La Vitamine K (VK) est largement reconnue pour son rôle dans la coagulation sanguine toutefois, de plus en plus de travaux indiquent son implication dans la fonction cérébrale. La VK est requise pour l'activation de différentes protéines, par exemple la protéine Gas6, et la ménaquinone-4 (MK-4), le principal vitamère K dans le cerveau, est impliquée dans le métabolisme des sphingolipides. Dans un rapport précédent, nous avons montré qu'un régime alimentaire faible en VK tout au long de la vie était associé à des déficits cognitifs chez des rats âgés. La warfarine sodique est un puissant antagoniste de la VK qui agit en bloquant le cycle de la VK, provoquant un «déficit relatif de VK » au niveau cellulaire. À la lumière du rôle émergent de la VK dans le cerveau, la warfarine pourrait représenter un facteur de risque pour la fonction cérébrale. Ce travail est donc pertinente en raison de la forte proportion d'adultes traîtés à la warfarine sodique. Dans la présente étude, 14 rats mâles Wistar ont été traités avec 14 mg de warfarine/kg /jour (dans l'eau potable) et des injections sous-cutanées de VK (85 mg/kg), 3x/sem, pendant 10 semaines. Quatorze rats témoins ont été traités avec de l'eau normale et injectés avec une solution saline. Les rats ont été soumis à différents tests comportementaux après quoi les niveaux de phylloquinone, MK-4, sphingolipides (cérébroside, sulfatide, sphingomyéline, céramide et gangliosides), et les sous-types de gangliosides (GT1b, GD1a, GM1, GD1b), ont été évalués dans différentes régions du cerveau. Comparativement aux rats du groupe contrôle, les rats traités à la warfarine présentaient des latences plus longues au test de la piscine de Morris (p <0,05) ainsi qu'une hypoactivité et un comportement exploratoire plus faible au test de « l’open field » (p <0,05). Le traitement par warfarine a également entraîné une diminution spectaculaire du niveau de MK-4 dans toutes les régions du cerveau (p <0,001), une altération des concentrations de sphingolipidiques, en particulier dans le cortex frontal et le mésencéphale (p <0,05), et une perte de différences régionales sphingolipidiques, notamment pour les gangliosides. Le traitement par warfarine a été associé à un niveau inférieur de GD1a dans l'hippocampe et un niveau supérieur de GT1b dans le cortex préfrontal et le striatum. En conclusion, la déficience en VK induite par warfarine altère les niveaux de VK et sphingolipides dans le cerveau, avec de potentiels effets néfastes sur les fonctions cérébrales.

Let G be finite group and K a number field or a p-adic field with ring of integers O_K. In the first part of the manuscript we present an algorithm that computes the relative algebraic K-group K_0(O_K[G],K) as an abstract abelian group. We solve the discrete logarithm problem, both in K_0(O_K[G],K) and the locally free class group cl(O_K[G]). All algorithms have been implemented in MAGMA for the case K = \IQ. In the second part of the manuscript we prove formulae for the torsion subgroup of K_0(\IZ[G],\IQ) for large classes of dihedral and quaternion groups.

One among the most influential and popular data mining methods is the k-Means algorithm for cluster analysis. Techniques for improving the efficiency of k-Means have been largely explored in two main directions. The amount of computation can be significantly reduced by adopting geometrical constraints and an efficient data structure, notably a multidimensional binary search tree (KD-Tree). These techniques allow to reduce the number of distance computations the algorithm performs at each iteration. A second direction is parallel processing, where data and computation loads are distributed over many processing nodes. However, little work has been done to provide a parallel formulation of the efficient sequential techniques based on KD-Trees. Such approaches are expected to have an irregular distribution of computation load and can suffer from load imbalance. This issue has so far limited the adoption of these efficient k-Means variants in parallel computing environments. In this work, we provide a parallel formulation of the KD-Tree based k-Means algorithm for distributed memory systems and address its load balancing issue. Three solutions have been developed and tested. Two approaches are based on a static partitioning of the data set and a third solution incorporates a dynamic load balancing policy.

Tumor cell survival and proliferation is attributable in part to suppression of apoptotic pathways, yet the mechanisms by which cancer cells resist apoptosis are not fully understood. Many cancer cells constitutively express heme oxygenase-1 (HO-1), which catabolizes heme to generate biliverdin, Fe(2+), and carbon monoxide (CO). These breakdown products may play a role in the ability of cancer cells to suppress apoptotic signals. K(+) channels also play a crucial role in apoptosis, permitting K(+) efflux which is required to initiate caspase activation. Here, we demonstrate that HO-1 is constitutively expressed in human medulloblastoma tissue, and can be induced in the medulloblastoma cell line DAOY either chemically or by hypoxia. Induction of HO-1 markedly increases the resistance of DAOY cells to oxidant-induced apoptosis. This effect was mimicked by exogenous application of the heme degradation product CO. Furthermore we demonstrate the presence of the pro-apoptotic K(+) channel, Kv2.1, in both human medulloblastoma tissue and DAOY cells. CO inhibited the voltage-gated K(+) currents in DAOY cells, and largely reversed the oxidant-induced increase in K(+) channel activity. p38 MAPK inhibition prevented the oxidant-induced increase of K(+) channel activity in DAOY cells, and enhanced their resistance to apoptosis. Our findings suggest that CO-mediated inhibition of K(+) channels represents an important mechanism by which HO-1 can increase the resistance to apoptosis of medulloblastoma cells, and support the idea that HO-1 inhibition may enhance the effectiveness of current chemo- and radiotherapies.

Background

Patients with end-stage renal failure (ESRF) exhibit grossly impaired maximal exercise performance. This study investigated whether K+ regulation during exercise is impaired in ESRF and whether this is related to reduced exercise performance.

Methods

Nine stable hemodialysis patients and eight controls (CON) performed incremental cycling exercise to volitional fatigue, with measurement of peak oxygen consumption (VdotO2 peak). Arterial blood was sampled during and following exercise and analyzed for plasma [K+] (PK).

Results

The VdotO2 peak was approximately 44% less in ESRF than in CON (P < 0.001), whereas peak exercise PK was greater (7.23 plusminus 0.38 vs. 6.23 plusminus 0.14 mmol dot L-1, respectively, P < 0.001). In ESRF, the rate of rise in PK during exercise was twofold greater (0.43 plusminus 0.05 vs. 0.23 plusminus 0.03 mmol dot L-1dotmin-1, P < 0.005) and the ratio of rise in PK relative to work performed was 3.7-fold higher (90.1 plusminus 13.5 vs. 24.7 plusminus 3.3 nmol dot L-1dot J-1, P < 0.001). A strong inverse relationship was found between VdotO2 peak and the DeltaPKdot work-1 ratio (r = -0.80, N = 17, P < 0.001).

Conclusions

Patients with ESRF exhibit grossly impaired extrarenal K+ regulation during exercise, demonstrated by an excessive rise in PK relative to work performed. We further show that K+ regulation during exercise was correlated with aerobic exercise performance. These results suggest that disturbed K+ regulation in ESRF contributes to early muscle fatigue during exercise, thus causing reduced exercise performance.

A deficiência de guanidino acetato metiltransferase (GAMT) é um erro inato do metabolismo da creatina caracterizado por hipotonia muscular, movimentos extrapiramidais involuntários e epilepsia. A doença é bioquimicamente caracterizada por acúmulo de guanidino acetato e deficiência de creatina e fosfocreatina nos tecidos dos pacientes afetados. Os mecanismos de disfunção neurológica que ocorrem nessa doença ainda são desconhecidos. A Na+,K+-ATPase desempenha um papel fundamental no sistema nervoso central (SNC), sendo responsável pela manutenção dos gradientes iônicos e pela propagação do impulso nervoso, consumindo cerca de 50% do ATP formado no cérebro. A acetilcolinesterase (AChE) é uma importante enzima regulatória que controla a transmissão de impulsos nervosos através de sinapses colinérgicas pela hidrólise da acetilcolina, e apresenta um papel fundamental na cognição. Com o propósito de ampliar o conhecimento sobre os mecanismos fisiopatológicos da deficiência de GAMT, esse trabalho teve como objetivo investigar o efeito do guanidino acetato, o principal metabólito acumulado na deficiência de GAMT, sobre as atividades das enzimas Na+,K+-ATPase, Mg2+-ATPase e AChE em estriado de ratos. A cinética de inibição da Na+,K+-ATPase causada pelo guanidino acetato também foi estudada. Além disso, investigamos o efeito in vitro do guanidino acetato sobre as atividades da Na+,K+-ATPase, Mg2+-ATPase e da AChE de hipocampo de ratos. Nossos resultados mostraram que o guanidino acetato não altera as atividades da AChE e Mg2+-ATPase. No entanto, a atividade da Na+,K+-ATPase foi inibida por esse composto guanidínico (CG), e a análise cinética mostrou uma inibição do tipo acompetitiva. Também foi demonstrada uma interação entre o guanidino acetato e o ácido arginínico, sugerindo um sítio comum de ligação entre esses dois compostos na Na+,K+-ATPase. Os resultados mostraram que, o guanidino acetato inibiu a atividade da Na+,K+-ATPase in vitro mas não alterou as atividades da Mg2+-ATPase e da AChE. Considerando que o guanidino acetato e outros compostos guanidínicos (CG) induzem a formação de espécies reativas de oxigênio e que a Na+,K+-ATPase e a AChE são inibidas por radicais livres, estudamos o efeito da pré-incubação de homogeneizado de hipocampo de ratos na presença de guanidino acetato sobre a atividade dessas enzimas. Além disso, o efeito da pré-incubação de homogeneizado de ratos com guanidino acetato foi investigado na presença e ausência de antioxidantes, tais como glutationa (GSH), trolox, taurina e L-NAME. A pré-incubação de homogeneizado de hipocampos na presença de guanidino acetato inibiu a atividade da Na+,K+-ATPase, mas não alterou a atividade da Mg2+-ATPase. No entanto, L-NAME e taurina foram capazes de prevenir tal efeito. Dessa forma, propõem-se que a inibição da atividade da Na+,K+-ATPase pelo guanidino acetato seja um dos mecanismos envolvidos na disfunção neuronal observada em pacientes com deficiência de GAMT.

Subjects taking a hydrogen pump blocking agent (omeprazole) develop bacterial overgrowth of the small intestine. We tested the hypothesis that this bacterial overgrowth produces menaquinones, which would meet the Vitamin requirement in situations of vitamin K deficiency. In a crossover-type design, 13 healthy Volunteers eating a phylloquinone-restricted diet for 35 d were randomly assigned to take omeprazole during the first period of study or starting on day 15 until the end of the study. Coagulation times, serum osteocalcin [total osteocalcin and undercarboxylated osteocalcin (ucOC)], plasma phylloquinone, urinary gamma-carboxyglutamic acid, and plasma undercarboxylated prothrombin (PIVKA-II) were measured. Plasma phylloquinone concentrations declined 82% with dietary phylloquinone restriction (P < 0.05) and were not significantly different in the period when the diet was combined with omeprazole treatment (P > 0.05), the mean value for PIVKA-II during the phylloquinone-restricted diet significantly increased 5.7-fold from baseline (P < 0.05); however, the combination of omeprazole treatment and the phylloquinone-restricted diet significantly reduced PIVKA-II values by 21% (P < 0.05) compared with the diet period alone. There were no alterations in total or percentage ucOC concentrations during the phylloquinone-restricted diet or during the period of diet plus omeprazole treatment. Our data support the hypothesis that bacterial overgrowth results in the synthesis and absorption of menaquinones. These menaquinones contribute to vitamin K nutriture during dietary phylloquinone restriction, but not enough to restore normal vitamin K status.

Type II Bartter's syndrome is a hereditary hypokalemic renal salt-wasting disorder caused by mutations in the ROMK channel (Kir1.1; Kcnj1), mediating potassium recycling in the thick ascending limb of Henle's loop (TAL) and potassium secretion in the distal tubule and cortical collecting duct (CCT). Newborns with Type II Bartter are transiently hyperkalemic, consistent with loss of ROMK channel function in potassium secretion in distal convoluted tubule and CCT. Yet, these infants rapidly develop persistent hypokalemia owing to increased renal potassium excretion mediated by unknown mechanisms. Here, we used free-flow micropuncture and stationary microperfusion of the late distal tubule to explore the mechanism of renal potassium wasting in the Romk-deficient, Type II Bartter's mouse. We show that potassium absorption in the loop of Henle is reduced in Romk-deficient mice and can account for a significant fraction of renal potassium loss. In addition, we show that iberiotoxin (IBTX)-sensitive, flow-stimulated maxi-K channels account for sustained potassium secretion in the late distal tubule, despite loss of ROMK function. IBTX-sensitive potassium secretion is also increased in high-potassium-adapted wild-type mice. Thus, renal potassium wasting in Type II Bartter is due to both reduced reabsorption in the TAL and K secretion by max-K channels in the late distal tubule. © 2006 International Society of Nephrology.

A challenge in mesonic three-body decays of heavy mesons is to quantify the contribution of re-scattering between the final mesons. D decays have the unique feature that make them a key to light meson spectroscopy, in particular to access the Kn S-wave phase-shifts. We built a relativis-tic three-body model for the final state interaction in D+ → K -π+π+ decay based on the ladder approximation of the Bethe-Salpeter equation projected on the light-front. The decay amplitude is separated in a smooth term, given by the direct partonic decay amplitude, and a three-body fully interacting contribution, that is factorized in the standard two-meson resonant amplitude times a reduced complex amplitude that carries the effect of the three-body rescattering mechanism. The off-shell reduced amplitude is a solution of an inhomogeneous Faddeev type three-dimensional integral equation, that includes only isospin 1/2 K -π+ interaction in the S-wave channel. The elastic K-π+ scattering amplitude is parameterized according to the LASS data[1]. The integral equation is solved numerically and preliminary results are presented and compared to the experimental data from the E791 Collaboration[2, 3] and FOCUS Collaboration[4, 5].

We investigate the decay Bs0→J/ψK +K - for invariant masses of the K +K - pair in the range 1.35integrated luminosity of 10.4fb -1 of pp̄ collisions at √s=1.96TeV accumulated with the D0 detector at the Fermilab Tevatron collider. From the study of the invariant mass and spin of the K +K - system, we find evidence for the two-body decay Bs0→J/ψf2′(1525) and measure the relative branching fraction of the decays Bs0→J/ψf2′(1525) and Bs0→J/ψ to be R f2′/=0.19±0.05(stat)±0.04(syst). © 2012 American Physical Society.

We extend some results of Rosenthal, Cembranos, Freniche, E. Saab-P. Saab and Ryan to study the geometry of copies and complemented copies of c(0)(Gamma) in the classical Banach spaces C(K, X) in terms of the carclinality of the set Gamma, of the density and caliber of K and of the geometry of X and its dual space X*. Here are two sample consequences of our results: (1) If C([0, 1], X) contains a copy of c(0)(N-1), then X contains a copy of c(0)(N-1). (2) C(beta N, X) contains a complemented copy of c(0)(N-1) if and only if X contains a copy of c(0)(N-1). Some of our results depend on set-theoretic assumptions. For example, we prove that it is relatively consistent with ZFC that if C(K) contains a copy of c(0)(N-1) and X has dimension NI, then C(K, X) contains a complemented copy of cc(0)(N-1).

̀ qui presentato lo studio della produzione della risonanza K∗0 in collisioni p-Pb con l’esperimento ALICE presso LHC. L’elaborato si compone di una introduzione sulla natura del fenomeno studiato: la formazione del Quark Gluon Plasma (QGP), uno stato della materia fortemente interagente ad alte temperatura e densità d’energia. Vengono descritte le segnature studiate ai fini di identificare il suddetto fenomeno, riportando come esempio concreto i risultati sperimentali. Successivamente l’acceleratore di particelle, LHC, e l’esperimento, ALICE, vengono brevemente introdotti. Più in dettaglio ven- gono descritti i rivelatori di ALICE effettivamente usati per l’analisi, a cui sono dedicate sezioni approfondite. Viene infine introdotta l’analisi e le sue motivazioni. Il metodo utilizzato e lo studio degli errori da associare alla misura sono illustrati in ogni loro passo e supportati dai risultati ottenuti. La discussione finale dei risultati include il confronto con i risultati preceden- temente ottenuti da ALICE in collisioni pp e Pb-Pb e da altri esperimenti.

Open web steel joists are designed in the United States following the governing specification published by the Steel Joist Institute. For compression members in joists, this specification employs an effective length factor, or K-factor, in confirming their adequacy. In most cases, these K-factors have been conservatively assumed equal to 1.0 for compression web members, regardless of the fact that intuition and limited experimental work indicate that smaller values could be justified. Given that smaller K-factors could result in more economical designs without a loss in safety, the research presented in this thesis aims to suggest procedures for obtaining more rational values. Three different methods for computing in-plane and out-of-plane K-factors are investigated, including (1) a hand calculation method based on the use of alignment charts, (2) computational critical load (eigenvalue) analyses using uniformly distributed loads, and (3) computational analyses using a compressive strain approach. The latter method is novel and allows for computing the individual buckling load of a specific member within a system, such as a joist. Four different joist configurations are investigated, including an 18K3, 28K10, and two variations of a 32LH06. Based on these methods and the very limited number of joists studied, it appears promising that in-plane and out-of-plane K-factors of 0.75 and 0.85, respectively, could be used in computing the flexural buckling strength of web members in routine steel joist design. Recommendations for future work, which include systematically investigating a wider range of joist configurations and connection restraint, are provided.
