A distribuição espaço-temporal dos invertebrados bentônicos, no estuário Tramandaí-Armazém, localizado no Litoral Norte do Rio Grande do Sul, Brasil, foi analisada em três sub-ambientes: laguna Armazém (3 áreas amostrais), laguna Tramandaí (3 áreas amostrais) e Canal de ligação com o Oceano Atlântico (1 área amostral). Na laguna Tramandaí, ocorre um aporte de água doce pelo rio Tramandaí, na laguna Armazém, ocorre a entrada mais freqüente da cunha salina e no Canal verifica-se uma maior influência do Oceano Atlântico. No ARTIGO I, foram analisados os organismos da meiofauna, obtidos a partir de amostragens sazonais em 2000. Em cada área foram tomadas seis amostras, até a profundidade de 5 cm no interior do sedimento, com um corer de 2,7cm de diâmetro. Nematoda foi o grupo taxonômico dominante em todos os ambientes. As análises uni e multivariadas mostraram que a estrutura da meiofauna na laguna Armazém, caracterizada pelas densidades mais elevadas, difere significativamente dos demais ambientes amostrados. A similaridade das amostras coletadas na laguna Tramandaí e no Canal ocorreu em quase todas as estações, com exceção da primavera. Picos de densidade da meiofauna foram encontrados no verão. Os resultados sugerem que os fatores abióticos, como salinidade, hidrodinâmica e temperatura, são importantes condicionantes desta variabilidade espaço-temporal detectada. No ARTIGO II, detalhou-se a identificação de Nematoda, em gênero, e sua classificação em grupo trófico. As análises multivariadas mostraram que a associação de Nematoda é distinta entre os três ambientes analisados. Os atributos densidade, diversidade e riqueza de gêneros foram significativamente mais elevados na laguna Armazém. Nesta laguna, os nematódeos comedores de epistrato e de bactérias foram os dominantes, enquanto que no Canal e Tramandaí prevaleceram os comedores de depósito e predadores facultativos. Uma variabilidade temporal foi detectada tanto para os gêneros, como para os grupos tróficos, com densidades mais elevadas no verão. A identificação de Nematoda e de seu comportamento alimentar mostraram que além da salinidade, hidrodinâmica e temperatura, a disponibilidade de alimento e a forma como está disponível, determinam a estrutura desta associação. No ARTIGO III, foi analisada a distribuição espaço-temporal da macrofauna, a partir de amostragens sazonais entre o outono de 2002 e o verão de 2003. Em cada área foram obtidas 6 amostras com um corer de 10cm de diâmetro enterrado até a profundidade de 20 cm no interior do sedimento. No Canal, a comunidade da macrofauna, foi caracterizada pelas menores densidades e riqueza específica, em todas as estações do ano. Diferente do verificado para Nematoda, a estrutura da macrofauna, encontrada na laguna Armazém e laguna Tramandaí foi similar. Este resultado se deve a baixa riqueza específica da macrofauna e a dominância de espécies oportunistas. Em situações de maior oferta de alimento, oriunda da decomposição de detritos (no inverno) e ao aparecimento da macrófita Ruppia maritima (na primavera), observou-se uma diferenciação da estrutura da macrofauna encontrada na laguna Armazém da laguna Tramandaí. Na laguna Armazém densidades mais elevadas ocorreram no inverno, nos demais ambientes não foram constatadas diferenças significativas entre as estações. Provavelmente as características geomorfológicas do estuário e a hidrodinâmica atuante promovam uma instabilidade em todo o estuário, que para a macrofauna se sobrepõem às diferenças ambientais encontradas entre as lagunas. No ARTIGO IV, avaliou-se a macro e meiofauna, frente a um derrame experimental de óleo, através de análises univariadas. No experimento foram considerados dois fatores: tratamento (controle, corer sem óleo, corer com óleo) e tempo (4 e 9 horas após a adição do óleo). Cada tratamento constou de cinco réplicas, nos dois períodos de coleta. Não foram detectados efeitos do óleo sobre a macrofauna, no tempo de 4 horas e 9 horas de exposição. Para a meiofauna e gêneros de Nematoda, não foram encontradas variações significativas de densidade, com exceção do gênero Theristus, que apresentou um maior número de organismos mortos na presença do óleo.

A pesca artesanal, assim como a agricultura, constitui um objeto de estudo complexo, sendo influenciada por determinantes ambientais, culturais, sócio-econômicas e técnico-produtivas. A combinação destes determinantes com as variáveis tempo e espaço, produz, para cada localidade, uma diversidade de formas através das quais os pescadores artesanais buscam interagir com a natureza e extrair dela seu sustento. Estas constatações motivaram uma dissertação de mestrado, realizada em uma tradicional comunidade pesqueira do estuário da Lagoa dos Patos, no estado do Rio Grande do Sul, e cujos principais resultados buscaremos apresentar neste artigo. A utilização de uma abordagem sistêmica, através de uma adaptação conceitual e metodológica das teorias de evolução e diferenciação dos sistemas agrários, permitiu identificar a variabilidade existente nesta comunidade, em termos de tipos de pescadores e sistemas de produção na pesca, por eles praticados. Foi possível, partindo de categorias utilizadas pelos próprios pescadores, identificar cinco diferentes tipos, que interagem, através de relações de trabalho ou em função da estrutura de capital da qual dispõe, na implementação de três sistemas de produção na pesca distintos. Ao assumir esta variabilidade interna ao grupo social, a pesquisa pode revelar a dinâmica de diferenciação social proporcionada pelo impacto desigual de fatores como as políticas sociais e as políticas públicas voltadas ao fomento da produção, em inter-relação com a eficiência técnica e econômica dos diferentes sistemas de produção na pesca.

The genome of all organisms constantly suffers the influence of mutagenic factors from endogenous and/or exogenous origin, which may result in damage for the genome. In order to keep the genome integrity there are different DNA repair pathway to detect and correct these lesions. In relation to the plants as being sessile organisms, they are exposed to this damage frequently. The Base Excision DNA Repair (BER) is responsible to detect and repair oxidative lesions. Previous work in sugarcane identified two sequences that were homologous to Arabidopsis thaliana: ScARP1 ScARP3. These two sequences were homologous to AP endonuclease from BER pathway. Then, the aim of this work was to characterize these two sequence using different approaches: phylogenetic analysis, in silico protein organelle localization and by Nicotiana tabacum transgenic plants with overexpression cassette. The in silico data obtained showed a duplication of this sequence in sugarcane and Poaceae probably by a WGD event. Furthermore, in silico analysis showed a new localization in nuclei for ScARP1 protein. The data obtained with transgenic plants showed a change in development and morphology. Transgenic plants had slow development when compared to plants not transformed. Then, these results allowed us to understand better the potential role of this sequence in sugarcane and in plants in general. More work is important to be done in order to confirm the protein localization and protein characterization for ScARP1 and ScARP3

The screening for genes in metagenomic libraries from soil creates opportunities to explore the enormous genetic and metabolic diversity of microorganisms. Rivers are ecosystems with high biological diversity, but few were examined using the metagenomic approach. With this objective, a metagenomic library was constructed from DNA soil samples collected at three different points along the Jundiaí-river (Rio Grande do Norte-Brazil). The points sampled are from open area, rough terrain and with the direct incidence of sunlight. This library was analyzed functionally and based in sequence. For functional analysis Luria-Bertani solid medium (LB) with NaCl concentration varied from 0.17M to 0.85M was used for functional analysis. Positives clones resistant to hypersaline medium were obtained. The recombinant DNAs were extracted and transformed into Escherichia coli strain DH10B and survival curves were obtained for quantification of abiotic stress resistance. The sequences of clones were obtained and submitted to the BLASTX tool. Some clones were found to hypothetical proteins of microorganisms from both Archaea and Bacteria division. One of the clones showed a complete ORF with high similarity to glucose-6-phosphate isomerase which participates in the synthesis of glycerol pathway and serves as a compatible solute to balance the osmotic pressure inside and outside of cells. Subsequently, in order to identify genes encoding osmolytes or enzymes related halotolerance, environmental DNA samples from the river soil, from the water column of the estuary and ocean were collected and pyrosequenced. Sequences of osmolytes and enzymes of different microorganisms were obtained from the UniProt and used as RefSeqs for homology identification (TBLASTN) in metagenomic databases. The sequences were submitted to HMMER for the functional domains identification. Some enzymes were identified: alpha-trehalose-phosphate synthase, L-ectoina synthase (EctC), transaminase L-2 ,4-diaminobutyric acid (EctB), L-2 ,4-diaminobutyric acetyltransferase (EctA), L-threonine 3 dehydrogenase (sorbitol pathway), glycerol-3-phosphate dehydrogenase, inositol 3-phosphate dehydrogenase, chaperones, L-proline, glycine betaine binding ABC transporter, myo-inositol-1-phosphate synthase protein of proline simportadora / PutP sodium-and trehalose-6-phosphate phosphatase These proteins are commonly related to saline environments, however the identification of them in river environment is justified by the high salt concentration in the soil during prolonged dry seasons this river. Regarding the richness of the microbiota the river substrate has an abundance of halobacteria similar to the sea and more than the estuary. These data confirm the existence of a specialized response against salt stress by microorganisms in the environment of the Jundiaí river

This study analyzes the institucional environment and its changes around the carcinicultora activity in Rio Grande do Norte from the understanding of its trajectory and the macroinstitucional elements micron and that characterizes the activity in the period of 1999 the 2009. In the analysis of the occured institucional changes in the carcinicultura the Guaraíra Estuary is overcome as space reference, located in the eastern coast of the Rio Grande of the North. The analysis proposal has for base the adoption of the conceptual support of the innovative institucionalistas and neoshumpterianas theories, for understanding that these bring complementary and enriching elements that will allow an analysis, not static, but, dynamics of the carcinicultora activity. It was verified that the crisis brought up for the antidumping action, for the problems of the illnesses that had abated the shrimp, for the depreciation of the Real, among others it led to an overturn in the productive conjuncture of market and forced the alteration of the effective institutions. This institucional alteration, goes in the direction to search to take care of the new demands of the carcinicultura that was possible for the solidity of the institucional foundations constructed throughout the thirty previous years and the consequent consolidation of the composed institucional apparatus for rich a mix of organizations, not only for agencies of government, as well as for organizations with social representation

This study is an environmental diagnosis of the Jundiaí-Potengi/RN estuarine system waters, using calculations of pollution indicator indices such as the Water Quality Index (WQI) and the Toxicity Index (TI). The samples were collected at twelve points on the estuary, at high and low tide, between August and November 2007, over four campaigns. The study area, located in a high impact region, has various activities on its banks such as: discharge of untreated or undertreated domestic and industrial sewage, shrimp farming, immunizer stabilization lakes, riverside communities, etc. All the parameters analyzed were compared to the limits of CONAMA Resolution No. 357 of 2005 for healthy and saline Class 1 waters. The results found prove the impact caused by various activities, mainly the parameters related to the presence of organic material, such as DQO, DBO, COT and thermotolerant colliforms. The IQA for most of the collection points was of medium quality. For the metals, although values above the Resolution limits were found, most of them were lower than the detection limits of ICP-OES used, indicating that they tend to be transported by the dynamic of the tides or rainfall and are deposited in bottom sediments, resulting in a TI of 1.0 in this water, when they are absent, which occurs in most cases, or 0.0, when heavy metals are found in these waters

The aim of this study was to quantify the presence of major and minor elements in the sediments of the Jundiaí-Potengi estuary, bordering the municipalities of Macaíba, Natal and São Gonçalo do Amarante in the state of Rio Grande do Norte, Brazil, considering the evidence of anthropic activity along this estuary, resulting in pollution by treated and untreated wastewaters as well as those produced by industry and shrimp farming. The assessment of the environmental status of this estuary was based on physical-chemical parameters, such as granulometry, organic matter and the major (Al, Fe, Mn and P) and minor (Cd, Cr, Cu, Ni, Pb and Zn) elements found in sediment samples. Two collections were made at 12 georeferenced points along the channel of the estuary at different times. Granulometric analysis was carried out and the percentage of organic matter was determined. The sediments were microwave digested using acid digestion and quantitative analyses of the elements Al, Fe, Cd, Cr, Cu, Mn, Ni, P, Pb and Zn were performed by inductively coupled plasma optical emission spectrometry. Sediment analysis followed the methodology proposed by US EPA 3051A for total elements analysis. The small correlation between the organic matter and the elements studied here suggests that they are of geochemical rather than anthropic origin. The analytical results obtained from the bottom sediment samples in the study area show that the concentration of elements was below the reference values for standard shales (Turekian & Wedepohl, 1961) at the vast majority of points analyzed, ruling out a possible contamination by metals

The aim of this study is to quantify the presence of major and minor elements in the sediments of estuary Potengi. Four georeferenced sampling points were used in the study, at which sediment samples were collected in the channel of the river and on the right and left banks. In addition, dissolved oxygen, salinity and water conductivity were taken in situ at the time of sample collection. The percentage of organic matter, determined by gravimetry, and granulometric analysis of the sediment samples were conducted in the laboratory. To quantify the major and minor elements a prior test to open the sample was conducted with standard NIST 1646ª estuarine sediment to choose the best methodology to be adopted. The sediment samples were dissolved in microwaves with nitric acid and chloridric acid, according to methodology proposed by US EPA 3051ª. Quantitative analyses of the elements Al, Fe, Cd, Cr, Cu, Mn, Ni, Pb and Zn were conducted by inductively coupled plasma optical emission spectrometry (ICPOES). The results showed that the partial concentrations of the elements analyzed are below average worldwide shale levels, the standard described by Turekian and Wedepohl (1961)

The importance of non-functional requirements for computer systems is increasing. Satisfying these requirements requires special attention to the software architecture, since an unsuitable architecture introduces greater complexity in addition to the intrinsic complexity of the system. Some studies have shown that, despite requirements engineering and software architecture activities act on different aspects of development, they must be performed iteratively and intertwined to produce satisfactory software systems. The STREAM process presents a systematic approach to reduce the gap between requirements and architecture development, emphasizing the functional requirements, but using the non-functional requirements in an ad hoc way. However, non-functional requirements typically influence the system as a whole. Thus, the STREAM uses Architectural Patterns to refine the software architecture. These patterns are chosen by using non-functional requirements in an ad hoc way. This master thesis presents a process to improve STREAM in making the choice of architectural patterns systematic by using non-functional requirements, in order to guide the refinement of a software architecture

The estuaries are important investigation zones of the actual morphodynamic and of depositional facies of recent geological history. They are constituted in important receptor means of the coastal area sediments, where the evolutionary processes occur quickly. They are also attractive means for the development of anthropic activities, which in a disordered way interfere in the active processes in the sedimentary balance of the coastal areas. Among the human interventions, the alterations of the depositional environment of mangroves in areas of tropical estuary is deserving relevance, whose implications for the environment estuarine and the coastal adjacent, they are still far to be known. Due to the interest of the sedimentologic component in the comprehension of the processes linked to the evolution of the environments estuarine and coastal adjacent, this work, aimed at the understanding of the morphodynamic coastal phenomena that comprise the region of estuarine influence of the River Curimataú / RN. It was also evaluated in the morphodynamic context the implications due to alterations of the depositional environment of mangrove by anthropic activity. The Curimataú Estuary, located in the south portion of the oriental coast of Rio Grande do Norte, in the last decades has been objective of the overwhelming occupation of the shrimp farm in areas of mangroves, which were implanted with perspectives of development in a short to medium period. On the other hand, the estuary and its region of coastal influence lacks enough information to subsidize the planning and reorganization more effective of the surrounding activities. Thus, it was intended with this work to give a contribution target tothe maintainable use of the coastal resources of this region. A series of studies using data of orbital and acoustic remote sensing, as of sediments sampling, were executed in the gutter of the estuary. The obtained results starting from the interpretation of bathymetric maps, echo sounder graphics and of distribution of sediments made possible the location of the estuary based in morpho-sedimentar criteria. The estuarine tidal flat was dissected in environments of intertidal mangroves, supratidal mangroves and apicuns with base in the integration of data of sensor optic and of radar following by the field control. The adjacent coast that is influenced by the Curimataú estuary, was segmented according to their geomorphologic characteristics, where each segment had a point of observation of the beach morphodynamic, during the period from january/2001 to february/2002. Once every month, beaches profiles, collections of sediments in the beach zones, as measurement of hydrodynamic parameters were executed. The results of the observations of the tidal environment showed that the area of estuarine influence of the Curimataú begins to suffer negative sedimentary taxes, where in some beaches, the erosive processes are already observed. The granulometric characteristics of the beach sediments start to tend for the increase of thin sand in the erosive periods. The destruction of the depositional environments of mangroves of the Curimataú estuary, to the construction of shrimp farms, can be providing the diminution of the tidal prism of the estuary, enlarging the effects of the local increasing of the sea level, through the smaller supplying of sediments to the adjacent coast. Besides this, it was verified the possibility of the sanding of the tidal channel in the margins of the destroyed areas of mangroves, where very high taxes of sedimentation of thin materials were estimated in case of these areas were preserve

The genome of all organisms is subject to injuries that can be caused by endogenous and environmental factors. If these lesions are not corrected, it can be fixed generating a mutation which can be lethal to the organisms. In order to prevent this, there are different DNA repair mechanisms. These mechanisms are well known in bacteria, yeast, human, but not in plants. Two plant models Oriza sativa and Arabidopsis thaliana had the genome sequenced and due to this some DNA repair genes have been characterized. The aim of this work is to characterized two sugarcane cDNAs that had homology to AP endonuclease: scARP1 and scARP3. In silico has been done with these two sequences and other from plants. It has been observed domain conservation on these sequences, but the cystein at 65 position that is a characteristic from the redox domain in APE1 protein was not so conservated in plants. Phylogenetic relationship showed two branches, one branch with dicots and monocots sequence and the other branch with only monocots sequences. Another approach in order to characterized these two cDNAs was to construct overexpression cassettes (sense and antisense orientation) using the 35S promoter. After that, these cassettes were transferred to the binary vector pPZP211. Furthermore, previously in the laboratory was obtained a plant from nicotiana tabacum containing the overexpression cassette in anti-sense orientation. It has been observed that this plant had a slow development and problems in setting seeds. After some manual crossing, some seeds were obtained (T2) and it was analyzed the T2 segregation. The third approach used in this work was to clone the promoter region from these two cDNAs by PCR walking. The sequences obtained were analyzed using the program PLANTCARE. It was observed in these sequences some motives that may be related to oxidative stress response

The aim of this study was the seasonal characterization of the morphology, sedimentology and hydrodynamic of the Açu, Cavalos and Conchas estuaries. These estuaries are inserted in a semi-arid climate area and form the mouth of the hydrographic basin of the Piranhas-Açu river, that represent the discharge of the largest watershed in the state. They are embedded in an environment consisting of a fluvial-marine floodplain, mangrove ecosystem, sandbanks, fields of dunes, spits and sandy beaches. Adjacent to the natural units are the main local socioeconomic activities (oil industry, salt industry, shrimp farming, fishing and tourism) are dependent on this river and its conservation. The environmental monitoring is necessary because it is an area under constant action of coastal processes and at high risk of oil spill. The acquisition and interpretation of hydrodynamic, sonographic and sediment data was conducted in two campaigns, dry season (2010) and rainy season (2011), using respectively the current profiler ADCP Doppler effect, the side-scan sonar and Van Veen sampler. In these estuaries: Açu, Cavalos and Conchas were identified the following types of bedforms: flatbed and Dunes 2-D and 3-D (small to medium size), generated at lower flow regime (Froude number <1). Structures such as ripples were observed in the Açu estuary mouth. The higher values of flow discharge and velocity were recorded in the Açu estuary (434,992 m³.s-¹ and 0,554 m.s-¹). In rainy season, despite the record of highest values of discharge and flow velocities at the mouth, the energy rates upstream did not differ much from the data of the dry season. However, in all estuaries were recorded an increase in speed and flow, with reservation to the flow in the Açu estuary and flow at the mouth of the Conchas estuary. Sediment grain sizes tend to increase towards the mouth of the estuary and these ranged from very fine sand to very coarse sand, medium sand fraction being the most recurrent. Based on the data acquired and analyzed, the estuaries Açu, Cavalos and Conchas are classified as mixed , dominated by waves and tides. According to their morphology, they are classified as estuaries constructed by bar and according to the classification by salinity, estuaries Conchas and Cavalos were ranked as hypersaline estuaries, and Açu as hypersaline and vertically well mixed type C

From August 2000 to December 2003, 185 colonies of Polistes versicolor were observed at the Bioterio of the Instituto de Biociencias, UNESP, Campus de Rio Claro, SP. In 144 colonies (77,84%) didn't emerge wasps, and of others 41 (22,16%), in 16 it was not possible to obtain the number of meconia, because they were attacked in several degrees of injury by natural enemies such as, ants, birds and parasitic wasps. The colonies productivity was measured through the meconia count in the cells of 25 colonies, which ranged from 1 to 6 meconia by cell.
