This paper gives a brief survey of research and development work done on hand pumps in India as well as elsewhere and sets out the approach adopted by ASTRA Working Group. Ten ways in which a hand pump breakdown in practice have been identified. The physical reasons behind each type of breakdown analysed. Remedial measures have been developed from this analysis. Laboratory test rigs fabricated to evaluate these measures have been described and some experimental results presented. The course of further work has been charted.

An indigenous electron energy loss spectrometer has been designed and fabricated for the study of free molecules. The spectrometer enables the recording of low-resolution electronic spectra of molecules inthe vapour phase with ready access to the vacuum ultraviolet region. Electron energy loss spectra of aliphatic alcohols and carbonyl compounds as wellas of benzene derivatives have been recorded with the indigenous spectrometer and the electronic transitions in these molecules discussed.

Using elementary comparison geometry, we prove: Let (M, g) be a simply-connected complete Riemannian manifold of dimension >= 3. Suppose that the sectional curvature K satisfies -1-s(r) <= K <= -1, where r denotes distance to a fixed point in M. If lim(r ->infinity) e(2r) s(r) = 0, then (M, g) has to be isometric to H-n.The same proof also yields that if K satisfies -s(r) <= K <= 0 where lim(r ->infinity) r(2) s(r) = 0, then (M, g) is isometric to R-n, a result due to Greene and Wu.Our second result is a local one: Let (M, g) be any Riemannian manifold. For a E R, if K < a on a geodesic ball Bp (R) in M and K = a on partial derivative B-p (R), then K = a on B-p (R).

Wavenumber-frequency spectral analysis of different atmospheric variables has been carried Out using 25 years of data. The area considered is the tropical belt 25 degrees S-25 degrees N. A combined FFT wavelet analysis method has been used for this purpose. Variables considered are outgoing long wave radiation (OLR), 850 hPa divergence, zonal and meridional winds at 850, 500 and 200 hPa levels, sea level pressure and 850 hPa geopotential height. It is shown that the spectra of different variables have some common properties, but each variable also has few features diffe:rent from the rest. While Kelvin mode is prominent in OLR, and zonal winds, it is not clearly observed in pressure and geopotential height fields; the latter two have a dominant wavenumber zero mode not seen in other variables except in meridional wind at 200 hPa and 850 hPa divergences. Different dominant modes in the tropics show significant variations on sub-seasonal time scales.

The effect of the addition of different concentratons of cystine and cysteine on sporulation and parasporal crystal formation in Bacillus thuringiensis var. thuringiensis was studied. The effect was well pronounced when the systine/cysteine additions were made after the stationary phase. Heat stable spores and crystals were formed when the culture was provided with a low concentration of cystine/cysteine (0.05 per cent w/v). At a moderate concentration of cystine or cysteine (0.15%), only heat labile spores were formed without the production of the crystal. When the cystine/cysteine concentration was high (0.25%), spore and crystal formation were completely inhibited. Partial reversal of inhibition of sporulation was brought about by sodium sulphate or zinc sulphate and lead, copper, cadmium or cobalt acetate at 0.2 mM or at 0.2% of sodium or potassium pyruvate, citrate, isaconitate, oxalosuccinate, ∝ -keto-glutarate, succinate, fumarate, malate, or oxalacetate. Glutamate (0.2%) overcame the inhibitory effect of cystine/cysteine completely. The structural changes observed using phase contrast microscopy were dependent upon the concentration of cystine/cysteine.

The growth patterns of Mycobacterium smegmatis SN2 in a minimal medium and in nutrient broth have been compared. The growth was monitored by absorbancy (Klett readings), colony forming units, wet weight and content of DNA, RNA and protein. During the early part of the growth cycle, the bacteria had higher wet weight and macromolecular content in nutrient broth than in minimal media. During the latter half of the growth cycle however, biosynthesis stopped much earlier in nutrient broth and the bacteria had a much lower content of macromolecules than in the minimal medium. In both the media, a general pattern of completing biosynthesis rapidly in the initial phase and a certain amount of cell division at a later time involving the distribution of preformed macromolecules was seen. The possible adaptive significance of this observation has been discussed.

The technology scene in India is at one and the same time promising, frustrating and fascinating. Three broad areas in technology development can be distinguished. The first is relatively small scale; it is typified by the absorption of products of the industrial revolution into the repertoire of the Indian artisan and craftsman, examples being diesel engines from Kolhapur and centrifugal pumps from Coimbatore. The second class is essentially 'state technology', developed at public expense by national commissions: agriculture, atomic energy and space are examples. There is a vast third area in both private and public sector, covering products for the urban consumer and the state (e.g. r defence); this area has largely remained colonial. The factors affecting the three areas of technology are described and analysed from the point of view of an Indian scientistengineer; and it is concluded that the enormous potential of the country's human and mat.erial resources is not only unrealized, but even unrecognized as yet.

The administration of a potent antiestrogen, tamoxifen at a dose of 3 mg/kg body weight/day orally post-coitally to cycling mated bonnet monkeys(Macaca radiata) from days 18–30 of cycle resulted in inhibition of establishment of pregnancy in 9 out of 10 monkeys. Tamoxifen effect was not due to interference with luteal function. The effect was specific to tamoxifen as exogenously administered progesterone could not reverse it. In addition to suggesting a role for estrogen in maintenance of early pregnancy in the primate the present study could be a prelude to the development of an effective post-ovulatory approach for regulation of fertility in the human female

Intracranial artery aneurysms (IAs) are estimated to be present in 2.3% of the population. A rupture of an IA causes subarachnoid hemorrhage, with up to 50% mortality. The annual low rupture risk of an IA indicates that most IAs never rupture. The current treatment options are invasive and somewhat risky. Thus rupture-prone IAs should be identified and this requires a better understanding of the IA wall pathobiology. Inflammatory cell infiltrations have been found to precede IA rupture, indicating the role of inflammation in IA wall degeneration and rupture. The complement system is a key mediator of inflammation and house-hold processing of injured tissue. This study aimed at identifying the role of complement activation in IA wall degeneration and the complement activators involved and determining how the complement system is regulated in the IA wall. In immunostainings, the end-product of complement activation, the terminal complement complex (TCC), was located mainly in the outer part of the IA wall, in areas that had also sustained loss of cells. In electron microscopy, the area of maximum TCC accumulation contained cellular debris and evidence of both apoptotic and necrotic cell death. Complement activation correlated with IA wall degeneration and rupture, de-endothelialization, and T-cell and CD163-positive macrophage infiltration. The complement system was found to become activated in all IAs by the classical pathway, with recruitment of alternative pathway amplification. Of the potential activators immunoglobulins G and M and oxidatively modified lipids were found in large areas. Lipid accumulation was observed to clearly colocalize with TCC and C-reactive protein. In the luminal parts of the IA wall, complement activation was limited by cellular expression of protectin (CD59) and extracellular matrix-bound inhibitors, C4b binding protein and factor H whereas the outer part of the wall lacked cells expressing protectin as well as matrix-bound factor H. In single nucleotide polymorphism-analysis, age-related macular degeneration-associated factor H Y402H polymorphism did not associate with the presence of IAs or their rupture The data suggest that complement activation and TCC formation are involved in IA wall degeneration and rupture. Complement seems to become activated by more than one specific activator. The association of complement with de-endothelialization and expression of several complement activators indicate a possible role of endothelial dysfunction and/or impaired clearance mechanisms. Impaired complement regulation seems to be associated with increased complement activation in IA walls. These results stress the role of chronic inflammation in IA wall pathobiology and the regulatory role of complement within this process. Imaging inflammation would possibly enhance the diagnostics of rupture-prone IAs, and targeting IA treatment to prevent chronic inflammation might improve IA treatment in the future.
