First published October 15, 2009; doi:10.1152/ajpgi.00245.2009.-The nutritional status influences gastric growth, and interestingly, whereas cell proliferation is stimulated by fasting in suckling rats, it is inhibited in adult animals. Corticosterone takes part in the mechanisms that govern development, and its effects are regulated in particular by corticosterone-binding globulin (CBG) and glucocorticoid receptor (GR). To investigate whether corticosterone activity responds to fasting and how possible changes might control gastric epithelial cell cycle, we evaluated different parameters during the progression of fasting in 18- and 40-day-old rats. Food restriction induced higher corticosterone plasma concentration at both ages, but only in pups did CBG binding increase after short-and long-term treatments. Fasting also increased gastric GR at transcriptional and protein levels, but the effect was more pronounced in 40-day-old animals. Moreover, in pups, GR was observed in the cytoplasm, whereas, in adults, it accumulated in the nucleus after the onset of fasting. Heat shock protein (HSP) 70 and HSP 90 were differentially regulated and might contribute to the stability of GR and to the high cytoplasmic levels in pups and elevated shuttling in adult rats. As for gastric epithelial cell cycle, whereas cyclin D1 and p21 increased during fasting in pups, in adults, cyclin E slowly decreased, concomitant with higher p27. In summary, we demonstrated that corticosterone function is differentially regulated by fasting in 18-and 40-day-old rats, and such variation might attenuate any possible suppressive effects during postnatal development. We suggest that this mechanism could ultimately increase cell proliferation and allow regular gastric growth during adverse nutritional conditions.

Objectives: Early weaning (EW) increases proliferation of the gastric epithelium in parallel with higher expression of transforming growth factor alpha and its receptor epidermal growth factor receptor (EGFR). The primary objective of the present study was to examine involvement of EGFR signalling in regulating mucosal cell proliferation during the early weaning period. Materials and methods: Fifteen-day-old rats were split into two groups: suckling (control) and EW, in which pups were separated from the dam. Animals were killed daily until the 18th day, 3 days after onset of treatment. To investigate the role of EGFR in proliferation control, EW pups were injected with AG1478, an EGFR inhibitor; signalling molecules, proliferative indices and cell cycle-related proteins were evaluated. Results: EW increased ERK1/2 and Src phosphorylation at 17 days, but p-Akt levels were unchanged. Moreover, at 17 days, AG1478 administration impaired ERK phosphorylation, whereas p-Src and p-Akt were not altered. AG1478 treatment reduced mitotic and DNA synthesis indices, which were determined on HE-stained and BrdU-labelled sections. Finally, AG1478 injection decreased p21 levels in the gastric mucosa at 17 days, while no changes were detected in p27, cyclin E, CDK2, cyclin D1 and CDK4 concentrations. Conclusions: EGFR is part of the mechanism that regulates cell proliferation in rat gastric mucosa during early weaning. We suggest that such responses might depend on activation of MAPK and/or Src signalling pathways and regulation of p21 levels.

Recombinant Bacillus subtilis strains, either spores or vegetative cells, may be employed as safe and low cost orally delivered live vaccine vehicles. In this study, we report the use of an orally delivered B. subtilis vaccine strain to boost systemic and secreted antibody responses in mice i.m. primed with a DNA vaccine encoding the structural subunit (CfaB) of the CFA/I fimbriae encoded by enterotoxigenic Escherichia coli (ETEC), an important etiological agent of diarrhea among travelers and children living in endemic regions. DBA/2 female mice submitted to the prime-boost immunization regimen developed synergic serum (IgG) and mucosal (IgA) antibody responses to the target CfaB antigen. Moreover, in contrast to mice immunized only with one vaccine formulation, sera harvested from prime-boosted vaccinated individuals inhibited adhesion of ETEC cells to human red blood cells. Additionally, vaccinated dams conferred full passive protection to suckling newborn mice challenged with a virulent ETEC strain. Taken together the present results further demonstrate the potential use of recombinant B. subtilis strains as an alternative live vaccine vehicle. (C) 2008 Elsevier Ltd. All rights reserved.

Com o objetivo de verificar a capacidade enterotoxigênica de cepas de Aeromonas sp. isoladas em diferentes produtos e locais no fluxograma de abate bovino, foram testadas 102 cepas (18 da espécie A. hydrophila, 65 da espécie A. caviae e 19 atípicas) ante os testes de inoculação intragástrica em camundongo lactente e em alça intestinal ligada de coelho. Revelaram-se como produtoras de enterotoxinas três (16,7%) cepas da espécie A. hydrophila, originárias das mãos do manipulador antes que ele iniciasse seus trabalhos e da carne desossada pronta para o consumo, e uma (1,5%) da espécie A. caviae, também isolada das mãos. Os resultados são preocupantes pela presença de cepas enterotoxigênicas de bactérias do gênero Aeromonas em indústria de alto nível higiênico-sanitário.

Oropouche, Caraparu, Guama, Guaroa and Tacaiuma viruses (Orthobunyavirus genus) cause human febrile illnesses and/or encephalitis. To achieve a therapeutical agent to prevent and/or treat these diseases we evaluated the antiviral action of Interferon-alpha (IFN-alpha) on these orthobunyaviruses. In vitro results showed that all the studied orthobunyaviruses are susceptible to antiviral action of IFN-alpha, but this susceptibility is limited and dependent on both concentration of drug and treatment period. In vivo results demonstrated that IFN-alpha present antiviral action on Oropouche and Guaroa viruses when used as a prophylactic treatment. Moreover, a treatment initiated 3 It after infection prevented the death of Guaroa virus infected-mice. Additionally, mortality of mice was related to the migration and replication of viruses in their brains. Our results suggest that IFN-alpha could be potentially useful in the prevention of diseases caused by Oropouche virus and in the prevention and/or treatment of diseases caused by Guaroa virus. Foram estudados a produção de leite de vacas Nelore e o comportamento de amamentação em diferentes sistemas de produção: NR-Nelore Referência, sob manejo extensivo (manejo tradicional); NI-Nelore, sob manejo intensivo; e três cruzamentos CN-Canchim x Nelore, AN-Angus x Nelore e SN-Simental x Nelore, sob manejo intensivo. em três momentos da lactação (60, 120 e 180 dias após o parto), foram medidos, nos bezerros, o número e a duração das mamadas, o ganho diário de peso (kg/dia) e o peso à desmama. O momento da lactação e a interação sistema de produção x momento da lactação apresentaram efeito significativo sobre a produção de leite. A produção de leite não apresentou corrrelação com o comportamento de amamentação nem com o ganho de peso dos bezerros dos diferentes sistemas de produção. Condições deficientes de alimentação não resultaram em menores produções de leite de vacas Nelore, mas sim em acentuadas perdas de peso (80 kg) durante a estação de monta no sistema NR. O tempo diário de amamentação apresentou diminuições significativas no sistema extensivo com o decorrer da lactação, enquanto os sistemas intensivos não mudaram ou aumentaram os minutos de amamentação por dia. Para as condições nas quais o experimento foi desenvolvido, os bezerros cruzados apresentaram os melhores desempenhos durante a fase pré-desmama, em comparação com os bezerros Nelore.

Foram utilizados 24 bezerros da raça Holandesa distribuídos em delineamento inteiramente casualizado com oito repetições e três tratamentos: leite integral (LI) e dois tipos de sucedâneos, DestetorÒ (DES) e LactalÒ (LAC), durante o período de aleitamento (60 dias), sendo avaliados: ganho de peso, consumo de matéria seca e mensurações corporais (altura na cernelha, circunferência toráxica e ventral). Os ganhos médios diários de peso, a altura na cernelha e a circunferência torácica e ventral dos bezerros não diferiram (P>0,05) entre os tratamentos. Foram obtidas as médias de ganho de peso de 0,32, 0,41 e 0,26 kg/bezerro/dia nos tratamentos LI, DES e LAC, respectivamente. O consumo de matéria seca total na fase de aleitamento, durante 60 dias, diferiu entre os tratamentos, sendo que os bezerros que receberam o sucedâneo DES consumiram mais matéria seca total, comparados àqueles que receberam o sucedâneo LAC (P<0,05). As médias de consumo diário de matéria seca foram: 0,79; 0,94 e 0,68kg/bezerro respectivamente para os tratamentos LI, DES e LAC.

The objective was to contribute to a reading of the characteristics and diversity of production systems from bovine milk in the microregion Seridó of Rio Grande do Norte, discuss the social, economic, environmental and husbandry-related primary activity of milk production. We randomly selected 28 agricultural establishments that performed the activity of dairy bovine culture with subsequent applicat ion of a structured questionnaire during September and October 2011. Data were analyzed with application of measures of descriptive analysis and determination of the index rural development (IRD). The results showed that 53.57% of the interviewees were owners of the land, the median area of the properties amounted to 135 hectares, the median number of animals in the herd was 51 head, with minimum 11 and maximum of 350 heads establishments in the sample, 85.72% of establishments had maximum 23 cows in lactation, 100% performed manual milking with suckling calves, average productivity of 3.91 liters / cow / day, 92.86% of the interviewees did not produce silage and / or hay, 64.29% had no access to technical assistance, the average age of interviewees was 51 ± 10.85 years, 78.57% had only elementary education. The average of the IRD computed in the test sample amounted to 0.43 ± 0.11 on a scale 0-1. The sites were grouped into 04 classes, based on your IRD. The average IRD in each dimension was 0.43, 0.55 and 0.34, respectively for economic / husbandry, social and environmental. The determination of the rate of rural development - IDR found for the sample could add establishments hierarchically. It is necessary that the inclusion of best practices in herd management, bookkeeping zootechnical, technical assistance and reorganization practices in land and environmental preservation

Studies indicate that spinner dolphins use the Baía dos Golfinhos, in Fernando de Noronha Archipelago, for resting, reproduction, parental care and protection against shark attacks. The present study had the purpose of verifying the seasonality of spinner dolphin newborns and calves in relation to the months of the year and the pluviometric seasons (dry and rainy), as well as their interaction with the number and gender of accompanying adults and their positioning in relation to the adults (vertical, horizontal, and depth) in the above mentioned bay. The analysis were made out of photo records of dolphins collected between 2000 and 2006 (seasonality) and between 1995 and 2006 (interaction) both using ad libitum sampling method during free dives. To determine the age category, the reason between the smaller dolphin s total body length and the bigger dolphin s total body length was calculated. The dolphins were then divided into three age groups: adults, newborns and calves. Those with total body length ≥ 170cm were considered adults, newborns up to 105cm, and calves from 106cm to 128cm. In addition, the secondary characters described in literature were used to identify newborns and calves. The adults had dimensions of total lenght ≥170cm , the newborns until 105cm and calves between 106cm and 128cm. I addition, secondary characteristics described in the literature were used to indentify newborns and calves. The number of spinner dolphin newborns was greater in the month of April and higher during the rainy season. Throughout the months and pluviometric seasons (pluviometer/pluvial metric), the number of calves did not have a significant difference. Concerning to the presence of newborns and calves age groups at Baía dos Golfinhos, there was not a significant difference. It was possible to identify the gender of the escorting adults as (42), 95.24% being females and 4.76% males. Newborns were more frequently seen in the company of two adults, whereas calves were more often accompanied by more than two adults However, there was not a significant difference for the newborns, whereas for the calves there was a significant difference for those classified as loners and those accompanied by more than two adults. When in vertical positioning, the newborns and calves were more frequently observed in inferior position with some difference demonstrated between some of that. While in horizontal positioning both age groups were more often seen in posterior position, also with differences between them. In the depth perspective newborns and calves were positioned anterior, however with significant difference for the calves. The occurrence of a peak of newborns in the months of April may indicate the existence of a birth seasonality pattern for the beginning of the rainy season, with births scattered throughout the year. The results for the positions and escorting of newborns and calves are related to protection and suckling. These conditions reinforce the importance of the area when it comes to the care for offspring, which calls for the creation of conservation rules to the area, especially during those months with greater occurrence of newborns

Toxocara vitulorum, a nematode parasite in the small intestine of cattle and water buffaloes, causes high morbidity and mortality of 1-3 months old buffalo calves. This research evaluated the specific perieneteric antigens (Pe) reactivity of anti-T. vitulorum-Pe antibody (Tv-Pe-Ab) in both immune sera and colostrum from buffalo cows immediately post-partum from buffalo cows. The presence of Tv-Pe-Ab in sera of buffalo newborn calves was also examined at 1 day before and after suckling the colostrum as well as in sera from naturally infected calves at the beginning and peak of the maximum infection and then again during the period of rejection and post-rejection of the parasite. Pe antigens were characterized for Tv-Pe-Ab by SDS-PAGE and Western blot (WB). The SDS-PAGE showed that Pe contained nine protein bands (11, 14, 31, 38, 58, 76, 88,112 and 165 kDa). All Pe bands were recognized by Tv-Pe-Ab in sera and colostrum of buffalo cows. Only the serum antibodies of buffalo calves at 1 day of age after suckling the colostrum and during the beginning of T. vitulorum infection recognized Pe antigen's nine bands. In contrast, serum antibodies from 1-day-old buffalo calves, taken before suckling colostrum, did not react with any protein band. In suckling calves, which reached peak egg output, rejection and post-rejection stages of the infection, serum Tv-Pe-Ab reactivity with lower molecular weight protein bands (11-76 kDa) was lost and only reactivity with the Pe protein bands of higher molecular weight (88, 112 and 165 kDa) remained. Toxocara vitulorum is a nematode parasite of the small intestine of cattle and water buffalo, particularly buffalo calves between one and three months of age, causing high morbidity and mortality. The purpose of this research was to characterize the excretory/secretory (ES) antigens of T vitulorum larvae by SDS-polyacrylamide gel electrophoresis (PAGE) and Western blot (WB), using immune sera and colostrum of buffalo naturally infected by T vitulorum. The parasitological status of the buffalo calves was also evaluated using sequential fecal examinations. The results showed that the ES antigen revealed eight (190, 150, 110, 90, 64, 56, 48, and 19 kDa) protein bands by SDS-PAGE. The majority of these bands were recognized in the sera and colostrum of infected buffalo with T vitulorum when analyzed by WB. However, particularly fractions of high molecular weight (190, 150, 110, and 90 kDa) were represented in more prominent bands and persisted in the groups of buffalo calves at the peak of egg output, as well as during the period of rejection of T vitulorum by the feces of the calves. During the period of post-rejection of the worms (between the day 118 and 210 of age) the serum antibodies did not react with any protein bands. on the other hand, sera from buffalo calves at one day of age (after suckling the colostrum and at the beginning of infection) reacted with the same bands detected in the serum and colostrum of the buffalo cows.

Objetivou-se, neste trabalho, pesquisar a relação entre os microrganismos patogênicos isolados e identificados em água utilizada na ordenha, com o isolamento e identificação dos mesmos em amostras de leite, de quartos mamários apresentando mastite clínica ou subclínica nas mesmas propriedades. Foram utilizadas 16 propriedades rurais leiteiras, escolhidas aleatoriamente, na região de Cerqueira César - SP, que utilizavam ordenha mecânica. A água utilizada na ordenha foi classificada em relação à presença de coliformes totais e fecais, como dentro dos padrões ou fora dos padrões de potabilidade humana. Nos resultados obtidos, 94% das amostras foram classificadas como fora dos padrões em relação a coliformes totais e fecais. Os microrganismos identificados foram: Escherichia coli (51%), Enterobacter spp. (25%), Enterobacter cloacae (8%) Edwardsiella tarda (8%) e Klebsiella oxytoca (8%). em relação ao leite, foram analisadas 373 amostras provenientes de vacas em lactação, com mastite clínica (n=19; 5%) e subclínica (n=354; 95%). Os animais com mastite subclínica foram identificados pela contagem de células somáticas (CCS), utilizando-se o aparelho eletrônico (Somacount 300, Bentley), onde a média observada foi de 1.631 x 10³ células/mL. Os principais microrganismos identificados foram: Staphylococcus aureus (30%), Corynebacterium bovis (23%) e Staphylococcus spp. (15%). Conforme os dados obtidos, os agentes coliformes encontrados na água, utilizada na ordenha, não estavam presentes nas análises das amostras de leite dos quartos mamários com mastite clínica ou subclínica das respectivas propriedades, demonstrando não haver associação entre a qualidade da água e a ocorrência de mastite.

Um experimento foi conduzido no intuito de avaliar ovelhas de descarte abatidas em diferentes estágios fisiológicos e terminadas em confinamento, quanto aos rendimentos de carcaça e seus cortes, assim como os percentuais de cada não-componente da carcaça. Utilizaram-se 21 ovelhas da raça Santa Inês com idade média de 6 anos, distribuídas nos tratamentos: OL = ovelhas mantidas por 60 dias em lactação com seus cordeiros e abatidas um dia após o desmame dos mesmos; OSC = ovelhas mantidas por 60 dias em lactação com seus cordeiros e mais um período de aproximadamente 30 dias sem os cordeiros, no intuito de recuperar o peso corporal perdido na amamentação, e posteriormente abatidas; e ONP = ovelhas mantidas por 60 dias em confinamento e que não pariram durante o ano. As ovelhas que permaneceram por 60 dias em confinamento e que não pariram durante o ano apresentaram maiores rendimentos de carcaça quente 45,00 e fria 44,73% em comparação àquelas mantidas por 60 dias em lactação com seus cordeiros e abatidas um dia após o desmame: 40,80 e 40,66%. Os rendimentos dos cortes da carcaça (perna, lombo, paleta, costelas e pescoço) foram similares entre todas as ovelhas abatidas nos distintos estágios fisiológicos. Os rendimentos dos não-componentes da carcaça, com exceção dos valores do fígado, do sangue, do úbere e da gordura interna, foram similares entre as ovelhas.

Os efeitos da remoção temporária de bezerros (RB) sobre características foliculares e taxas de ovulação e de prenhez de vacas Nelore foram avaliados em dois experimentos quando da inseminação artificial em tempo fixo (IATF), utilizando-se um protocolo à base de GnRH-PGF2α-BE. No experimento 1, 139 vacas Nelore, lactantes, cíclicas ou acíclicas, receberam um protocolo hormonal envolvendo a aplicação de 100±g de análogo GnRH no dia zero (D0-GnRH), 25mg de PGF2α no dia 7 (D7-PGF2α) e 1,0mg de benzoato de estradiol no dia 8 (D8-BE). Os animais foram aleatoriamente distribuídos para serem submetidas ou não à RB (48 horas) antes da aplicação de GnRH (RB1) ou após a aplicação de PGF2α (RB2). Avaliaram-se o diâmetro folicular (DF) nos dias 0 (D0) e 9 (D9) do protocolo e as taxas de ovulação (T0), advindas dos tratamentos. As vacas acíclicas que receberam RB1 apresentaram maiores DF no D0 (P<0,05) e taxa de ovulação (P<0,05) quando da aplicação de GnRH, em relação às que não receberam RB1. Nas vacas cíclicas, não houve efeito da RB1 sobre as variáveis analisadas. As vacas que receberam RB2 apresentaram maiores DF no D9 (P<0,05) e taxa de ovulação (P<0,01) quando da aplicação de BE, em relação às que não receberam a RB2. No experimento 2, 376 vacas da raça Nelore, lactantes, foram submetidas ao mesmo protocolo hormonal e aos mesmos tratamentos do experimento 1, mas foram inseminadas 24 a 30 horas após a aplicação de BE no D8, visando avaliar a taxa de prenhez após a IATF. A taxa de prenhez foi maior nos animais que receberam as duas RB em relação às que não receberam RB (29,8% vs 10,6%; P<0,05). A remoção de bezerros associada ao protocolo hormonal aplicado pode ter aumentado as taxas de prenhez devido às maiores taxas de ovulação, em resposta ao GnRH ou ao benzoato de estradiol.
