Beginning the study of chronic pathologic changes in pancreas of hamsters experimentally infected with Trypanosoma cruzi Vic strain, hepatocyte metaplasia was observed in one animal from infected group. This is the first report of oncocytes in Chagas' disease, which could be due to aberrant regenerative response to pancreas inflammatory process.

We have studied the role of the immune response in the morphology of the leishmaniotic granuloma induced in the cheek pouch of hamsters, an immunologically privileged site, after inoculation of 3 x 10(5) Leishmania mexicana. Animals were histologically and immunologically evaluated until 120 days after inoculation. Independent of the time of sacrifice, the animals were always non-reactors to the footpad test (FPT). At histology, the introduction of L. mexicana in the cheek pouch leads to an abscess that evolves to a granulomatous reaction rich in amastigote forms, and later it leads to resolution, even in the absence of immune response detectable by FPT. Our results demonstrate that the development of immune response is not preponderant for the control of infection induced by L. mexicana inoculated subcutaneously in the cheek pouch of the hamster. It also suggests that the macrophages present in the leishmaniotic granuloma are capable of eliminating this parasite, even in the absence of immune response evaluated by FPT.

In order to investigate the pathogenicity of the virus strain GOI 4191 that was isolated from a fatal adverse event after yellow fever virus (YFV) vaccination, an experimental assay using hamsters (Mesocricetus auratus) as animal model and YFV 17DD vaccine strain as virus reference was accomplished. The two virus strains were inoculated by intracerebral, intrahepatic and subcutaneous routes. The levels of viremia, antibody response, and aminotransferases were determined in sera; while virus, antigen and histopathological changes were determined in the viscera. No viremia was detected for either strain following infection; the immune response was demonstrated to be more effective to strain GOI 4191; and no significant aminotransferase levels alterations were detected. Strain GOI 4191 was recovered only from the brain of animals inoculated by the IC route. Viral antigens were detected in liver and brain by immunohistochemical assay. Histothological changes in the viscera were characterized by inflammatory infiltrate, hepatocellular necrosis, and viral encephalitis. Histological alterations and detection of viral antigen were observed in the liver of animals inoculated by the intrahepatic route. These findings were similar for both strains used in the experiment; however, significant differences were observed from those results previously reported for wild type YFV strains.

No município de Paracambi, Estado do Rio de Janeiro, foi realizado um inquérito epidemiológico sobre a leishmaniose tegumentar americana na população canina residente em áreas endêmicas rural e semiurbana. Foram cadastrados 179 cães e 138 (77,1%) foram examinados, segundo seus aspectos clínicos e desenvolvimento de hipersensibilidade tardia ao antígeno Imunoleish® e respostas sorológicas à reação de imunofluorescência indireta e ao ensaio imunoenzimático. Dos 9 (6,5.%) animais portadores de lesões/cicatrizes suspeitas, 66,7% foram causadas por Leishmania sp; 44,4% produziram infecção em hamsters e apresentaram crescimento em meio de cultura, compatíveis com o comportamento de Leishmania do complexo braziliensis. A caracterização molecular (análises isoenzimáticas e do perfil de restrição do KDNA) identificou 2 amostras como similares à Leishmania (Viannia) braziliensis. A prevalência da infecção canina observada através do teste cutâneo, RIFI e ELISA foi, respectivamente, 10,1%, 16,7% e 27,8%. A presença das formas clínica/subclínica da LTA na população canina associada à infecção humana sugere que o cão pode atuar como possível fonte de infecção, assim como na disseminação da doença.

New therapeutic alternatives against leishmaniasis remain a priority. The activity of azithromycin against Leishmania (Leishmania) major has been previously demonstrated. Different responses among species of Leishmania make species-specific drug screening necessary. The activity of azithromycin against Leishmania (Viannia) braziliensis and Leishmania (Leishmania) amazonensis was evaluated in golden hamsters infected through footpad injections of metacyclic promastigotes, and compared with untreated controls and animals treated with meglumine antimoniate. Footpad thickness, lesion cultures and dissemination sites were analyzed. Treatment of golden hamsters with oral azithromycin at 450mg/kg had no activity against infections with Leishmania (Leishmania) amazonensis. For infections due to Leishmania (Viannia) braziliensis, azithromycin demonstrated significant activity relative to untreated controls, but inferior to meglumine antimoniate, for controlling lesion size. Neither drug was able to totally eliminate parasites from the lesions. It was concluded that azithromycin has activity against Leishmania (Viannia) braziliensis but not against Leishmania (Leishmania) amazonensis in this model.

INTRODUCTION: The work was conducted to study phlebotomine fauna (Diptera: Psychodidae) and aspects of American cutaneous leishmaniasis transmission in a forested area where Leishmania (Leishmania) amazonensis occurs, situated in the municipality of Bela Vista, State of Mato Grosso do Sul, Brazil. METHODS: The captures were conducted with modified Disney traps, using hamster (Mesocricetus auratus) as bait, from May 2004 to January 2006. RESULTS: Ten species of phlebotomine sandflies were captured: Brumptomyia avellari, Brumptomyia brumpti, Bichromomyia flaviscutellata, Evandromyia bourrouli, Evandromyia lenti, Lutzomyia longipalpis, Psathyromyia campograndensis, Psathyromyia punctigeniculata, Psathyromyia shannoni and Sciopemyia sordellii. The two predominant species were Ev bourrouli (57.3%) and Bi flaviscutellata (41.4%), present at all sampling sites. Two of the 36 hamsters used as bait presented natural infection with Leishmania. The parasite was identified as Leishmania (Leishmania) amazonensis. CONCLUSIONS: Analysis of the results revealed the efficiency of Disney traps for capturing Bichromomyia flaviscutellata and the simultaneous presence of both vector and the Leishmania species transmitted by the same can be considered a predictive factor of the occurrence of leishmaniasis outbreaks for the human population that occupies the location.

The sand fly Lutzomyia cruzi is considered as one of vectors of visceral leishmaniasis in Brazil. This work examined optimum feeding age, feeding time, host preference, fecundity rates, and female blood meal volume taken by single females from a closed colony of L. cruzi. Mean feeding time was longer on hamsters, 6.6 minutes, than on humans, 5.7 minutes. 49.1% of the 48h-old flies fed on humans and 43.3% of 72h-old flies fed on hamsters. Of a total of 120 females, 61% fed on humans and 25% fed on hamsters. Total fecundity was significantly higher in females fed on hamster than on human or opossum. Laboratory-reared L. cruzi females fed earlier, more promptly, and preferably on humans than on hamsters when offered these blood-meal sources simultaneously. The blood-meal volume is higher in females fed on hamsters than other hosts (human and opossum).

Passando em revista as experiencias de infecção que effectuamos e que acabamos de relatar, vemos que, as culturas isoladas de casos de Leishmaniose visceral americana quer do homem quer do cão, são capazes de infectar hamsters, rhesus e cães de maneira identica ao que acontece com as outras formas de Kala-Azar. Notamos ainda que a evolução da doença e as lesões observadas nos animaes assim infectados, se assemelham ao que tem sido observado nessa enfermidade aproximando-a de maneira singular do Kala-Azar mediterraneo. É sobretudo no cão que a semelhança se torna mais patente. A infecção da pelle e as lesões por ella provocadas que observamos, não differem em nada das que tem sido descriptas na infecção natural do cão e já assignaladas tambem na infecção experimental desse animal, embora de maneira menos completa do que fazemos agora aqui. As lesões oculares com formação de placas de keratite, a infeccção massiça do intestino nos casos fataes de infecção e até mesmo as lesões do figado descriptas por Adler como peculiares á infecção experimental, (infiltração em torno da veia central do lobulo) constituem outros tantos caracteres que mostram a completa analogia entre as infecções do cão que obtivemos e as já observadas no Kala-Azar mediterraneo. Além disso, a presença de Leishmanias na pelle, as vezes em grande numero e a constancia dessa localisação parasitaria, vem mostrar que o cão apresenta as condições necessarias para desempenhar o papel de depositario de virus tal como acontece no Kala-Azar mediterraneo, tanto mais que é elle encontrado naturalmente infectado, nas regiões em que grassa a doença. Mas não é só a infecção do cão que mostra essa semelhança; nos outros animaes tambem se verifica o mesmo facto e até pequenos signaes, taes como a tumefacção das patas assignalada nos hamsters infectados com Leishmania infantum, foram tambem observados aqui. Por outro lado, a reacção de sôro-agglutinação, conforme mostramos em trabalho anterior, não permite a separação das especies do genero Leishmania, pois todas ellas, quando recentemente isoladas, possuem identica constituição antigenica, que se modifica depois, pela conservação longo tempo em cultura. É esse facto, que deu logar ás conclusões contradictorias a que chegaram os autores que se ocuparam do assumpto bem como os primeiros resultados que obtivemos. Deante de todos esses factos, nos julgamos autorizados a concluir como já fizemos anteriormente, que o agente da Leishmaniose visceral americana é identico á Leishmania infantum. Ao terminar, queremos deixar consignados nossos agradecimentos ao Dr. E. chagas, por ter posto a nossa disposição as culturas de Leishmania por elle isoladas, tornando possivel deste modo, a execução do presente trabalho.

A esterase, aliesterase, foi determinada no sôro de 38 ratos de ambos os sexos, 6 hamsters, 34 cobaias e S coelhos, pela técnica de Huggins e Lapides. Os resultados obtidos para os machos e fêmeas foram tabelados em separado. As fêmeas apresentaram uma esterasemia maior que os machos, com uma média global de 11.6 U/ml ± 4.01 e um erro padrão de 0.83 enquanto que para os machos a média foi de 6.9 U ml ± 0.99 com um erro padrão de 0.26. A significância calculada pclo t foi de 4.4 Isto demonstra a interferência da aliesterase na produção ou no metabolismo dos estrogênios. Os valores médios encontrados para as cobaias foram 4.50 U/ ml ± 0.15, com um êrro padrão de 0.036 para as fêmeas e 4.28 U / ml ± 0.30 com um êrro padrão de 0.073 para os machos, sendo a significância (t) de 2.6. Para os coelhos (machos) a média foi de 4.41 U / ml ± 0.16, com um erro padrão de 0.058 enquanto para os hamsters os valores foram de 4.01 U/l ± 0.085, êrro padrão de 0.049 para os machos e 3.81 U/ml ± 0.227, êrro padrão de 0.133 para as fêmeas. Os animais castrados mostram uma diminuição progressiva da esterase no sôro, enquanto que nos castrados e tratados com estrogênios, êstes valores atingem o teôr normal. Os resultados obtidos nestes casos serão objeto de publicação ulterior.

In two experiments, 8 Hamsters inoculated with material from yaws lesions (Treponema pertenue), developed skin lesions considered specific by their clinical and histopathological aspects and by the presence of treponemae. These lesions appeared on the scrotumm, testicle, prepuce, anus, tail, muzzle, back and hinders paws (palm surface). In the internal organs no treponemae were found in direct examinations and inoculation of brain, spleen and lymph node. The incubation period was of 35 days for the testicle, 55 days for the scrotum and 107 days for peritoneal cavity inoculation. Positive sub-inoculations were obtained. The serum reactions (Qasserman's and Kahn's) were negative in all 5 tested Hamsters. Out of 4 normal females matched to infected males two developed nasal lesions resulting from direct contact. Apparently the genital lesions hindered copulation. Hamsters are very well suited for an experimental study of yaws.

O autor estudou, pela reação de fixação do complemento, amostras de vírus da gripe isoladas no Rio de janeiro, durante a epidemia de 1973. Preparou imunesoros em hamsters pela inoculação do líquido alantóide de embriões de galinha infectados. o antígeno solúvel foi preparado com líquido obtido da mesma proveniência. As reações foram positivas, em grau variável, com as amostras clássicas PR8, FM1 e Ásia dos subtipos A0,A1 e A2 e as mais recentes A2/Hong Kong/68 e A2/England/72 e negativa com o anticorpo B/Mass/66. Para as duas variantes do subtipo A2, acima assinaladas e para as 7 amostras isoladas o comportamento foi praticamente o mesmo, não deixando de ser uma reação tipo específica se encararmos, também, as reações obtidas com as demais variantes do tipo A.

Circulating antigens were detected in sera of mice experimentally infected with a high close of Trypanosoma cruzi by reaction with sera from chronically infected mice. The immunodiffusion reaction between homologous acute and chronic sera produced four precipitation lines. By reaction with chronic mouse serum, circulating antingens were detected in sera from heavily infected hamsters, dogs, rabbits and in sera from chagasic patients. A reaction was also found in urine from acutely infected mice and dogs. Trypanosoma cruzi exoantigen was detected in trypanosome culture medium and in the supernatant of infected cell cultures. Attempts to isolate the antigens are described.

Infective stages of Leishmania (Leishmania) amazonensis, capable of producing amastigote infections in hamster skin, were shown to be present in the experimentally infected sandfly vector Lutzomyia flaviscutellata 15, 25, 40, 49, 70, 96 and 120 hours after the flies had received their infective blood-meal. Similarly, infective stages of Leishmania (L.) chagasi were demonstrated in the experimentally infected vector Lu. longipalpis examined 38, 50, 63, 87, 110, 135, 171 and 221 hours following the infective blood-meal, by the intraperitoneal inoculation of the flagellates into hamsters. The question of whether or not transmission by the bite of the sandfly is dependent on the presence of [quot ]metacyclic[quot ] promastigotes in the mouthparts of the vector is discussed.
