A mechanistic model is developed to present the photosynthetic response of phytoplankton to irradiance at the physiological level. The model is operated on photosynthetic units (PSU), and each PSU is assumed to have two states: reactive and activated. Light absorption that drives a reactive PSU into the activated state results from the effective absorption of the PSU. Transitions between the two states are asymmetrical in rate. A PSU in the reactive state becomes activated much faster than it recovers from the activated state to the reactive one. The turnover time for an activated PSU to transit into the reactive one is defined by the turnover time of the electron transport chain. The present model yields a photosynthesis-irradiance curve (PE-curve) in a hyperbola, which is described by three physiological parameters: effective cross-section (sigma (PSII)), turnover time of electron transport chain (tau) and number of PSUs (N). The PE-curve has an initial slope of sigma (PSII) x N, a half-saturated irradiance of 1/(sigma (PSII)), and a maximal photosynthetic rate of Nlc at the saturated irradiance. The PE-curve from the present model is comparable to the empirical function based on the target theory described by the Poisson distribution. (C) 2001 Academic Press.

This paper focuses on urban road pricing as a demand management policy that is often regarded as radical and generally unacceptable. Road pricing often gets delayed or abandoned due to low acceptability. This may be due to the fact that complex interactions and drivers of change affect road transport management and require cooperation within implementation networks. The implementation network is a group of people (referred to as partners and actors) who co-ordinate the introduction of policy tools. The drivers of change include any internal or external influences that have an effect on the time, place, or ‘shape’ of the policy measures being introduced. Demand management measures that focus on 'sustainable transport' usually address a limited set of objectives and are often implemented alone i.e. are not necessarily combined with other policy measures. When combined with other measures, it is not always clear whether the multiple interactions between policy tools and implementation networks have been sufficiently considered. Examples of ongoing implementation of policy package in the UK are the support of road pricing initiatives combined with public transport improvements by the Transport Innovation Fund. The objectives of the paper are twofold. First, we present a review of the UK urban road pricing situation. Second, we contrast the emerging issues against six key implementation factors. The analysis of three existing UK road pricing examples - London, Edinburgh and Durham – shows the importance of combining policy tools. Furthermore, through the above examples and theoretical arguments, we emphasise the additional need of creating and maintaining strong networks when implementing policy packages.

NO (nitric oxide) can affect mitochondrial function by interacting with the cytochrome c oxidase (complex IV) of the electron transport chain in a manner that is reversible and in competition with oxygen. Concentrations of NO too low to inhibit respiration can trigger cell defence response mechanisms involving reactive oxygen species and various signalling molecules such as nuclear factor kappa B and AMP kinase. Inhibition of mitochondrial respiration by NO at low oxygen concentrations can cause so-called metabolic hypoxia and divert oxygen towards other oxygen-dependent systems. Such a diversion reactivates prolyl hydroxylases and thus accounts for the prevention by NO of the stabilization of hypoxia-inducible transcription factor. In certain circumstances NO interacts with superoxide radical to form peroxynitrite, which can affect the action of key enzymes, such as mitochondrial complex I, by S-nitrosation. This chapter discusses the physiological and pathophysiological implications of the interactions of NO with the cytochrome c oxidase.

This paper examines the applicability of a digital manufacturing framework to the implementation of a Value Driven Design (VDD) approach for the development of a stiffened composite panel. It presents a means by which environmental considerations can be integrated with conventional product and process design drivers within a customized, digital environment. A composite forming process is used as an exemplar for the work which creates a collaborative environment for the integration of more traditional design drivers with parameters related to manufacturability as well as more sustainable processes and products. The environmental stakeholder is introduced to the VDD process through a customized product/process/resource (PPR) environment where application specific power consumption and material waste data has been measured and characterised in the process design interface. This allows the manufacturing planner to consider power consumption as a concurrent design driver and the inclusion of energy as a parameter in a VDD approach to the development of efficiently manufactured, sustainable transport systems.

Introduction: Individuals carrying pathogenic mutations in the BRCA1 and BRCA2 genes have a high lifetime risk of breast cancer. BRCA1 and BRCA2 are involved in DNA double-strand break repair, DNA alterations that can be caused by exposure to reactive oxygen species, a main source of which are mitochondria. Mitochondrial genome variations affect electron transport chain efficiency and reactive oxygen species production. Individuals with different mitochondrial haplogroups differ in their metabolism and sensitivity to oxidative stress. Variability in mitochondrial genetic background can alter reactive oxygen species production, leading to cancer risk. In the present study, we tested the hypothesis that mitochondrial haplogroups modify breast cancer risk in BRCA1/2 mutation carriers.

Methods: We genotyped 22,214 (11,421 affected, 10,793 unaffected) mutation carriers belonging to the Consortium of Investigators of Modifiers of BRCA1/2 for 129 mitochondrial polymorphisms using the iCOGS array. Haplogroup inference and association detection were performed using a phylogenetic approach. ALTree was applied to explore the reference mitochondrial evolutionary tree and detect subclades enriched in affected or unaffected individuals.

Results: We discovered that subclade T1a1 was depleted in affected BRCA2 mutation carriers compared with the rest of clade T (hazard ratio (HR) = 0.55; 95% confidence interval (CI), 0.34 to 0.88; P = 0.01). Compared with the most frequent haplogroup in the general population (that is, H and T clades), the T1a1 haplogroup has a HR of 0.62 (95% CI, 0.40 to 0.95; P = 0.03). We also identified three potential susceptibility loci, including G13708A/rs28359178, which has demonstrated an inverse association with familial breast cancer risk.

Conclusions: This study illustrates how original approaches such as the phylogeny-based method we used can empower classical molecular epidemiological studies aimed at identifying association or risk modification effects.

Ao longo da última década, a crescente preocupação com as ameaças suscitadas pelas mudanças climáticas e o esgotamento dos recursos naturais tem-se tornado evidente em diversas indústrias e na população global. Além da salvaguarda do ambiente natural, espera-se, das empresas, que respeitem princípios de equidade social nas suas práticas e processos produtivos. Existe uma perceção generalizada de que as empresas e os diversos governos nacionais devem ser mais eficientes na utilização dos recursos naturais e humanos, de modo a promover um desenvolvimento ambiental, social e económico equilibrado e sustentável. Consequentemente, todas as indústrias serão desafiadas a reorganizar as suas cadeias de abastecimento, preservando o ambiente natural e respeitando as comunidades locais. É neste contexto que se insere a presente investigação, que visa contribuir para a consolidação da teoria da gestão sustentável da cadeia de abastecimento. O objetivo geral deste trabalho é o de identificar e analisar os fatores que induzem e instigam as empresas à implementação de práticas ambientais e sociais, identificar e caracterizar as práticas sustentáveis utilizadas e perceber a relação destas práticas com o desempenho económico, ambiental e social. Para responder aos propósitos fixados para o estudo fez-se uso de uma abordagem qualitativa que integrou estudos de caso múltiplos, constituídos a partir de oito empresas de diferentes setores de atividade, que operam em Portugal, destinados a analisar as estratégias de sustentabilidade desenvolvidas e implementadas por essas das organizações. Ao longo da investigação empírica, identificam-se as práticas ambientais e sociais implementadas nas empresas e nas suas cadeias de abastecimento e os diferentes indicadores que as mesmas utilizam para medição do seu desempenho. Nos casos analisados, são apresentadas evidências de que a gestão sustentável da cadeia de abastecimento requer das empresas a criação de mecanismos formais de cooperação entre os vários membros da cadeia de abastecimento e de que existe uma relação entre a aplicação de práticas sustentáveis e o desempenho económico, ambiental e social.

The article presents the “LungoSolofrana” project, carried out during the course “Urban and Mobility” in the academic year 2009/2010, held during the bachelor in Environmental Engineering at the University of Naples “Federico II”. The work has also been chosen as a finalist at the “UrbanPromo 2010” contest, the urban and territorial marketing event sponsored by the National Institute of Urban Planning and Urbit which was held in Venice in 2010. The project consists in a green mobility proposal, developed with an approach based on the integration of the environmental redevelopment of a portion of river Solofrana, located in the Salerno Province, and of the renewal of seven local stations of the railway line Mercato San Severino – Nocera Inferiore, including the realization of a cycle-path network for the natural environment fruition. Furthermore the work drew attention to the local and regional administration. The main intent of the project is to integrate sustainable mobility themes with the environment recovery in a territory affected by high environmental troubles. The area includes the municipalities of Nocera Inferiore, Nocera Superiore, Mercato San Severino, Castel San Giorgio and Roccapiemonte, situated in Salerno’s province, with a total population about 114.000 (font Demo ISTAT 2010). The area extension is about 84,30 sqkm and it is crossed by river Solofrana that is the central point of the project idea. The intervention strategy is defined in two kinds of actions: internal and external rail station interventions. The external rail station interventions regard the construction of pedestrian-cycle paths with the scope of increasing the spaces dedicated to cyclists and to pedestrians along the river Solofrana sides and to connect the urban areas with the railway station. In this way, it’s also possible to achieve an urban requalification of the interested area. On the other side, the interventions inside the station , according to Transit Oriented Development principles, aim at redeveloping common spaces with the insertion of new activities and at realizing new automatic cycle parks covered by photovoltaic panels. The project proposal consists of the urban regeneration of small railway stations along the route-Nocera-Codola Mercato San Severino in the province of Salerno, through interventions aimed at improving pedestrian accessibility. The project involves in particular the construction of pedestrian paths protected access to the station and connecting with neighboring towns and installation of innovative bike parking stations in elevation, covering surfaces coated with solar panels and spaces information. The project is aimed to propose a new model of sustainable transport for small and medium shifts as an alternative to private transportation

O Short Sea Shipping (SSS) é um conceito de transporte marítimo de difícil definição. No entanto, considera-se neste trabalho, que corresponde ao movimento de carga e passageiros por mar entre portos situados geográficamente na Europa ou entre outros portos que não estejam situados na Europa, mas que partilham costa com mares que banham a Europa. Esta definição é sem dúvida aquela que melhor se adequa à inclusão do Short SSS como elo de uma cadeia multimodal de transporte de mercadorias no espaço Europeu, e como alternativa ao transporte de mercadorias apenas por estrada. Este trabalho aborda alguns conceitos importantes subjacentes ao transporte marítimo de forma a permitir uma contextualização que possibilite uma melhor compreensão da abordagem feita ao SSS. A sua promoção e implementação, como alternativa viável para o transporte de mercadorias dentro do espaço Europeu, são objetivos da política de transportes da União Europeia (UE). Este trabalho reflete sobre algumas das políticas estratégicas de promoção da UE para impulsionar o SSS como alternativa ao transporte rodoviário. O objetivo deste trabalho é analisar se o SSS é uma alternativa ao transporte terrestre, no contexto de uma distribuição no Reino Unido de produtos de uma Industria localizada nos arredores da cidade do Porto. Neste trabalho analisou-se as vantagens e desvantagens do SSS, foi efetuada uma descrição do Porto de Leixões como porto escolhido para as expedições, assim como uma análise descritiva dos serviços de Short Sea disponiveis neste porto para as zonas de distribuição desta empresa. Foram descritos os fatores chave da mudança de transporte terrestre para SSS, efectuada uma caracterização da distribuição desta empresa e da solução que esta adotou para distribuir os seus produtos no Reino Unido. A anállise económica da operação, dos tempos de trânsito são dois temas fundamentais para a viabilidade da solução como alternativa ao transporte terrestre.

Cytoch ro me c oxidase (ferrocytochrome c : 02 oxidoreductase ; EC 1.9. 3.1) is the terminal enzyme in the mitochondrial electron transport chain, catalyzing the transfer of electrons from ferrocytochrome c to molecular oxygen. The effects of two large amphiphilic molecules .. valinomycin and dibucaine upon the spectra of the isolated enzyme and upon the activity of both isolated enzyme and enzyme in membrane systems are investigated by using spectrophotometric and oxygen electrode techniques. The results show that both valinomycin and dibucaine change the Soret region of the spectrum and cause a partial inhibition in a concentration range higher than that in which they act as ionophores. It is concluded that both valinomycin and dibucain~ binding induce a conformational change of the protein structure which modifies the spectrum of the a3 CUB centre and diminishes the rate of electron transfer between cytochrome a and the binuclear centre.

Contrairement à la plupart des eucaryotes non-photosynthétiques, les végétaux doivent assurer la stabilité d’un génome additionnel contenu dans le plastide, un organite d’origine endosymbiotique. Malgré la taille modeste de ce génome et le faible nombre de gènes qu’il encode, celui-ci est absolument essentiel au processus de photosynthèse. Pourtant, même si ce génome est d’une importance cruciale pour le développement de la plante, les principales menaces à son intégrité, ainsi que les conséquences d’une déstabilisation généralisée de sa séquence d’ADN, demeurent largement inconnues. Dans l’objectif d’élucider les conséquences de l’instabilité génomique chloroplastique, nous avons utilisé le mutant why1why3polIb d’Arabidopsis thaliana, qui présente d’importants niveaux de réarrangements génomiques chloroplastiques, ainsi que la ciprofloxacine, un composé induisant des brisures double-brins dans l’ADN des organites. Ceci nous a permis d’établir qu’une quantité importante de réarrangements génomiques provoque une déstabilisation de la chaîne de transport des électrons photosynthétique et un grave stress oxydatif associé au processus de photosynthèse. Étonnamment, chez why1why3polIb, ces hautes concentrations d’espèces oxygénées réactives ne mènent ni à la perte de fonction des chloroplastes affectés, ni à la mort cellulaire des tissus. Bien au contraire, ce déséquilibre rédox semble être à l’origine d’une reprogrammation génique nucléaire permettant de faire face à ce stress photosynthétique et conférant une tolérance aux stress oxydatifs subséquents. Grâce à une nouvelle méthode d’analyse des données de séquençage de nouvelle génération, nous montrons également qu’un type particulier d’instabilité génomique, demeuré peu caractérisé jusqu’à maintenant, constitue une des principales menaces au maintien de l’intégrité génomique des organites, et ce, tant chez Arabidopsis que chez l’humain. Ce type d’instabilité génomique est dénommé réarrangement de type U-turn et est vraisemblablement associé au processus de réplication. Par une approche génétique, nous démontrons que les protéines chloroplastiques WHY1, WHY3 et RECA1 empêchent la formation de ce type d’instabilité génomique, probablement en favorisant la stabilisation et le redémarrage des fourches de réplication bloquées. Une forte accumulation de réarrangements de type U-turn semble d’ailleurs être à l’origine d’un sévère trouble développemental chez le mutant why1why3reca1. Ceci soulève de nombreuses questions quant à l’implication de ce type d’instabilité génomique dans de nombreux troubles et pathologies possédant une composante mitochondriale.

La résistance bactérienne aux antibiotiques est de nos jours une préoccupation majeure aux acteurs du monde de la santé publique. L’identification de nouvelles cibles bactériennes en vue de développer de nouveaux antibiotiques est donc nécessaire. La paroi bactérienne est une bonne cible car l’inhibition de sa biosynthèse cause la mort des bactéries. De récents travaux de notre laboratoire ont identifié de nombreux nouveaux facteurs importants pour la biosynthèse de la paroi chez Escherichia coli. L’un de ces facteurs renommé ElyC a un domaine DUF218 extrêmement conservé à travers les espèces bactériennes. L’absence du gène elyC entraîne la lyse bactérienne à température pièce. Des études bioinformatiques indiquent qu’ElyC est une protéine membranaire avec deux domaines transmembranaires et un domaine conservé DUF218 de fonction inconnue. Étant donné que les protéines agissent souvent en complexes, nous avons émis l’hypothèse qu’ElyC interagit avec d’autres protéines afin d'exécuter sa fonction biologique. Le but de mon projet est de déterminer la topologie d’ElyC et d’identifier ses partenaires protéiques. L’étude de la topologie a été faite par l’essai de modification de cystéine sur des souches exprimant individuellement le facteur ElyC avec un résidu cystéine en position N-terminale, dans la boucle ou en position C-terminale. Les partenaires protéiques d’ElyC ont été isolés par immuno-précipitation et identifiés par spectrométrie de masse. Les résultats obtenus ont révélé qu’ElyC est une protéine membranaire chez E. coli et est impliquée dans l'assemblage de l'enveloppe bactérienne, dans la chaîne de transport d'électrons et la phosphorylation oxydative. Ils ont permis aussi de confirmer l’existence d’un lien entre ElyC et le stress oxydatif. Cependant les résultats pour la détermination de la topologie restent à être clarifiés.
