Background: Fumonisins produced by Fusarium verticillioides are among the most important medical mycotoxins known. The intake of concentrate based on corn and corn by-products contaminated with fumonisins can cause severe poisoning in horses. The injuries are observed mainly in the white matter of the brain, and the disease is known as Equine Leukoencephalomalacia (ELEM). This study aims to describe and discuss the epidemiological, clinical and diagnostic aspects of an outbreak of ELEM occurred in three farms in the municipalities of Canarana and Agua Boa, in the eastern region of Mato Grosso, Brazil.Materials, Methods & Results: The outbreak occurred between May and August 2010. The disease affected six horses and four mules of different ages and sex. Clinical examination was only possible in animals with chronic evolution of the disease. All the affected animals showed neurological clinical signs such as ataxia and recumbency, which progressed to death or sudden death. Histopathological analysis showed foci of necrosis that predominantly affected the white matter, and the presence of gitter cells. Degenerative lesions were observed in the liver of the animals. Mortality rate ranged from 12.5 to 71%, and lethality reached 100%. The cases were preceded by sudden drops in the weather temperature. Fumonisins levels of 6.6 ppm were detected in the feed of the animals.Discussion: The presumptive diagnosis of leukoencephalomalacia was consistent and based on clinical and epidemiological studies. However, the definitive diagnosis was based upon the histological features of the brain including the presence of extensive areas of malacia. Moreover, the animals were being fed with corn or corn by-products contaminated with fumonisins levels considered to be toxic to equids. The mortality and lethality rates are in agreement with outbreaks described in previous studies. The animals showed neurological signs as the predominant clinical manifestation, with gait ataxia followed by recumbency, prostration and death between 24 h and 29 days. Similarly to other reports, the disease was more frequent in adult animals, which succumbed in 24-48 h. Conversely, the evolution of the disease in young animals was of 10 to 29 days. Sudden death was more prevalent in the mules. Previous studies have shown a predominance of cerebral and brainstem lesions in horses, whereas in mules the clinical signs are related to brainstem lesions. Corn and corn by-products are commonly used as energy supplementation to horses in the southern of Mato Grosso state, but outbreaks of the disease are uncommon. This may be influenced by the prevalence of hot climate conditions, which does not favor the production of toxin by the fungus. The atypical low weather temperatures (9-11 degrees C) observed prior to the outbreak could have contributed to the mycotoxin production by F. verticillioides, which requires temperatures between 8-12 degrees C to produce toxins. However, the disease in the region may be underestimated, considering that the practice of necropsies is not common among field technicians, mainly in the occurrence of sporadic deaths among horses intended for work. Preventive measures include avoiding the use of corn and corn by-products for horses after periods of sudden drops in temperature in the region. Furthermore, clinical and epidemiological surveys and post-mortem and histopathological analyses are undoubtedly important for appropriate differential diagnosis, especially in equids with neurological signs.

As micotoxinas são metabólitos secundários produzidos por fungos. Com o intuito de aumentar a produção leiteira, cada vez mais usam-se alimentos ricos em grãos na dieta de bovinos, o que eleva significativamente o risco de exposição às micotoxinas. As condições climáticas brasileiras favorecem o desenvolvimento de fungos e a produção de micotoxinas. As micotoxinas ingeridas pelos bovinos podem ser transferidas para o leite que, ao ser consumido, causará a contaminação do homem. Este é um fator preocupante, pois o leite é um alimento altamente consumido em todas as fases da vida e é extremamente importante para o desenvolvimento de crianças. O Brasil é o 3º maior produtor mundial de leite, com a intenção de se colocar no mercado externo. Com base nesses fatos, o objetivo deste trabalho foi levantar informações a respeitos das principais micotoxinas encontradas no leite, seja este cru, refrigerado ou esterilizado. Para tanto, foi realizado um levantamento bibliográfico em três bases de dados, utilizando-se os seguintes descritores: (mycotoxins OR micotoxinas) AND (milk OR leite). Foram selecionados 10 trabalhos que realizaram quantificação de micotoxinas no leite, sendo concluído que as micotoxinas de maior importância no leite são as aflatoxinas, que representam um grande risco à saúde humana e animal, sendo importante a prevenção dos fungos produtores da mesma.

Aflatoxin M-1 (AFM(1)) is a hepatocarcinogen found in milk of animals that have consumed feeds with aflatoxin B-1. The carry-over of AFM(1) from milk to Minas Frescal cheese produced with or without starter cultures was determined. 40 L of milk were divided into 10 L each and assigned to the following treatments for cheese manufacture: 0.250 rig AFM(1) mL(-1), 0.500 rig AFM(1) mL(-1), 0.250 ng AFM(1) mL(-1) + starter, 0.500 ng AFM(1) mL(-1) + starter. Quantification of AFM(1) was achieved by high performance liquid chromatography. The carry-over of AFM(1) from milk to cheese ranged from 30.64% to 42.26%. There was no effect of storage time on AFM(1). Milk with AFM(1) in levels studied may concentrate the toxin in Minas Frescal cheese, but at concentrations below the Brazilian tolerance limit. The addition of starter cultures did not influence concentration or stability of the AFM(1) in cheese over 30 days storage. (C) 2011 Elsevier Ltd. All rights reserved.

This study investigated the ability of weevils to transmit Aspergillus flavus and Fusarium verticillioides fungal spores and the consequent production of mycotoxins. For this purpose, corn grain samples were stored in flasks connected to a hose to form a closed system (flasks A and B). Flasks A were inoculated with the following groups: group 1 (corn + weevil); group 2 (corn + A. flavus); group 3 (corn + A. flavus + weevil); group 4 (corn + F. verticillioides); group 5 (corn + E verticillioides + weevil), and group 6 (corn + A. flavus + E verticillioides + weevil). Flasks B contained sterile grains. The samples were incubated for 10, 20 and 30 days, posteriorly, weight, water activity, mycoflora, aflatoxins and fumonisins. The corn grain samples were also submitted to scanning electron microscopy. Our results showed that weevils could enhance corn grains contamination by these fungi, hence, could increase mycotoxins production. These findings demonstrate the importance of weevils as fungal vectors and the need for good manipulation and storage practices of grains. (C) 2012 Elsevier Ltd. All rights reserved.

The occurrence of ochratoxin A (OTA) in wine from 2002 to 2008 harvest, traded in Rio de Janeiro State, was evaluated by analysing 43 national and 37 imported wines from Argentina (32) and Chile (5), adding up to 80 samples in total. OTA determination was performed using immunoaffinity columns and high-performance liquid chromatography. In 80 wine samples analysed, 25 (31.3%) were positive, presenting levels greater than 0.020 ng OTA mL(-1). It was not detected in imported wines. Within national wines, 58.1% of the samples were contaminated, with levels ranging from 0.020 to 0.050 ng mL(-1). The toxin was detected in 18 (69.2%) of 26 samples analysed of red table wine. Wines from 2008 harvest presented 84.6% of samples contaminated in 13 samples analysed. Despite the levels found in this study, they are below Brazilian tolerance limits. Nevertheless, the presence of OTA as found contributes to the human exposure to this toxin.

Brazil nuts are an important export market in its main producing countries, including Brazil, Bolivia, and Peru. Approximately 30,000 tons of Brazil nuts are harvested each year. However, substantial nut contamination by Aspergillus section Flavi occurs with subsequent production of aflatoxins. In our study, Aspergillus section Flavi were isolated from Brazil nuts (Bertholletia excelsa), and identified by morphological and molecular means. We obtained 241 isolates from nut samples, 41% positive for aflatoxin production. Eighty-one isolates were selected for molecular investigation. Pairwise genetic distances among isolates and phylogenetic relationships were assessed. The following Aspergillus species were identified: A. flavus, A. caelatus, A. nomius, A. tamarii, A. bombycis, and A. arachidicola. Additionally, molecular profiles indicated a high level of nucleotide variation within beta-tubulin and calmodulin gene sequences associated with high genetic divergence from RAPD data. Among the 81 isolates analyzed by molecular means, three of them were phylogenetically distinct from all other isolates representing the six species of section Flavi. A putative novel species was identified based on molecular profiles.

The mycotoxin aflatoxin B1 (AFB1) is a carcinogenic food contaminant which is metabolically activated by epoxydation. The metabolism of mycotoxins via the mercapturate metabolic pathway was shown, in general, to lead to their detoxication. Mercapturic acids thus formed (S-substitued-N-acetyl-L-cysteines) may be accumulated in the kidney and either excreted in the urine or desacetylated by Acylase 1 (ACY1) to yield cysteine S-conjugates. To be toxic, the N-acetyl-L-cysteine-S-conjugates first have to undergo deacetylation by ACY 1. The specificity and rate of mercapturic acid deacetylation may determine the toxicity, however the exact deacetylation processes involved are not well known. The aim of this study was to investigate the role of ACY1 in the toxicity of some bioactive epoxides from Aflatoxin B1. We characterized the kinetic parameters of porcine kidney and human recombinant aminoacylase-1 towards some aromatic and aliphatic-derived mercapturates analogue of mycotoxin mercapturic acids and 3,4-epoxyprecocene, a bioactive epoxide derivated from aflatoxin. The deacetylation of mercapturated substrates was followed both by reverse phase HPLC and by TNBS method. Catalytic activity was discussed in a structure function relationship. Ours results indicate for the first time that aminoacylase-1 could play an important role in deacetylating mercapturate metabolites of aflatoxin analogues and this process may be in relation with their cyto- and nephrotoxicity in human. This study evaluated the presence of fungi and mycotoxins [aflatoxins (AFs), cyclopiazonic acid (CPA), and aspergillic acid] in stored samples of peanut cultivar Runner IAC Caiapó and cultivar Runner IAC 886 during 6 months. A total of 70 pod and 70 kernel samples were directly seeded onto Aspergillus flavus and Aspergillus parasiticus agar for fungi isolation and aspergillic acid detection, and AFs and CPA were analyzed by high-performance liquid chromatography. The results showed the predominance of Aspergillus section Flavi strains, Aspergillus section Nigri strains, Fusarium spp., Penicillium spp. and Rhizopus spp. from both peanut cultivars. AFs were detected in 11.4% of kernel samples of the two cultivars and in 5.7% and 8.6% of pod samples of the Caiapó and 886 cultivars, respectively. CPA was detected in 60.0% and 74.3% of kernel samples of the Caiapó and 886 cultivars, respectively. Co-occurrence of both mycotoxins was observed in 11.4% of kernel samples of the two cultivars. These results indicate a potential risk of aflatoxin production if good storage practices are not applied. In addition, the large number of samples contaminated with CPA and the simultaneous detection of AFs and CPA highlight the need to investigate factors related to the control and co-occurrence of these toxins in peanuts.
