Tässä diplomityössä on selvitetty Stora Enso Publication Papers Oy Ltd:n Summan tehtaiden ympäristömelun leviämistä tehtaiden ympärillä oleviin häiriintyviin kohteisiin. Ympäristömelua tutkittiin ympäristömelumittauksilla ja mallintamalla SoundPLAN –ohjelmalla. Työssä on ympäristömelun lisäksi tarkastelu työsuojelumielessä Summan tehtaiden voimalaitoksen melua. Työssä selvitettiin Summan tehtaiden merkittävimpien äänilähteiden äänitehotasot. Maastotietojen ja äänitehotasojen perusteella luotiin melumallit koko tehtaalle ja melua tuottaville osastoille. Melumallien avulla saatiin selville, miten melu leviää ympäristöön. Suurimpiin melulähteisiin, kuorimoon ja PK2:n ilmastointikoneisiin, laadittiin meluntorjuntaehdotukset ja tarkasteltiin uusien mallien avulla toimenpiteiden vaikutusta melun leviämiseen. Tulosten perusteella voidaan todeta, että Summan tehtaiden ympäristömelu ei ylitä lupa-arvoja häiriintyvissä kohteissa. Summan tehtaiden voimalaitoksella suoritettiin annosmelumittaukset ja laadittiin meluntorjuntaohjelma. Meluntorjuntaohjelmassa selvitettiin melun syyt, esitettiin meluntorjuntatoimenpiteitä ja niiden toteuttamissuunnitelma sekä ohjelman uusimisajankohta.

Aims:We searched for very high energy (VHE) γ-ray emission from the supernova remnant Cassiopeia A Methods: The shell-type supernova remnant Cassiopeia A was observed with the 17 m MAGIC telescope between July 2006 and January 2007 for a total time of 47 h. Results: The source was detected above an energy of 250 GeV with a significance of 5.2σ and a photon flux above 1 TeV of (7.3 ± 0.7_stat ± 2.2_sys) × 10-13 cm-2s-1. The photon spectrum is compatible with a power law dN/dE ∝ E-Γ with a photon index Γ = 2.3 ± 0.2_stat ± 0.2_sys. The source is point-like within the angular resolution of the telescope.

Background: Bone morphogenetic proteins (BMPs) have been shown to participate in the patterning and specification of several tissues and organs during development and to regulate cell growth, differentiation and migration in different cell types. BMP-mediated cell migration requires activation of the small GTPase Cdc42 and LIMK1 activities. In our earlier report we showed that activation of LIMK1 also requires the activation of PAKs through Cdc42 and PI3K. However, the requirement of additional signaling is not clearly known. Methodology/Principal Findings: Activation of p38 MAPK has been shown to be relevant for a number of BMP-2¿s physiological effects. We report here that BMP-2 regulation of cell migration and actin cytoskeleton remodelling are dependent on p38 activity. BMP-2 treatment of mesenchymal cells results in activation of the p38/MK2/Hsp25 signaling pathway downstream from the BMP receptors. Moreover, chemical inhibition of p38 signaling or genetic ablation of either p38¿ or MK2 blocks the ability to activate the downstream effectors of the pathway and abolishes BMP-2-induction of cell migration. These signaling effects on p38/MK2/Hsp25 do not require the activity of either Cdc42 or PAK, whereas p38/MK2 activities do not significantly modify the BMP-2-dependent activation of LIMK1, measured by either kinase activity or with an antibody raised against phospho-threonine 508 at its activation loop. Finally, phosphorylated Hsp25 colocalizes with the BMP receptor complexes in lamellipodia and overexpression of a phosphorylation mutant form of Hsp25 is able to abolish the migration of cells in response to BMP-2. Conclusions: These results indicate that Cdc42/PAK/LIMK1 and p38/MK2/Hsp25 pathways, acting in parallel and modulating specific actin regulatory proteins, play a critical role in integrating responses during BMP-induced actin reorganization and cell migration.

Large Hadron Collider (LHC) is the main particle accelerator at CERN. LHC is created with main goal to search elementary particles and help science investigate our universe. Radiation in LHC is caused by charged particles circular acceleration, therefore detectors tracing particles in existed severe conditions during the experiments must be radiation tolerant. Moreover, further upgrade of luminosity (up to 1035 cm-2s-1) requires development of particle detector’s structure. This work is dedicated to show the new type 3D stripixel detector with serious structural improvement. The new type of radiation-hard detector has a three-dimensional (3D) array of the p+ and n+ electrodes that penetrate into the detector bulk. The electrons and holes are then collected at oppositely biased electrodes. Proposed 3D stripixel detector demonstrates that full depletion voltage is lower that that for planar detectors. Low depletion voltage is one of the main advantages because only depleted part of the device is active are. Because of small spacing between electrodes, charge collection distances are smaller which results in high speed of the detector’s response. In this work is also briefly discussed dual-column type detectors, meaning consisting both n+ and p+ type columnar electrodes in its structure, and was declared that dual-column detectors show better electric filed distribution then single sided radiation detectors. The dead space or in other words low electric field region in significantly suppressed. Simulations were carried out by using Atlas device simulation software. As a simulation results in this work are represented the electric field distribution under different bias voltages.

Two new compounds, 5 and 8, and an epimeric mixture 4a/4b were isolated from hardwood of Auxemma glazioviana. This study describes the use of three (-)-alpha-pinene derivatives, one diol-1,2 [(-)-(1R, 2R, 3S, 5R)-2,6,6-trimethylbicyclo[3.1.1]heptane-2,3-diol 4] and two piridine-hydroxy derivatives [(+)-(1R,2S,3R,5S)-2,6,6-trimethyl-3-(2-pyridinylmethyl)bicyclo[3.1.1]heptan-3-ol 7 and (-)-(1R,2S,3R,5S)-2,6,6-trimethyl-3-[2-(2-pyridinyl) ethyl]bicyclo[3.1.1]heptan-3-ol 8]; one diol-1,3 [(-)-(1S,2R,5S)-2-(1-hydroxy-1-methylethyl)-5-methylcyclohexanol 5] derived from (+)-isopulegol 2 and one diol-1,3 [(+)-(1R,2R,5R)-2-(1-hydroxy-1-methylethyl)-5-methylcyclohexanol 6] derived from (+)-neo-isopulegol 3, as ligands in the asymmetric Reformatsky reaction. The best enantiomeric excess of beta-hydroxy ester obtained in the Reformatsky asymmetric reaction was 18% using ligand 6, and the chemical yield of the reactions was 65% on average.

A new flavone named 3,4',5,6-tetramethoxy-[2'',3'':7,8]furanoflavone besides the known flavonoids (2S,3R,4S)-3,4,5,8-tetramethoxy-[2'',3'':6,7]-furanoflavan, 3,6-dimethoxy-2'',2''-dimethylcromene-[2'',3'':7,8] -flavone, 3,5,6-trimethoxy-[2'',3'':7,8]-furanoflavone, 2,4',4,5-tetramethoxy-[2'',3'':6,7]-furanodihydroaurone, (2R,3S,4S)-3,4,5,6-tetramethoxy-[2'',3'':7,8]-furanoflavan and 3',4'-methylenodioxy-5,6-dimethoxy-[2'',3'':7,8]-furanoflavone were isolated from the root barks of Lonchocarpus campestris. The complete ¹H and 13C NMR assignments of the new furan flavonoid was performed using 1D and 2D pulse sequences, including COSY, HMQC and HMBC experiments.

Five monomers 5-[4-(5-cyano-4,5-dihydroisoxazol-3-yl)phenoxy]undecyl acrylate (7a); n-alkyl 3-{4-[5-(acryloyloxyundecyl)oxyphenyl]}-4,5-dihydroisoxazole-5-carboxylate (7b,c for n-butyl and n-hexyl, respectively); 3-{4-[5-(acryloyloxyundecyl) oxyphenyl]}-4,5-dihydroisoxazole-5-carboxylic acid (7d) and (1R,2S,5R)-2-isopropyl-5-methylcyclohexyl acrylate (9) and the corresponding copolymers 10a-d,11 and homopolymers 12 from 7a and 13 from 9 were designed and synthesized. Except for acrylate 9 which is derived from (-)-menthol, all of the monomers belong to the series containing the isoxazoline ring linked to the acrylate unit by a flexible spacer chain of eleven methylene units. They presented low glass temperature and despite birefringence behavior, these copolymers showed no mesomorphic properties.

O setor automotivo é um dos mais competitivos e um grande consumidor de materiais ferrosos, que exigem uma eficiente proteção contra a corrosão. Esta proteção é alcançada aplicando revestimentos adequados. Para se aplicar revestimentos se requer uma cuidadosa preparação da superfície que pode ser feita por ataque químico. Neste trabalho, determinou-se a velocidade de reação do aço extra-doce, SAE-AISI 1005, laminado a frio (CFF) utilizando várias soluções ácidas, de baixo custo, baixa agressividade ao meio ambiente e que geram pouco hidrogênio. O estudo cinético foi realizado medindo-se a perda de massa (deltam/mg cm-2) de chapas de aço CFF em função do tempo (s) de reação nas diferentes soluções, com e sem agitação, a 30ºC. Os resultados foram interpretados considerando-se os efeitos de transporte de massa, o efeito oxidante dos íons Me3+ e inibidor dos íons Fe2+. Dentre as várias soluções estudadas a que apresentou maior velocidade de reação foi a solução de H2SO4 2 M + MeCl3 1 M, v = 15,44 x 10-6 g cm-2s-1 para tempos até 1800 s.

O estudo foi realizado na floresta de galeria do Córrego Bacaba, no Parque Municipal do Bacaba (14º43'12,2S e 52º21'36,7"W), em Nova Xavantina, MT. A dispersão de sementes foi analisada e comparada entre três porções da floresta (alto, meio e baixo), distantes cerca de 200 m entre si, em um gradiente topográfico. Foram distribuídos, aleatoriamente, 20 coletores circulares de 0,3 m² em cada porção de floresta, sendo as coletas de sementes realizadas quinzenalmente, entre junho de 2007 e maio de 2008. O material foi submetido à secagem em estufa até peso constante e as sementes, separadas em duas categorias, baseando-se na presença de estruturas de voo (V) e na ausência dessas estruturas (NV). O número de morfoespécies e de sementes e a biomassa foram comparados entre as porções de floresta e entre os períodos seco e chuvoso. Coletaram-se 20.965 sementes em 24 quinzenas, sendo 777 do tipo V e 20.188 do tipo NV. Na porção do alto ocorreram 52 morfoespécies de sementes, no meio 55 e no baixo 49. A diversidade de morfoespécies de sementes foi inferior à diversidade de espécies de plantas lenhosas na mesma área. Considerando o padrão sazonal de dispersão de sementes, tanto para as V (número, morfoespécies e biomassa) quanto para as NV (biomassa), recomenda-se um controle no fluxo de visitantes no Parque do Bacaba, especialmente nos meses de pico de dispersão de sementes, no final do período seco e início do chuvoso (setembro-dezembro). Medidas de excursionismo de mínimo impacto poderão trazer benefícios para o estabelecimento de sementes e plântulas e garantir a sustentabilidade da floresta.

ABSTRACT The Paratudo (Tabebuia aurea) is a species occurring in the Pantanal of Miranda, Mato Grosso do Sul, Brazil, an area characterized by seasonal flooding. To evaluate the tolerance of this plant to flooding, plants aged four months were grown in flooded soil and in non-flooded soil (control group). Stomatal conductance, transpiration and CO2 assimilation were measured during the stress (48 days) and recovery (11 days) period, totalling 59 days. The values of stomatal conductance of the control group and stressed plants at the beginning of the flooded were 0.33 mol m-2s-1 and reached 0.02 mol m-2 s-1 (46th day) at the end of this event. For the transpiration parameter, the initial rate was 3.1 mol m s-1, and the final rate reached 0.2 or 0.3 mol m-2 s-1 (47/48 th day). The initial photosynthesis rate was 8.9 mmol m-2s-1 and oscillated after the sixth day, and the rate reached zero on the 48th day. When the photosynthesis rate reached zero, the potted plants were dried, and the rate was analyzed (11th day). The following values were obtained for dried plants: stomatal conductance = 0.26 mol m-2 s-1, transpiration rate = 2.5 mol m-2 s-1 and photosynthesis rate = 7.8 mmol m-2 s-1. Flooded soil reduced photosynthesis and stomatal conductance, leading to the hypertrophy of the lenticels. These parameters recovered and after this period, and plants exhibited tolerance to flooding stress by reducing their physiological activities.

O objetivo do experimento consistiu na simulação in vitro da soja transgênica tolerante ao glyphosate, através do cultivo de explantes em meios de cultura contendo aminoácidos aromáticos. As avaliações basearam-se nos efeitos do glyphosate sobre sementes oriundas de explantes de soja (Glycine max (L.) Merr.) cv. UFV-16. Para tanto, explantes de soja foram cultivados em meios de cultura líquidos com pH em torno de 5,0. Cada explante constou de um legume completamente expandido contendo duas sementes de aproximadamente 100 mg, conectada a um segmento de caule de 45 mm de comprimento. Os tratamentos testados foram: A = glutamina (Gln); B = Gln + fenilalanina (Phe) + tirosina (Tyr) + triptofano (Trp); C = Gln + glyphosate; D = Gln + Phe + Tyr + Trp + glyphosate. O experimento foi conduzido sob irradiância de 80 mmol-2s-1 a 25oC por 204 horas. Nos tratamentos que receberam aminoácidos aromáticos e glutamina, o herbicida não afetou as massas fresca e seca das sementes, como também, não afetou seus constituintes bioquímicos (proteínas, óleo, ácidos graxos, carboidratos e clorofilas). Portanto, a suplementação exógena de aminoácidos aromáticos suprime os efeitos fitotóxicos do glyphosate sobre explantes de soja, permitindo estudos sobre o seu modo de ação e metabolismo nas sementes, uma vez que os explantes se comportaram analogamente à soja transgênica não suscetível ao herbicida.

A enzima nitrato redutase (NR) catalisa a redução do nitrato a nitrito e controla a taxa de assimilação do nitrato. O ensaio in vitro da nitrato redutase foi otimizado para a linhagem selvagem (marrom, MA) e para a linhagem deficiente em ficoeritrina (verde-clara, VC) de Hypnea musciformis. As duas linhagens foram cultivadas em temperatura de 23 ± 2°C, fotoperíodo de 14 horas, irradiância de 60-90µmol fótons m-2s-1, e meio composto por água do mar esterilizada (30ups) enriquecida com a solução de von Stosch na concentração de 50% (VSES/2). As condições ótimas de ensaio para ambas as linhagens foram: 40µM de NADH; 10min de incubação do extrato bruto (EB) e 100µL de EB. A atividade ótima da NR ocorreu em 4 e 2mM de nitrato para a linhagem VC e MA, respectivamente. As linhagens VC e MA apresentaram, respectivamente, constante aparente de Michaelis-Menten (K M) para NADH de 0,2068 e 0,0837 µM, e K M para nitrato de 0,0492 e 0,0294mM. Os resultados indicam que a NR da linhagem MA tem maior afinidade pelo substrato do que a NR da linhagem VC de H. musciformis. Os experimentos para avaliar os efeitos da disponibilidade de nitrato (5 a 105µM) e nitrato e fosfato (0,5 a 25,5µM, com a relação N:P de 4:1) mostraram que a atividade da NR das linhagens VC e MA não aumentou com a adição de nitrato no meio, o que pode estar relacionado com o estado nutricional dessas algas. A atividade da NR foi maior nos tratamentos com adição de fosfato do que naqueles com adição de apenas nitrato, indicando que esse nutriente é importante para os processos metabólicos relacionados a atividade da NR.

The use of hydroxyurea (HU) can improve the clinical course of sickle cell disease. However, several features of HU treatment remain unclear, including the predictability of drug response and determination of adequate doses, considering positive responses and minimal side effects. In order to identify adequate doses of HU for treatment of sickle cell disease, 10 patients, 8 with sickle cell anemia and 2 with Sß thalassemia (8SS, 2Sß), were studied for a period of 6 to 19 months in an open label dose escalation trial (10 to 20 mg kg-1 day-1). Hemoglobin (Hb), fetal hemoglobin (Hb F) and mean corpuscular volume (MCV) values and reticulocyte, neutrophil and platelet counts were performed every two weeks during the increase of the HU dose and every 4 weeks when the maximum HU dose was established. Reduction in the number of vasoocclusive episodes was also considered in order to evaluate the efficiency of the treatment. The final Hb and Hb F concentrations, and MCV values were significantly higher than the initial values, while the final reticulocyte and neutrophil counts were significantly lower. There was an improvement in the concentration of Hb (range: 0.7-2.0 g/dl) at 15 mg HU kg-1 day-1, but this concentration did not increase significantly when the HU dose was raised to 20 mg kg-1 day-1. The concentration of Hb F increased significantly (range: 1.0-18.1%) when 15 mg HU was used, and continued to increase when the dose was raised to 20 mg kg-1 day-1. The final MCV values increased 11-28 fl (femtoliters). However, reticulocyte (range: 51-205 x 109/l) and neutrophil counts (range: 9.5-1.3 x 109/l) obtained at this dose were significantly lower than those obtained with 15 mg kg-1 day-1. All patients reported a decrease in frequency or severity of vasoocclusive episodes. These results suggest that a hydroxyurea dose of 15 mg kg-1 day-1 seems to be adequate for treatment of sickle cell disease in view of the minimal side effects observed and the improvement in laboratory and clinical parameters.

We describe here the isolation and characterization of a major albumin from the seeds of Opuntia ficus-indica (Cactaceae). This protein has a molecular mass of 6.5 kDa and was isolated by a combination of gel filtration chromatography and reverse-phase HPLC. The amino acid composition of this protein was determined and it was shown to have similarities with the amino acid composition of several proteins from the 2S albumin storage protein family. The N-terminal amino acid sequence of this protein is Asp-Pro-Tyr-Trp-Glu-Gln-Arg.
