northeast (NE) Atlantic primary productivity, Atlantic meridional overturning circulation (AMOC), and drought in the Sahel region during Heinrich Stadial 1 (HS1), we used dinoflagellate cyst (dinocyst) assemblages, Mg/Ca based reconstructed temperatures, stable carbon isotopes (d13C) and geochemical parameters of a marine sediment core (GeoB 9508-5) from the continental slope offshore Senegal. Our results show a two-phase productivity pattern within HS1 that progressed from an interval of low marine productivity between ~ 19 and 16 kyr BP to a phase with an abrupt and large productivity increase from ~ 16 to 15 kyr BP. The second phase is characterized by distinct heavy planktonic d13C values and high concentrations of heterotrophic dinocysts in addition to a significant cooling signal based on reconstructions of past sea surface temperatures (SST). We conclude that productivity variations within HS1 can be attributed to a substantial shift of West African atmospheric processes. Taken together our results indicate a significant intensification of the North East (NE) trade winds over West Africa leading to more intense upwelling during the last millennium of HS1 between ~ 16 and 15 kyr BP, thus leaving a strong imprint on the dinocyst assemblages and sea surface conditions. Therefore, the two-phase productivity pattern indicates a complex hydrographic setting suggesting that HS1 cannot be regarded as uniform as previously thought.

Multidecadal variations in Atlantic sea surface temperatures (SST) influence the climate of the Northern Hemisphere. However, prior to the instrumental time period, information on multidecadal climate variability becomes limited, and there is a particular scarcity of sufficiently resolved SST reconstructions. Here, we present an eastern tropical North Atlantic reconstruction of SSTs based on foraminiferal (Globigerinoides ruber pink) Mg/Ca ratios that resolves multidecadal variability over the past 1700 years. Spectral power in the multidecadal band (50 to 70 years period) is significant over several time intervals suggesting that the Atlantic Multidecadal Oscillation (AMO) has been influencing local SST. Since our data exhibit high scatter the absence of multidecadal variability in the remaining record does not exclude the possibility that SST variations on this time scale might have been present without being detected in our data. Cooling by ~0.5 °C takes place between about AD 1250 and AD 1500; while this corresponds to the inception of the Little Ice Age (LIA), the end of the LIA is not reflected in our record and SST remains relatively low. This transition to cooler SSTs parallels the previously reconstructed shift in the North Atlantic Oscillation towards a low pre-20th century mean state and possibly reflects common solar forcing.
