Prokaryotes represent one-half of the living biomass on Earth, with the vast majority remaining elusive to culture and study within the laboratory. As a result, we lack a basic understanding of the functions that many species perform in the natural world. To address this issue, we developed complementary population and single-cell stable isotope (C-13)-linked analyses to determine microbial identity and function in situ. We demonstrated that the use of rRNA/mRNA stable isotope probing (SIP) recovered the key phylogenetic and functional RNAs. This was followed by single-cell physiological analyses of these populations to determine and quantify in situ functions within an aerobic naphthalene-degrading groundwater microbial community. Using these culture-independent approaches, we identified three prokaryote species capable of naphthalene biodegradation within the groundwater system: two taxa were isolated in the laboratory (Pseudomonas fluorescens and Pseudomonas putida), whereas the third eluded culture (an Acidovorax sp.). Using parallel population and single-cell stable isotope technologies, we were able to identify an unculturable Acidovorax sp. which played the key role in naphthalene biodegradation in situ, rather than the culturable naphthalene-biodegrading Pseudomonas sp. isolated from the same groundwater. The Pseudomonas isolates actively degraded naphthalene only at naphthalene concentrations higher than 30 mu M. This study demonstrated that unculturable microorganisms could play important roles in biodegradation in the ecosystem. It also showed that the combined RNA SIP-Raman-fluorescence in situ hybridization approach may be a significant tool in resolving ecology, functionality, and niche specialization within the unculturable fraction of organisms residing in the natural environment.

Rhizosphere microorganisms play an important role in soil carbon flow, through turnover of root exudates, but there is little information on which organisms are actively involved or on the influence of environmental conditions on active communities. In this study, a (CO2)-C-13 pulse labelling field experiment was performed in an upland grassland soil, followed by RNA-stable isotope probing (SIP) analysis, to determine the effect of liming on the structure of the rhizosphere microbial community metabolizing root exudates. The lower limit of detection for SIP was determined in soil samples inoculated with a range of concentrations of C-13-labelled Pseudomonas fluorescens and was found to lie between 10(5) and 10(6) cells per gram of soil. The technique was capable of detecting microbial communities actively assimilating root exudates derived from recent photo-assimilate in the field. Denaturing gradient gel electrophoresis (DGGE) profiles of bacteria, archaea and fungi derived from fractions obtained from caesium trifluoroacetate (CsTFA) density gradient ultracentrifugation indicated that active communities in limed soils were more complex than those in unlimed soils and were more active in utilization of recently exuded C-13 compounds. In limed soils, the majority of the community detected by standard RNA-DGGE analysis appeared to be utilizing root exudates. In unlimed soils, DGGE profiles from C-12 and C-13 RNA fractions differed, suggesting that a proportion of the active community was utilizing other sources of organic carbon. These differences may reflect differences in the amount of root exudation under the different conditions.

The microbial contribution to soil organic matter (SOM) has recently been shown to be much larger than previously thought and thus its role in carbon sequestration may also be underestimated. In this study we employ C-13 ((CO2)-C-13) to assess the potential CO2 sequestration capacity of soil chemoautotrophic bacteria and combine nuclear magnetic resonance (NMR) with stable isotope probing (SIP), techniques that independently make use of the isotopic enrichment of soil microbial biomass. In this way molecular information generated from NMR is linked with identification of microbes responsible for carbon capture. A mathematical model is developed to determine real-time CO2 flux so that net sequestration can be calculated. Twenty-eight groups of bacteria showing close homologies with existing species were identified. Surprisingly, Ralstonia eutropha was the dominant group. Through NMR we observed the formation of lipids, carbohydrates, and proteins produced directly from CO2 utilized by microbial biomass. The component of SOM directly associated with CO2 capture was calculated at 2.86 mg C (89.21 mg kg(-1)) after 48 h. This approach can,differentiate between SOM derived through microbial uptake of CO2 and other SOM constituents and represents a first step in tracking the fate and dynamics of microbial biomass in soil.

Objectives: This study aimed to gather data on the nutritional status of older patients attending Cork University Dental School and Hospital for treatment in the Restorative Department. Information was also collected about the medical status of the patients including the prevalence of self-reported xerostomia.
Methods: Data was collected by a self-completion questionnaire followed by a brief clinical examination. Nutritional Status was measured using the short version of the Mini Nutritional Assessment (MNA) which recorded patients’ Body Mass Index (BMI). The MNA consists of 6 parameters (including questions relating to patients’ history and anthropometric data) with a maximum total of 14 points. Scores of 12-14 indicate “normal nutritional status” whilst those between 8 and 11 indicate a patient “at risk of malnutrition”. Scores lower than 8 are an indication a patient who is “malnourished”. All patients attending Cork University Dental School and Hospital aged 65 years and older were invited to participate in the study.

Results: A total of 22 subjects participated in this study. Twelve patients were partially dentate with 10 edentulous. The results from the MNA indicate that 11 patients were of “normal nutritional status” with 11 patients identified as being “at risk of malnutrition”. None of the subjects were “malnourished”. Edentate patients generally recorded lower MNA scores than partially dentate patients. In total, 9 patients reported experiencing xerostomia with 8 indicating that they needed to sip liquids to aid swallowing but only 3 had difficulty swallowing food.

Conclusion: This small study indicates that a number of the older patients attending Cork University Dental School and Hospital for dental care may be “at risk of malnutrition”. These findings suggest that nutritional advice and dental care should both be included in an overall package of care for older patients.

In this case, an individual was suspected of attempting to burn materials potentially relating to a murder case. A number of spent and unspent matches were seized at the scene by police for forensic examination. Coincidentally, a police raid at the suspect's house revealed a number of matchboxes, all of the same brand, containing matches that had a visual similarity to those recovered at the scene. Stable Isotope Profiling (SIP) was used to assess whether matches could either be distinguished or shown to be indistinguishable by 13C and 2H isotopic composition. These results were then compared to those from the X-ray diffraction (XRD) analysis of match heads and microscopy of the wood. SIP showed the scene matches and seized matches to be different, which was confirmed by XRD and microscopy analyses.

A evolução observada nas redes de comunicações durante a última década traduziu-se na diversificação de serviços que utilizam a rede, no aumento das taxas de transferência e na massificação da utilização de serviços de acesso à Internet e de comunicações celulares. Durante esta década, várias organizações, das quais se destacam os operadores de telecomunicações, têm dedicado consideráveis esforços no sentido de definir e normalizar arquitecturas de redes de próxima geração. A principal característica deste tipo de rede reside no facto de possuir uma arquitectura modular capaz de fornecer serviços multimédia a clientes de uma rede de acesso com características tecnológicas heterogéneas. Os trabalhos de normalização das arquitecturas de rede NGN têm-se limitado, até ao momento, a especificar detalhes relativos ao funcionamento da rede não tendo ainda sido definida a arquitectura de gestão. Em termos de tecnologias de gestão de redes, foram propostos nas últimas duas décadas novos paradigmas de gestão, novos modelos de dados, novos protocolos de transporte e várias linguagens de definição de informação de gestão. Os modelos de dados têm vindo a ser enriquecidos, os protocolos são mais flexíveis e poderosos, as soluções de gestão oferecem interoperabilidade acrescida e as linguagens permitem definir formatos de configuração mais ricos. Simultaneamente tem crescido a complexidade das soluções de gestão, aumentado a sobrecarga causada pelo aumento de complexidade nos equipamentos bem como nas plataformas computacionais que suportam os sistemas de gestão. O presente trabalho propõe uma solução de gestão para redes NGN capaz de gerir os recursos de rede garantindo Qualidade de Serviço. A solução de gestão proposta inclui uma plataforma de execução de políticas que utiliza os eventos ocorridos na rede para empreender acções de configuração, autonomizando o processo de gestão. Inclui uma avaliação da complexidade de várias tecnologias de gestão estudando a sobrecarga causada pela tecnologia tanto no processo de gestão como na operação da rede. É ainda estudada a escalabilidade das várias tecnologias e analisado o seu comportamento num cenário da rede de um operador de telecomunicações. O trabalho propõe ainda uma metodologia de configuração integrada dos elementos de gestão, através de uma interface de configuração amigável para o administrador do sistema.

In this paper we consider the uplink transmission within CP-assisted (Cyclic Pre¯x) DS-CDMA (Direct Sequence Code Division Multiple Access) systems and we present a frequency-domain MUD (MultiUser Detection) receiver with iterative estimation and compensation of residual frequency errors. The proposed receiver is suitable for broadband wireless systems, with performances that can be close to the single-user MFB (Matched Filter Bound), even for fully loaded systems and/or in the presence of strong interfering signals. The receiver is powerful enough for typical asynchronous scenarios, requiring only a coarse synchronization between users.

In this paper, we consider low-PMEPR (Peak-to-Mean Envelope Power Ratio) MC-CDMA (Multicarrier Coded Division Multiple Access) schemes. We develop frequencydomain turbo equalizers combined with an iterative estimation and cancellation of nonlinear distortion effects. Our receivers have relatively low complexity, since they allow FFT-based (Fast Fourier Transform) implementations. The proposed turbo receivers allow significant performance improvements at low and moderate SNR (Signal-to-Noise Ratio), even when a low-PMEPR MC-CDMA transmission is intended.

Na presente dissertação é proposto o desenvolvimento de um novo sistema de calibração de roscados de exteriores através de visão computacional. A calibração de roscados de exterior consiste na obtenção do diâmetro efectivo, do diâmetro exterior e do passo, e no cálculo da incerteza expandida correspondente. Actualmente, a calibração é efectuada com o auxílio de máquinas universais (SIP), na qual o diâmetro efectivo é obtido através de um modelo matemático, pois não se consegue obtê-lo directamente. O sistema de calibração por visão computacional tem como objectivo obter-se o diâmetro efectivo directamente, assim como as restantes características. A vantagem deste novo sistema será para roscados com dimensões inferiores a 2 mm, que não se conseguem medir utilizando a SIP. A desvantagem é referente a diâmetros superiores a 2 mm, devido à resolução obtida com a câmara utilizada. Este sistema foi validado por comparação com a calibração utilizando como equipamento calibrador a SIP. Ao longo da dissertação irão ser explicados todos os passos dados para a calibração de roscados de exterior.

House Finches (CarpQdacqs mexiCAnuS) were introduced to Long Island, New York from southern'California in 1940. Apparently, an initial sample of less than 100 birds has given rise to a population that now occupies much of the eastern United States. This study was to determine if morphological and reproductive changes have taken place in introduced eastern birds, which have colonized a novel environment. A study area in Goleta, California (CAL) represented the parental population whereas for comparison, House Finches in St. Catharines, Ontario (ONT) represented the introduced population. Interlocality variation in 25 morphometric characters of 100 adult House Finches was examined statistically. Singleclassification analysis of variance revealed significant interlocality differentiation in seven characters of males and nine of females. Females showed differentiation in more limb elements than males. Analysis of character variation using discriminant and principal component analysis distinguished samples on the basis of variation in shape. Compared to CAL, aNT birds (especially females) had smaller extremities relative to certain core parts and weight. Females showed similar patterns of character covariation in each locality on the second principal component, which suggests that differentiation of the ONT population may not be solely environmentally induced. Sexual dimorphism was evident in four charaoters in aNT and five in CAL. Disoriminant analysis distinguished sex on the basis of variation in shape. Males possessed a relatively larger flying apparatus and small.er hind limbs than females. The dearee of sexual dimorphism did not vary sicnifioantly between looalities. 3 Data on reproduotive parameters were oolleoted in 1983 and 1984 in ONT, and 1984 in CAL. In 1984, Bouse Finohes began breedina approximately three months earlier in CAL than in ONT. In ONT, there was no sianifioant differenoe in mean olutoh initiation date between 1983 and 1984. In both looalities most nests oontained either four or five ea",s, and olutoh size differenoes between looalites were not signifioant. Seasonal deolines in olutch size were evident in ONT but not in CAL. Intralooality variation in e.g weight and size was not related to clutch size. E",g weiaht showed no seasonal trend in ONT, but inoreased sianifioantly with breed ina season in OAL. In both looalities e8'''' weiaht increased sipifioantly with order of layina in olutohes of four but not in clutohes of five. Eag's in ONT in 1983 and 1984 were sip.ificantly larser than in CAL in 1984. The modal inoubation period was 13 days and did not vary sip.ifioantly between localites. In both looalities nestling weiaht on the day of hatohing was oorrelated to fresh ega welaht. For muoh of the period between hatohing and 14 days post-hatoh, ONT nestlinas were signifioantly laraer than CAL nestlings in terms of weiaht. bill length, bill depth, and manus length.

L’anémie est fréquente chez les patients pédiatriques en postopératoire de chirurgie cardiaque. Malgré le nombre important de patients transfusés, le taux d’hémoglobine (Hb) pour lequel les bénéfices surpassent les risques est inconnu chez ces patients. Récemment, Lacroix et al. ont démontré qu’une stratégie transfusionnelle restrictive n’était pas inférieure à une stratégie libérale en ce qui concerne le développement ou la progression du syndrome de défaillance multiviscérale (SDMV) et la mortalité chez les patients de soins intensifs pédiatriques (SIP).Devant le manque d’évidence, une analyse de sous-groupes des patients en postopératoire de chirurgie cardiaque de l’étude Transfusion Requirements in Pediatric Intensive Care (TRIPICU) a été réalisée. L’objectif de cette étude était de déterminer l’impact d’une stratégie transfusionnelle restrictive comparée à une stratégie libérale sur l’acquisition ou l’aggravation du syndrome de défaillance multiviscérale (SDMV) chez les enfants en postopératoire de chirurgie cardiaque. Cette étude n’a pas démontré de différences statistiquement, ni cliniquement significatives du nombre de patients ayant acquis ou aggravés un SDMV, ni des issues secondaires entre les stratégies transfusionnelles restrictive et libérale. L’analyse de sous-groupes permet de générer une hypothèse de recherche et les résultats devraient être confirmés par un essai randomisé contrôlé.
