The aim of this work is to analyze the short-story The Fall of the House of Usher by Edgar Allan Poe, and two of its versions into Portuguese: A Queda da Casa de Uhser, a translation by Brenno Silveira, published in 1959 by Abril, and the adaptation by Clarice Lispector, with the same title, published in 1975 by Ediouro. The original short-story was first published in 1840, in Tales of the Grotesque and Arabesque, translated into Portuguese as Histórias Extraordinárias. In this work, we will identify the differences between a translation and an adaptation, as well as the construction of the grotesque universe in those different texts. By means of a comparative analysis from the selected corpus, we will deploy the ways each one constructs the effect wanted by Poe, highlighting the main semantic and structural differences present in the translation and the adaptation of the short-story in relation with the original. Lastly, we will try to show the losses and gains of an adaptation

β-(1→3)-Glucanases were produced by Trichoderma harzianum Rifai PAMB-86 cultivated on botryosphaeran in a bench-fermenter and optimised by the response surface method. Maximal enzyme titres occurred at 5 days, initial pH 5.5 and aeration of 1.5vvm. β-(1→3)-The β-glucanolytic enzyme complex produced by T. harzianum Rifai PAMB- 86 was fractionated by gel filtration into 2 fractions (F-I, F-II), and employed to produce gluco-oligosaccharides from algal paramylon ((1→3)-β-D-glucan) and lichen pustulan ((1→6)-β-D-glucan). Both enzymes attacked paramylon to the extent of ~15-20% in 30 min releasing glucose and laminaribiose as major end-products, and laminarioligosaccharides of degree of polymerization (DP) ≥3. Only F-I degraded pustulan resulting in ~2% degradation at 30 min, with glucose, gentiobiose and gentio-oligosaccharides of DP ≥4 as major products. The difference in the nature of the hydrolysis products can be explained by the substrate specificities of each enzyme fraction, and the structural differences of the β-D-glucans attacked.

Angiosperm and gymnosperm plants evolved from a common ancestor about 300 million years ago. Apart from morphological and structural differences in embryogenesis and seed origin, a set of embryogenesis-regulating genes and the molecular mechanisms involved in embryo development seem to have been conserved alike in both taxa. Few studies have covered molecular aspects of embryogenesis in the Brazilian pine, the only economically important native conifer in Brazil. Thus eight embryogenesis-regulating genes, viz.,ARGONAUTE 1, CUP-SHAPED COTYLEDON 1, WUSCHEL-related WOX, S-LOCUS LECTIN PROTEIN KINASE, SCARECROW-like, VICILIN 7S, LEAFY COTYLEDON 1, and REVERSIBLE GLYCOSYLATED POLYPEPTIDE 1, were analyzed through semi-quantitative RT-PCR during embryo development and germination. All the eight were found to be differentially expressed in the various developmental stages of zygotic embryos, seeds and seedling tissues. To our knowledge, this is the first report on embryogenesis-regulating gene expression in members of the Araucariaceae family, as well as in plants with recalcitrant seeds.

Cathepsin L-like proteinases (CAL) are major digestive proteinases in the beetle Tenebrio molitor. Procathepsin Ls 2 (pCAL2) and 3 (pCAL3) were expressed as recombinant proteins in Escherichia coil, purified and activated under acidic conditions. Immunoblot analyses of different T. molitor larval tissues demonstrated that a polyclonal antibody to pCAL3 recognized pCAL3 and cathepsin L 3 (CAD) only in the anterior two-thirds of midgut tissue and midgut luminal contents of T. molitor larvae. Furthermore, immunocytolocalization data indicated that pCAL3 occurs in secretory vesicles and microvilli in anterior midgut Therefore CAL3, like cathepsin L 2 (CAL2), is a digestive enzyme secreted by T. molitor anterior midgut CAD hydrolyses Z-FR-MCA and Z-RR-MCA (typical cathepsin substrates), whereas CAL2 hydrolyses only Z-FR-MCA. Active site mutants (pCAL2C25S and pCAL3C265) were constructed by replacing the catalytic cysteine with serine to prevent autocatalytic processing. Recombinant pCAL2 and pCAL3 mutants (pCAL2C25S and pCAL3C26S) were prepared, crystallized and their 3D structures determined at 1.85 and 2.1 angstrom, respectively. While the overall structure of these enzymes is similar to other members of the papain superfamily, structural differences in the S2 subsite explain their substrate specificities. The data also supported models for CAL trafficking to lysosomes and to secretory vesicles to be discharged into midgut contents. (C) 2012 Elsevier Ltd. All rights reserved.

O músculo diafragma, encontrado apenas nos mamíferos, é o principal músculo no processo respiratório, servindo de fronteira entre as cavidades torácica e abdominal. Sua importância também ganha destaque em pesquisas realizadas no âmbito dos enxertos, empregando-se diversos tipos de membranas biológicas para o reparo de defeitos diafragmáticos, os quais podem gerar hérnias diafragmáticas. Apesar de muitos estudos já conduzidos para com os primatas não humanos, especialmente no que tange a espécie do novo mundo Callithrix jacchus (Sagui-de-tufo-branco), oriundo do nordeste brasileiro, as pesquisas envolvendo o uso do diafragma em tal espécie é inexistente. Deste modo objetivou-se caracterizar a morfologia e a biometria do diafragma na espécie Callithrix jacchus de ambos os sexos, analisando possíveis divergências estruturais entre machos e fêmeas. Para tal foram utilizados quatros animais, 2 machos e 2 fêmeas, adultos, que vieram a óbito por causas naturais, provenientes de um criadouro comercial. Após fixação em solução de formaldeído 10% os animais foram devidamente dissecados para fotodocumentação e em seguida o diafragma coletado para efetuação da biometria (comprimento e largura) com o uso de um paquímetro e para o processamento histológico por meio da coloração de hematoxilina-eosina e tricrômio de masson, da porção muscular. As mensurações feitas permitiram concluir que não houve diferenças signifcativas entre machos e femeas. A topografia e a presença de três aberturas (forame da veia cava caudal, hiato aórtico e esofágico) na extensão do diafragma corroboram com descrições na literatura classica para outros mamíferos. A presença de um centro tendíneo em "V" difere do encontrado para animais como o peixe-boi e porquinho-da-india, mas é similar ao encontrado para o gambá-de-orelhas-brancas e rato albino. No que diz respeito aos achados histológicos conclui-se que as fibras musculares estão dispostas de forma organizada, apresentam diâmetro grande e núcleos basais, tendo, portanto, características similares do músculo estriado esquelético tanto nos animais machos como nas fêmeas.

This work reports the investigation on the structural differences between InAs quantum rings and their precursor quantum dots species as well as on the presence of piezoelectric fields and asymmetries in these nanostructures. The experimental results show significant reduction in the ring dimensions when the sizes of capped and uncapped ring and dot samples are compared. The iso-lattice parameter mapped by grazing-incidence x-ray diffraction has revealed the lateral extent of strained regions in the buried rings. A comparison between strain and composition of dot and ring structures allows inferring on how the ring formation and its final configuration may affect optical response parameters. Based on the experimental observations, a discussion has been introduced on the effective potential profile to emulate theoretically the ring-shape confinement. The effects of confinement and strain field modulation on electron and hole band structures are simulated by a multiband k.p calculation. (C) 2012 American Institute of Physics. Structural differences such as abnormalities, damage and free spaces in seeds may affect germination. The aim of this study was to study the relationship between eggplant seed morphology and seed germination. Ten seed lots of the eggplant cultivar Embu were evaluated by X-ray image analysis and the germination test. Seed image analysis was performed by Image Pro Plus® software and the whole seed area and free space between the embryo and endosperm were measured. The internal seed area filled by the embryo and endosperm was calculated from the difference between the whole seed and free space areas. Based on these results and visual seed analysis, seeds were classified into three categories and information on germination was obtained for each one. X-ray image analysis provides a perfect view of the internal seed parts and for seed morphology studies. An increase in seed area filled by the endosperm and embryo does not improve seed germination. Mechanical seed damage and deteriorated tissues can adversely affect seed germination.

[EN] Global warming is affecting all major ecosystems, including temperate reefs where canopy-forming seaweeds provide biogenic habitat. In contrast to the rapidly growing recognition of how climate affects the performance and distribution of individuals and populations, relatively little is known about possible links between climate and biogenic habitat structure. We examined the relationship between several ocean temperature characteristics, expressed on time-scales of days, months and years, on habitat patch characteristics on 24 subtidal temperate reefs along a latitudinal gradient (Western Australia; ca 34 to 27º S). Significant climate related variation in habitat structure was observed, even though the landscape cover of kelp and fucalean canopies did not change across the climate gradient: monospecific patches of kelp became increasingly dominant in warmer climates, at the expense of mixed kelp-fucalean canopies. The decline in mixed canopies was associated with an increase in the abundance of Sargassum spp., replacing a more diverse canopy assemblage of Scytothalia doryocarpa and several other large fucoids. There were no observed differences in the proportion of open gaps or gap characteristics. These habitat changes were closely related to patterns in minimum temperatures and temperature thresholds (days > 20 °C), presumably because temperate algae require cool periods for successful reproduction and recruitment (even if the adults can survive warmer temperatures). Although the observed habitat variation may appear subtle, similar structural differences have been linked to a range of effects on canopy-associated organisms through the provision of habitat and ecosystem engineering. Consequently, our study suggests that the magnitude of projected temperature increase is likely to cause changes in habitat structure and thereby indirectly affect numerous habitat-dependent plants and animals

Molekulardynamik-Simulationen zu OberflächeneigenschaftenvonSiliziumdioxid-Schmelzen In der vorliegenden Arbeit werdenMolekulardynamik-Computersimulationenzur Untersuchung der statischen und dynamischenOberflächeneigenschafteneiner Siliziumdioxid (SiO$_2$)-Schmelze durchgeführt.Als Modellpotential verwenden wirein in der Literatur als BKS-Potential bezeichnetesPaarpotential.Wir betrachten in dieser Arbeit zwei Systemgeometrien: zumeinenSysteme aus 432, 1536 und 4608 Atomen ohne periodischeRandbedingungen(Tropfen), zum anderen ein System aus 1152 Atomen mitperiodischen Randbedingungen in zwei Richtungen (dünnerFilm).Für beide Geometrien finden wir im Inneren der Systemekonstante Dichten.Direktan der Oberfläche halten sich ausschließlich Sauerstoffatomeauf.Die Strukturan der Oberfläche erscheint weniger ausgeprägt mit mehrDefekten, als im Inneren des Systems.Es erweist sich,daß viele Eigenschaften statischer Größen, die wir an derOberfläche im Vergleich zum Inneren finden, aufdas Vorhandensein von Zweierringen zurückzuführen sind.Bei der Betrachtung der dynamischen Größen finden wir eineerhöhteBeweglichkeit der Atome an der Oberfläche gegenüber demSysteminneren.

Das Membranprotein LHCII ist ein Lichtsammelkomplex der höheren Pflanzen, der in vitro, ausgehend von bakteriell überexprimiertem Apoprotein, als Monomer und als Trimer rekonstituiert werden kann. Um Strukturunterschiede zwischen Monomeren und Trimeren zu bestimmen, wurden ortsspezifische Derivatisierungen des Proteins durchgeführt. Dazu wurden verschiedene Mutationen am LHCII vorgenommen. Das einzige Cystein des nativen, maturen LHCII wurde zunächst in ein Serin umgewandelt. Ausgehend von dieser Mutante wurden an fünf Positionen singuläre Cysteine eingefügt. Zugänglichkeitsuntersuchungen mit dem thiolreaktiven Farbstoff Rhodamine Red-Maleimid zeigten zum Teil Unterschiede zwischen Monomeren und Trimeren auf. Außerdem deutete eine zweiphasige Markierungskinetik eines der rekombinanten LHCII auf mindestens zwei konformelle Populationen in Detergenslösung. Die Beobachtungen dieser Arbeit wurden zudem genutzt, um im Strukturmodell des LHCII unklare Positionen näher zu beschreiben. Schließlich wurden einige der LHCII mit angekoppeltem Fluoreszenzfarbstoff spektroskopisch charakterisiert.
