O Brasil possui cerca de 32 milhões de hectares de áreas com aptidão para a expansão da cultura da palma de óleo e mais de 90% destas estão na Amazônia. Há necessidade de compreender a interação de novos plantios e de seu desenvolvimento com o meio ambiente. Este estudo foi conduzido em plantios comerciais da empresa Dendê do Pará SA (DENPASA) em Castanhal – Pará e avaliou o efeito da variação sazonal da precipitação sobre a fertilidade do solo e desta sobre a biomassa microbiana do solo em uma cronosseqüencia de plantios de palma de óleo com 5, 8 e 12 anos e floresta secundária. Foram mensurados/estimados e correlacionados os atributos do solo carbono da biomassa microbiana (CBMS), carbono total (CTOTAL), nitrogênio (NTOTAL), respiração basal (RBS), quociente metabólico (qCO2), relação carbono da biomassa microbiana/ carbono total (CBMS:CTOTAL), relação carbono/nitrogênio (C/N), umidade gravimétrica (Ug), fósforo (P), potássio (K), cálcio (Ca), magnésio (Mg), alumínio (Al), capacidade de troca efetiva de cátions (CTC) e saturação por alumínio (m). O CBMS foi o atributo mais sensível para diferenciar as áreas do estudo e os períodos seco e chuvoso. Os atributos químicos de fertilidade do solo e a biomassa microbiana do solo apresentaram correlações significativas mais fortes e em maior número no período chuvoso. Os índices microbianos qCO2 e CBMS:CTOTAL comprovaram que áreas de plantio convencional podem ser relativamente eficientes em relação à dinâmica do C em comparação a área de floresta secundária.

Currently the mobile services represent an essential tool in daily life of the population. However, while offering greater convenience to its users, there is growing concern about the harmful effects to human health, derived from daily exposure of the public to electromagnetic fields from radio base stations (RBS), since even today, there is no study proving that longterm exposure to low-level fields are not harmful to health. In Presidente Prudente has not been a study reporting values of measurements of electromagnetic fields from base stations installed in the city. Based on these data, this study aimed to assess the levels of electromagnetic exposure in the city of Presidente Prudente regarding recommended by international bodies, as well as propose measures that can reduce public exposure to electromagnetic fields. For measuring values of electromagnetic fields, we used appliance Electromagnetic Field Meter Portable Digital - DRE-050, the Instrutherm, following the methodology suggested and adapted from the Adilza Condessa Dode’study. In total, 49 points were mapped corresponding to the areas at risk of exposure to electromagnetic fields generated by the substations of power grid, transmission towers and telecommunication towers located in the city of Presidente Prudente (SP)

Amorphous SiC(x)N(y) films have been deposited on (100) Si substrates by RF magnetron sputtering of a SiC target in a variable nitrogen-argon atmosphere. The as-deposited films were submitted to thermal anneling in a furnace under argon atmosphere at 1000 degrees C for 1 hour. Composition and structure of unannealed and annealed samples were investigated by RBS and FTIR. To study the electrical characteristics of SiC(x)N(y) films, Metal-insulator-semiconductor (MIS) structures were fabricated. Elastic modulus and hardness of the films were determined by nanoindentation. The results of these studies showed that nitrogen content and thermal annealing affect the electrical, mechanical and structural properties of SiC(x)N(y) films.

The present work is focused on the study of a seasonal anthropogenic influence on the beach of Itamambuca (Ubatuba, SP, Brazil) carried out using Atherinella brasiliensis as biomonitor. In total 84 fish were caught between July 2004 and February 2005 in different locations at the beach and inside the Itamambuca river. The fish were pooled according to catch and their musculature was analyzed by Particle-Induced X-ray Emission (PIXE) and Rutherford Backscattering (RBS) techniques. While the concentration of light (matrix) elements like C and O were obtained using the RBS technique, major (Na, Mg, P, S, Cl, K and Ca) and trace (Si, Al, Ti, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br and Sr) elements were measured by PIXE. The results show that differences were observed for several elemental concentrations of fish tissue between high season (spring-summer) and low season (winter-fall), indicating that increased human activity in the beach during high season may have some impact on the beach ecosystem. The role of the water salinity in the results is also discussed. (C) 2009 Elsevier B.V. All rights reserved.

Die vorgestellten Arbeiten bezüglich der Biosynthese von pflanzlichen Indolalkaloiden können in einen molekularbiologischen und einen proteinche-mischen Teil aufgegliedert werden. Im molekularbiologischen Abschnitt stand die Entwicklung eines Vektorsystems im Vordergrund, das die gleichzeitige Expression mehrerer Enzyme in bakteriellen Kulturen erlaubte. Hierfür konnte zunächst die cDNA der Strictosidin-Synthase aus Rauvolfia serpentina in den Expressionsvektor pQE-70 einkloniert und das Protein aktiv exprimiert werden. Bevor es zum Einbau des Strictosidin-β-D-Glucosidase-Gens –ebenfalls aus Rauvolfia serpentina– kam, musste dessen Aktivität mit einer vorgeschalteten Ribosomenbindestelle sichergestellt werden. Diese zusätzliche Binderegion wurde an das 5’-Ende der cDNA angefügt, um die ungestörte Expression des Enzyms im späteren Coexpressions-System zu gewährleisten. Im Hinblick auf weiterführende Arbeiten in Bezug auf die komplette in vitro-Synthese bereits bekannter Alkaloide, wie z.B. das antiarrhythmisch wirkende Ajmalin oder das antihypertensiv wirkende Heteroyohimbin-Alkaloid Raubasin, wurde die ursprüngliche multiple-clonig-site des verwendeten Expressionsvektors pQE-70 um 27 zusätzliche Restriktionsschnittstellen (verteilt auf 253 bp) erweitert. Nach erfolgreicher Ligation der Strictosidin-β-D-Glucosidase-cDNA mit vorgeschalteter Ribosomenbindestelle an das 3’-Ende des Strictosidin-Synthase-Gens gelang die heterologe Coexpression beider Enzyme in einer homogenen Suspensionskultur des E. coli-Expressionsstamms M15. Dafür wurde das Vektorkonstrukt pQE-70bh-STR-RBS-SG entwickelt. Das Endprodukt der anschließenden enzymatischen Umsetzung von Tryptamin und Secologanin wurde über das Zwischenrodukt Strictosidin gebildet und konnte als Cathenamin identifiziert werden. Im proteinchemischen Teil der Dissertation wurde die Reinigung einer Cathenamin-Reduktase aus Zellsuspensionskulturen von Catharanthus roseus RC mit dem Ziel der partiellen Bestimmung der Aminosäuresequenz bearbeitet. Das gesuchte Enzym wandelte in einer NADPH-abhängigen Reaktion das Edukt Cathenamin in Raubasin um. Des weiteren wurde untersucht, wie viele Enzyme insgesamt an der Umwandlung von Cathenamin zu Raubasin und den eng verwandten Produkten Tetrahydroalstonin und 19-Epi-Raubasin beteiligt waren. Unter Anwendung eines hierfür entwickelten säulenchromatographischen Protokolls gelang die Reinigung einer Raubasin-bildenden Reduktase, deren Teilsequenz jedoch noch nicht bestimmt werden konnte. Die Anzahl der beteiligten Enzyme bei der Ausbildung von Raubasin, Tetrahydroalstonin und 19-Epi-Raubasin konnte auf mindestens zwei beziffert werden.

Ocean drilling has revealed that, although a minor mineral phase, native Cu ubiquitously occurs in the oceanic crust. Cu isotope systematics for native Cu from a set of occurrences from volcanic basement and sediment cover of the oceanic crust drilled at several sites in the Pacific, Atlantic and Indian oceans constrains the sources of Cu and processes that produced Cu**0. We propose that both hydrothermally-released Cu and seawater were the sources of Cu at these sites. Phase stability diagrams suggest that Cu**0 precipitation is favored only under strictly anoxic, but not sulfidic conditions at circum-neutral pH even at low temperature. In the basaltic basement, dissolution of primary igneous and potentially hydrothermal Cu-sulfides leads to Cu**0 precipitation along veins. The restricted Cu-isotope variations (delta 65Cu = 0.02-0.19 per mil) similar to host volcanic rocks suggest that Cu**0 precipitation occurred under conditions where Cu+-species were dominant, precluding Cu redox fractionation. In contrast, the Cu-isotope variations observed in the Cu**0 from sedimentary layers yield larger Cu-isotope fractionation (delta 65Cu = 0.41-0.95 per mil) suggesting that Cu**0 precipitation involved redox processes during the diagenesis, with potentially seawater as the primary Cu source. We interpret that native Cu precipitation in the basaltic basement is a result of low temperature (20°-65 °C) hydrothermal processes under anoxic, but not H2S-rich conditions. Consistent with positive delta 65Cu signatures, the sediment cover receives major Cu contribution from hydrogenous (i.e., seawater) sources, although hydrothermal contribution from plume fallout cannot be entirely discarded. In this case, disseminated hydrogenous and/or hydrothermal Cu might be diagenetically remobilized and reprecipitated as Cu**0 in reducing microenvironment.

The thermal annealing of amorphous tracks of nanometer-size diameter generated in lithium niobate (LiNbO3) by Bromine ions at 45 MeV, i.e., in the electronic stopping regime, has been investigated by RBS/C spectrometry in the temperature range from 250°C to 350°C. Relatively low fluences have been used (<1012 cm−2) to produce isolated tracks. However, the possible effect of track overlapping has been investigated by varying the fluence between 3×1011 cm−2 and 1012 cm−2. The annealing process follows a two-step kinetics. In a first stage (I) the track radius decreases linearly with the annealing time. It obeys an Arrhenius-type dependence on annealing temperature with activation energy around 1.5 eV. The second stage (II) operates after the track radius has decreased down to around 2.5 nm and shows a much lower radial velocity. The data for stage I appear consistent with a solid-phase epitaxial process that yields a constant recrystallization rate at the amorphous-crystalline boundary. HRTEM has been used to monitor the existence and the size of the annealed isolated tracks in the second stage. On the other hand, the thermal annealing of homogeneous (buried) amorphous layers has been investigated within the same temperature range, on samples irradiated with Fluorine at 20 MeV and fluences of ∼1014 cm−2. Optical techniques are very suitable for this case and have been used to monitor the recrystallization of the layers. The annealing process induces a displacement of the crystalline-amorphous boundary that is also linear with annealing time, and the recrystallization rates are consistent with those measured for tracks. The comparison of these data with those previously obtained for the heavily damaged (amorphous) layers produced by elastic nuclear collisions is summarily discussed.

The lattice order degree and the strain in as-grown, Mn-implanted and post-implantedannealedInAsthinfilms were investigated with depth resolution by means of Rutherford backscattering spectrometry in channeling conditions (RBS/C). Three main crystallographic axes were analyzed for both In and As sublattices. The behaviour of the induced defects was evaluated in two regions with different native defects: the interface and the surface. The results show that Mn implantation and post-implantation annealing are anisotropic processes, affecting in a different way the In and As sublattices. The mechanisms influencing the enhancement and deterioration of the crystal quality during the implantation are discussed in relation to the as-grown defects and the segregation of the elements

The damage induced on quartz (c-SiO2) by heavy ions (F, O, Br) at MeV energies, where electronic stopping is dominant, has been investigated by RBS/C and optical methods. The two techniques indicate the formation of amorphous layers with an isotropic refractive index (n = 1.475) at fluences around 1014 cm−2 that are associated to electronic mechanisms. The kinetics of the process can be described as the superposition of linear (possibly initial Poisson curve) and sigmoidal (Avrami-type) contributions. The coexistence of the two kinetic regimes may be associated to the differential roles of the amorphous track cores and preamorphous halos. By using ions and energies whose maximum stopping power lies inside the crystal (O at 13 MeV, F at 15 MeV and F at 30 MeV) buried amorphous layer are formed and optical waveguides at the sample surface have been generated.
