The Atlantic Forest biome, reduced to 12% of its original coverage, holds a significant portion of brazilian biodiversity, with high levels of endemism. Among the few remaining areas of the Atlantic, there is the region between the Complex of Cantareira and the municipality of Nazaré Paulista, which will be the object of study of this work. Using the concepts of Landscape Ecology, and analysis of thematic maps, this study aims to characterize the region in a ecologically scaled perspective, identifying two types of hotspots: i) for conservation and ii) for maintain habitat connectivity. Concepts of Landscape Ecology as landscape structure, patches, matrix, corridors (structural and functional) and connectivity, as well as their effects on local biodiversity was adopted and applied in the study region. We also analyzed the effects of roads, hydrographic system, Permanent Protection Areas (PPAs), and Conservation Units for flora and fauna maintenance at regional scale. This allowed us to define strategies and priority areas for the conservation and restoration of forest fragments of Atlantic Forest remnants within the study region. This study generated relevant knowledge for a better planning the region in order to create best conditions for the maintenance of regional biodiversity, and consequently, allowing to improve the quality of life for local population

Com a disseminação da agricultura industrial, com grande utilização de insumos tecnológicos, mecanizada e fundamentada num número reduzido de espécies e variedades de plantas cultivadas, os modos de produção autóctone têm sofrido pressões em todo mundo. Entre estas, citam-se a modernização do campo, a perda de terras para agricultura comercial em larga escala e a emigração dos jovens em busca de melhores condições de vida, que acarretam grandes mudanças para estes sistemas agrícolas. As comunidades de agricultores de subsistência são detentoras de uma gama de variedades de diversas espécies cultivadas, sendo que essa diversidade é um fator adaptativo importante para o agricultor ante as variações ambientais, climáticas, biológicas e socioeconômicas, além de ser importante para a segurança do sistema alimentar mundial. Isto faz com que estes agricultores se tornem importantes parceiros para programas de conservação in situ de plantas cultivadas. No Brasil, assentamentos rurais são formados por agricultores de diversas origens, e são locais onde a agricultura é parcialmente voltada para a subsistência. Dentre a grande diversidade de plantas cultivadas para subsistência, as raízes e os tubérculos constituem, depois dos cereais, a principal fonte de alimento para a população humana. O presente estudo visou levantar as espécies de raízes e tubérculos cultivados nos Assentamentos Rurais Araras I, II, III e IV, no município de Araras, São Paulo, e analisou a potencialidade dos pequenos agricultores das assentamentos como mantedores de agrobiodiversidade. A pesquisa se deu através de entrevistas estruturadas e semi-estruturadas, onde os agricultores entrevistados forma selecionados de maneira aleatória e estratificada entre os quatro assentamentos. En el año 2003 Brasil incorporó a su Estrategia Nacional ¨Fome Zero¨ el Programa Cisternas, iniciativa que comenzó a nivel local y cuyo objetivo es garantizar el acceso a agua para el consumo humano por medio de la construcción de un millón de cisternas rurales en todo el semiárido brasileño. Las cisternas de captación de agua de lluvia son una tecnología social de bajo coste y alto valor técnico, cultural y ambiental, destinada a garantizar agua de calidad a la población rural de baja renta y a mitigar los efectos que sobre ella tienen las sequias recurrentes y prolongadas que afectan al semiárido. Como parte del Programa Cisternas, la ONG brasileña “Instituto Ambiental Brasil Sustentável” (IABS) está apoyando el proyecto Cisternas Escolares, cuyo objetivo es construir cisternas de 52.000 litros de captación de agua de lluvia en 110 escuelas rurales del semiárido alagoano, y capacitar a las comunidades escolares para su correcta gestión. Este Trabajo Fin de Máster consiste en la evaluación de los efectos que dicho proyecto está teniendo sobre las condiciones de vida de las comunidades escolares rurales beneficiarias, aspirando a aportar información sobre los potenciales beneficios sociales de este tipo de actuaciones y a incrementar dichos beneficios proporcionando recomendaciones prácticas. De esta forma se pretende dar respuesta a la necesidad de conocimiento y mejora de los efectos que los proyectos de cisternas de captación de agua de lluvia tienen sobre sus beneficiarios, pero también a la demanda que está surgiendo desde la agenda del desarrollo, de evaluaciones de impacto más prácticas que sirvan para mejorar los proyectos que evalúan. La metodología propuesta en este trabajo adopta un enfoque mixto orientado a combinar el rigor de los datos cuantitativos, con el análisis de la cadena de efectos del proyecto y las percepciones de los beneficiarios. Dicha metodología consta de cuatro partes diferenciadas: i) la elaboración de la línea de base del proyecto; ii) un análisis causa – efecto cualitativo del proyecto, incluyendo las percepciones de los beneficiarios; iii) una evaluación de impacto que no pudo llevarse a cabo debido a retrasos en la ejecución del proyecto que desplomaron el potencial estadístico; y iv) un estudio de caso múltiple, realizado como alternativa a la evaluación de impacto, orientado a identificar diferencias entre las escuelas que reciben una cisternas y las que no. Los resultados de esta evaluación de efectos muestran que el proyecto Cisternas Escolares tiene potencial para producir mejoras en la salud, el funcionamiento, la autonomía, los usos del agua, la preocupación de las comunidades escolares y los conflictos con los vecinos de las escuelas beneficiarias. Dicho potencial es variable en cada una de las escuelas y depende de múltiples factores, los cuales son identificados y analizados en este trabajo. Fruto de este trabajo son también una serie de recomendaciones orientadas a que el proyecto Cisternas Escolares consiga alcanzar su máximo potencial en todas las escuelas beneficiarias. Estas recomendaciones pretenden mejorar las capacitaciones del proyecto y los consejos gestores del agua de las escuelas, así como incluir nuevas actividades relevantes en el proyecto, como la creación de espacios de participación para los actores y la involucración de las alcaldías y las secretarías de educación.

The general objective of this thesis was to stablish protocols to obtain and conserve agouti (Dasyprocta leporina) sperm bred in captivity in the Brazilian semi-arid, aiming its sustainable production. The thesis was divided in three experiments. In the first one, we studied the influence of the interaction between two probes (quadratics and sine waves) and two stimulation protocols (continuous and in series) on the agouti sperm collection by electroejaculation efficiency. The most efficient interaction on this obtainment was the one with probes with rings associated with stimuli in series (4/7; 57%, P<0.05). In the second experiment we compared the cryoprotectant effects of different substances (glycerol, ethyleneglycol, dimethylsulfoxide, dimethylformamide) on epididymis sperm cryopreservation. The highest values on motility (39.5±4.6%), vigor (2.9±0.2) and membrane integrity (30.6±4.5%) were observed on the samples cryopreserved using glycerol when compared to those with ethyleneglycol and dimethylformamide, but there was no difference (P>0.05) when compared to the samples cryopreserved with dimethylsulfoxide. At last, we studied the effects of the methods to obtain sperm (electroejaculation vs epididymal collection) on post thawing sperm quality. The samples obtained by epididymal retrograde flushing showed values for motility of 25.0±10.9% and vigor 2.4±0.8, and those obtained by electroejaculation had 31.2±14.2% of motility and vigor of 2.2±0.7, however, without statistical difference (P>0.05), which shows the possibility to successfully use the epididymal sperm cryopreservation protocol on agouti ejaculated sperm. In conclusion, significant advances on obtainment and processing of agouti sperm were made, allowing the establishment of germplasm banks from sperm samples obtained from the epididymis or by electroejaculation.

The umbu tree (Spondias tuberosa Arruda) is a fruit native to the northeast of Brazil with great economic, social and ecological importance for the northeastern semi-arid region. Despite its role, the umbu tree has suffered negative pressure thanks to cluttered extractivism and to negative selection of its fruits, which as the deforestation and the dormancy of seeds contribute to the decrease of its production year after year, making necessary studies that contribute to the improvement of this specie and its conservation. Given the risks to the conservation of the specie and its usefulness to the population, the association between plant biotechnology, for being a tool that can be used to increase its production. and the perception of gathering communities, by valuing the point of view and the knowledge of the population, can facilitate its conservation. This work aimed to develop methods of propagation for umbu tree as well as contribute to its conservation by using biotechnology, with specific objectives to contribute to the conservation of this species; determine concentrations of BAP and ANA in the formation of buds; testing the efficiency of different substrates and concentrations of gibberellic acid on germination in vitro and ex vitro, as well as capture the perception of families in communities that engage in the gathering of umbu. To study the germination, the seeds were inoculated in different substrates (vermiculite, vermiculite + clay, clay, clay + manure and manure + vermiculite) and in different concentrations of gibberellic acid (0 mg, 250 g and 500 mg). For the formation of buds BAP to 0.1 mg-1 was associated with different concentrations of ANA (0.2; 0.4; 0.8mg.L-1). The study of perception was conducted by applying semi-structured questionnaire with Malhada Vermelha community. The experiments resulted in vermiculite and concentration of 500 mg gibberellic acid as the best for germination. The association of 0.1 mg.L-1 of BAP to 0.2 mg.L-1 of ANA provided better formation of buds. As to the application of questionnaires, they revealed that the population understands the decreased amount of umbu plants and umbu fruit in the region, as well as shows concern for its conservation.

The spatial and temporal distribution of the population reflects the adjustment of their biological characteristics to environmental conditions and biotic interactions as adaptive and phylogenetic precursors elements. The habitat’s heterogeneity and alternating seasons tend to cause patterns of activity of organisms and species diversity. However, these seasonal and spatial patterns in butterfly communities in dry environments are not yet clear. We studied a community of frugivorous butterflies in ESEC Seridó, in northeastern Brazil, aiming to characterize the guild in semiarid and check the relative contribution of climate and vegetation variables on its composition, diversity and phenofaunistic. The butterflies were sampled monthly during one year, and the distribution of species was associated with structural characteristics of three vegetation types (eg. richness and abundance of tree and shrub species, canopy cover, herbaceous cover, litter) and climatological data (temperature, rainfall and humidity). We captured 9580 individuals of 16 species of butterflies belonging to four subfamilies (Biblidinae, Charaxinae, Nymphalinae and Satyrinae). The richness, abundance and diversity varied in different scales, especially in time, being higher in the rainy season, while the β-diversity and turnover was higher in the dry. The distribution of species mainly followed the changes in humidity, rainfall and vegetation phenology, with no defined boundaries between habitats. The flight period was shared within subfamilies, which should have distinct response to environmental stimuli, as well as respond to the phenology of host plants and have different reproductive strategies. There is even evidence of physiological and behavioral adaptations as seasonal reproduction and aestivation. So there was environmental control over the distribution and diversity of species, with the key role climate Association and vegetation structure in the community of differentiation in the seasons, and the availability and quality of resources on the variation of species abundance in small scales. These results may support the biomonitoring and conservation preserved areas, particularly in environments under human pressure and extreme environmental conditions such as semi-arid.

While the carnivores are considered regulators and structuring of natural communities are also extremely threatened by human activities. Endangered little-spotted-cat (Leopardus tigrinus) is one of the lesser known species from the Neotropical cats. In this work we investigate the occupancy and the activity pattern of L. tigrinus in Caatinga of Rio Grande do Norte testing: 1) how environmental and anthropogenic factors influence their occupation and 2) how biotic and abiotic factors influence their activity pattern. For this we raised occurrence data of species in 10 priority areas for conservation. We built hierarchical models of occupancy based on maximum likelihood to represent biological hypotheses which were ranked using the Akaike Information Criterion (AIC). According to the results the feline occupancy is more likely away from rural settlements and in areas with a higher proportion of woody vegetation. The opportunistic killing of L. tigrinus and in retaliation for poultry predation close to residential areas can explain this result; as well as more complex vegetation structure can better serve as refuge and ensure more food. Analyzing the records of the species through circular statistics we conclude that the activity pattern is mostly nocturnal, although considerable crepuscular and a small diurnal activity. L. tigrinus activity was directly affected by the availability of small terrestrial mammals, which are essentially nocturnal. In addition, the temperatures recorded in the environment directly and indirectly affect the activity of the little-spotted-cat, as also influence the activity of their potential prey. Generally, the cats were more active when possible prey were active, and this happened at night when lower temperatures are recorded. Moreover, the different lunar phases did not affect the activity pattern. The results improve the understanding of an endangered feline inhabiting the Caatinga biome, and thus can help develop conservation and management strategies, as well as in planning future research in this semi-arid ecosystem.

The farm’s rural dwellings of creation from the Seridó Potiguar microregion, built in the nineteenth century, became a reference by its vernacular character, i.e. these buildings, besides having recognized relevance to the identity of the region, they are adapted to the conditions of the place in many aspects (economic, cultural, construction, physical, et.) and consist in protective spaces in relation to hostile characteristics of Seridó’s climate. Considering the above premise, the following question arises: What characteristics of the nineteenth century Seridó Potiguar’s cattle farms are crucial for them to be a protective space in relation to the semiarid climate? In order to answer the question, this research aim to identify which particularities of the Seridó’s farmhouses that contribute to adaptability in these buildings to semiarid climate, as protection environments; and contribute to the stock valuation of the architectural heritage concerned. Therefore, procedures were adopted divided into two stages. Were first identified the recurring characteristics in the studied buildings, through typological study performed from existing inventories (DINIZ, 2008; FEIJÓ, 2002; IPHAN, 2012). To define the type it worked up with the concept that merges Durand’s analytical typology that identifies the similarities and differences to classify buildings, having the character of historical survey and architectural documentation, with the definition proposed by Argan (1963) that the type is not defined a priory, but the deduction from a number of illustrative cases which have formal and functional similarity with each other. Then worked up in a sample of five different types with each other, defined by the possibility of access to the interior of the houses, proximity to other copies, good state of conservation and preservation. The contemplated farms were: Pitombeiras, Agenus e Garrotes in Acari’s town, and the municipality of Caicó, Palma and Penedo. The second stage consists of the architectural survey, photographic record, digital three-dimensional modeling (aiming to expand the existing documentation and registration) and thermal monitoring over approximately a representative day in five farmhouses, relating the thermal performance of the houses with their individual characteristics. The selected variables for analysis monitoring are based on the thermal comfort adaptive model (SPAGNOLO and DE DEAR, 2003 apud NEGREIROS, 2010). The characteristics of the houses were analyzed as meeting the passive thermal conditioning strategies recommended by NBR 15220 (ABNT, 2005), for the bioclimatic zone 7 where the municipalities of Caicó and Acari are located. The house’s analysis of the operating temperatures revealed that 90% of the times of day the environments are within the comfort range. The farmhouses, which had a higher degree of compliance with recommended bioclimatic strategies, had the best thermal performance. In environments (usually the kitchen and rooms with low ceiling heights, exposed to west radiation) which still had discomfort hours, the thermal comfort can be reached with air movement approximately 1,0 m/s.

The human activities responsible for the ambient degradation in the modern world are diverse. The industrial activities are preponderant in the question of the impact consequences for brazilian ecosystems. Amongst the human activities, the petroliferous industry in operation in Potiguar Petroliferous Basin (PPB) displays the constant risk of ambient impacts in the integrant cities, not only for the human populations and the environment, but also it reaches the native microorganisms of Caatinga ground and in the mangrove sediment. Not hindering, the elaboration of strategies of bioremediation for impacted areas pass through the knowledge of microbiota and its relations with the environment. Moreover, in the microorganism groups associated to oil, are emphasized the sulfate-reducing prokaryotes (SRP) that, in its anaerobic metabolism, these organisms participate of the sulfate reduction, discharging H2S, causing ambient risks and causing the corrosion of surfaces, as pipelines and tanks, resulting in damages for the industry. Some ancestries of PRS integrate the Archaea domain, group of microorganisms whose sequenced genomes present predominance of extremophilic adaptations, including surrounding with oil presence. This work has two correlated objectives: i) the detection and monitoring of the gene dsrB, gift in sulfate-reducing prokaryotes, through DGGE analysis in samples of mDNA of a mangrove sediment and semiarid soil, both in the BPP; ii) to relate genomic characteristics to the ecological aspects of Archaea through in silico studies, standing out the importance to the oil and gas industry. The results of the first work suggest that the petrodegraders communities of SRP persist after the contamination with oil in mangrove sediment and in semiarid soil. Comparing the populations of both sites, it reveals that there are variations in the size and composition during one year of experiments. In the second work, functional and structural factors are the probable cause to the pressure in maintenance of the conservation of the sequences in the multiple copies of the 16S rDNA gene. Is verified also the discrepancy established between total content GC and content GC of the same gene. Such results relating ribosomal genes and the ambient factors are important for metagenomic evaluations using PCR-DGGE. The knowledge of microbiota associated to the oil can contribute for a better destination of resources by the petroliferous industry and the development of bioremediation strategies. Likewise, search to lead to the best agreement of the performance of native microbiota in biogeochemical cycles in Potiguar Petroliferous Basin ecosystem

O estado de Santa Catarina possuía sua área inteiramente coberta por Mata Atlântica, um dos biomas mais ameaçados mundialmente. Mesmo com o crescente aumento de informações sobre os anfíbios no estado, ainda existe muitas lacunas de informações para este grupo e o desconhecimento é alto mesmo no interior das Unidades de Conservação. O Parque Estadual da Serra do Tabuleiro é a maior unidade de conservação de proteção integral de Santa Catarina e ainda assim possui inúmeras ameaças e pressões antrópicas. Os objetivos deste estudo foram estudar os anfíbios anuros em duas fitofisionomias, restinga e floresta ombrófila densa submontana, do Parque Estadual da Serra do Tabuleiro, verificar as diferenças dos parâmetros da comunidade entre três mesohábitats na restinga, compreender em que extensão algumas variáveis estruturais do hábitat afetam a riqueza e abundância dos anuros na restinga e na floresta ombrófila densa submontana, conhecer os anuros de folhiço na área de floresta e, adicionalmente, verificar o atual conhecimento sobre os anuros de folhiço no mundo. Os anuros no parque foram amostrados por meio dos métodos de procura ativa em transecções, armadilhas de queda com cerca guia e parcelas no chão da floresta durante três anos consecutivos com amostragens trimestrais, de julho de 2007 a abril de 2010. As variáves estruturais foram medidas nos locais amostrados enquanto que os dados climáticos foram obtidos da estação meteorológica mais próxima da área de estudo. A revisão sobre os anuros de folhiço no mundo foi realizada por uma busca na base de dados ISI Web if Science entre os anos de 1945 e 2008. Foram registradas 39 espécies de anuros no Parque, sendo 15 espécies para a restinga, 31 espécies na floresta. No folhiço do chão da floresta, a comunidade foi composta por 13 espécies de anuros. No total, 66% dos anuros registrados aqui, foram endêmicos da Mata Atlântica, 17% possuíram distribuição restrita à Santa Catarina e quatro espécies foram consideradas como vulneráveis na lista de espécies ameaçadas de extinção de Santa Catarina. Na restinga, a área aberta e a mata de restinga foram as mais dissimilares com relação à composição e abundância de anuros e também para as variáveis estruturais do hábitat. Na floresta ombrófila densa, a cobertura de dossel e o número de corpos dágua foram as variáveis mais importantes para a riqueza e abundância das espécies de anuros. Também para os anuros de folhiço do parque, a cobertura de dossel foi importante, conjuntamente com a umidade do ar e a profundidade de folhiço. O atual conhecimento sobre os anuros de folhiço no mundo se concentra nas regiões tropicais, sendo que o Brasil possui o maior número de estudos. Algumas tendências para as comunidades de anuros de folhiço puderam ser identificadas, mas estudos adicionais são necessários para que mais inferências possam ser feitas. O presente estudo contribuiu para preencher parte das lacunas de conhecimento existentes para a biodiversidade dos anuros do estado de Santa Catarina e em especial para o Parque Estadual da Serra do Tabuleiro.
