Imbalance of IL-2, IFN-gamma and IL-10 secretion in the immunosuppression associated with human paracoccidioidomycosis

Patients with paracoccidioidomycosis (PCM) display a certain degree of immunecompromise characterized by lymphocyte hyporesponsiveness to the main Paracoccidioides brasiliensis antigen (gp43). To determine whether cytokines are involved in this state, we evaluated the secretion of IL-2, IL-10 and IFN-gamma by peripheral blood mononuclear cells (PBMC) from patients with the acute (AF) and chronic (CF) forms of PCM and from healthy, P. brasiliensis-sensitized subjects. gp43-stimulated PBMC from healthy subjects produced substantial amounts of IL-2, IFN-gamma and IL-10, whereas PBMC from AF and CF patients produced low levels of IL-2 and IFN-gamma but substantial amounts of IL-10, Phytohaemagglutinin-induced cytokine secretion was comparable among AF and CF patients and healthy subjects, suggesting integrity of non-specific cellular immune mechanisms in PCM. gp43-pulsed adherent cells, but not non-adherent cells, mere the main source of IL-10, Moreover, IL-2 and IFN-gamma secretion correlated inversely with the amount of specific antibodies produced by patients and healthy subjects. Our results suggest that the imbalance in cytokine production of patients with PCM plays a role in the gp43-hyporesponsiveness and the marked (non-protective) antibody production of these patients. Controlled hydrolysis of cheese whey proteins using trypsin and alpha-chymotrypsin

This study examined the production of protein hydrolysates with controlled composition from cheese whey proteins. Cheese whey was characterized and several hydrolysis experiments were made using whey proteins and purified beta -lactoglobulin, as substrates, and trypsin and a-chymotrypsin, as catalysts, at two temperatures and several enzyme concentrations. Maximum degrees of hydrolysis obtained experimentally were compared to the theoretical values and peptide compositions were calculated. For trypsin, 100% of yield was achieved; for alpha -chymotrypsin, hydrolysis seemed to be dependent on the oligopeptide size. The results showed that the two proteases could hydrolyze beta -lactoglobulin. Trypsin and alpha -chymotrypsin were stable at 40 degreesC, but a sharp decrease in the protease activity was observed at 55 degreesC.

Z gamma production and limits on anomalous ZZ gamma and Z gamma gamma couplings in pp collisions at root s=1.96 TeV

We present a study of eeγ and μμγ events using 1109 (1009) pb-1 of data in the electron (muon) channel, respectively. These data were collected with the DØ detector at the Fermilab Tevatron p over(p, ̄) collider at sqrt(s) = 1.96   TeV. Having observed 453 (515) candidates in the eeγ (μμγ) final state, we measure the Zγ production cross section for a photon with transverse energy ET > 7   GeV, separation between the photon and leptons Δ Rℓ γ > 0.7, and invariant mass of the di-lepton pair Mℓ ℓ > 30   GeV / c2, to be 4.96 ± 0.30 (stat . + syst .) ± 0.30 (lumi .)   pb, in agreement with the Standard Model prediction of 4.74 ± 0.22   pb. This is the most precise Zγ cross section measurement at a hadron collider. We set limits on anomalous trilinear Zγγ and ZZγ gauge boson couplings of - 0.085 < h30 γ < 0.084, - 0.0053 < h40 γ < 0.0054 and - 0.083 < h30 Z < 0.082, - 0.0053 < h40 Z < 0.0054 at the 95% C.L. for the form-factor scale Λ = 1.2   TeV.

Polymorphisms in Oxytocin and alpha(1a) Adrenergic Receptor Genes and Their Effects on Production Traits in Dairy Buffaloes

The use of molecular markers may auxiliary the buffalo breeding. The oxytocin (OXT) and the adrenergic receptor alpha(1A) (ADRA1A) may be involved in milk ejection in ruminants. The aim of this study was to verify the existence of polymorphisms in the OXT and ADRA1A genes and their associations with milk production traits. A total of 220 buffaloes were genotyped using PCR-RFLP for both genes. The SNP identified in the ADRA1A gene was associated with protein percentage in dairy buffaloes. This is the first report of such association in the literature, which has not been studied in other species.
