Sweet orange is considered a very important species in the citrus world market and presents wide morphological variability. However, its characterization at the molecular level by random amplified polymorphic DNA (RAPD) and isozyme markers is not appropriate. Microsatellite or simple sequence repeats (SSRs) have been suggested as ideal for studies in cultures of vegetative propagation and as value markers for mapping in several species. However, information on microsatellite polymorphism in citrus species is scarce. In this work, microsatellite markers (AG-repeats) were developed from an enrichment library of genomic DNA of sweet orange cv. Pera (Citrus sinensis [L.] Osbeck), and 31 cultivars of sweet orange were evaluated. Evaluation of 18 microsatellite primers did not permit differentiation of the varieties studied. New microsatellite primers are being evaluated with the aim of detecting polymorphisms among the cultivars and closely related species to be used in genetic mapping programs.

Objective: The aim of this study was to assess the existence of myofunctional alterations before and after first premolar extraction in Class II/1 malocclusion patients that could endanger the long-term dental arch stability. Materials and Methods: The study was performed by means of morphological, functional and electromyographic analyses in 17 Class II/1 malocclusion patients (group T) and 17 Class I malocclusion patients (group C), both groups with 12-30-year age range (mean age: 20.93 ± 4.94 years). Results: Data analyzed statistically by Student's t-test showed a significant decrease in the maxillary and mandibular dental arch perimeters after orthodontic treatment (p<0.05). The Kruskal-Wallis test analyzed data from tongue posture at rest and during swallowing, not showing significant differences after treatment (groups Tb and Ta) (p>0.05). However, group T differed significantly from group C (p<0.05). The electromyographic data showed that the anterior right and left suprahyoid muscles acted synergistically in both groups, while having a lower myoelectric activity in group T during swallowing. Conclusions: Myofunctional alterations observed after the orthodontic treatment in Class II/1 malocclusion seemed to jeopardize the long-term orthodontic stability, making recurrence possible. Further research should be conducted to compare electromyographic data before and after orthodontic treatment in order to corroborate the results of the present investigation.

The evaluation of diversity in germplasm collections is important for both plant breeders and germplasm curators to optimize the use of the variability available. Diversity can be estimated by different genetic markers. The purpose of this study was to estimate the genetic divergence of 30 morphological and agronomic traits in 108 sesame genotypes by multivariate analysis. The Cole-Rodgers index was used to establish the dissimilarity matrices. The principal component analysis identified the traits that contributed most to the divergence and the genotypes were clustered by Tocher's optimization. Despite the narrow genetic basis, the markers were efficient to characterize the genotypes and identify the most similar groups or duplicate and divergent genotypes. Greatest variation was found for the traits number of capsules per plant and grain yield.

The purpose of this in vitro study was to evaluate the effect of neodymium:yttrium-aluminum-garnet (Nd:YAG) laser irradiation on intracanal dentin surface by SEM analysis and its interference in the apical seal of filled canals. After endodontic treatment procedures, 34 maxillary human incisors were randomly assigned to 2 groups. In the negative control group (n=17), no additional treatment was performed and teeth were filled with vertically condensed gutta-percha; in the laser-treated group (n=17), the root canals were irradiated with Nd:YAG laser (1.5 W, 100 mJ, 15 Hz) before filling as described for the control group. Two specimens of each group were prepared for SEM analysis to evaluate the presence and extent of morphological changes and removal of debris; the other specimens were immersed in 0.5% methylene blue dye (pH 7.2) for 24 h for evaluation of the linear dye leakage at the apical third. SEM analysis of the laser-treated group showed dentin fusion and resolidification without smear layer or debris. The Student's t-test showed that the laser-treated group had significantly less leakage in apical third than the control group. Within the limitations of this study, it may be concluded that the morphological changes on the apical intraradicular dentin surface caused by Nd:YAG laser resulted in less linear dye apical leakage.

Introduction: Aging skin is a condition that affects (or will affect) all people, and its treatment is considered a clinical challenge. Growth factors and their analogues are emerging as a promising therapeutic option. Objectives: To evaluate the safety profile of some dermocosmetic products with formulations based on growth factors - or their analogs intended for that purpose - using in vitro human skin cell culture models. Methods: Two types of cell cultures were studied, and the effects of the study products on the proliferation of melanoma cells and normal human fibroblasts were evaluated. Results: No significant morphological alterations were found in the cultured human melanoma, and no significant decrease in the number of healthy cells was verified in the normal fibroblasts culture. In some cases there was even a proliferation of those cells. Conclusions: These preliminary data demonstrate that cosmeceutical products containing growth factors as an active principle can be considered safe for topical application.

Bats present unique features among mammals with respect to reproduction, and although neotropical bats do not have a hibernation period, many of their reproductive characteristics vary seasonally and interspecifically. Thus, this work aimed to examine the reproductive structures of 18 species belonging to five families of Brazilian bats. The gross anatomy of the testes varied little; however, the epididymis of Emballonuridae and Vespertilionidae showed exceptional structures with a large elongation of the caudal region. We observed a wide variation in the positioning of the testes: Phyllostomidae and Noctilionidae presented external testes; Emballonuridae and Molossidae presented migratory testes that may be in intra-abdominal or external positions; and Vespertilionidae displayed scrotal testes. In the histological evaluation, we observed a different pattern in vespertilionid species, with testicular regression and sperm retention/storage in the cauda epididymis in the five species analyzed. Similar testicular regression was observed in Molossops temminckii; however, sperm retention/storage was not observed in this species. These data suggest that although the species analyzed are tropical species that do not present a prolonged period of torpor (hibernation), they still maintain a period of seminiferous tubule regression and sperm storage very similar to that observed in hibernating bats. © 2012 Wiley Periodicals, Inc.

Objective: To evaluate the correlations between clinical-radiographical aspects and histomorphometric-molecular parameters of endosseous dental implant sites in humans. Material and methods: The study sample consisted of bone implant sites from the jawbones of 32 volunteers, which were classified according to two different systems: (1) based only on periapical and panoramic images (PP); (2) as proposed by Lekholm & Zarb (L&Z). Bone biopsies were removed using trephine during the first drilling for implant placement. Samples were stained with haematoxylin-eosin (HE), and histomorphometric analysis was performed to obtain the following parameters: trabecular thickness (Tb.Th), trabecular number, bone volume density (BV/TV), bone specific surface (BS/BV), bone surface density and trabecular separation (Tb.Sp). In addition, immunohistochemistry analysis was performed on bone tissue samples for the proteins, Receptor activator of nuclear factor kappa-B (RANK), RANK ligand (RANKL), osteoprotegerin (OPG) and Osteocalcin (OC). Also, the determination of the relative levels of gene expression was performed using Reverse transcription-real-time Polymerase Chain Reaction (RT-PCR). Results: PP and L&Z classification systems revealed a moderate correlation with BV/TV, BS/BV, Tb.Th and Tb.Sp. L&Z's system identified differences among bone types when BV/TV, BS/BV, Tb.Th and Tb.Sp were compared. A weak correlation between PP/L&Z classifications and the expression of bone metabolism regulators (RANK, RANKL, OPG e OC) was found. The analysis of mRNA expression showed no difference between the bone types evaluated. Conclusions: Our results suggest that PP and L&Z subjective bone-type classification systems are related to histomorphometric aspects. These data may contribute to the validation of these classifications. This paper presents a novel approach to the computed assessment of a mammographic phantom device. The approach shown here is fully automated and is based on the automatic selection of the region of interest, in the use of the discrete wavelet transform (DWT) and morphological operators to assess the quality of the American College of Radiology (ACR) mammographic phantom images. The algorithms developed here have succesfully scored 30 images obtained with different combinations of voltage applied to the tube and exposure and could notice the differences in the radiographs due to the different level of exposure to radiation. © 2013 Springer-Verlag.

Casearia sylvestris Swartz (Salicaceae) é uma planta utilizada na medicina tradicional, cujos extratos de folhas demonstraram importantes ações farmacológicas. A espécie apresenta variação morfológica, genética e química. Duas variedades são consideradas devido a diferenças morfológicas: C. sylvestris var. sylvestris e var. lingua. Há dificuldades na definição destas variedades. O objetivo deste trabalho é avaliar diferenças morfo-anatômicas e químicas entre as variedades de C. sylvestris que permitam sua diferenciação com aplicação farmacêutica ou botânica. Seções transversais e paradérmicas de folhas foram preparadas para análises morfo-anatômicas, histoquímicas e microscopia quantitativa (indices de estômatos e paliçada). Análises cromatográficas (CLAE-DAD e CCD) foram realizadas para obter o perfil de diterpenos clerodânicos. Os resultados das análises morfo-anatômicas demonstraram diferenças significativas entre as variedades somente em cortes paradérmicos: var. sylvestris - paredes celulares epidérmicas poligonais e hipoestomática, var. lingua - paredes celulares epidérmicas arredondadas e anfiestomática. Os índices de estômatos não revelaram diferenças; os valores dos índices de paliçada foram de 2,8 para var. lingua e 3,9 para var. sylvestris. As análises cromatográficas confirmaram resultados prévios, demonstrando predomínio de diterpenos na var. sylvestris. Este trabalho sugere que análises cromatográficas e morfo-anatômicas podem ser ferramentas aplicáveis na distinção das variedades da espécie.

This study evaluated the influence of finishing and polishing procedures and differ-ent fluoride solutions on superficial morphology and chemistry of the nanofilled composite resinSupreme XT (3M) through the EDX analysis and SEM evaluation. Circular specimens (n 5 30) of 10mm diameter and 2 mm thickness were prepared, with half of the sample assays finished and polishedwith Super-Snap1sandpaper. The experimental groups were divided according to the presence or ab-sence of finishing and polishing and solutions (arti ficial saliva, 0.0 5% of manipulated sodium fluoridesolution, Fluordent Re ach, Oral B, Fluorgard). Specimens were immersed in each respective solutionfor 1 min per day, during 60 days and stored in artificial saliva at 37 6 18C between immersion peri-ods. Topography and chemical analysis was qualitative. It was observed that specimens submitted tofinishing and polishing procedures had lower superficial degradation. Fluoride solutio ns promoted su-perficial alterations on specimens, being the highest degradation obtained with Fluordent Reach. Itca n be concluded that finishing and polishing procedures and the immersion media influence the su-perficial morphology of composite resin tested; the Fluordent Reach was the flu oride solution thatmo st affected the material’s surface.

Aims: The study evaluated the influence of light curing units and immersionmedia on superficial morphology and chemistry of the nanofilled composite resin Supreme XT (3M)through the EDX analysis and SEM evaluation. Light curing units with different power densitiesand mode of application used were XL 3000 (480 mW/cm2), Jet Lite 4000 Plus (1230mW/cm2), andUltralume Led 5 (790 mW/cm2) and immersion media were artificial saliva, Coke1, tea and coffee,totaling 12 experimental groups. Specimens (10 mm 3 2 mm) were immersed in each respectivesolution for 5 min, three times a day, during 60 days and stored in artificial saliva at 378C 6 18Cbetween immersion periods. Topography and chemical analysis was qualitative. Findings: Groupsimmersed in artificial saliva, showed homogeneous degradation of matrix and deposition of calciumat the material surface. Regarding coffee, there was a reasonable chemical degradation with loss ofload particles and deposition of ions. For tea, superficial degradation occurred in specific areaswith deposition of calcium, carbon, potassium and phosphorus. For Coke1, excessive matrix degra-dation and loss of load particles with deposition of calcium, sodium, and potassium. Conclusion:Light curing units did not influence the superficial morphology of composite resin tested, but theimmersion beverages did. Cisplatin (CPL) is one of the most widely used and effective chemotherapeutic agents for the treatment of several human malignancies. However, it causes serious side effects, especially on reproduction. In order to reduce the undesirable effects caused by many drugs, liposomes have been used as a good system for drug delivery. The aim of this study was to investigate, for the first time, the effects of CPL incorporated into the dipalmitoyl phosphatidylcholine liposome (DPPC) on the testicular tissue of adult Wistar rats. The animals (n = 20) were distributed into four experimental groups: (a) control (distillated water); (b) liposome (DPPC, 1 mL), (c) cisplatin incorporated into liposome (CPL/DPPC), and (d) CPL (8 mg/kg body weight). The animals received a single intraperitoneal injection and were killed 10 days after each treatment for histopathological analysis of testes. The results showed that the testicular histomorphometric parameters in rats of DPPC and CPL/DPPC groups were similar to those of the control group. Meanwhile, rats of the CPL-treated group showed a variety of morphological alterations, including atrophy of seminiferous tubules and presence of multinucleated cells in the germinal epithelium. The incorporation of CPL into the liposome had no influence on the testicular weight or any other stereological parameters, but it was beneficial in maintaining the body weight of the animals. In conclusion, the liposome suppressed the cytotoxic effects caused by cisplatin in the testes of rats, suggesting a possible use in chemotherapy against cancer to reduce the side effects seen on reproduction.
