A solid-phase extraction and chromatography-flame ionization detection (GC-FID) method has been developed for the routine analysis of psoralen, bergapten, isopimpinellin and pimpinellin in creams and pomades employed in Brazil for the treatment of vitiligo. The calibration curve for psoralen was linear in the range 10-100 mu g ml(-1), for bergapten 5-90 mu g ml(-1), for pimpinellin 10-90 mu g ml(-1) and for isopimpinellin 5-100 mu g ml(-1). The best recoveries of the furanocoumarins in the creams analysed were 94-97%, whereas in the pomades, recoveries were 94-96%. The R.S.D. of the quantitative analysis of the furanocoumarins in the products analyses were within 5%. (C) 2000 Elsevier B.V. B.V. All rights reserved.

Organotin compounds, largely used as biocides in antifouling paints, are among the most toxic materials introduced into the aquatic environment. Sensitive analytical methods are thus required to characterize their occurrence in environmental and biological matrices. The comparison between two different photometric detectors in terms of analytical performance was carried out for the analysis of organotin compounds. A flame photometric detector (FPD) and a pulsed flame photometric detector (PFPD) were optimized. Their respective sensitivity, linearity range and selectivity were evaluated. Limits of detection obtained for a tributyltin compound (TBT) were 5.0 and 0.9 pg (as Sn) for the FPD and PFPD, respectively, using a 390 nm filter. The PFPD showed higher selectivity, besides reduced gas consumption in the flame, and is very attractive for organotin compound speciation in complex environmental matrices.

In this communication we report a proof of concept study of the use of cyclic voltammetry with a polyeugenol-modified glassy carbon (GC) electrode to selectively detect L-cysteine in the presence of both DL-homocysteine and glutathione in perchloric acid. The formation of a polyeugenol-modified gold electrode is also reported for the first time.

The aim of this paper was to apply a multiresidue method using Supercritical Fluid Extraction (SFE) and capillary gas chromatography with electron capture and mass spectrometry detections in the analysis of the levels of pesticide residues in fruits and vegetables. Single laboratory validation of both solid-liquid and supercritical fluid extraction methods was carried out for 32 compounds selected from four pesticide classes (organochlorine, organonitrogen, organophosphorus and pyretroid) in blank and fortified samples of fresh lettuce, potato, apple and tomato. Recoveries for the majority of pesticides from fortified samples at fortification level of 0.04-0.10 mg kg -1 ranged 74-96% for both methods and confirmation of pesticide identity was performed by gas-chromatography-mass spectrometry in a selected-ion monitoring mode. Both methods showed good limits of detection (less 0.01 mg kg-1, depending on the pesticide and matrix) and the SFE method minimized environmental concerns, time, and laboratory work. ©2005 Sociedade Brasileira de Química.

A study on the presence of herbicides, namely simazine, metribuzin, metolachlor, trifluralin, atrazine and two metabolites, deisopropylatrazine (DIA) and deethylatrazine (DEA), was performed in ground and surface waters from Primavera do Leste region, Mato Grosso state (Middle West of Brazil). The analytical procedure was based on solid-phase extraction (SPE) with Sep-Pak C18 disposable cartridges and ethyl acetate for elution solvent. Residue levels were determined by gas chromatography with nitrogen-phosphorus detection. For most of the pesticides average recoveries at different fortification levels were >with relative standard deviation < The recoveries of DIA and trifluralin in water were 25% and 56%, respectively, which were attributed to the incomplete retention of DIA and strong retention on the sorbing material and high volatility of trifluralin. Detection limits ranged from 0.023 to 0.088 μg L-1. This method was applied for the analysis of 5 superficial water samples and 28 groundwater samples, in places used for human consumption without previous treatment, collected in Primavera do Leste, Mato Grosso, Brazil. Results indicated that the highest level of contamination in a water sample was 1.732 μg L-1 for metolachlor, while metribuzin was the most frequently detected herbicide with maximum concentration of 0.351 μg L-1. ©2006 Sociedade Brasileira de Química.

The use of inks containing organic solvents by the offset printing process implies in the release of volatile organic compounds to the work environment. Many of these compounds such as benzene, toluene, ethylbenzene, and the xylene isomers (well known by the acronym BTEX) are extremely toxic. In this study, the BTEX concentrations were determined in two different printing plants that use distinct types of inks: the conventional and the so-called ecological, which is manufactured based on vegetal oil. Concentration ranges were 43-84, 15-3,480, 2-133, 5-459, and 2-236 μg m-3 for benzene, toluene, ethylbenzene, m + p-xylene, and o-xylene, respectively, for the conventional printing plant. Butyltin (BTs) quantification in environmental matrices can be affected by interfering species found primarily in complex matrices,such as sediment and biota tissues. This study investigated matrix effects in analytical procedures for butyltin (TBT,DBT and MBT) quantification and speciation in sediments and in two fish tissues (gill and liver) by gas chromatography with pulsed flame photometric detection (GC-PFPD) analysis. Unlike sediment samples,tissues exhibited a significant matrix effect,thus,the quantification should be made by curve over matrix to avoid quantification errors and loss of analytical accuracy. © 2013 Sociedade Brasileira de Química.

Esse estudo descreve o desenvolvimento e otimização de um método de extração em fase solida (SPE) para análise dos filtros ultravioletas (UV): benzofenona-3 (BP-3), etilhexil salicilato (ES), etilhexil metoxinamato (EHMC) e octocrileno (OC) em matrizes ambientais. Um planejamento fatorial fracionário (PFF) 25-1 foi empregado na avaliação das variáveis significativas do método de extração. As condições experimentais otimizadas da avaliação estatística foram: capacidade do cartucho de 500 mL, eluente acetato de etila, metanol como solvente de lavagem (10% em água, v/v) and volume do eluente de 3 × 2 mL e pH 3. Os parâmetros analíticos avaliados foram satisfatõrios, apresentando linearidade de 100 a 4000 ng L -1, recuperaç ões para os quatro níveis de fortificação (Limite de Quantificação do Método, 200, 1000 e 2000 ng L-1) entre 62 e 107% com desvio padrão relativo menor que 14%. Os limites de quantificação foram encontrados na faixa de ng L-1, variando entre 10 e 100 ng L-1. O método proposto foi aplicado para a determinação dos quatro filtros UV em amostras de águas naturais. This study describes the development and optimization of a solid-phase extraction (SPE) method for analysis of ultraviolet (UV) filters, benzophenone-3 (BP-3), ethylhexyl methoxycinnamate (EHMC), ethylhexyl salicylate (ES) and octocrylene (OC), in environmental matrices. A 25-1 fractional factorial design (FFD) was used to evaluate the significant variables for the extraction method. The optimized experimental conditions determined from the statistical evaluation were: breakthrough volume of 500 mL, eluent of ethyl acetate, wash solvent of methanol (10% in water, v/v), eluent volume of 3 × 2 mL and pH 3. The evaluated analytical parameters were satisfactory for the analytes and showed linearity between 100 and 4000 ng L-1, recoveries for four fortification levels (Method Quantification Limit, 200, 1000 and 2000 ng L-1) were between 62 and 107% with relative standard deviations less than 14%. Limits of quantification were in the ng L-1 range and were between 10 and 100 ng L-1. Neste estudo, um protocolo para induzir maiores quantidades de calos friáveis de Boerhaavia paniculata RICH e uma técnica de lipidômica foram aplicados para investigar o perfil dos lipídios e serem comparados com aqueles presentes nas raízes desta planta, os quais apresentaram atividade anti-inflamatória no extrato hexânico bruto. A cultura de calo foi induzida a partir de sementes em meio sólido contendo sais de Murashige e Skoog com diferentes quantidades de glicose e diferentes concentrações de ácido 2,4-diclorofenoxiacético. Os frascos foram mantidos em câmara de germinação a 30 ± 2°C, com fotoperíodo de 16h sob intensidade luminosa de 27 µmol m–2 s–1 durante 4 semanas. Os melhores resultados para a formação de calo friável e desenvolvimento da biomassa foram obtidos no tratamento contendo 2,26 µM de 2,4-D e de glicose (1,5 %; m/v). Técnica de lipidômica foi aplicada na fração hexânica mostrando as concentrações mais elevadas de esteróides β-sitosterol (3,53 mg/100 g de cs - células seca ), e ácidos graxos; especialmente o ácido 2-hidroxi-tetracosanóico (0,34 mg/100 g cs), ácido eicosanoico (86,25 mg/100 g cs), ácido esteárico (420,83 mg/100 g cs), ácido tetradecanóico (10,74 mg/100 g cs) e ácido linoléico (100,61 mg/100 g cs). O perfil lipídico do calo versus aquele encontrado nas raízes da planta silvestre é descrito neste trabalho.

O surgimento das plataformas de sequenciamento de nova geração (NGS) proporcionou o aumento do volume de dados produzidos, tornando possível a obtenção de genomas completos. Apesar das vantagens alcançadas com estas plataformas, são observadas regiões de elevada ou baixa cobertura, em relação à média, associadas diretamente ao conteúdo GC. Este viés GC pode afetar análises genômicas e dificultar a montagem de genomas através da abordagem de novo, além de afetar as análises baseadas em referência. Além do que, as maneiras de avaliar o viés GC deve ser adequada para dados com diferentes perfis de relação/associação entre GC e cobertura, tais como linear e quadrático. Desta forma, este trabalho propõe o uso do Coeficiente de Correlação de Pearson (r) para analisar a correlação entre conteúdo GC e Cobertura, permitindo identificar aintensidade da correlação linear e detectar associações não-lineares, além de identificar a relação entre viés GC e as plataformas de sequenciamento. Os sinais positivos e negativos de r também permitem inferir relações diretamente proporcionais e inversamente proporcionais respectivamente. Utilizou-se dados da espécie Corynebacterium pseudotuberculosis, conhecido por serem genomas clonais obtidas através de diferentes tecnologias de sequenciamento para identificar se há relação do viés GC com as plataformas utilizadas.
