Ferritins are nearly ubiquitous iron storage proteins playing a fundamental role in iron metabolism. They are composed of 24 subunits forming a spherical protein shell encompassing a central iron storage cavity. The iron storage mechanism involves the initial binding and subsequent O-2-dependent oxidation of two Fe2+ ions located at sites A and B within the highly conserved dinuclear "ferroxidase center" in individual subunits. Unlike animal ferritins and the heme-containing bacterioferritins, the Escherichia coli ferritin possesses an additional iron-binding site (site C) located on the inner surface of the protein shell close to the ferroxidase center. We report the structures of five E. coli ferritin variants and their Fe3+ and Zn2+ (a redox-stable alternative for Fe2+) derivatives. Single carboxyl ligand replacements in sites A, B, and C gave unique effects on metal binding, which explain the observed changes in Fe2+ oxidation rates. Binding of Fe2+ at both A and B sites is clearly essential for rapid Fe2+ oxidation, and the linking of Fe-B(2+) to Fe-C(2+) enables the oxidation of three Fe2+ ions. The transient binding of Fe2+ at one of three newly observed Zn2+ sites may allow the oxidation of four Fe2+ by one dioxygen molecule.

Single-cell analysis is essential for understanding the processes of cell differentiation and metabolic specialisation in rare cell types. The amount of single proteins in single cells can be as low as one copy per cell and is for most proteins in the attomole range or below; usually considered as insufficient for proteomic analysis. The development of modern mass spectrometers possessing increased sensitivity and mass accuracy in combination with nano-LC-MS/MS now enables the analysis of single-cell contents. In Arabidopsis thaliana, we have successfully identified nine unique proteins in a single-cell sample and 56 proteins from a pool of 15 single-cell samples from glucosinolate-rich S-cells by nanoLC-MS/MS proteomic analysis, thus establishing the proof-of-concept for true single-cell proteomic analysis. Dehydrin (ERD14_ARATH), two myrosinases (BGL37_ARATH and BGL38_ARATH), annexin (ANXD1_ARATH), vegetative storage proteins (VSP1_ARATH and VSP2_ARATH) and four proteins belonging to the S-adenosyl-l-methionine cycle (METE_ARATH, SAHH1_ARATH, METK4_ARATH and METK1/3_ARATH) with associated adenosine kinase (ADK1_ARATH), were amongst the proteins identified in these single-S-cell samples. Comparison of the functional groups of proteins identified in S-cells with epidermal/cortical cells and whole tissue provided a unique insight into the metabolism of S-cells. We conclude that S-cells are metabolically active and contain the machinery for de novo biosynthesis of methionine, a precursor for the most abundant glucosinolate glucoraphanine in these cells. Moreover, since abundant TGG2 and TGG1 peptides were consistently found in single-S-cell samples, previously shown to have high amounts of glucosinolates, we suggest that both myrosinases and glucosinolates can be localised in the same cells, but in separate subcellular compartments. The complex membrane structure of S-cells was reflected by the presence of a number of proteins involved in membrane maintenance and cellular organisation.

Bacterioferritin (BFR) from Escherichia coli is a member of the ferritin family of iron storage proteins and has the capacity to store very large amounts of iron as an Fe(3+) mineral inside its central cavity. The ability of organisms to tap into their cellular stores in times of iron deprivation requires that iron must be released from ferritin mineral stores. Currently, relatively little is known about the mechanisms by which this occurs, particularly in prokaryotic ferritins. Here we show that the bis-Met-coordinated heme groups of E. coli BFR, which are not found in other members of the ferritin family, play an important role in iron release from the BFR iron biomineral: kinetic iron release experiments revealed that the transfer of electrons into the internal cavity is the rate-limiting step of the release reaction and that the rate and extent of iron release were significantly increased in the presence of heme. Despite previous reports that a high affinity Fe(2+) chelator is required for iron release, we show that a large proportion of BFR core iron is released in the absence of such a chelator and further that chelators are not passive participants in iron release reactions. Finally, we show that the catalytic ferroxidase center, which is central to the mechanism of mineralization, is not involved in iron release; thus, core mineralization and release processes utilize distinct pathways.

The mobilization of food reserves in storage tissues and allocation of their hydrolysis products in the growing axis are critical processes for the establishment of seedlings after germination. Therefore, it is crucial for mobilization of reserves to be synchronized with the growing axis, so that photosynthetic activity can be started before depletion of reserves. For this, integrative approaches involving different reserves, different hydrolysis products and interaction between storage and growing axis tissues, either through hormones or metabolites with signaling role, can contribute greatly to the elucidation of the regulation mechanisms for reserve mobilization. In this study, was hypothesized that hormones and metabolites have different actions on reserve mobilization, and there must be a crossed effect of sugars on the mobilization of proteins and amino acids on lipids and starch mobilization in sunflower seedlings. This study was conducted with seeds of sunflower (Helianthus annuus L.) hybrid Helio 253 using in vitro culture system. Seeds were germinated on Germitest® paper and grown on agar-water 4 g/L without addition of nutrients during 9 days after imbibition (DAI) for growth curve. To verify the effect of metabolites and hormones, seedlings were transferred in the 2nd DAI to agar-water 4 g/L supplemented with increasing concentrations of sucrose or L-glutamine, abscisic acid, gibberellic acid or indolebutyric acid. The results of this study confirm that the mobilization of lipids and storage proteins occurs in a coordinated manner during post-germination growth in sunflower, corroborating the hypothesis that the application of external carbon (sucrose) and nitrogen (L-glutamine) sources can delay the mobilization of these reserves in a crossed way. Moreover, considering the changes in the patterns of reserve mobilization and partition of their products in seedlings treated with different growth regulators, it is evident that the effects of metabolites and hormones must involve, at least in part, distinct mechanisms of action

Grains and legume seeds are foods that form the basis of the diets of many cultures around the world, winch contritbute to the daily nutrient requirements of humans. Vicilins (7S globulin) are storage proteins found in legume seeds, and may have an additional function constitutive defense of the embryo against pests and pathogens. In this work the vicilin from Anadenanthera macrocarpa - AmV (red-angico), was purified and partially characterized, its effect on development and larval survival and adult emergence of Callosobruchus maculatus was evaluated by determination of LD50, WD50 and ED50 in system bioassay. Purification of vicilin was initiated by the chitin affinity chromatography and then gel filtration (Superdex 75 Tricorn 10x300 mm) FPLC system followed by reverse phase chromatography (C8 phenomenex) on HPLC system. Bioassays WD50 and LD50 for larvae were 0.32% and 0.33% (w:w) respectively, since the ED50 for adults was 0.096%. The probable mechanism of action was evaluated by testing digestibility of AmV in vitro, and observed for the involvement of two fragments vicilins immunoreactive against polyclonal Anti-vicilin from Erythrina velutina (Anti-EvV) about of 22 and 13 kDa chitin binding. The AmV in its native form has been recognized by the anti-EvV, indicating that there is a conserved region in the vicilin and is probably corresponding to the chitin binding domains. These results point to a new vicilin chitin binding that can subsequently be used as a possible biopesticide protein source, in order to control insect pest C. maculatus and confirm literature findings that demonstrate vicilin in the presence of different kinds of ligands to conserved regions chitin not yet characterized

O cultivo da soja fora da época convencional pode ser uma alternativa de rotação de cultura além de proporcionar sementes de melhor qualidade fisiológica, que podem ser utilizadas na semeadura da próxima safra diminuindo o período de armazenamento. O trabalho de pesquisa teve por objetivo avaliar o comportamento de sete cultivares de soja, em três densidades populacionais, quanto ao porte e altura de inserção da primeira vagem, assim como quanto à produção e qualidade fisiológica das sementes, semeadas no período de inverno, na região de Selvíria-MS. O delineamento experimental foi em blocos casualizados, com quatro repetições, dispostos em esquema fatorial 7 ×3. Os tratamentos constaram de sete cultivares (IAC-16, IAC-Foscarin 31, FT-2, IAC-17, IAC-8, Doko e FT-Cristalina) e três densidades de plantas (300, 400 e 500 mil plantas ha-1). O ciclo da soja foi reduzido no cultivo de inverno, principalmente nos cultivares considerados tardios. O fotoperíodo no cultivo de inverno reduziu o período entre o florescimento e a maturação. O cultivar IAC-8 foi o menos sensível ao fotoperíodo, possuindo maturação em época semelhante aos cultivares tardios, e apresentou as melhores características agronômicas para o cultivo no período de inverno. É aconselhável o aumento de densidade de semeadura quando do uso de cultivares precoces em cultivos de inverno. Não é aconselhável armazenar sementes em condições ambientes, principalmente com valores de germinação próximo ao limite inferior desejável (80%).

Considerando que o armazenamento desempenha papel decisivo na manutenção da qualidade da semente, desenvolveu-se este trabalho com o objetivo de avaliar a qualidade fisiológica e fitossanitária de sementes de algodão, tratadas quimicamente e armazenadas por doze meses. Foram utilizados dois lotes de sementes de algodão cv. DeltaPine-AC90, deslintados quimicamente, que foram submetidos aos tratamentos fungicidas e inseticidas: testemunha; Disulfoton + Carboxin + Thiram; Carbofuran + Carboxin + Thiram; Imidacloprid + Tolylfluanid + Pencycuron. As sementes foram armazenadas em armazém sem controle de temperatura e umidade relativa do ar. Foram retiradas amostras de sementes no início do armazenamento e a cada dois meses e avaliadas quanto ao teor de água, à porcentagem de germinação, ao vigor (testes de envelhecimento acelerado e de germinação à baixa temperatura), à sanidade e à emergência das plântulas. O delineamento utilizado foi o inteiramente casualizado, em esquema fatorial, com quatro repetições. Verificou-se redução da germinação e do vigor de sementes em função do armazenamento. A redução da qualidade fisiológica associou-se com o aumento na ocorrência de Aspergillus sp. e Penicillium sp. nas sementes. A manutenção da qualidade de sementes de algodão foi obtida até o oitavo mês de armazenagem, podendo-se concluir que: a eficiência do tratamento químico de sementes de algodão depende da combinação de produtos utilizados; não se deve tratar com fungicida sementes de algodão com baixo nível de vigor; a ocorrência dos fungos Aspergillus sp. e Penicillium sp. aumentou com o período de armazenamento nas sementes sem tratamento e que a manutenção da qualidade de sementes de algodão para comercialização depende da sua qualidade inicial e do período de armazenamento.

Caesalpinia leiostachya (Benth.) Ducke (pau-ferro) é uma planta arbórea nativa do Brasil, cujas sementes possuem dormência causada pela impermeabilidade do tegumento à água. Neste trabalho foram conduzidos dois experimentos, nos quais foram utilizados diferentes períodos de escarificação em ácido sulfúrico concentrado para superar a dormência das sementes. No primeiro experimento, sementes coletadas em agosto de 1997 foram armazenadas por oito meses em ambiente não controlado no interior do próprio fruto, e em câmara seca após serem extraídas dos frutos; a seguir, elas foram imersas em ácido sulfúrico por 0, 10, 20, 40, 60 e 80min e colocadas para germinar nas temperaturas constante de 25ºC e alternada de 20-30°C, sob fotoperíodo de 8h. No segundo experimento, sementes extraídas de frutos recém-coletados em agosto de 1998 foram imersas em ácido sulfúrico por 0, 10, 20, 30, 40 e 60min, seguido do teste de germinação conduzido nas mesmas temperaturas do experimento anterior, na ausência e presença de luz. Foram avaliados a porcentagem final e o índice de velocidade de germinação das sementes. Os resultados mostraram que (a) a manutenção das sementes no interior dos frutos é uma alternativa viável para o armazenamento durante o período adotado; (b) as sementes recém-coletadas são indiferentes à luz, nas duas temperaturas testadas; (c) as sementes recém-coletadas e as armazenadas germinam em maior velocidade a 25ºC; (d) em sementes armazenadas, a imersão em ácido sulfúrico por 10min é suficiente para superar a dormência; (e) em sementes recém-coletadas, a imersão em ácido sulfúrico por 20 a 30min favorece a porcentagem e a velocidade de germinação.

During bean seed storage, yield can be lost due to infestations of Acanthoscelides obtectus Say, the bean weevil. The use of resistant varieties has shown promising results in fighting these insects, reducing infestation levels and eliminating chemical residues from the beans. The expression of resistance to A. obtectus in bean varieties is frequently attributed to the presence of phytohemagglutinins, protease inhibitors and alpha-amylase, and especially to variants of the protein arcelin, which reduce the larval viability of these insects. To evaluate the effect of bean seed storage time on the resistance expression of bean varieties to A. obtectus, tests with seeds of three ages (freshly-harvested, 4-month-old, and 8-month-old) were conducted in the laboratory, using four commercial varieties: Carioca Pitoco, Ipa 6, Porrillo 70, Onix; four improved varieties containing arcelin protein: Are. 1, Arc.2, Arc. 3, Arc.4; and three wild varieties also containing arcelin protein: Arc. IS, Arc.3S, and Arc. 5S. The Arc.5S, Arc. IS, and Arc.2 varieties expressed high antibiosis levels against the weevil; Arc. I and Arc3S expressed the same mechanism, but at lower levels. The occurrence of oviposition non-preference was also observed in Arc.5S and Arc. IS. The Arc.3 and Arc. 4 varieties expressed low feeding non-preference levels against A. obtectus. The expression of resistance in arcelin-bearing, wild or improved varieties was affected during the storage of seeds, and was high under some parameters but low in others. The results showed that addition of chemical resistance factors such as protein arcelin via genetic breeding may be beneficial in improving the performance of bean crops.

Para avaliar os efeitos da idade e do repouso pós-colheita de frutos de pepino (Cucumis sativus L.) de duas cultivares, Pérola (espinhos escuros) e Rubi (espinhos claros), sobre as alterações na qualidade fisiológica das sementes durante o armazenamento, conduziu-se um experimento em Botucatu, SP e Bandeirantes, PR, durante quatro anos. Os frutos foram colhidos desde os 15 ('Rubi') ou 20 ('Pérola') dias até 45 dias após a antese, permanecendo em repouso pós-colheita por 0, 5, 10 e 15 dias, antes da extração das sementes. Antes e após dois e três anos de armazenamento (sacos de papel em ambiente de laboratório), foram retiradas amostras para avaliação do potencial germinativo e do vigor. Os resultados revelaram que sementes imaturas, retiradas de frutos colhidos precocemente (até 30 dias após a antese) e sem repouso pós-colheita, perderam totalmente a capacidade germinativa no segundo ano de armazenamento. Sementes que apresentavam valores iniciais de germinação mais elevados, as colhidas de frutos maduros (com ou sem repouso pós-colheita) ou de imaturos (30 e 35 dias de idade) com 10 a 15 dias de repouso pós-colheita, mantiveram valores acima de 70% após três anos de armazenamento.

Brazil has been considered one of the diversity centers of Gossypium barbadense species. It is believed that a relatively big erosion genetic process occurs with the species, due to economic, cultural and agricultural problems. A local diagnostic about species situation is the first step for reducing the diversity loss and establishing conservation strategies in situ. This research aimed the identification of the presence of Gossypium populations, characterization, determination of the main risks and collection of the accesses to store in germoplam banks, in Para and Amapa States. Expeditions were conducted in November 2004. An interview was carried out with the plant proprietor for characterizing in situ of G. barbadense species and of the environment where the plants were inserted. On hundred seventy nine plants in 22 municipal districts were collected in Para State and 117 plants in nine municipal districts in Amapa State. The majority of plants belong to G. barbadense species (98% in Amapa and 94% in Para). Plants occur in back yards, beside roads and spontaneously. That ones from back yards were more abundant (97% in Amapa and 95% in Para) and maintained as medicinal plants as the principal reason. Plants in natural environments in both states evaluated were not found, therefore, the creation of reserves and the application of others conventional methods of maintenance in situ are not applicable. The plant proprietors do not use to store or process seeds. Seed storage was reported as a practice by only 1% of the plant proprietors from Para and 11% from Amapa. The most plants collected were from two to three years of age (58% in Amapa and 93% in Para). As conclusions G. barbadense is the species most spread in the two studied states and are found in back yards. In Amapa State the botanical variety barbadense or Quebradinho is predominant, whereas in Para State the predominant variety is brasiliense or Rim-de-boi. O trabalho é um estudo das condições de germinação das sementes de Cassia tora L., planta invasora das culturas em geral e, principalmente, da cultura da soja. Nesta pesquisa, estudou se a quebra da dormência, o efeito da profundidade de semeadura sobre a emergência de plântulas e o potencial de armazenamento, considerando o envelhecimento natural e precoce com o objetivo de um controle mais racional desta espécie. As profundidades de semeadura de 4 a 6 cm apresentaram as porcentagens mais altas de emergência das plântulas. A porcentagem de germinação de sementes, logo após a colheita, encontra-se próxima de 42%, decrescendo para 40%, 39% e 30%, aproximadamente, nos períodos de armazenamento por 90, 180 e 270 dias, respectivamente, evidenciando uma porcentagem média de germinação próxima de 38% nos períodos considerados. O processo de escarificação mecânica mostrou-se o mais eficiente para quebra da dormência das sementes

O presente trabalho teve como objetivo, verificar o tempo de armazenamento necessário para que seja atingida a viabilidade máxima e o tempo de embebição máxima de sementes de macadâmia (Macadamia integrifolia Maiden & Betche var. IAC-4-20). As sementes foram pesadas e colocadas em recipientes com água para que ocorresse o processo da embebição, sendo realizada a pesagem a cada 6 horas. Pode-se concluir que a embebição foi máxima ao redor de 90 horas após o início da embebição. Para a análise da viabilidade das sementes, estas foram semeadas mensalmente, durante 12 meses a partir da colheita. Verificou-se que a viabilidade máxima das sementes é atingida, 4 meses após o início do armazenamento.
