从栽培蕃茄(Lycopersilon esculentum Mill.)和秘鲁蕃茄(L. peruvianum Mill.)叶子中提取核DNA用EcoRI、BamHI、 HindⅢ限制性内切酶酶切,经Southern转移与digoxigenin标记的渗透调节蛋白cDNA探针进行杂交。从杂交图谱上可以看出两种蕃茄中都至少有两类渗调蛋白基因,同时两种蕃茄在杂交图谱上有差异。栽培蕃茄核DNA经HindⅢ酶切后的5kb和10kb两个片段从电泳凝胶中回收,插入到puc-19载体质粒中,宿主菌为JM109。然后以渗调蛋白cDNA为探针,进行菌落原位杂交,得到几个阳性克隆。

近年来,植物耐盐生物技术研究取得了可喜的进展,特别是通过抗盐基因转化在一定程度上使植物的耐盐性得到了提高。然而,植物的耐盐性是一个多基因控制的复杂性状,依赖于多个基因之间的相互作用。因此,只是将单个基因导入植物获得的抗逆性还是远不能达到满意的效果。一般认为,将多个与耐盐相关的基因转入到同一个植物(即所谓的“复合基因转化”)将会大大提高转基因植物的耐盐能力。 渗透调节是植物抵御盐胁迫的主要方式。植物渗透调节的方式分为两类:一是在细胞中吸收和积累无机盐,如通过离子通道、Na+/H+逆向运输蛋白和ATP酶/H+泵;二是在细胞中合成有机溶质,如脯氨酸和甘氨酸甜菜碱。 我们通过农杆菌介导法向转AtNHX1(拟南芥Na+/H+逆向运输蛋白编码基因)的番茄(Lycopersicum esculentum L. ‘Moneymaker’)株系X1OEA1自交二代植株(T2)中转入山菠菜甜菜碱醛脱氢酶基因(BADH)。PCR、Southern、RT-PCR和甜菜碱含量分析结果证明,BADH已经整合到目标植物基因组,并在转基因植株中转录和翻译表达。叶绿素荧光(Fv/Fm)、相对电导率(Rc/Rc’)、叶绿素含量(Chla+b)、叶绿素a/b比(Chla/b)和光合速率(Pn)测定结果表明,在200 mM NaCl 胁迫下,二次转化的番茄植株各项生理指标均优于转单基因AtNHX1的番茄。初步证明“复合基因转化”有助于进一步提高植物的耐盐性。同时对番茄的转化系统进行了优化,结果表明使用抗生素‘特美汀’作为抑制农杆菌的抗生素的转化效率明显高于使用头孢霉素。

A murcha-de-fusário, causada por Fusarium oxysporum f. sp. lycopersici (FOL), é uma importante doença do tomateiro (Lycopersicon esculentum Mill.) no mundo. Existem três raças identificadas do patógeno, sendo que a raça 3 ainda não havia sido registrada no Brasil. Este trabalho teve dois objetivos: comunicar a presença da raça 3 de FOL no Brasil e selecionar fontes de resistência às três raças do patógeno. Nove isolados de FOL foram obtidos de dois híbridos de tomate (Carmen e Alambra) com sintomas de mrucha, provenientes de três lavouras localizadas nos municípios de Venda Nova do Imigrante - Espírito Santo e Domingos Martins 9ES). Estes dois híbridos comerciais de tomate são considerados resistentes ás raças 1 e 2 de FOL. O teste de virulência foi feito com as cultivares: Ponderosa (suscetível a todasa as raças), IPA-5 (resistente à raça 1), Floradade (resistente às raças 1 e 2) e BHRS-2,3 (resistente às raças 1, 2 e 3). Todos os isolados foram virulentos às cultivares Ponderosa, IPA-R e Floradade e ainda infectaram algumas plantas de BHRS-2,3. O teste de virulência foi repetido com as mesmas cultivares mas também incluindo o acesso 'LA 716' da espécie selvagem L. pennellii. Foram obtidos resultados semelhantes para as cultivares, enquanto L. pennellii apresentou uma reação de imunidade ao patógeno. Estes resultadaos comprovam que os novos isolados de ES pertencem à raça 3 de FOL. Uma coleção de germoplasma de acessos de Lycopersicon spp. da Embrapa Hortaliças foi inicialmente avaliada quanto à reação de um dos isolados da raça 3 e uma parte deles às raças 1 e 2. Novas fontes de resistência múltipla foram identificadas em acessos de L. chilense, l. hirsutum e L. peruvianum, sendo dez genótipos imunes às raças 2 e 3 e cinco às três raças. A identificação destas fontes de resistência peermite que os programas de melhoramento de tomate antecipem potenciais problemas, inclusive a emergência de novas raças de FOL, além das raças 1, 2 e 3.

Una nueva teoría debe ir más allá de la ventaja comparativa y llegar a la ventaja competitiva de una nación. Debe explicar por qué las empresas de una nación consiguen ventajas competitivas en todas sus formas, que no solamente en los limitados tipos de ventajas basadas en los factores que se contemplan en la teoría de la ventaja comparativa. La mayoría de las teorías del comercio se fijan exclusivamente en el costo y tratan como nota a pie de página la calidad y los productos diferenciados. Una nueva teoría debe reflejar un rico concepto de la competencia que comprenda los mercados segmentados, los productos diferenciados, las diferencias en las tecnologías y las economías de escala. La calidad, las características y la innovación en los nuevos productos son determinantes en los sectores y segmentos avanzados. Además, la ventaja en los costos se deriva tanto de los diseños que permiten una fabricación eficiente y de las tecnologías avanzadas de proceso cuanto de los costos de los factores o incluso de las economías de escala. Debemos comprender las razones de que algunas naciones sean mejores que otras en la creación de estas ventajas, tan esenciales para la alta y creciente productividad1. En el presente estudio se interrelacionó la investigación documental y la investigación de campo; permitiendo demostrar que la producción de tomate riñón (o Lycopersicum Esculentum, pertenece a la familia de las solanáceas) bajo invernadero, tal como se plantea es técnica y económicamente factible; ya que el cultivo de tomate riñón es uno de los más representativos en cuanto a hortalizas, además, dado que el tomate constituye uno de los principales productos que la gente consume diariamente en sus comidas, lo hace acreedor a una gran demanda diaria sea en su presentación natural o ya procesado, por ejemplo tomate enlatado, pasta de tomate, salsa de tomate, entre otras.

'DRK') were grown hydroponically in two experiments to determine the effects of nutrient concentration and distribution in the root zone on yield, quality and blossom end rot (BER). The plants were grown in rockwool with their root systems divided into two portions. Each portion was irrigated with nutrient solutions with either the same or different electrical conductivity (EC) in the range 0 to 6 dS m(-1). In both experiments, fruit yields decreased as EC increased from moderate to high when solutions of equal concentration were applied to both portions of the root system. However, higher yields were obtained when a solution with high EC was applied to one portion of the root system and a solution of low EC to the other portion. For example, the fresh weight of mature fruits in the 6/6 treatment was only 20% that of the 3/3 treatment but the 6/0 treatment had a yield that was 40% higher. The reduction in yield in the high EC treatments was due to an increase in the number of fruits with BER and smaller fruit size. BER increased from 12% to 88% of total fruits as EC increased from 6/0 to 6/6 and fruit length decreased from 67 mm to 52 mm. Fruit quality (expressed as titratable acidity and soluble solids) increased as EC increased. In summary, high yields of high quality tomatoes with minimal incidence of BER were obtained when one portion of the root system was supplied with a solution of high EC and the other portion with a solution of moderate or zero EC.

Tomato plants (Lycopersicon esculentum Mill. var. DRK) were grown with a split root system to determine the effect of an unequal distribution of salinity in the root zone on yield and quality. The roots of the plant were divided into two portions and each portion was irrigated with nutrient solutions differing in EC levels achieved by adding Na or K. The maximum yield was observed in treatments with unequal EC when one portion of the roots received only water and the lowest in the high EC treatments. The reduced yield in the high EC treatment was due to the incidence of blossom-end rot and reduced fruit size. Fruit size in the treatments receiving solutions of unequal EC was up to 12% greater than that in the control. No significant differences were found in soluble solids and acidity between control and all other unequal EC treatments. Ca concentration was significantly higher in the treatments where one portion of the root system received water. It was concluded that high salinity had positive effects on yield and quality provided that one portion of the root system were placed in low EC or only water.

The concentration of K in the mature fruits of the uneven EC treatments was lower than that of the high EC treatments and higher or similar that of the 3/3 or 2.5/2.5 EC treatments ( controls). A clear relationship was found between the incidence of BER and the exudation rate. High rate of xylem exudation was observed in the uneven EC treatments. Reduction of BER in the uneven EC treatments is most likely to be the effect of high exudation rate on Ca status in the young fruits. It was concluded that high EC of solution had positive effects on Ca concentration and incidence of BER provided that nutrient solution with low EC or water is supplied to the one portion of the root system.

O experimento foi realizado em Jaboticabal-SP, sob ambiente protegido, objetivando avaliar quatro substratos (Rendimax-Estufa®, areia, solo coberto com filme de polietileno preto e solo descoberto) e quatro híbridos de tomateiro tipo cereja ('Mascot', 'Gisela', 'Cheri' e 'Sweet Million'), sendo os substratos comercial e areia acondicionados em sacos plásticos. O delineamento experimental foi em blocos casualizados, em esquema fatorial 4 x 4, com quatro repetições. Utilizou-se irrigação por gotejamento, sendo a dotação hídrica realizada em função dos dados obtidos em um tanque Classe A. A solução nutritiva utilizada foi a recomendada por Castellane & Araújo (1995) para a cultura do tomateiro. Os frutos foram colhidos semanalmente, durante o período de 24/11/2000 a 24/01/2001, sendo avaliados o número e produtividade diária de frutos. Os cultivos em solo proporcionaram maior produção diária que no substrato comercial e em areia, para os híbridos Gisele e 'Mascot'. O híbrido 'Gisela' mostrou-se mais produtivo nos cultivos em solo, enquanto o híbrido 'Cheri', embora tenha proporcionado menores produções em peso, produziu maior número de frutos por planta. A produtividade dos tratamentos mais produtivos foi satisfatória, estando de acordo com os padrões de produção para a cultura no Brasil.

O crescimento e desenvolvimento de uma planta dependem da intensidade, qualidade e duração da radiação solar. Por esse fator ser de importância vital às plantas, o presente trabalho objetivou fazer uma avaliação sobre sua variação, bem como sobre a sua disponibilidade no interior do ambiente protegido durante o ciclo do tomateiro nas estações verão-outono em Pelotas, Rio Grande do Sul. O experimento foi conduzido de janeiro a junho de 2003 no Campus da Universidade Federal de Pelotas (latitude 31°52'S; longitude 52°21'W e altitude de 13m), em estufa plástica disposta no sentido Leste-Oeste, com área de 180m². A cultivar utilizada foi Flora-dade, semeada em 24/01/03, transplantada no dia 28/02/03, sendo a última colheita em 12/06/03. Avaliou-se a radiação solar global externa (Rgext) e interna (Rgint), transmitância, radiação fotossinteticamente ativa (RFA) e o albedo da cultura a partir de sensores eletrônicos conectados a um datalloger. Durante o ciclo da cultura, o total de Rgext foi 1161,21MJ m-2, enquanto a Rgint foi 881,85MJ m-2. A Rgint e a RFA apresentaram valores médios diários de 8,5MJ m-2 dia-1 e 3,4MJ m-2 dia-1, respectivamente. A transmitância média da cobertura plástica à radiação solar global foi de 76%. O albedo médio diário da cultura foi 0,23, com albedo de 0,17 nos estádios iniciais, 0,26 no período de máximo crescimento e 0,23 no final do ciclo.

The production and quality of tomatoes has increased with the emergence of new genotypes and cropping systems such as hydroponics, however, there are few studies on the nutrition of plants. The objective was to evaluate the growth and motion of absorption of nutrients by Raisa tomato cultivar, grown in hydroponic system. The experimental design was randomized blocks with eight treatments consisting of the times of sampling: 15, 25, 35, 45, 55, 65, 75 and 85 days after transplanting (DAT) and five replications. The tomato seedlings were transplanted to pots of 8 dm(3) (on 31-03-2008), filled with the base substrate of coconut fiber, constantly irrigated with Hoagland and Arnon nutrient solution. During the experimental period and focuses on the development of plants and dry leaves, stems, fruits and roots. The dry matter accumulation of leaves and roots of tomato cultivar Raisa was relatively faster than the national stem and fruit, over cultivation, with predominance of dry fruits (45%) on the leaves (27%), the stem (24%) and roots (3%) at the end of the cycle. The accumulation of nutrients and micronutrients by the tomato cultivar Raisa was fit with linear increase throughout the cultivation, except for Mn which was quadratic. Most of the tomato nutritional requirement for nutrients was K, N and Ca and micronutrients was Fe, Zn and Mn.
