Adult fertile male bonnet monkeys (Macaca radiata) were continuously deprived of endogenous follicle stimulating hormone (FSH) support for 240 days by injecting them with 1 ml of characterized monkey antiserum to oFSH every 48 hr; control monkeys received during the same period normal monkey serum instead. Testicular function was assessed at periodic intervals by (a) carrying out differential counting of sperm in the ejaculate obtained and (b) determining the hyaluronidase activity as well as in vitro 3H thymidine incorporation into DNA of testicular tissue removed at biopsy. Both the quality (viability and motility) of the sperms voided and the total sperm counts showed marked decreases as a function of time of immunization, the first significant reduction being noted by 100 days. FSH deprivation affected both the biochemical parameters used to test testicular functionality they being reduced at ∼200 days by 50%-60%. The fertility of these monkeys was evaluated at periodic times after 90 days of treatment by means of mating studies. FSH deprivation had rendered the monkeys incapable of impregnating any of the females used. Testosterone and luteinizing hormone (LH) levels remained unchanged following FSH antiserum injection. With cessation of antiserum treatment testicular function and fertility were completely restored to normalcy, indicating that the observed effect was specifically due to FSH deprivation. This study thus provides conclusive evidence for the involvement of FSH in maintenance of testicular function and fertility in the adult male primate.

The aim of this study was to estimate the development of fertility in North-Central Namibia, former Ovamboland, from 1960 to 2001. Special attention was given to the onset of fertility decline and to the impact of the HIV epidemic on fertility. An additional aim was to introduce parish registers as a source of data for fertility research in Africa. Data used consisted of parish registers from Evangelical Lutheran congregations, the 1991 and 2001 Population and Housing Censuses, the 1992 and 2000 Namibia Demographic and Health Surveys, and the HIV sentinel surveillances of 1992-2004. Both period and cohort fertility were analysed. The P/F ratio method was used when analysing census data. The impact of HIV infection on fertility was estimated indirectly by comparing the fertility histories of women who died at an age of less than 50 years with the fertility of other women. The impact of the HIV epidemic on fertility was assessed both among infected women and in the general population. Fertility in the study population began to decline in 1980. The decline was rapid during the 1980s, levelled off in the early 1990s at the end of war of independence and then continued to decline until the end of the study period. According to parish registers, total fertility was 6.4 in the 1960s and 6.5 in the 1970s, and declined to 5.1 in the 1980s and 4.2 in the 1990s. Adjustment of these total fertility rates to correspond to levels of fertility based on data from the 1991 and 2001 censuses resulted in total fertility declining from 7.6 in 1960-79 to 6.0 in 1980-89, and to 4.9 in 1990-99. The decline was associated with increased age at first marriage, declining marital fertility and increasing premarital fertility. Fertility among adolescents increased, whereas the fertility of women in all other age groups declined. During the 1980s, the war of independence contributed to declining fertility through spousal separation and delayed marriages. Contraception has been employed in the study region since the 1980s, but in the early 1990s, use of contraceptives was still so limited that fertility was higher in North-Central Namibia than in other regions of the country. In the 1990s, fertility decline was largely a result of the increased prevalence of contraception. HIV prevalence among pregnant women increased from 4% in 1992 to 25% in 2001. In 2001, total fertility among HIV-infected women (3.7) was lower than that among other women (4.8), resulting in total fertility of 4.4 among the general population in 2001. The HIV epidemic explained more than a quarter of the decline in total fertility at population level during most of the 1990s. The HIV epidemic also reduced the number of children born by reducing the number of potential mothers. In the future, HIV will have an extensive influence on both the size and age structure of the Namibian population. Although HIV influences demographic development through both fertility and mortality, the effect through changes in fertility will be smaller than the effect through mortality. In the study region, as in some other regions of southern Africa, a new type of demographic transition is under way, one in which population growth stagnates or even reverses because of the combined effects of declining fertility and increasing mortality, both of which are consequences of the HIV pandemic.

Seminal plasma (SP) is the fluid portion of semen, secreted by the epididymides and the accessory glands before and during ejaculation. The stallion s ejaculate is a series of jets that differ in sperm concentration, semen volume and biochemical composition. Before the actual ejaculation, a clear and watery pre-sperm fluid is secreted. The first three jets form the sperm-rich fractions, and contain ¾ of the total number of sperm. The semen volume and sperm concentration in each of the jets decrease towards the end of the ejaculation, and the last jets are sperm-poor fractions with a low sperm concentration. The aims of these studies were to examine the effects of the different SP fractions, and the presence of SP, on sperm survival during storage. Pre-sperm fluid, and semen fractions with a high (sperm-rich) and low (sperm-poor) sperm concentration were collected in five experiments. The levels of selected enzymes, electrolytes and proteins in different SP fractions were determined. These studies also aimed at assessing the individual variation in the levels of the selected SP components and in the effects of SP on spermatozoa. The association between the components of SP and semen quality, sperm longevity, and fertility was examined with a stepwise linear regression analysis. Compared to samples containing SP during storage, centrifugation and the subsequent removal of SP reduced sperm motility parameters during 24 h of cooled storage in all SP fractions, but sperm membrane integrity was not affected. Some of the measured post-thaw motility parameters were also higher in samples containing SP compared to samples stored without SP. In contrast, the proportion of DNA-damaged spermatozoa was greater in the samples stored with SP than those without SP, and this effect was seen in both sperm-rich and sperm-poor fractions. There were no differences in DNA integrity between fractions stored with SP, but the sperm-rich fraction showed less DNA damage than the sperm-poor fraction after SP removal. The differences between fractions in sperm motility after cooled storage were non-significant. The levels of alkaline phosphatase, acid phosphatase and β-glucuronidase were higher in the sperm-rich fractions compared to the sperm-poor fractions, while the concentrations of calcium and magnesium were higher in sperm-poor fractions than in sperm-rich fractions. The concentrations of sodium and chloride were highest in pre-sperm fluid. In the sperm-poor fraction, the level of potassium was associated with the maintenance of sperm motility during storage. The levels of alkaline and acid phosphatase were associated with sperm concentration and the total number of spermatozoa in the ejaculates. None of the measured SP components were correlated to the first cycle pregnancy rate. In summary, the removal of SP improved DNA integrity after cooled storage compared with samples containing SP. There were no differences in the maintenance of sperm motility between the sperm-rich and sperm-poor fractions and whole ejaculates during cooled storage, irrespective of the presence of SP. The lowest rate of DNA damage was found in the sperm-rich fractions stored without SP. In practice, the results presented in this thesis support the use of individual modifications of semen processing techniques for cooled transported semen from subfertile stallions.

Maintenance of breeding efficiency and high semen quality is essential for reproductive success in farm animals. Early recognition of possible inheritable factors causing infertility requires constant attention. This thesis focuses on describing different manifestations of impaired spermatogenesis, their impact on fertility and partly also their incidence in populations. The reasons for spermatogenic failure are various. An interruption of germ cell differentiation, spermatogenic arrest, can lead to infertility. The incidence of azoospermia was investigated in the 1996 2005 survey of Finnish AI and farm breeding boars. We focused on the diagnosis, testicular morphometry and the possible reasons for the condition. The incidence of azoospermia was significantly higher in Yorkshire boars than in the Landrace breed. The most common diagnosis in Yorkshire boars was germ cell arrest at the primary spermatocyte level. The second most frequent diagnosis in Yorkshire boars was segmental aplasia of the Wolffian ducts with idiopathic epididymal obstruction. Other reasons for azoospermia were infrequent. In the second study we investigated the incidence of two relatively well-defined specific sperm defects in Finnish Yorkshire and Landrace boars during the same survey, the immotile short-tail sperm (ISTS) defect and the knobbed acrosome (KA) defect. In the Finnish Yorkshire boars the inherited ISTS defect, and the probably inherited KA defect, were important causes of infertility during 1996 2005. The ISTS defect was found in 7.6% and the KA defect in 0.8% of the Yorkshire boars. No Landrace boars were diagnosed with either of these two defects. In the third study we described a new sterilizing sperm defect in an oligoasthenoterazoospermic bull. Because of its morphological characteristics this defect was termed the multinuclear-multiflagellar sperm (MNMFS) defect. The number of Sertoli cells in the seminiferous tubuli was highly increased in the MNMFS bull compared with the number in normal bulls. In the following two studies we used a combined approach of fluorescence in situ hybridization (FISH), flow cytometry and morphometric studies to provide information on the cytogenetic background of macrocephalic bull spermatozoa. We described cellular features of diploid spermatozoa and compared the failures in the first and second meiotic divisions. In the last study we describe how the transplantation of testicular cells was used to determine whether spermatogonia derived from donor animals are able to colonize and produce motile spermatozoa in immune-competent unrelated boars suffering the ISTS defect. Transplantation resulted in complete focal spermatogenesis, indicated by the appearance of motile spermatozoa and confirmed by genotyping.

Unlike queens of typical primitively eusocial species, Ropalidia marginata queens are docile and non-interactive, and hence cannot be using dominance to maintain their status. It appears that the queen maintains reproductive monopoly through a pheromone, of which the Dufour's gland is at least one source. Here, we reconfirm earlier results showing that queens and workers can be correctly classified on a discriminant function using the compositions of their respective Dufour's glands, and also demonstrate consistent queen-worker differences based on categories of compounds and on single compounds also in some cases. Since the queen pheromone is expected to be an honest signal of the fecundity of a queen, we investigate the correlation of Dufour's gland compounds with ovarian activation of queens. Our study shows that Dufour's gland compounds in R. marginata correlate with the state of ovarian activation of queens, suggesting that such compounds may portray the fecundity of a queen, and may indeed function as honest signals of fertility.

Administration of Tamoxifen via the Alzet pump at a rate of 50 mu g hr(-1) for 90 days in the adult male bonnet monkeys Macaca radiata had no effect on the serum testosterone concentration determined at 10 AM and 10 PM as well as total sperm count determined at 15-day intervals over a period of 260 days. However, a significant reduction in sperm motility was observed beyond 90 days up until the 225th day. Breeding studies conducted from day 90 to 260 revealed that these males were infertile.

GH3 proteins control auxin homeostasis by inactivating excess auxin as conjugates of amino acids and sugars and thereby controlling cellular bioactive auxin. Since auxin regulates many aspects of plant growth and development, regulated expression of these genes offers a mechanism to control various developmental processes. OsMGH3/OsGH3-8 is expressed abundantly in rice florets and is regulated by two related and redundant transcription factors, OsMADS1 and OsMADS6, but its contribution to flower development is not known. We functionally characterize OsMGH3 by overexpression and knock-down analysis and show a partial overlap in these phenotypes with that of mutants in OsMADS1 and OsMADS6. The overexpression of OsMGH3 during the vegetative phase affects the overall plant architecture, whereas its inflorescence-specific overexpression creates short panicles with reduced branching, resembling in part the effects of OsMADS1 overexpression. In contrast, the down-regulation of endogenous OsMGH3 caused phenotypes consistent with auxin overproduction or activated signaling, such as ectopic rooting from aerial nodes. Florets in OsMGH3 knock-down plants were affected in carpel development and pollen viability, both of which reduced fertility. Some of these floret phenotypes are similar to osmads6 mutants. Taken together, we provide evidence for the functional significance of auxin homeostasis and its transcriptional regulation during rice panicle branching and floret organ development.

Queens of many social insect species are known to maintain reproductive monopoly by pheromonal signalling of fecundity. Queens of the primitively eusocial wasp Ropalidia marginata appear to do so using secretions from their Dufour's glands, whose hydrocarbon composition is correlated with fertility. Solitary nest foundresses of R. marginata are without nestmates; hence expressing a queen signal can be redundant, since there is no one to receive the signal. But if queen pheromone is an honest signal inextricably linked with fertility, it should correlate with fertility and be expressed irrespective of the presence or absence of receivers of the signal, by virtue of being a byproduct of the state of fertility. Hence we compared the Dufour's gland hydrocarbons and ovaries of solitary foundresses with queens and workers of post-emergence nests. Our results suggest that queen pheromone composition in R. marginata is a byproduct of fertility and hence can honestly signal fertility. This provides important new evidence for the honest signalling hypothesis.

Compost, vermicompost and biochar amendments are thought to improve soil quality and plant yield. However, little is known about their long-term impact on crop yield and the environment in tropical agro-ecosystems. In this study we investigated the effect of organic amendments (buffalo manure, compost and verrnicompost) and biochar (applied alone or with vermicompost) on plant yield, soil fertility, soil erosion and water dynamics in a degraded Acrisol in Vietnam. Maize growth and yield, as well as weed growth, were examined for three years in terrestrial mesocosms under natural rainfall. Maize yield and growth showed high inter-annual variability depending on the organic amendment. Vermicompost improved maize growth and yield but its effect was rather small and was only significant when water availability was limited (year 2). This suggests that vermicompost could be a promising substrate for improving the resistance of agrosystems to water stress. When the vermicompost biochar mixture was applied, further growth and yield improvements were recorded in some cases. When applied alone, biochar had a positive influence on maize yield and growth, thus confirming its interest for improving long-term soil productivity. All organic amendments reduced water runoff, soil detachment and NH4+ and NO3- transfer to water. These effects were more significant with vermicompost than with buffalo manure and compost, highlighting that the beneficial influence of vermicompost is not limited to its influence on plant yield. In addition, this study showed for the first time that the combination of vermicompost and biochar may not only improve plant productivity but also reduce the negative impact of agriculture on water quality. El presente trabajo de tesis se realizó en la granja experimental porcina MAGFOR-Misión China de la república de Taiwán, ubicada en Cofradía. El estudio consistió en la evaluación de los parámetros reproductivos en grupos de cerdas obtenidos por inseminación artificial y grupos obtenidos a través de la monta natural, también se aborda el tema de la consanguinidad y sus consecuencias como un factor negativo en la fijación de caracteres indeseables, esto sucede en las poblaciones en las que existe una alta homocigosis debido al origen común del material genético (padres emparentados). Las variables evaluadas en el presente trabajo fueron: tamaño de la camada al nacimiento, peso promedio de los lechones al nacimiento, peso promedio de los lechones al destete e intervalo parto parto. para el análisis estadístico se elaboraron tablas de contingencias por cada una de las variables con base en los promedios. Para la variable tamaño de la camada al nacimiento se obtuvo para el tratamiento inseminación artificial un promedio de 9.16 lechones y para la monta natural un promedio de 9.89 lechones al contrastar las medias de los dos tratamientos se obtuvo un valor de t de 0.88 que comparado con el valor tabulado al 5% resulto no significativo al observar el comportamiento de las diferentes razas ,la yorkshire obtuvo los mayores promedios en inseminación el caso especifico de esta habían muy pocos ejemplares 6 . Se trabajo en condiciones normales de producción lo cual pudo influir en estos resultados debido al tamaiio de la muestra en esta raza. Los resultados para la variable peso promedio de los lechones al nacimiento, arrojaron un promedio de 1.76 Kg. para las camadas obtenidas por inseminación artificial y 1.67kg para las obtenidas por el método de monta natural en las observaciones de los promedios por razas, se obtuvo el mayor peso a favor de la raza Landrace con 1.99 Kg. seguido de la raza duroc con 1.85 Kg. en el tratamiento de inseminación artificial, y para la monta natural el menor peso lo obtuvo la raza duroc con 1.44 Kg. y elmayor peso fue también la raza Landrace. la raza Yorkshire mostró un comportamiento similar tanto en inseminacion artificial, como en monta natural. Para la variable peso promedio de los lechones al destete, se obtuvo un peso promedio de 7.39 Kg. para los lechones obtenidos por el método de inseminacion artificial y un peso promedio de 6.71 Kg. en el caso de lechones obtenidos por monta natural al evaluar las razas, la raza Landrace obtuvo el mayor peso al destete con 8.01 Kg. como era de esperar al obtener los mayores promedios en peso al nacimiento esto fue para las camadas obtenidas por inseminacion artificial. en el caso. de los lechones obtenidos mediante la monta natural el mayor peso correspondió a la raza Hampshire pudiendo haber influido el numero de lechones, ya que esta fue la que presento el menor numero la raza Yorkshire tuvo en este caso un comportamiento similar en los dos tratamientos. Para la variable intervalo parto-parto, en el caso de las cerdas servidas por el método de inseminacion artificial presentaron un IPP de 5.52 meses o sea 167 días y las cerdas servidas por monta natural obtuvieron un IPP de5.01 meses o 152 días. En la prueba de hipótesis tanto como el contraste de varianza, los resultados fueron significativos y una diferencia de 15 días vacíos en una hembra eleva los costos de producción de una manera considerable por esta razón un IPP como el de la inseminación artificial solo se justifica en la granja experimental porcina en un numero especifico de cerdas elite y como apoyo al mejoramiento genético del hato porcino de esta así como del hato nacional.

La inseminación artificial es una técnica que fue desarrollada en la década de los años treinta utilizando el método descrito por Buno\VS y Quinn (1937), consistía en practicarle masajes abdominales y en la cloaca tanto al ave macho como a la hembra Este trabajo de investigación que tiene por título: Establecimiento de técnicas de extracción de semen en gallos criollos e inseminación artificial en gallinas criollas, fue investigado con los tines de realizar la técnica inseminación artificial en gallinas criollas que no se utiliza en nuestro país, y también comprobar que esta tiene resultado positivo. En la investigación se trabajo con 10 gallinas y 2 gallos en donde se divide dos tratamientos: 1er tratamiento monta natural que se utilizaron 5 gallinas y un gallo, 2do tratamiento Inseminación artificial que se utilizaron 5 gallinas y un gallo Los cuales fueron seleccionados con Jónne parámetros establecidos En el tratamiento en la monta natural las gallinas estaban en manejo tradicional y la alimenticio ad-libitum y copulacion natural de gallo. Mientras que en 2do tratamiento para el gallo de la inseminación se le realizo un adiestramiento durante 6 meses para lograr la obtención de semen puro posteriormente inseminar a las gallinas destinadas para el 2do tratamiento. En los resultados de la variable de incubación de cada tratamiento se valoró la diferencia de los tratamiento empleados, mostrando que en el tratamiento 1 por la monta natural (T1 M.N) fue de margen inferior de 75%, donde para el tratamiento 2 de inseminación artificial (T2 LA) un margen de superioridad reflejado en 83% Se encontró un Chi cuadrado de 1.7562 no significativo al5% con 1 grado de libertad,-- (filas-uno)*(columnas-1) - entendiéndose que las diferencias observadas en el porcentaje de huevos nacidos de cada grupo de tratamiento, así como el porcentaje de huevos no nacidos de cada grupo de tratamiento están en el rango posible de diferencias atribuibles a la casualidad propias de unidades experimentales biológicas empleando un nivel de significación del 5 % También se rellejo el porcentaje de huevos incubables fue para(T1 M.N) 35% y ("12 LA) 31%, para mortalidad embrionaria se determino un 32%(T1 M.N)y el 20% para elpara(T2I.A). En elcomportamiento reproductivo se constato que para (T1 M.N) un margen de 28- 15 huevos y para (T2 LA) un margen de 24- 13 huevos. Concluyendo que la técnica de inseminación attiticial se puede realizar, y puede ser beneficiosa, pero a niveles industriales o en aves de razas debido a que los costos son significativamente costosos no siendo viable para los pequeños productores de gallinas de patio
