This article, about reflections on the condition of Brazilian women from the Colony to the first decades of the twentieth century, reveals the historical position of them and the attitudes and behaviors related to gender and sexuality. Subdued, it was treated as a sexual object, arousing all sorts of misogyny by men. Rebel, veiled or ostensibly, could serve their own desires. Throughout history, the Church and medical institutions which jointly accounted for, significantly, established the meaning and place of women. In Colony period, the woman is a ward from the Catholic ideology, but from the nineteenth century, after Independence, this power control arises to Medicine. The physician submits the religious discourse, naturalizing the status of women as one that breeds, namely the insertion of the medical issues of family scientifically legitimate colonial patriarchy. This is accentuated in the early twentieth century, when medicine consolidated setting standards and rules for marriage, to motherhood and family life. We note how the feminine universe was (and it is nowadays) ambivalent, with "one foot" in virtue and another in sin, with a tendency to contain and another to trespass. On the one hand we have the home and motherhood, validated in marriage, in which the woman is cared for and dependent on her husband. Reflecting on the motherhood of Virgin Mary, comes to the sacred dimension of the idealized woman saint by the Church. At the same time, however, feels the need for freedom, identity and independence, needing to give a voice to the desire to have their sexuality and all that it is due in full. The manifestation of the desire and the call for sexual satisfaction, and put in permanent conflict personal, psychological and social split between moral entrenched across generations and cultural transformations resulting from decades of the 20th Century.

The microstructural behavior of industrial standardized cocoa butter samples and cocoa butter samples from three different Brazilian states is compared. The cocoa butters were characterized by their microstructural patterns, crystallization kinetics and polymorphic habits. The evaluation of these parameters aided in establishing relationships between the chemical compositions and crystallization behavior of the samples, as well as differentiating them in terms of technological and industrial potential for use in tropical regions.

A comparative study of the primary properties of six cocoa butter samples, representative of industrial blends and cocoa butter extracted from fruits cultivated in different geographical areas in Brazil is presented. The samples were evaluated according to fatty acid composition, triacylglycerol composition, regiospecific distribution, melting point, solid fat content and consistency. The results allowed for differentiating the samples according to their chemical compositions, thermal resistance properties, hardness characteristics, as well as technological adequacies and potential use in regions with tropical climates.

Se cultivó alga roja Gratelupia doryphora en agua de mar enriquecida con Provasoli y en glicerol. La incubación en agua de mar condujo a un aumento en el porcentaje de los ácidos grasos polünsaturados, mientras que el glicerol incrementó el contenido de lípidos totales. Por tanto, si la alga está siendo producida como un cultivo heterotrófico, es posible aumentar la biomasa de la misma así como el contenido de ácidos grasos biológicamente activos. The red alga Gratelupia doryphora was cultivated in Provasoli enricher (plain) seawater and in a glycerol media. The incubation in seawater leads to an increase in the percentage of polyunsaturated acids, while the glycerol increase the total lipid content. If the alga is being grown as a heterotrophic culture, it is possible to increase the alga biomass as well as the content of biologically-active fatty acids.

Synthetic biology is a young field of applicative research aiming to design and build up artificial biological devices, useful for human applications. How synthetic biology emerged in past years and how the development of the Registry of Standard Biological Parts aimed to introduce one practical starting solution to apply the basics of engineering to molecular biology is presented in chapter 1 in the thesis The same chapter recalls how biological parts can make up a genetic program, the molecular cloning tecnique useful for this purpose, and an overview of the mathematical modeling adopted to describe gene circuit behavior. Although the design of gene circuits has become feasible the increasing complexity of gene networks asks for a rational approach to design gene circuits. A bottom-up approach was proposed, suggesting that the behavior of a complicated system can be predicted from the features of its parts. The option to use modular parts in large-scale networks will be facilitated by a detailed and shared characterization of their functional properties. Such a prediction, requires well-characterized mathematical models of the parts and of how they behave when assembled together. In chapter 2, the feasibility of the bottom-up approach in the design of a synthetic program in Escherichia coli bacterial cells is described. The rational design of gene networks is however far from being established. The synthetic biology approach can used the mathematical formalism to identify biological information not assessable with experimental measurements. In this context, chapter 3 describes the design of a synthetic sensor for identifying molecules of interest inside eukaryotic cells. The Registry of Standard parts collects standard and modular biological parts. To spread the use of BioBricks the iGEM competition was started. The ICM Laboratory, where Francesca Ceroni completed her Ph.D, partecipated with teams of students and Chapter 4 summarizes the projects developed.

Melittin, Hauptbestandteil des Bienengifts, ist ein kationisches Peptid, welches in der Lage ist, die biophysikalischen Eigenschaften der Zellmembran zu beeinflussen. Melittin werden unter anderem auch entzündungshemmende, schmerzlindernde, anti-rheumatische und anti-arthritische Wirkungen zugeschrieben. rnIn dieser Arbeit wurde nachgewiesen, dass Melittin die Proteolyse von ADAM10- und ADAM17-Substraten in verschiedenen Zellen stimuliert. Durch das Sheddingvon TGF-α wurde in HaCaT-Keratinozyten eine Transaktivierung des EGF-Rezeptors und eine daraus resultierende Phosphorylierung der Kinase ERK1/2 beobachtet. Die durch Melittin gesteigerte Aktivität der ADAMs ist calciumunabhängig und wird nicht durch Änderungen in der Membranfluidität verursacht. Eine Beteiligung der P2-Rezeptoren an der Melittin-induzierten ADAM-Aktivierung konnte sowohl durch Inhibition der Rezeptoren als auch durch Transfektion von HEK-Zellen mit dem P2X7-Rezeptor nachgewiesen werden. In diesen wurde nach der Behandlung mit Melittin eine Phosphorylierung von ERK1/2 beobachtet, welche durch ATPasen und P2-Rezeptor-Inhibitoren unterdrückt werden konnte. rnMit Hilfe des Kaninchenerythrozyten-Modells wurde nachgewiesen, dass eine Translokation von Phosphatidylserin von der Innen- zur Außenseite der Membran unmittelbar mit einer erhöhten ADAM-Aktivität korreliert. Sowohl durch Aktivierung des P2X7-Rezeptors als auch durch die Behandlung der Zellen mit dem Ionophor A23187 konnte ein Phosphatidylserin-Flip induziert werden. Dieser Flip führte zu einer erhöhten Aktivität von ADAM10, die durch eine gesteigerte Hämolyse und Spaltung von pVCC nachgewiesen werden konnte. Wurde der Phosphatidylserin-Flip durch Inhibitoren des P2X7-Rezeptors bzw. die Chelation von Ca2+ und Hemmung der Ionenfluxe unterdrückt, blieb auch die erhöhte ADAM-Aktivität aus. Wurde dagegen der Phosphatidylserin-Flip erst induziert und nachträglich die Inhibition des P2X7-Rezeptors bzw. die Chelation von Ca2+ und Hemmung der Ionenfluxe durchgeführt, zeigte dies keine Inhibition der ADAM-Aktivität.rnZusammenfassend zeigen diese Ergebnisse, dass eine Exposition von Phosphatidylserin auf der Außenseite der Membran in einem kausalen Zusammenhang mit einer gesteigerten ADAM-Aktivität steht.rn
