In this work, the influence of modifications of the cp-Ti and Ti 6Al 4V alloy, by treating the surface with NaOH and depositing titanium oxide and hydroxyapatite by sol-gel method, on their biocompatibility was studied. The coatings were characterized by scanning electron microscopy and X-ray diffraction which showed that the coatings on Ti 6Al 4V are better than on cp-Ti. Adhesion tests showed that adhesion strength of the coatings on cp-Ti substrate is less than on Ti 6Al 4V as well as cytotoxicity for L929 fibroblast cells is higher

In this study, a composite of titanium oxide (TixOy) and carbon nanotubes multi-walled (MWCNT) was synthesized on a titanium substrate using the sol-gel method. The electrode obtained (TixOy-MWCNT/Ti) was used to the photodegradation of Carbaryl. The morphology and structure of the TixOy-MWCNT composite were characterized by scanning electron microscopy (SEM), scanning electron microscopy by field emission (FEG-SEM) and X-ray diffraction (XRD). The electrode was evaluated for degradation of Carbaryl (0.9 mmol L-1) in phosphate buffer pH 6, and using chronoamperometry by applying a potential of +1,5 V for 1 h. Using the Ultraviolet-Visible test, the absorbance at 220 nm was collected every 15 min to calculate the percentage of Cabaryl´s degradation. Can be evaluated that the Carbaryl degradation using the TixOy-MWCNT/Ti electrode was 22% more efficient when compared with the electrode without the presence of titanium oxides (MWCNT / Ti)

Three nanostructured platinum-niobium supported on Vulcan XC-72R carbon black materials were prepared as catalysts for the ethanol electroxidation: (i) deposition of platinum and niobium on Vulcan XC-72R carbon black, (ii) platinum decorated on a mixture of commercial amorphous Nb2O5/carbon black, and (iii) the same than ii but using crystalline Nb2O5, by reduction of the precursors with sodium borohydride in ethanol. All the catalysts showed platinum crystal sizes in the range of 3-4 nm, with no or little modification of the lattice parameter. The analyses of the electronic structure from the XANES region of the XAS spectra displayed some interactions between platinum and niobium, despite the niobium was primarily in the form of pentoxide in all the catalysts. CO stripping exhibited a promising low onset potential and a large current density, especially in the case of the deposited catalyst. Ethanol electroxidation experiments revealed that the Pt-Nb(2)O(5)crystalline/C generated the largest current. However it was not effective to completely oxidize ethanol, leading to acetic acid as the main product. In this sense, the highest efficiency for the complete oxidation of ethanol was obtained for the deposited catalyst. These results were interpreted in terms of the physico-chemical characteristic displayed by the different catalysts. (C) 2012 The Electrochemical Society. This work presents a methodology to the morphology analysis and characterization of nanostructured material images acquired from FEG-SEM (Field Emission Gun-Scanning Electron Microscopy) technique. The metrics were extracted from the image texture (mathematical surface) by the volumetric fractal descriptors, a methodology based on the Bouligand-Minkowski fractal dimension, which considers the properties of the Minkowski dilation of the surface points. An experiment with galvanostatic anodic titanium oxide samples prepared in oxalyc acid solution using different conditions of applied current, oxalyc acid concentration and solution temperature was performed. The results demonstrate that the approach is capable of characterizing complex morphology characteristics such as those present in the anodic titanium oxide.

This work proposes the application of fractal descriptors to the analysis of nanoscale materials under different experimental conditions. We obtain descriptors for images from the sample applying a multiscale transform to the calculation of fractal dimension of a surface map of such image. Particularly, we have used the Bouligand-Minkowski fractal dimension. We applied these descriptors to discriminate between two titanium oxide films prepared under different experimental conditions. Results demonstrate the discrimination power of proposed descriptors in such kind of application.

The dramatic impact that vascular diseases have on human life quality and expectancy nowadays is the reason why both medical and scientific communities put great effort in discovering new and effective ways to fight vascular pathologies. Among the many different treatments, endovascular surgery is a minimally-invasive technique that makes use of X-ray fluoroscopy to obtain real-time images of the patient during interventions. In this context radiopaque biomaterials, i.e. materials able to absorb X-ray radiation, play a fundamental role as they are employed both to enhance visibility of devices during interventions and to protect medical staff and patients from X-ray radiations. Organic-inorganic hybrids are materials that combine characteristics of organic polymers with those of inorganic metal oxides. These materials can be synthesized via the sol-gel process and can be easily applied as thin coatings on different kinds of substrates. Good radiopacity of organic-inorganic hybrids has been recently reported suggesting that these materials might find applications in medical fields where X-ray absorption and visibility is required. The present PhD thesis aimed at developing and characterizing new radiopaque organic-inorganic hybrid materials that can find application in the vascular surgery field as coatings for the improvement of medical devices traceability as well as for the production of X-ray shielding objects and garments. Novel organic-inorganic hybrids based on different polyesters (poly-lactic acid and poly-ε-caprolactone) and polycarbonate (poly-trimethylene carbonate) as the polymeric phase and on titanium oxide as the inorganic phase were synthesized. Study of the phase interactions in these materials allowed to demonstrate that Class II hybrids (where covalent bonds exists between the two phases) can be obtained starting from any kind of polyester or polycarbonate, without the need of polymer pre-functionalization, thanks to the occurrence of transesterification reactions operated by inorganic molecules on ester and carbonate moieties. Polyester based hybrids were successfully coated via dip coating on different kinds of textiles. Coated textiles showed improved radiopacity with respect to the plain fabric while remaining soft to the touch. The hybrid was able to coat single fibers of the yarn rather than coating the yarn as a whole. Openings between yarns were maintained and therefore fabric breathability was preserved. Such coatings are promising for the production of light-weight garments for X-ray protection of medical staff during interventional fluoroscopy, which will help preventing pathologies that stem from chronic X-ray exposure. A means to increase the protection capacity of hybrid-coated fabrics was also investigated and implemented in this thesis. By synthesizing the hybrid in the presence of a suspension of radiopaque tantalum nanoparticles, PDMS-titania hybrid materials with tunable radiopacity were developed and were successfully applied as coatings. A solution for enhancing medical device radiopacity was also successfully investigated. High metal radiopacity was associated with good mechanical and protective properties of organic-inorganic hybrids in the form of a double-layer coating. Tantalum was employed as the constituent of the first layer deposited on sample substrates by means of a sputtering technique. The second layer was composed of a hybrid whose constituents are well-known biocompatible organic and inorganic components, such as the two polymers PCL and PDMS, and titanium oxide, respectively. The metallic layer conferred to the substrate good X-ray visibility. A correlation between radiopacity and coating thickness derived during this study allows to tailor radiopacity simply by controlling the metal layer sputtering deposition time. The applied metal deposition technique also permits easy shaping of the radiopaque layer, allowing production of radiopaque markers for medical devices that can be unambiguously identified by surgeons during implantation and in subsequent radiological investigations. Synthesized PCL-titania and PDMS-titania hybrids strongly adhered to substrates and show good biocompatibility as highlighted by cytotoxicity tests. The PDMS-titania hybrid coating was also characterized by high flexibility that allows it to stand large substrate deformations without detaching nor cracking, thus being suitable for application on flexible medical devices.

Die zentrale Funktion des Hauptlichtsammlerkomplexes des Photosystems II, LHCII, besteht in der Absorption von Sonnenlicht und der Bereitstellung von Energie für die photosynthetische Ladungstrennung im Reaktionszentrum des Photosystems. Auch in der Regulation der Photosynthese spielt der LHCII eine wichtige Rolle, da die Energieverteilung zwischen Photosystem I und Photosystem II im Rahmen des sog. „State Transition“-Prozesses über die Verteilung der Lichtsammlerkomplexe zwischen den beiden Photosystemen gesteuert wird. Im Blickfeld des ersten Teils dieser Arbeit stand die konformative Dynamik der N-terminalen Domäne des LHCII, die wahrscheinlich in die Regulation der Lichtsammlung involviert ist. Gemeinsam mit Mitarbeitern des 3. Physikalischen Instituts der Universität Stuttgart wurde an der Etablierung einer Methode zur einzelmolekülspektroskopischen Untersuchung der Dynamik des N-Terminus gearbeitet. Als Messgröße diente der Energietransfer zwischen einem Fluoreszenzfarbstoff, der an die N-terminale Domäne gekoppelt war, und den Chlorophyllen des Komplexes. Die Funktion des LHCII als effiziente Lichtantenne bildete die Grundlage für den zweiten Teil dieser Arbeit. Hier wurde untersucht, in wie weit LHCII als Lichtsammler in eine elektrochemische Solarzelle integriert werden kann. In der potentiellen Solarzelle sollte die Anregungsenergie des LHCII auf Akzeptorfarbstoffe übertragen werden, die in der Folge Elektronen in das Leitungsband einer aus Titandioxid oder Zinndioxid bestehenden porösen Halbleiterelektrode injizierten, auf der Komplexe und Farbstoffe immobilisiert waren.

In der hier vorliegenden Dissertation wird die Entwicklung und Charakterisierung einer biomimetischen Beschichtung für Titanimplantatoberflächen, insbesondere Dentalimplantate, beschrieben. Ziel war es, die Adhäsion und Aktivität von Osteoblasten auf Titanoberflächen zu steigern und so eine Beschleunigung der Implantatintegration in das Knochengewebe zu erreichen. Hierfür wurde eine spezielle Art der biomimetischen Beschichtung entwickelt, bei der biotinyliertes Fibronektin (bFn) über Streptavidin auf eine biotinylierte TiOX-Modelloberfläche immobilisiert wurde. Die Biotinmodifizierung der TiOX-Oberfläche erfolgte hierbei über einen „Self-Assembly-Prozess“ durch sequenzielle Chemiesorption von N-(6-aminohexyl)aminopropyltrimethoxysilan sowie verschiedenen Sulfo-NHS-Biotin-Derivaten, welche den Aufbau einer Streptavidin-Monolage ermöglichten. Als ein wichtiges Resultat zeigte sich, dass die Streptavidin-Monolage effektiv die unspezifische Adsorption von Proteinen an die TiOX-Oberfläche unterbindet und hierdurch die Adhäsion von Osteoblasten auf dieser unterdrückt. Dies hat den Vorteil, dass auf eine antiadhäsive Basisbeschichtung, welche für eine spezifische Zellreaktion wichtig ist, verzichtet werden kann. Dieses osteoblastere Adhäsionsverhalten änderte sich signifikant nach Anbindung von bFn an die Streptavidin-Monolage, mit dem Ergebnis, einer drastischen Steigerung der Osteoblastenadhäsion. Weiterhin besaßen Osteoblasten auf diesen Oberflächen ein Proteinexpressionsmuster, das auf eine erhöhte Osteoinduktion schließen lässt. Es zeigte sich darüber hinaus eine verstärkte Zelladhäsion sowie eine Steigerung des osteoinduktiven Effekts auf Substraten, bei denen bFn über eine Streptavidin-Monolage immobilisiert wurde, gegenüber mit nativem Fibronektin (Fn) modifizierten TiOX-Oberflächen. Ein wesentlicher Schwerpunkt bestand daher in der Analyse der Zusammensetzung und Struktur der biomimetischen Beschichtung über „Surface Plasmon Spectroscopy“ und „Atomic Force Microscopy“. Diese ergab, dass bFn und natives Fn auf den jeweiligen Oberflächen eine unterschiedliche Konformation einnimmt. Im Gegensatz zu nativem Fn, das bei der Adsorption unter physiologischen Bedingungen auf TiOX-Oberflächen eine kompakte Konformation besitzt, nimmt bFn auf einer Streptavidin-Monolage eine entfaltete Konformation ein. Bei letzterer handelt es sich um dieselbe, welche Fn in vivo innerhalb der extrazellulären Matrix besitzt. Sie unterscheidet sich von der kompakten Fn-Konformation dahingehend, dass entlang der Fn-Achse weitere Proteinbindestellen zugänglich werden und hierdurch die Zellaffinität von Fn gesteigert wird. Die nachgewiesene Konformationsänderung kann somit als Grund für die gesteigerte Osteoblasten-Adhäsion und Aktivität auf Oberflächen mit bFn angenommen werden. Diese Kenntnisse konnten weiterhin für die Optimierung des biomimetischen Schichtsystems genutzt werden. So war es möglich, durch alternierendes Inkubieren der Biotin-aktivierten Oberfläche mit Streptavidin und bFn, ein Multilayersystem gezielt aufzubauen. Der Vorteil dieses Multilayersystems gegenüber einer einfachen Monolage aus bFn besteht in einer erhöhten Stabilität der biomimetischen Beschichtung, wodurch eine Anwendung in der Praxis erleichtert würde.

Titanium oxide is an important semiconductor, which is widely applied for solar cells. In this research, titanium oxide nanotube arrays were synthesized by anodization of Ti foil in the electrolyte composed of ethylene glycol containing 2 vol % H2O and 0.3 wt % NH4F. The voltages of 40V-50V were employed for the anodizing process. Pore diameters and lengths of the TiO2 nanotubes were evaluated by field emission scanning electron microscope (FESEM). The obtained highly-ordered titanium nanotube arrays were exploited to fabricate photoelectrode for the Dye-sensitized solar cells (DSSCS). The TiO2 nanotubes based DSSCS exhibited an excellent performance with a high short circuit current and open circuit voltage as well as a good power conversion efficiency. Those can be attributed to the high surface area and one dimensional structure of TiO2 nanotubes, which could hold a large amount of dyes to absorb light and help electron percolation process to hinder the recombination during the electrons diffusion in the electrolyte.
