One of the major questions concerning Giardia is the understanding of pathophysiological processes associated with small intestine abnormalities. There are evidences that Giardia trophozoites contain and/or release proteolytic enzymes that may be implicated in the host intestinal epithelium. The present investigation was undertaken to examine the protease activity in excretory/secretory (E/S) products of Giardia duodenalis trophozoites of an axenic Brazilian strain (BTU-11) and the reference strain Portland 1 (P1). E/S products from trophozoites of each strain in conditioned medium were tested with sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) for the protein profiles, and the protease activity was analyzed using substrate-impregnated SDS-PAGE (gelatin and collagen) and hemoglobin assay. The proteases characterization was based on inhibition assays including synthetic inhibitors. Electrophoresis analysis of E/S products revealed a banding pattern composed by few bands (4 to 6 bands) in the migration region of 123 to 28 kDa. Proteolytic products were detected in the conditioned medium by trophozoites of both assayed strains. In the gels containing copolymerized gelatin and collagen, E/S products promoted substrate degradation and the most evident proteolysis zones were distributed in the migration regions of 77 to 18 kDa and 145 to 18 kDa, respectively, in the patterns of gelatinolytic and collagenolytic activities. Degradation of hemoglobin was also observed, and the pattern of hydrolysis was similar in both E/S products assayed. Inhibitor assays showed that the main proteolytic activity in both E/S products is due to cysteine proteases, although the presence of serine proteases was also indicated. Degradation of substrates including collagen and hemoglobin could lead us to speculate different functions of Giardia excreted/secreted proteases in vivo, but to confirm this possibility and to elucidate its implication on host-parasite interactions, further experiments applying protocols for the purification of proteases are necessary. Even so, our observations are relevant and hold the perspective for the understanding about protease activity in Giardia trophozoites of axenic strain isolated in an endemic area.

Proteolytic activity in excretory/secretory products (ESP) of first- (L1), second- (L2) and third-instar (L3) larvae of Dermatobia hominis was analyzed through gelatin-gel and colorimetric enzyme assays with the chromogenic substrates azocasein and BApNA. The functional characterization of proteases was based on inhibition assays including synthetic inhibitors. ESP were obtained from new-hatched larvae reared in the laboratory and from second- and third-instar larvae removed from naturally infested cattle. Gelatin-gel analysis evidenced few bands of proteolysis, predominantly of high apparent molecular masses, in ESP of L1, whereas in the gel of L2 and U ESP there was a wide range of proteolytic activity most of them not resolved in a single species. Azocasein assays revealed a progressive increase of protease activity from first- to third-instar larvae. Protease inhibitor assays revealed a predominance of metalloproteases in L1 ESP that could be related to a skin penetration process and to a diversion of host immune response. The predominance of serine proteases in L2 and L3 and the great tryptic activity presented by L3 ESP were attributed to an increasing trophic activity by the growing larvae, since the viability of adult flies strictly depends on larval abilities to assimilate nutrients from the host. Taking together, these results suggest that Dematobia larvae secrete/excrete different proteases that may be related to diverse functions during host penetration and infestation, which reinforces the relevance of the study of such proteolytic enzymes. This study examined the production of protein hydrolysates with controlled composition from cheese whey proteins. Cheese whey was characterized and several hydrolysis experiments were made using whey proteins and purified beta -lactoglobulin, as substrates, and trypsin and a-chymotrypsin, as catalysts, at two temperatures and several enzyme concentrations. Maximum degrees of hydrolysis obtained experimentally were compared to the theoretical values and peptide compositions were calculated. For trypsin, 100% of yield was achieved; for alpha -chymotrypsin, hydrolysis seemed to be dependent on the oligopeptide size. The results showed that the two proteases could hydrolyze beta -lactoglobulin. Trypsin and alpha -chymotrypsin were stable at 40 degreesC, but a sharp decrease in the protease activity was observed at 55 degreesC.

Trichomonas vaginalis is the flagellate that causes trichomonadiasis, a sexually transmitted disease. Immunological methods have been proposed for the study of antigenic characterization using strains isolated from different patients. This work compares protease profiles from the different strains using gelatin containing polyacrylamide gels to analyse the protease activity. High molecular weight proteases (20 to 100 kDa) were found on gels showing quantitative differences. Human IgG antiproteases were detected by immunoblotting using the same extracts. These proteases could be related with T. vaginalis pathogenesis.

Os flavivírus são conhecidos por seu complexo ciclo biológico e importância na saúde pública e na economia mundial. Os aspectos ecológicos e quadros clínicos estão estreitamente relacionados à filogenia e evolução dos flavivírus. Este trabalho objetiva a caracterização molecular dos genomas dos flavivírus Bussuquara (VBSQ), Iguape (VIGU), Ilhéus (VILH) e Rocio (VROC), determinando relações filogenéticas com os demais integrantes do gênero Flavivirus. Foi realizado o seqüenciamento completo da região codificadora (ORF) e regiões não codificantes (RNC) 5’ e 3’; análise da estrutura secundária do RNA viral e das sequências conservadas da 3’RNC; determinação dos sítios de clivagem, glicosilação, resíduos Cis e motivos conservados na poliproteína; e as análises de similaridade e filogenética. Os genomas dos VBSQ, VIGU, VILH e VROC apresentaram a mesma organização que os demais flavivírus, medindo 10.815 nt, 10.922 nt, 10.775 nt, 10.794 nt, respectivamente. O padrão das sequências conservadas da 3’RNC do VBSQ foi RCS2-CS2-CS1, enquanto que para os VIGU, VILH e VROC foram CS3-RCS2-CS2-CS1. As características das estruturas secundárias do RNAs dos flavivírus em estudo foram similares aos demais flavivírus. O número dos sítios de glicosilação das proteínas PrM, E e NS1 foi distinto entre os flavivírus brasileiros, porém o padrão 6,12,12 dos resíduos de Cis e do sítios de clivagem permaneceram conservados. Na proteína E, alterações aminoacídicas pontuais foram observadas no peptídeo de fusão dos VBSQ, VIGU e VROC, e a sequência do tripepídeo RGD foi distinta para os quatro vírus em estudo. Os motivos determinantes das atividades de MTase-SAM da NS5, bem como da helicase e protease da NS3, permanecem conservados. Dentre os oito motivos da polimerase viral (NS5), somente os motivos V, VI e VII possuem alguma substituição nucleotídica para o VILH e VROC. As análises de similaridade mostram que VBSQ apresenta maior relação com VIGU enquanto que o VILH e VROC são mais relacionados entre si, porém sendo consideradas espécies virais distintas. Com base nas análises filogenéticas, características moleculares do genoma e biológicas, propõem-se a formação de três grupos genéticos: o grupo Rocio, que agrupa VROC e VILH; o grupo Bussuquara formado pelos VBSQ e Vírus naranjal e o grupo Aroa que inclui o Vírus Aroa e VIGU.

Background: Plasmodium has a complex cell biology and it is essential to dissect the cell-signalling pathways underlying its survival within the host. Methods: Using the fluorescence resonance energy transfer (FRET) peptide substrate Abz-AIKFFARQ-EDDnp and Fluo4/AM, the effects of extracellular ATP on triggering proteolysis and Ca2+ signalling in Plasmodium berghei and Plasmodium yoelii malaria parasites were investigated. Results: The protease activity was blocked in the presence of the purinergic receptor blockers suramin (50 mu M) and PPADS (50 mu M) or the extracellular and intracellular calcium chelators EGTA (5 mM) and BAPTA/AM (25, 100, 200 and 500 mu M), respectively for P. yoelii and P. berghei. Addition of ATP (50, 70, 200 and 250 mu M) to isolated parasites previously loaded with Fluo4/AM in a Ca2+-containing medium led to an increase in cytosolic calcium. This rise was blocked by pre-incubating the parasites with either purinergic antagonists PPADS (50 mu M), TNP-ATP (50 mu M) or the purinergic blockers KN-62 (10 mu M) and Ip5I (10 mu M). Incubating P. berghei infected cells with KN-62 (200 mu M) resulted in a changed profile of merozoite surface protein 1 (MSP1) processing as revealed by western blot assays. Moreover incubating P. berghei for 17 h with KN-62 (10 mu M) led to an increase in rings forms (82% +/- 4, n = 11) and a decrease in trophozoite forms (18% +/- 4, n = 11). Conclusions: The data clearly show that purinergic signalling modulates P. berghei protease(s) activity and that MSP1 is one target in this pathway.

The wide spectrum of candidiasis and its clinical importance encourage the research with the purpose of clarifying the mechanisms of pathogenicity and identification of virulence factors of Candida sp. Therefore, the aim of this study was to verify the adhesion capacity, protease activity and genotypic diversity of oral C. albicans and C. tropicalis isolates. The adhesion ability to the extracellular matrix glycoproteins laminin and fibronectin was evaluated using the ELISA technique. The research of proteases was carried out in agar plate containing bovine albumin and through a quantitative method in buffer solution containing haemoglobin. Intra and interspecies polymorphisms was verified through random amplified polymorphic DNA (RAPD) technique. All C. albicans and C. tropicalis isolates binded to immobilised laminin and fibronectin. Ca33 and Ct13 isolates had relative adhesion index significantly higher than the other isolates for both glycoproteins (P < 0.001). Protease activity was observed in all isolates of C. albicans using either the semi-quantitative or quantitative assay. The protease activity of C. tropicalis was better detected through the quantitative assay. The genotypic diversity by RAPD revealed a heterogeneous population in both species. Nevertheless, C. tropicalis presented higher genetic variability than C. albicans strains.

Abstract Background Citrus bacterial canker is a disease that has severe economic impact on citrus industries worldwide and is caused by a few species and pathotypes of Xanthomonas. X. citri subsp. citri strain 306 (XccA306) is a type A (Asiatic) strain with a wide host range, whereas its variant X. citri subsp. citri strain Aw12879 (Xcaw12879, Wellington strain) is restricted to Mexican lime. Results To characterize the mechanism for the differences in host range of XccA and Xcaw, the genome of Xcaw12879 that was completed recently was compared with XccA306 genome. Effectors xopAF and avrGf1 are present in Xcaw12879, but were absent in XccA306. AvrGf1 was shown previously for Xcaw to cause hypersensitive response in Duncan grapefruit. Mutation analysis of xopAF indicates that the gene contributes to Xcaw growth in Mexican lime but does not contribute to the limited host range of Xcaw. RNA-Seq analysis was conducted to compare the expression profiles of Xcaw12879 and XccA306 in Nutrient Broth (NB) medium and XVM2 medium, which induces hrp gene expression. Two hundred ninety two and 281 genes showed differential expression in XVM2 compared to in NB for XccA306 and Xcaw12879, respectively. Twenty-five type 3 secretion system genes were up-regulated in XVM2 for both XccA and Xcaw. Among the 4,370 common genes of Xcaw12879 compared to XccA306, 603 genes in NB and 450 genes in XVM2 conditions were differentially regulated. Xcaw12879 showed higher protease activity than XccA306 whereas Xcaw12879 showed lower pectate lyase activity in comparison to XccA306. Conclusions Comparative genomic analysis of XccA306 and Xcaw12879 identified strain specific genes. Our study indicated that AvrGf1 contributes to the host range limitation of Xcaw12879 whereas XopAF contributes to virulence. Transcriptome analyses of XccA306 and Xcaw12879 presented insights into the expression of the two closely related strains of X. citri subsp. citri. Virulence genes including genes encoding T3SS components and effectors are induced in XVM2 medium. Numerous genes with differential expression in Xcaw12879 and XccA306 were identified. This study provided the foundation to further characterize the mechanisms for virulence and host range of pathotypes of X. citri subsp. citri.

The lipase produced by a newly isolate Sporidiobolus pararoseus strain has potential catalysis ability for esterification reactions. In order to improve its synthetic activity, this work aimed at optimizing 'synthetic lipase' production by submerged fermentation of a conventional media based on peptone, yeast extract, NaCl and olive oil using experimental design technique. According to the results obtained in the first experimental design (2(4-1)), yeast extract and NaCl concentrations were tested to further optimization by response surface methodology. The maximum 'synthetic lipase' activity obtained was 26.9 U/mL in the optimized media (5.0, 6.8, 7.0 and 1.0% (wt/v) of peptone, yeast extract, NaCl and olive oil, respectively), representing a 6.36-fold increase compared to the initial medium. The time course of 'synthetic lipase' production in the optimized condition was evaluated in terms of synthetic activity, protease activity, biomass and total carbon and the maximum synthetic activity was observed during the stationary phase of growth.

Mikroorganismen spielen eine wichtige Rolle in der Weinherstellung. Neben ihren positiven Stoffwechselaktivitäten wie die Bildung von Ethanol während der alkoholischen Gärung sind vor allem Bakterien in der Lage, Weinfehler zu verursachen. Einer dieser Weinfehler ist die Produktion von biogenen Aminen. Diese niedermolekularen Stickstoffverbindungen können zu verschiedenen Gesundheitsproblemen wie Bluthochdruck und Migräne führen. Aufgrund von hohen Ethanolgehalten und dem Vorkommen verschiedener biogener Amine kommt es im Wein zu einer Verstärkung dieser physiologischen Effekte. Um die Bildung dieser Verbindungen zu verhindern, ist es von speziellem Interesse, die verantwortlichen Mikroorganismen zu identifizieren und sie in ihrem Wachstum zu hemmen.In einem Teil der Dissertation stand die Isolierung und Identifizierung biogener Amine produzierender Bakterien aus deutschen Jungweinen und Mosten im Vordergrund. Es konnte gezeigt werden, dass hauptsächlich Milchsäurebakterien als potenzielle Produzenten in Frage kommen. Diese Bakteriengruppe war in hohen Titern in nahezu allen Proben vorhanden und stellt somit eine potentielle Gefahr für die Weinbereitung dar. Zur Identifizierung der Isolate wurden verschiedene molekularbiologische Methoden wie specifically amplified DNA polymorphic-PCR (Fingerprintmethode), Multiplex-PCR oder 16S rDNA-Sequenzierung angewandt. Das Screening bezüglich der Bildung von biogenen Aminen erfolgte mit Hilfe einer im Rahmen dieser Arbeit entwickelten hochauflösenden Dünnschichtchromatographie gefolgt von der Quantifizierung mittels HPLC.Zur Wachstumshemmung dieser Schadbakterien wurden zwei Exoenzyme aus Streptomyces albidoflavus B578 isolieren. Diese Enzyme wurden gereinigt und als eine Muramidase und eine Protease identifiziert. Aktivitätstests konnten zeigen, dass diese Enzyme eine hohe lytische Wirkung gegen weinrelevante Mikroorganismen aufweisen. Ebenso war die Aktivität der Enzyme unter Weinbedingungen sehr stabil. Aufgrund dieser Ergebnisse könnten diese Enzyme eine mögliche Alternative zur Zugabe von Lysozym oder Schwefeldioxid sein, welche konventionell in der Weinbereitung ihren Einsatz finden.

Zu den Liganden des Zelloberflächenrezeptors RAGE gehören AGEs, S100-Proteine, HMGB1 und Aβ. RAGE wird daher eine Rolle bei verschiedenen neurologischen Erkrankungen sowie Diabetes, Arteriosklerose und Krebs zugesprochen. Des Weiteren geht eine Verringerung der Menge an sRAGE häufig mit diesen Krankheiten einher. Aus diesen Gründen stellt die pharmakologische Stimulierung der Proteolyse von RAGE eine vielversprechende Therapieform dar. Im Rahmen dieser Arbeit konnte gezeigt werden, dass eine Steigerung der sRAGE-Bildung über PAC1-, V2- und OT-Rezeptoren möglich ist. Die Untersuchung der PAC1-Signalwege zeigte, dass PKCα/PKCβI, CaMKII, Ca2+-Ionen, PI3-Kinase und der MAP-Kinase-Weg wichtig für die Stimulierung sind und dass der PKA-Weg nicht beteiligt ist. Die dreimonatige Behandlung von Mäusen mit PACAP-38 weist darauf hin, dass eine Stimulierung des Ectodomain Sheddings von RAGE auch in vivo erfolgen kann. Die Untersuchung der Signalwege, ausgehend von den V2- und OT-Rezeptoren, zeigte, dass ebenfalls PKCα/PKCβI, CaMKII, Ca2+-Ionen zur Aktivierung der Proteasen führen, dagegen konnte weder ein Einfluss des PKA- noch des MAP-Kinase-Weges festgestellt werden. Außerdem wurden sowohl MMP-9 als auch ADAM-10 als RAGE-spaltende Proteasen identifiziert. Die nähere Untersuchung der RAGE-Spaltstelle erbrachte, dass keine spezifische Sequenz, sondern vielmehr die Sekundärstruktur eine Rolle bei der Erkennung durch die Proteasen spielt. Im Rahmen der vorliegenden Arbeit wurde weiterhin ein anti-RAGE Antikörper anhand einer neu entwickelten Methode zunächst gereinigt und dann erfolgreich an ein mit dem Fluoreszenzfarbstoff Rhodamin markiertes Polymer gekoppelt. Die Stimulierung der Proteolyse von Meprin β wurde auch untersucht und es konnte ebenfalls eine Beteiligung von ADAM-10 an der Spaltung nachgewiesen werden.
