OBJECTIVE: A cross-sectional population-based study was conducted to assess, in active smokers, the relationship of number of cigarettes smoked and other characteristics to salivary cotinine concentrations. METHODS: A random sample of active smokers aged 15 years or older was selected using a stepwise cluster sample strategy, in the year 2000 in Rio de Janeiro, Brazil. The study included 401 subjects. Salivary cotinine concentration was determined using gas chromatography with nitrogen-phosphorus detection. A standard questionnaire was used to collect demographic and smoking behavioral data. The relation between the number of cigarettes smoked in the last 24h and cotinine level was examined by means of a nonparametric fitting technique of robust locally weighted regression. RESULTS: Significantly (p<0.05) higher adjusted mean cotinine levels were found in subjects smoking their first cigarette within five minutes after waking up, and in those smoking 1-20 cigarettes in the last 24h who reported inhaling more than ½ the time. In those smoking 1-20 cigarettes, the slope was significantly higher for those subjects waiting for more than five minutes before smoking their first cigarette after waking up, and those smoking "light" cigarettes when compared with their counterparts. These heterogeneities became negligible and non-significant when subjects with cotinine >40 ng/mL per cigarette were excluded. CONCLUSIONS: There was found a positive association between self-reporting smoking five minutes after waking up, and inhaling more than ½ the time are consistent and higher cotinine levels. These can be markers of dependence and higher nicotine intake. Salivary cotinine proved to be a useful biomarker of recent smoking and can be used in epidemiological studies and smoking cessation programs.

Objective - To evaluate the effect of prepregnancy body mass index (BMI), energy and macronutrient intakes during pregnancy, and gestational weight gain (GWG) on the body composition of full-term appropriate-for-gestational age neonates. Study Design - This is a cross-sectional study of a systematically recruited convenience sample of mother-infant pairs. Food intake during pregnancy was assessed by food frequency questionnaire and its nutritional value by the Food Processor Plus (ESHA Research Inc, Salem, OR). Neonatal body composition was assessed both by anthropometry and air displacement plethysmography. Explanatory models for neonatal body composition were tested by multiple linear regression analysis. Results - A total of 100 mother-infant pairs were included. Prepregnancy overweight was positively associated with offspring weight, weight/length, BMI, and fat-free mass in the whole sample; in males, it was also positively associated with midarm circumference, ponderal index, and fat mass. Higher energy intake from carbohydrate was positively associated with midarm circumference and weight/length in the whole sample. Higher GWG was positively associated with weight, length, and midarm circumference in females. Conclusion - Positive adjusted associations were found between both prepregnancy BMI and energy intake from carbohydrate and offspring body size in the whole sample. Positive adjusted associations were also found between prepregnancy overweight and adiposity in males, and between GWG and body size in females.

É fundamental compreender que os fumantes não são iguais e que determinados fumantes precisam ser conquistados como "potenciais clientes" de programas de intervenção voltados às suas necessidades específicas. O objetivo do estudo foi comparar os perfis de fumantes recrutados para um estudo de intervenção para cessação de fumar com os da população geral de fumantes no município do Rio de Janeiro, nos anos 2002-2003. As heterogeneidades encontradas indicam que diferentes estratégias de captação associadas às intervenções existentes devem ser elaboradas para motivar o maior e mais diversificado número possível de indivíduos elegíveis.

ABSTRACT OBJECTIVE To estimate the prevalences of tobacco use, tobacco experimentation, and frequent smoking among Brazilian adolescents. METHODS We evaluated participants of the cross-sectional, nation-wide, school-based Study of Cardiovascular Risks in Adolescents (ERICA), which included 12- to 17-year-old adolescents from municipalities of over 100 thousand inhabitants. The study sample had a clustered, stratified design and was representative of the whole country, its geographical regions, and all 27 state capitals. The information was obtained with self-administered questionnaires. Tobacco experimentation was defined as having tried cigarettes at least once in life. Adolescents who had smoked on at least one day over the previous 30 days were considered current cigarette smokers. Having smoked cigarettes for at least seven consecutive days was an indicator for regular consumption of tobacco. Considering the complex sampling design, prevalences and 95% confidence intervals were estimated according to sociodemographic and socio-environmental characteristics. RESULTS We evaluated 74,589 adolescents. Among these, 18.5% (95%CI 17.7-19.4) had smoked at least once in life, 5.7% (95%CI 5.3-6.2) smoked at the time of the research, and 2.5% (95%CI 2.2-2.8) smoked often. Adolescents aged 15 to 17 years had higher prevalences for all indicators than those aged 12 to 14 years. The prevalences did not differ significantly between sexes. The highest prevalences were found in the South region and the lowest ones, in the Northeast region. Regardless of sex, the prevalences were found to be higher for adolescents who had had paid jobs, who lived with only one parent, and who reported having been in contact with smokers either inside or outside their homes. Female public school adolescents were found to smoke more than the ones from private schools. CONCLUSIONS Tobacco use among adolescents is still a challenge. Intending to reduce the prevalence of tobacco use among young people, especially the ones under socioeconomic vulnerability conditions, Brazil must consolidate and increase effective public health care measures.

Previous experiments revealed that DHH1, a RNA helicase involved in the regulation of mRNA stability and translation, complemented the phenotype of a Saccharomyces cerevisiae mutant affected in the expression of genes coding for monocarboxylic-acids transporters, JEN1 and ADY2 (Paiva S, Althoff S, Casal M, Leao C. FEMS Microbiol Lett, 1999, 170∶301–306). In wild type cells, JEN1 expression had been shown to be undetectable in the presence of glucose or formic acid, and induced in the presence of lactate. In this work, we show that JEN1 mRNA accumulates in a dhh1 mutant, when formic acid was used as sole carbon source. Dhh1 interacts with the decapping activator Dcp1 and with the deadenylase complex. This led to the hypothesis that JEN1 expression is post-transcriptionally regulated by Dhh1 in formic acid. Analyses of JEN1 mRNAs decay in wild-type and dhh1 mutant strains confirmed this hypothesis. In these conditions, the stabilized JEN1 mRNA was associated to polysomes but no Jen1 protein could be detected, either by measurable lactate carrier activity, Jen1-GFP fluorescence detection or western blots. These results revealed the complexity of the expression regulation of JEN1 in S. cerevisiae and evidenced the importance of DHH1 in this process. Additionally, microarray analyses of dhh1 mutant indicated that Dhh1 plays a large role in metabolic adaptation, suggesting that carbon source changes triggers a complex interplay between transcriptional and post-transcriptional effects.

SUMMARYMycobacteria which are most commonly isolated in pulmonary secretions of patients with respiratory symptoms living in the State of Amazonas are given. The high percentage of isolates (25,4) and the species obtained indicate a strong relationship of contamination of man by the environment. Among the species isolated, seven of them are considered potentially pathogenic mycobacteria. Since some of these species may interfere with present and further vaccination programs against Tuberculosis and Leprosy, these investigations indicate a necessity of carrying out studies of population awareness against specific PPD's prepared from the same species.

RESUMOO pré-selecionamento de espécies madeireiras da Amazônia, determinou quais as mais adequadas ao curvamento de madeira maciça. Foram elaborados ensaios mecânicos de compressão paralela às fibras em amostras representativas de um determinado número de espécies, que aquecidas por vaporização, determinaram os níveis de compressibilidade das mesmas. Foram testadas amostras de todas as espécies pré-selecionadas também ao processo de curvamento e, determinados seus raios mínimos de curvatura.

De 1981 a 1985, foram atendidos 96 acientes nas unidades de saúde: Instituto de Dermatologia Troppical e Venereologia "Alfredo da Matta" IDTV-AM e Serviço Médico do Instituto nacional de Pesquisas da Amazônia (INPA), Manaus-AM.Os portadores de diagnóstico clínico de Tiena pedis (pé-de-atleta), foram submetidos ao exame micológico, os quais revelaram que dos segmentos podais mais afetados foram os da Região plantar (42,0%) e interdigital (26,0%). Das 99 espécies fúngicas, 74% eram dermatófitos e 26% de leveduras. Dentre as espécies observadas, verificou-se que as mais frequentes por número de ocorrência foram: Trichophyton rubrum (335,5%), Trichopthyton mentagrophytes (25,00% e Candida sp. (25,0%).

The Brazilian State of Amazonas has a high incidence of Tuberculosis, 91.4 in 10,000 habitants (SESAU, 1994) and resistant strains of Mycobacterium tuberculosis are frequently being found in the region (SALEM et.al, 1990). These problems have been associated with side effects caused by the antibiotics used to treat Tuberculosis, which have in rum been associated with treatment non-compliance (PATTISAPU, 1984). To resolve this problem a cost effective alternative treatment for Tuberculosis with few or no side effects, needs to be found. Amazonas has an abundance of plants, many of which are used by the lay population for medicinal purposes. A survey was carried out in five towns of the region, interviewing patients receiving treatment for Tuberculosis, to find out whether and which plants have been used to treat Tuberculosis. Results showed that the majority of patients in the sample had used medicinal plants before or after diagnosis of Tuberculoses. Thirteen different plants were recorded for this purpose. Chenopodium ambrosioides L, popularly known as Mastruz, was the most commonly used, followed by Caesalpinia ferrea Mart. Jucá and Spilanthes acmella DC. Jambu. This study concentrates on Mastruz as it was used more frequently than the other medicinal plants. No significant effects on baciloscopy test results were found when Mastruz was used before diagnosis. ln-vitro laboratory tests have also not shown any tuberculocidal effects for Mastruz. Further tests are being carried out on the other medicinal plants.

We elaborated an alternative culture method, which we denominated PKO (initials in tribute of respect to Petroff, Kudoh and Ogawa), for isolating Mycobacterium tuberculosis from sputum for diagnosis of pulmonary tuberculosis (TB), and to compare its performance with the Swab and Petroff methods. For the technique validation, sputum samples from patients suspected of pulmonary TB cases were examined by acid-fast microscopy (direct and concentrated smear), PKO, Swab and Petroff methods. We found that Petroff and PKO methods have parity in the effectiveness of M. tuberculosis isolation. However, by the PKO method, 65% of isolated strains were detected in a period of £15 days, while by the Petroff method the best detection was in an interval of 16-29 days (71%). In positive smear samples, the average time of PKO isolation is only superior to the one related for Bactec 460TB. In conclusion, the exclusion of the neutralization stage of pH in the PKO reduces the manipulation of the samples, diminishes the execution time of the culture according to the Petroff method and facilitates the qualification of professionals involved in the laboratorial diagnosis of Tuberculosis.

Investigou-se por métodos bacteriológicos (cultivo) e moleculares (PCR - Restriction Enzyme Analysis, PRA), a presença de micobactérias ambientais em águas de torneira, soluções e luvas cirúrgicas, utilizadas nas etapas dos procedimentos cirúrgicos executados no centro cirúrgico do Hospital Universitário Getulio Vargas (HUGV), na cidade de Manaus-Amazonas/Brasil. Foram colhidas e analisadas 105 amostras sendo: 24 de águas (colhidas das 2 torneiras existentes no centro cirúrgico), 8 de solução de Povidine e 7 de solução de Clorhexidina, que servem para a higienização das mãos dos cirurgiões; 39 de luvas cirúrgicas (superfícies internas e externas); e 27 de soluções que foram efetivamente utilizadas durante o ato cirúrgico. Por método bacteriológico obteve-se 41 isolados micobacterianos apenas de águas das torneiras. Pelo PRA obteve-se a detecção de DNA micobacteriano somente na amostra de água que forneceu acima de 100 colônias de micobactérias por tubo semeado. Os isolados foram identificados como sendo Mycobacterium celatum perfil 2, M. gordonae perfil 3, M. gordonae perfil 6, M. intracellulare perfil 1, M. lentiflavum perfil 3 e M. mucogenicum perfil 1. O encontro de M. mucogenicum, espécie já incriminada em surtos pós-cirúrgicos, indica que devem ser efetuados procedimentos de limpeza e monitoramento em todos os pontos de distribuição de águas, visando à prevenção de surtos de micobacterioses nosocomiais induzidos pelo uso das águas nas diferentes atividades de manuseio ou higienização dos pacientes submetidos a procedimentos invasivos.

In this study, a mathematical model for the production of Fructo-oligosaccharides (FOS) by Aureobasidium pullulans is developed. This model contains a relatively large set of unknown parameters, and the identification problem is analyzed using simulation data, as well as experimental data. Batch experiments were not sufficiently informative to uniquely estimate all the unknown parameters, thus, additional experiments have to be achieved in fed-batch mode to supplement the missing information. © 2015 IEEE.

El bajo rendimiento advertido en general en las aulas universitarias, sumado a la abulia y escasa motivación por aprender que parece caracterizar a muchos estudiantes, acentúan el desafío que enfrentan las universidades de intervenir para mejorar la calidad de la educación de los ciudadanos. Conocer más acerca de los recursos internos y externos con que cuentan nuestros estudiantes para aprender y analizar el modo en que repercuten en su motivación y aprendizaje es, sin duda, un modo de avanzar hacia contribuciones capaces de redundar en mejoras educativas. En este marco, proponemos brindar aportes teóricos y definir líneas de acción que contribuyan al diseño de contextos de evaluación óptimos para el surgimiento de creencias motivacionales y estados emocionales beneficiosos para los aprendizajes de estudiantes universitarios. Suponemos que un contexto de evaluación que propicie el uso del conocimiento por parte de los estudiantes, que favorezca el despliegue de estrategias de autorregulación y posibilite ricos procesos de feedback, favorecerá la emergencia de percepciones de autoeficacia positivas, de estados emocionales beneficiosos y la ampliación de las posibilidades de lograr éxito en los resultados obtenidos. Trabajaremos con estudiantes de la Universidad Nacional de Río Cuarto, con muestras accidentales compuestas por la totalidad de alumnos que cursen Tráfico de señales –asignatura de Ing. en Telecomunicaciones- y Psicología Educacional -materia de distintos profesorados de la Facultad de Ciencias Humanas y de la Facultad de Ciencias Exactas Físico-Químicas y Naturales. Los datos serán recabados mediante el Multidimensional Scales of Perceived Self-efficacy (Bandura, 1990); el Achievement Emotions Questionnaire (Pekrun et al., 2005); análisis de protocolos de evaluación, observación no participante de clases e instancias de evaluación; grabación en audio de las clases observadas y entrevistas semi-estructuradas a estudiantes. Los resultados obtenidos prometen aportes teóricos y orientaciones pedagógicas. Las contribuciones teóricas se vinculan con la profundización de los marcos teóricos y con los avances logrados en las interpretaciones realizadas. Las orientaciones pedagógicas, se vinculan con aportes al diseño de contextos de evaluación óptimos para el surgimiento de creencias motivacionales y estados emocionales beneficiosos para los aprendizajes. Este proyecto se estima importante y pertinente porque materializa uno de los desafíos que enfrentan actualmente los investigadores educacionales, esto es, conocer más acerca del modo en que los aspectos personales de los estudiantes interactúan con las características del contexto académico en situaciones genuinas de aprendizaje; porque considera un tópico de investigación que emerge con fuerza en la agenda de la Psicología Educacional, esto es, el estudio de las emociones académicas; porque integra tendencias metodológicas que marcan rumbos en el estudio de las variables consideradas.
