A simple enantioselective method for the determination of praziquantel (PZQ) and trans-4-hydroxypraziquantel (4-OHPZQ) in human plasma was developed and validated by high-performance liquid chromatography/mass spectrometry. The plasma samples were prepared by liquid-liquid extraction using a mixture of methyl-tert-butylether/dichloromethane (2:1, v/v) as extraction solvent. The direct resolution of PZQ and 4-OHPZQ enantiomers was performed on a Chiralpak AD column using hexane-isopropanol (75:25, v/v) as the mobile phase. Diazepam was used as internal standard. The method described here is simple and reproducible. The quantitation limit of 1.25 ng/ml for each PZQ enantiomer and of 12.5 ng/ml for each 4-OHPZQ enantiomer permits the use of the method in studies investigating the kinetic disposition of a single dose of 1.5g racemic PZQ. Enantioselectivity in the kinetic disposition of PZQ and 4-OHPZQ was observed in the clinical study. with the demonstration of a higher proportion of the (+)-(S)-PZQ and (-)-(R)-4-OHPZQ enantiomers in plasma. In order to combat a variety of pests, pesticides are widely used in fruits. Several extraction procedures (liquid extraction, single drop microextraction, microwave-assisted extraction, pressurized liquid extraction, supercritical fluid extraction, solid-phase extraction, solid-phase microextraction, matrix solid-phase dispersion, and stir bar sorptive extraction) have been reported to determine pesticide residues in fruits and fruit juices. The significant change in recent years is the introduction of the Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) methods in these matrices analysis. A combination of techniques reported the use of new extraction methods and chromatography to provide better quantitative recoveries at low levels. The use of mass spectrometric detectors in combination with liquid and gas chromatography has played a vital role to solve many problems related to food safety. The main attention in this review is on the achievements that have been possible because of the progress in extraction methods and the latest advances and novelties in mass spectrometry, and how these progresses have influenced the best control of food, allowing for an increase in the food safety and quality standards.

Pyrethroids are pesticides very used in agriculture, which tend to replace organophosfate and carbamate insecticides. These pesticides have shown to exhibit cardiotoxicity. The aim of this study was to assess if cardiotoxicity is due to direct or indirect effects (metabolites) of permethrin on hearts. There were studies 8 rats; three of them were sacrificed after 24 of the end of treatment with permethrin and the other four were sacrificed after 14 days of the end of treatment. Afterwards hearts and urines were collected. The amounts of permethrin and its main metabolite (3-PBA) were evaluated on hearts and urines of female rats which were treated with permethrin and sacrificed the day after and 14 days after the treatment. Moreover has been highlighted the difference of amount of permethrin and its metabolite between rats sacrificed immediately at the end of treatment and those sacrificed after 14 days. The study of permethrin was accomplished by liquid-liquid extraction and GC-ECD. The evaluation of 3-PBA was performed by SPE procedure with 2-PBA as internal standard and gas-chromatography GC-MS. The concentration of permethrin in hearts is basically the same in 24h and 14 days. The 3-PBA concentration in urines decreased 50 times from 24h to 14 days. In hearts the 3-PBA level also decrease but only 2.24 times and a high variation of results were achieved in rats after 14 days.

Este trabalho teve como principal objetivo o estudo da simulação da unidade de destilação atmosférica e a vácuo da fábrica de óleos base da Petrogal, permitindo verificar que, uma vez conseguido um modelo num programa de simulação que traduz o comportamento real de um dado processo, é possível sem riscos operacionais avaliar o efeito da alteração das condições normais de funcionamento desse processo. O trabalho foi orientado pelo Eng.º Carlos Reis, no que diz respeito à empresa, e pelo Prof. Dr. Luís Silva, pelo ISEP. O trabalho foi dividido em duas partes, sendo a primeira direcionada à obtenção do novo crude a partir de dois crudes pesados e caracterização dessa mistura. Já a segunda parte reside na refinação deste novo crude através da plataforma Aspen Plus para obtenção do resíduo de vácuo, para posterior processamento na coluna de discos rotativos usando um método de separação por extração liquido-liquido. Estudaram-se as propriedades físicas dos fluidos e verificou-se que na destilação atmosférica as curvas de destilação encontram-se muito próximas comparativamente com os resultados obtidos pela empresa, enquanto na destilação a vácuo os valores encontram-se mais afastados, apresentando uma variação de 30ºC a 100ºC em relação aos valores obtidos pelo Petro-Sim. Com a finalidade de cumprir um dos objetivos em falta, irão ser realizadas futuramente simulações no extrator de discos rotativos para otimização do processo para crudes pesados, sendo os resultados facultados, posteriormente, à Petrogal. Durante a realização do trabalho foi atingido um modelo real representativo da unidade de destilação atmosférica e a vácuo da Petrogal, podendo obter-se resultados para qualquer tipo de crude, fazendo apenas variar o assay da corrente de alimentação.

Synthetic pyrethroids are pesticides derived from naturally occurring pyrethrins, taken from pyrethrum of dried Chrysanthemum flowers. Pyrethroid insecticides are one of the most commonly used residential and agricultural insecticides. Many pyrethroids can significantly harm the nervous system. Permethrin could be one of the factors involved in the onset of neurodegenerative diseases. The present study aims to evaluate in brain, the effect that can induce the exposure to permethrin, during early life of female rats (from 6 to 21 days of life). Therefore, have been examined the concentrations of permethrin and its main metabolite (3-PBA) in the brain and urine in female rats sacrificed the day after and 14 days after treating. The different concentrations of permethrin and 3-PBA (after 24h and after 14 days in the end of treatment) were obtained using two different methods. The evaluation of permethrin by liquid-liquid extraction and GC-ECD was performed. The levels of the 3-PBA (in urine and brains) were obtained by SPE procedure and GC-MS using the 2- PBA as internal standard.

The use of natural pigments instead of synthetic colourants is receiving growing interest in the food industry. In this field, cactus pears (Opuntia spp.) have been identified to be a promising betalainic crops covering a wide coloured spectrum. The aim of this work was to develop adequate clean and mild methodologies for the isolation and encapsulation of betacyanins, from cactus pear fruits (Opuntia spp.). Firstly, two different emerging technologies, namely PLE (Pressurized Liquid Extraction) and HPCDAE (High Pressure Carbon Dioxide-Assisted Extraction), were exploited to isolation of betacyanins form cactus pear fruits. Different process conditions were tested for the maximum recovery of betacyanins. Results showed that highest extraction yields were achieved for HPCDAE and mass ratio of pressurized carbon dioxide vs. acidified water was the parameter that most affected the betacyanins extraction. At optimum conditions of HPCDAE, Opuntia spp. extract presented a total betacyanin content of 211 ± 10 mg/100 g whereas extracts obtained using conventional extraction, PLE in static and in dynamic mode presented a total betacyanin content of 85 ± 3, 191 ± 2 and 153 ± 5 mg/100 g, respectively. HPCDAE has proven to be a successful technology to extract betacyanins from Opuntia spp. fruits. Afterward, Supercritical Fluid Technology was exploited to develop lipidic particles of betalain-rich extract. A betacyanin-rich conventional extract was encapsulated by PGSS® (Particles from Gas Saturated Solutions) technique. Different process conditions were tested in order to model the encapsulation of betacyanins. The pressure had a negative effect on betacyanin encapsulation. Lower pressures leads to an increase in the betacyanin encapsulation. This effect was more pronounced at higher temperatures and lower equilibrium time. At these conditions, Opuntia spp. particles presented 64.4 ± 4.5 mg/100 g and high antioxidant capacity. When compared with the Opuntia spp. dried extract, lipidic particles contributed to a better homogenization of the pink colour after incorporation in ice cream.

Countries are currently faced with problems derived from changes in lifespan and an increase in lifestyle-related diseases. Neurodegenerative disorders such Parkinson’s (PD) and Alzheimer’s (AD) diseases are an increasing problem in aged societies. Data from World Alzheimer Report 2011 indicate that 36 million people worldwide are living with dementia. Oxidative stress has been associated with the development of AD and PD. Therefore there is interest to search for effective compounds or therapies to combat the oxidative damage in these diseases. Current evidence strongly supports a contribution of phenolic compounds present in fruits and vegetables to the prevention of neurodegenerative diseases such AD and PD. The industrial processing of a wide variety of fruits results in the accumulation of by-products without commercial value. Opuntia ficus-indica (cactus pear) is consumed fresh and processed like in juice. Prunnus avium (sweet cherry) is consumed fresh but the organoleptics characteristics of the fruits leads to the smaller and ragged fruits have no commercial value. Fruit extracts of both species has described to be rich in phenolic compounds and to have high antioxidant activities due to its composition. The aim of this work was assessing the efficacy of O. ficus-indica and P. avium by-products extracts obtained with conventional solvent extraction and pressurized liquid extraction in a neurodegeneration cell model. All extracts have protected neuroblastoma cells from H2O2-induced death at low, non-toxic levels, which approach to physiologically-relevant serum concentration. However, cherry extract has a slighter neuroprotective activity. The protective effect of Opuntia extracts are not conducted by a direct antioxidant activity since there are not decreases in intracellular ROS levels in cell treated with extracts and challenged with H2O2, while cherry extract neuroprotection seems to be due to a direct scavenging activity. Extracts from different biological matrixes seems to protect neuronal cells trough different cellular mechanisms.

Occurrence of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) was evaluated in sepiolite as a widely employed binder and anti-caking agent for animal feed. Also, naturally contaminated kaolinitic clay was used for comparative purposes. Since sepiolite shows remarkable adsorption properties, particular interest was paid to the extraction steps as they become critical for the final determination of these pollutants in such matrixes. Furthermore, classical Soxhlet extraction using different extracting strategies as well as acid treatment were carried out with simultaneous liquid-liquid extraction. Results obtained depended on the extraction procedure applied. Acid treatment or Soxhlet extraction using a mixture of toluene:ethanol as solvent allowed to reach the minimum requirements of recovery rates. However, Soxhlet extraction using a mixture cyclohexane:toluene as extracting solvent did not allow to comply with minimum specifications for recovery. Significant differences were obtained in TEQ units when acid treatment was applied in comparison to Soxhlet extraction. This fact can be explained because the use of drastic acid conditions allows removing strongly adsorbed analytes which can be uniquely extracted after a total destruction of the crystalline structure of sepiolite. On the contrary, Soxhlet extraction was not able to destroy the structure of sepiolite and as a consequence the PCDDs/Fs were strongly adsorbed in the internal structure of the mineral. From biological point of view the availability of these toxicants constitutes a critical aspect playing an important role in the final decision choosing particular analytical procedures. Then, acid conditions in the digestive tract should be taken into account. In this scenario, a bioaccumulation study was conducted to evaluate the transference of PCDDs/PCDFs from the sepiolite into the animal tissues when fed with feed containing sepiolite. To this end, chickens were used as a model to examine the bioavailability of PCDDs/PCDFs. Four groups of chickens were exposed through their diet to a control feed, feed with 3% w/w sepiolite as additive, feed contaminated with PCDDs/PCDFs at concentration around 2.8 pg WHO-TEQ/g and feed with 2% of a contaminated kaolinitic clay (460 pg TEQ/g mineral). Livers of the four studied groups were analyzed throughout the exposure period. Results of this trial showed that the performance of broilers was not affected by the presence of dioxins at levels tested, and chickens did not show any abnormal behaviour. Dioxins intentionally added to the diet were absorbed and accumulated in the liver in a significant manner, whereas the PCDDs/Fs from sepiolite were not available for chickens since livers from broilers fed 3% sepiolite presented similar WHO-TEQ values than those from broilers fed control diet.
