Hybrid materials with enhanced properties can now be obtained by combining nanomaterials such as carbon nanotubes and metallic nanoparticles, where the main challenge is to control fabrication conditions. In this study, we demonstrate that platinum nanoparticles (PtNps) can be electrogenerated within layer-by-layer (LbL) films of polyamidoamine (PAMAM) dendrimers and single-walled carbon nanotubes (SWCNTs), which serve as stabilizing matrices. The advantages of the possible control through electrogeneration were demonstrated with a homogeneous distribution of PtNps over the entire surface of the PAMAM/SWCNT LbL films, whose electroactive sites could be mapped using magnetic force microscopy. The Pt-containing films were used as catalysts for hydrogen peroxide reduction, with a decrease in the reduction potential of 60 mV compared to a Pt film deposited onto bare ITO. By analyzing the mechanisms responsible for hydrogen peroxide reduction, we ascribed the enhanced catalytic activity to synergistic effects between platinum and carbon in the LbL films, which are promising for sensing and fuel cell applications.

Das Hauptziel dieser Arbeit war die Identifizierung der Regulationsebenen auf denen die TPA-induzierte Matrix-Metalloproteinase-9 (MMP-9) durch das nitrose Gas Stickstoffmonoxid (NO) in MCF-7-Zellen verändert wird. Dabei konnte sowohl mit Hilfe der Zymographie als auch mit einem MMP-9-Aktivitäts-ELISA gezeigt werden, dass die extrazellulären MMP-9-Spiegel durch die Behandlung der Zellen mit NO reduziert werden. Gleichzeitig zeigte sich auch eine durch NO bedingte Abnahme der intrazellulären MMP-9-Spiegel, wie mit Hilfe von Western-Blot-Analyse nachgewiesen werden konnte. Experimente mit dem Proteasominhibitor Lactacystin und dem Proteinsynthesehemmstoff Cycloheximid ließen darüber hinaus eine NO-bedingte Veränderung der MMP-9-Proteinstabilität ausschließen. Im Gegensatz dazu konnte mittels der metabolischen Markierung mit radioaktiv markiertem Methionin und Cystein gezeigt werden, dass die Proteinneusynthese der MMP-9 durch eine Behandlung der Zellen mit NO stark beeinträchtigt wird. In Übereinstimmung mit diesen Daten finden sich reduzierte MMP-9-mRNA-Spiegel auch in der polysomalen Zellfraktion von MCF-7-Zellen. Wie mit Hilfe des Transkriptionshemmstoffes Actinomycin D und durch Reportergenstudien mit hybriden MMP-9-Promotorkonstrukten gezeigt werden konnte, ist die NO-induzierte Reduktion der MMP-9-mRNA-Spiegel nicht auf eine Verringerung der MMP-9-mRNA-Stabilität zurückzuführen. Reportergenstudien mit einem 670bp langen Promotorfragment des 5’flankierenden Bereichs des humanen MMP-9-Gens zeigten jedoch auf, dass der hemmende Effekt des NOs zum Teil auf eine NO-vermittelte Abnahme der TPA-induzierten MMP-9-Promotoraktivität zurückgeführt werden kann. Demzufolge wurde in den nachfolgenden Experimenten nach den für die MMP-9-Expression notwendigen und von NO modulierten Transkriptionsfaktoren in MCF-7-Zellen gesucht. Anhand von Western-Blot-Analysen und Gelshiftanalysen konnte gezeigt werden, dass die Aktivität des Transkriptionsfaktors AP-1 in MCF-7-Zellen durch NO gehemmt wird, während weder die Expressionspiegel noch die Bindungsaffinität der Transkriptionsfaktoren NFκB und Sp1 durch die NO-Behandlung verändert sind. Weiterhin konnte unter Verwendung von pharmakologischen Inhibitoren der MAPK-Signalwege mit Hilfe der Western-Blot-Analyse nachgewiesen werden, dass MAPK-vermittelte Signalwege zwar für die Induktion der MMP-9-Expression essenziell sind, diese jedoch nicht von NO beeinflusst sind. Im Unterschied hierzu konnte mit Hilfe eines PKC-Aktivitätsassays gezeigt werden, dass die Gesamtaktivität von PKCs nach Behandlung von MCF-7-Zellen mit NO signifikant gehemmt ist. Zusammenfassend zeigen diese Untersuchungen, dass die NO-vermittelte Hemmung der TPA-induzierten MMP-9-Expression in MCF-7-Zellen im Wesentlichen auf eine NO-abhängige Reduktion der Protein-Kinase-C-Aktivität und einer daraus resultierenden Aktivitätshemmung des Transkriptionsfaktors AP-1 zurückgeführt werden kann.

Das aSPECT Spektrometer wurde entworfen, um das Spektrum der Protonen beimrnZerfall freier Neutronen mit hoher Präzision zu messen. Aus diesem Spektrum kann dann der Elektron-Antineutrino Winkelkorrelationskoeffizient "a" mit hoher Genauigkeit bestimmt werden. Das Ziel dieses Experiments ist es, diesen Koeffizienten mit einem absoluten relativen Fehler von weniger als 0.3% zu ermitteln, d.h. deutlich unter dem aktuellen Literaturwert von 5%.rnrnErste Messungen mit dem aSPECT Spektrometer wurden an der Forschungsneutronenquelle Heinz Maier-Leibnitz in München durchgeführt. Jedoch verhinderten zeitabhängige Instabilitäten des Meßhintergrunds eine neue Bestimmung von "a".rnrnDie vorliegende Arbeit basiert hingegen auf den letzten Messungen mit dem aSPECTrnSpektrometer am Institut Laue-Langevin (ILL) in Grenoble, Frankreich. Bei diesen Messungen konnten die Instabilitäten des Meßhintergrunds bereits deutlich reduziert werden. Weiterhin wurden verschiedene Veränderungen vorgenommen, um systematische Fehler zu minimieren und um einen zuverlässigeren Betrieb des Experiments sicherzustellen. Leider konnte aber wegen zu hohen Sättigungseffekten der Empfängerelektronik kein brauchbares Ergebnis gemessen werden. Trotzdem konnten diese und weitere systematische Fehler identifiziert und verringert, bzw. sogar teilweise eliminiert werden, wovon zukünftigernStrahlzeiten an aSPECT profitieren werden.rnrnDer wesentliche Teil der vorliegenden Arbeit befasst sich mit der Analyse und Verbesserung der systematischen Fehler, die durch das elektromagnetische Feld aSPECTs hervorgerufen werden. Hieraus ergaben sich vielerlei Verbesserungen, insbesondere konnten die systematischen Fehler durch das elektrische Feld verringert werden. Die durch das Magnetfeld verursachten Fehler konnten sogar soweit minimiert werden, dass nun eine Verbesserung des aktuellen Literaturwerts von "a" möglich ist. Darüber hinaus wurde in dieser Arbeit ein für den Versuch maßgeschneidertes NMR-Magnetometer entwickelt und soweit verbessert, dass nun Unsicherheiten bei der Charakterisierung des Magnetfeldes soweit reduziert wurden, dass sie für die Bestimmung von "a" mit einer Genauigkeit von mindestens 0.3% vernachlässigbar sind.

Diese Arbeit befasst sich mit der Rolle der extrazellulären Matrix und insbesondere des Proteins Fibronektin bei der Leberfibrose und bei der Einnistung von Tumorzellen in die Stammzellnische im Knochenmark.rnrnHierfür wurde in einem Fibrosemodell das Peptid pUR4b verwendet, welches die Assemblierung einer Fibronektinmatrix verhindert. Bei der Verwendung dieses Peptids während und nach der Induktion einer Leberfibrose durch die Chemikalie Dimethylnitrosamin konnte eine Verminderung der Kollagenmenge (und damit des fibrotischen Narbengewebes) in der Leber im Vergleich zu fibrotischen Kontrolltieren beobachtet werden. Darüber hinaus konnte gezeigt werden, dass dieser Effekt unabhängig von der Aktivierung der hepatischen Stellatezellen ist, jedoch zum Teil von einer verminderten Anzahl entzündlicher Zellen abhängig sein könnte. Eine verminderte Bildung von Gesamt- und aktivem TGF-β, welche zum Teil auf Effekte der verringerten Zahl der inflammatorischen Zellen zurückzuführen sein könnte, unterstützt den Effekt des verminderten Aufbaus von Narbengewebe. In vitro Untersuchungen zeigten, dass hepatische Stellatezellen bei einer Behandlung mit pUR4b weniger Fibronektin in die extrazelluläre Matrix einbauten als unbehandelte hepatische Stellatezellen. Insgesamt sprechen die Daten dafür, dass das Peptid pUR4b den Aufbau einer Fibronektinmatrix verhinderte bzw. verminderte, wodurch die Ablagerung anderer Komponenten der extrazellulären Matrix wie z.B. Kollagen gestört war und es daher zu einem Rückgang des fibrotischen Narbengewebes kam.rnrnFür die Untersuchung des Einflusses der extrazellulären Matrix und des Fibronektins bei der Einnistung von Tumorzellen wurde zunächst das Fibronektin mit Hilfe konditioneller Knockout-Mäuse in verschiedenen Zellen bzw. Organen der Tiere ausgeschaltet. Weder die Ausschaltung des zirkulierenden, noch des durch Osteoblasten und Osteozyten gebildeten, noch des zirkulierenden und von Zellen des Knochenmarks gebildeten Fibronektins beeinträchtigte die Einnistung von Tumorzellen. Auch die Bildung eines Hämatoms im Knochen hatte weder einen Einfluss auf die Einnistung von Tumorzellen noch auf die spätere Tumorentwicklung. Die Ausschaltung des tumorzellendogenen Fibronektins führte hingegen zu einer signifikant verminderten Einnistung von Tumorzellen. Diese ist wahrscheinlich auf die verstärkte Affinität dieser Tumorzellen zu Zellen des Immunsystems zurückzuführen. Diese Beobachtung konnte zum Teil durch eine verstärkte eCadherin Expression erklärt werden, welche die Bindung an verschiedene Zellen des Immunsystems vermittelt. Eine Untersuchung der osteoblastischen Stammzellnische durch die kombinierte Gabe von Parathormon und Zoledronsäure führte zu keiner Veränderung der Fibronektinkonzentration innerhalb des Knochenmarks der behandelten Tiere. Dennoch nisteten sich in dem Knochenmark der mit Parathormon und Zoledronsäure behandelten Tiere signifikant mehr Tumorzellen ein als in dem von Kontrolltieren. Dieser Effekt konnte auf einen synergetischen Effekt von Parathormon und Zoledronsäure zurückgeführt werden, der zu einer gesteigerten Osteoblastenaktivität und Änderungen der Zytokinkonzentrationen im Knochenmark führte.rnZusammenfassend zeigte sich, dass eine Veränderung der extrazelluläre Matrix und insbesondere des Proteins Fibronektin bei Leberfibrose zu einem veränderten Krankheitsbild führt und die Einnistung von Tumorzellen in das Knochenmark beeinflusst.rn

BACKGROUND: There are still limited data on the outcomes of regenerative periodontal surgery using a combination of an enamel matrix protein derivative (EMD) and autogenous bone (AB). AIM: To evaluate the healing of deep intrabony defects treated with either a combination EMD+AB or EMD alone. MATERIALS AND METHODS: Forty patients with advanced chronic periodontitis, with one deep intrabony defect, were randomly treated with either EMD+AB (test) or EMD (control). Clinical assessments were performed at baseline and at 1 year after treatment. The primary outcome variable was relative attachment level (RAL). RESULTS: Healing was uneventful in all patients. The test sites showed a reduction in the mean probing pocket depth (PPD) of 5.6 +/- 0.9 mm (p<0.001), a gain in the mean RAL of 4.2 +/- 1.1 mm (p<0.001) and a gain in the mean probing bone level (PBL) of 3.9 +/- 1.0 mm (p<0.001). The control group displayed a mean PPD reduction of 4.6 +/- 0.4 mm (p<0.001), a mean RAL gain of 3.4 +/- 0.8 mm (p<0.001) and a mean PBL gain of 2.8 +/- 0.8 mm (p<0.001). RAL gains of > or =4 mm were measured in 90% of the test defects and in 55% of the controls. PBL gains of > or =4 mm were obtained in 85% of the test defects and in 25% of the control ones. The test treatment resulted in statistically higher PPD reductions, RAL gains and PBL gains compared with the control (p<0.01). CONCLUSIONS: Within their limits, the present results indicate that: (i) at 1 year after surgery, both therapies resulted in statistically significant clinical improvements compared with baseline and (ii) although the combination of EMD+AB resulted in statistically significant higher soft and hard tissue improvements compared with treatment with EMD, the clinical relevance of this finding is unclear.

Postmortem investigation is increasingly supported by computed tomography (CT) and magnetic resonance imaging, in which postmortem minimal invasive angiography has become important. The newly introduced approach using an aqueous contrast agent solution provided excellent vessel visualization but was suspected to possibly cause tissue edema artifacts in histological investigations. The aim of this study was to investigate on a porcine heart model whether it is possible to influence the contrast agent distribution within the soft tissue by changing its viscosity by dissolving the contrast agent in polyethylene glycol (PEG) as a matrix medium. High-resolution CT scans after injection showed that viscosities above c. 15 mPa s (65% PEG) prevented a contrast agent distribution within the capillary bed of the left ventricular myocardium. Thereby, the precondition of edema artifacts could be reduced. Its minimal invasive application on human corpses needs to be further adapted as the flow resistance is expected to differ between different tissues.

It is generally agreed that the mechanical environment of intervertebral disc cells plays an important role in maintaining a balanced matrix metabolism. The precise mechanism by which the signals are transduced into the cells is poorly understood. Osmotic changes in the extracellular matrix (ECM) are thought to be involved. Current in-vitro studies on this topic are mostly short-term and show conflicting data on the reaction of disc cells subjected to osmotic changes which is partially due to the heterogenous and often substantially-reduced culture systems. The aim of the study was therefore to investigate the effects of cyclic osmotic loading for 4 weeks on metabolism and matrix gene expression in a full-organ intervertebral disc culture system. Intervertebral disc/endplate units were isolated from New Zealand White Rabbits and cultured either in iso-osmotic media (335 mosmol/kg) or were diurnally exposed for 8 hours to hyper-osmotic conditions (485 mosmol/kg). Cell viability, metabolic activity, matrix composition and matrix gene expression profile (collagen types I/II and aggrecan) were monitored using Live/Dead cell viability assay, tetrazolium reduction test (WST 8), proteoglycan and DNA quantification assays and quantitative PCR. The results show that diurnal osmotic stimulation did not have significant effects on proteoglycan content, cellularity and disc cell viability after 28 days in culture. However, hyperosmolarity caused increased cell death in the early culture phase and counteracted up-regulation of type I collagen gene expression in nucleus and annulus cells. Moreover, the initially decreased cellular dehydrogenase activity recovered with osmotic stimulation after 4 weeks and aggrecan gene down-regulation was delayed, although the latter was not significant according to our statistical criteria. In contrast, collagen type II did not respond to the osmotic changes and was down-regulated in both groups. In conclusion, diurnal hyper-osmotic stimulation of a whole-organ disc/endplate culture partially inhibits a matrix gene expression profile as encountered in degenerative disc disease and counteracts cellular metabolic hypo-activity.

The purpose of this study was to evaluate the neuroimaging quality and accuracy of prospective real-time navigator-echo acquisition correction versus untriggered intrauterine magnetic resonance imaging (MRI) techniques. Twenty women in whom fetal motion artifacts compromised the neuroimaging quality of fetal MRI taken during the 28.7 +/- 4 week of pregnancy below diagnostic levels were additionally investigated using a navigator-triggered half-Fourier acquired single-shot turbo-spin echo (HASTE) sequence. Imaging quality was evaluated by two blinded readers applying a rating scale from 1 (not diagnostic) to 5 (excellent). Diagnostic criteria included depiction of the germinal matrix, grey and white matter, CSF, brain stem and cerebellum. Signal-difference-to-noise ratios (SDNRs) in the white matter and germinal zone were quantitatively evaluated. Imaging quality improved in 18/20 patients using the navigator echo technique (2.4 +/- 0.58 vs. 3.65 +/- 0.73 SD, p < 0.01 for all evaluation criteria). In 2/20 patients fetal movement severely impaired image quality in conventional and navigated HASTE. Navigator-echo imaging revealed additional structural brain abnormalities and confirmed diagnosis in 8/20 patients. The accuracy improved from 50% to 90%. Average SDNR increased from 0.7 +/- 7.27 to 19.83 +/- 15.71 (p < 0.01). Navigator-echo-based real-time triggering of fetal head movement is a reliable technique that can deliver diagnostic fetal MR image quality despite vigorous fetal movement.

Thrombospondin-5 (TSP5) is a large extracellular matrix glycoprotein found in musculoskeletal tissues. TSP5 mutations cause two skeletal dysplasias, pseudoachondroplasia and multiple epiphyseal dysplasia; both show a characteristic growth plate phenotype with retention of TSP5, type IX collagen (Col9), and matrillin-3 in the rough endoplasmic reticulum. Whereas most studies focus on defining the disease process, few functional studies have been performed. TSP5 knockout mice have no obvious skeletal abnormalities, suggesting that TSP5 is not essential in the growth plate and/or that other TSPs may compensate. In contrast, Col9 knockout mice have diminished matrillin-3 levels in the extracellular matrix and early-onset osteoarthritis. To define the roles of TSP1, TSP3, TSP5, and Col9 in the growth plate, all knockout and combinatorial strains were analyzed using histomorphometric techniques. While significant alterations in growth plate organization were found in certain single knockout mouse strains, skeletal growth was only mildly disturbed. In contrast, dramatic changes in growth plate organization in TSP3/5/Col9 knockout mice resulted in a 20% reduction in limb length, corresponding to similar short stature in humans. These studies show that type IX collagen may regulate growth plate width; TSP3, TSP5, and Col9 appear to contribute to growth plate organization; and TSP1 may help define the timing of growth plate closure when other extracellular proteins are absent.

Mutations in cartilage oligomeric matrix protein (COMP), a large extracellular glycoprotein expressed in musculoskeletal tissues, cause two skeletal dysplasias, pseudoachondroplasia and multiple epiphyseal dysplasia. These mutations lead to massive intracellular retention of COMP, chondrocyte death and loss of growth plate chondrocytes that are necessary for linear growth. In contrast, COMP null mice have only minor growth plate abnormalities, normal growth and longevity. This suggests that reducing mutant and wild-type COMP expression in chondrocytes may prevent the toxic cellular phenotype causing the skeletal dysplasias. We tested this hypothesis using RNA interference to reduce steady state levels of COMP mRNA. A panel of shRNAs directed against COMP was tested. One shRNA (3B) reduced endogenous and recombinant COMP mRNA dramatically, regardless of expression levels. The activity of the shRNA against COMP mRNA was maintained for up to 10 weeks. We also demonstrate that this treatment reduced ER stress. Moreover, we show that reducing steady state levels of COMP mRNA alleviates intracellular retention of other extracellular matrix proteins associated with the pseudoachondroplasia cellular pathology. These findings are a proof of principle and the foundation for the development of a therapeutic intervention based on reduction of COMP expression.

OBJECTIVE The aim of the present systematic review and meta-analysis was to assess the clinical efficacy of regenerative periodontal surgery of intrabony defects using a combination of enamel matrix derivative (EMD) and bone graft compared with that of EMD alone. MATERIALS AND METHODS The Cochrane Oral Health Group specialist trials, MEDLINE, and EMBASE databases were searched for entries up to February 2014. The primary outcome was gain of clinical attachment (CAL). Weighted means and forest plots were calculated for CAL gain, probing depth (PD), and gingival recession (REC). RESULTS Twelve studies reporting on 434 patients and 548 intrabony defects were selected for the analysis. Mean CAL gain amounted to 3.76 ± 1.07 mm (median 3.63 95 % CI 3.51-3.75) following treatment with a combination of EMD and bone graft and to 3.32 ± 1.04 mm (median 3.40; 95 % CI 3.28-3.52) following treatment with EMD alone. Mean PD reduction measured 4.22 ± 1.20 mm (median 4.10; 95 % CI 3.96-4.24) at sites treated with EMD and bone graft and yielded 4.12 ± 1.07 mm (median 4.00; 95 % CI 3.88-4.12) at sites treated with EMD alone. Mean REC increase amounted to 0.76 ± 0.42 mm (median 0.63; 95 % CI 0.58-0.68) at sites treated with EMD and bone graft and to 0.91 ± 0.26 mm (median 0.90; 95 % CI 0.87-0.93) at sites treated with EMD alone. CONCLUSIONS Within their limits, the present results indicate that the combination of EMD and bone grafts may result in additional clinical improvements in terms of CAL gain and PD reduction compared with those obtained with EMD alone. The potential influence of the chosen graft material or of the surgical procedure (i.e., flap design) on the clinical outcomes is unclear. CLINICAL RELEVANCE The present findings support the use of EMD and bone grafts for the treatment of intrabony periodontal defects.

This work provides the development of an antenna for satellite communications onboard systems based on the recommendations ITU-R S.580-6 [1] and ITU-R S.465-5 [2]. The antenna consists of printed elements grouped in an array, working in a frequency band from 7.25 up to 8.4 GHz (15% of bandwidth). In this working band, transmission and reception are included simultaneously. The antenna reaches a gain about 31 dBi, has a radiation pattern with a beam width smaller than 10oand dual circular polarization. It has the capability to steer in elevation through a Butler matrix to 45

A fully 3D iterative image reconstruction algorithm has been developed for high-resolution PET cameras composed of pixelated scintillator crystal arrays and rotating planar detectors, based on the ordered subsets approach. The associated system matrix is precalculated with Monte Carlo methods that incorporate physical effects not included in analytical models, such as positron range effects and interaction of the incident gammas with the scintillator material. Custom Monte Carlo methodologies have been developed and optimized for modelling of system matrices for fast iterative image reconstruction adapted to specific scanner geometries, without redundant calculations. According to the methodology proposed here, only one-eighth of the voxels within two central transaxial slices need to be modelled in detail. The rest of the system matrix elements can be obtained with the aid of axial symmetries and redundancies, as well as in-plane symmetries within transaxial slices. Sparse matrix techniques for the non-zero system matrix elements are employed, allowing for fast execution of the image reconstruction process. This 3D image reconstruction scheme has been compared in terms of image quality to a 2D fast implementation of the OSEM algorithm combined with Fourier rebinning approaches. This work confirms the superiority of fully 3D OSEM in terms of spatial resolution, contrast recovery and noise reduction as compared to conventional 2D approaches based on rebinning schemes. At the same time it demonstrates that fully 3D methodologies can be efficiently applied to the image reconstruction problem for high-resolution rotational PET cameras by applying accurate pre-calculated system models and taking advantage of the system's symmetries.

In this letter, a dual circular polarized steering antenna for satellite communications in X-band is presented. This antenna consists of printed elements grouped in an array, able to work from 7.25 up to 8.4 GHz in both polarizations: left-handed circular polarization (LHCP) and right-handed circular polarization (RHCP). The module antenna is compact, with narrow beamwidth, and reaches a gain of 16 dBi. It has the capability to steer in elevation to and electronically with a Butler matrix. In order to reduce the mutual coupling between adjacent patches, electromagnetic band-gap (EBG) structures are introduced. These EBGs combine double-layer and edge location via in order to reduce the size, without changing the low-permittivity substrate, and therefore maintaining the high radiation efficiency of the antenna.

The plant cell wall constitutes an essential protection barrier against pathogen attack. In addition, cell-wall disruption leads to accumulation of jasmonates (JAs), which are key signaling molecules for activation of plant inducible defense responses. However, whether JAs in return modulate the cell-wall composition to reinforce this defensive barrier remains unknown. The enzyme 13-allene oxide synthase (13-AOS) catalyzes the first committed step towards biosynthesis of JAs. In potato (Solanum tuberosum), there are two putative St13-AOS genes, which we show here to be differentially induced upon wounding. We also determine that both genes complement an Arabidopsis aos null mutant, indicating that they encode functional 13-AOS enzymes. Indeed, transgenic potato plants lacking both St13-AOS genes (CoAOS1/2 lines) exhibited a significant reduction of JAs, a concomitant decrease in wound-responsive gene activation, and an increased severity of soft rot disease symptoms caused by Dickeya dadantii. Intriguingly, a hypovirulent D. dadantii pel strain lacking the five major pectate lyases, which causes limited tissue maceration on wild-type plants, regained infectivity in CoAOS1/2 plants. In line with this, we found differences in pectin methyl esterase activity and cell-wall pectin composition between wild-type and CoAOS1/2 plants. Importantly, wild-type plants had pectins with a lower degree of methyl esterification, which are the substrates of the pectate lyases mutated in the pel strain. These results suggest that, during development of potato plants, JAs mediate modification of the pectin matrix to form a defensive barrier that is counteracted by pectinolytic virulence factors from D. dadantii.
