This is an Inter-Disciplinary Higher Degree (IHD) thesis about Water Pollution Control in the Iron and Steel Industry. After examining the compositions, and various treatment methods, for the major effluent streams from a typical Integrated Iron and Steel works, it was decided to concentrate investigative work on the activated-sludge treatment of coke-oven effluents. A mathematical model of this process was developed in an attempt to provide a tool for plant management that would enable improved performance, and enhanced control of Works Units. The model differs from conventional models in that allowance is made for the presence of two genera of microorganisms, each of which utilises a particular type of substrate as its energy source. Allowance is also made for the inhibitive effect of phenol on thiocyanate biodegradation, and for the self-toxicity of the bacteria when present in a high substrate concentration environment. The enumeration of the kinetic characteristics of the two groups of micro-organisms was shown to be of major importance. Laboratory experiments were instigated in an attempt to determine accurate values of these coefficients. The use of the Suspended Solids concentration was found to be too insensitive a measure of viable active mass. Other measures were investigated, and Adenosine Triphosphate concentration was chosen as the most effective measure of bacterial populations. Using this measure, a model was developed for phenol biodegradation from experimental results which implicated the possibility of storage of substate prior to metabolism. A model for thiocyanate biodegradation was also developed, although the experimental results indicate that much work is still required in this area.

The component spectra of a mixture of isomers with nearly identical diffusion coefficients cannot normally be distinguished in a standard diffusion-ordered spectroscopy (DOSY) experiment but can often be easily resolved using matrix-assisted DOSY, in which diffusion behaviour is manipulated by the addition of a co-solute such as a surfactant. Relatively little is currently known about the conditions required for such a separation, for example, how the choice between normal and reverse micelles affects separation or how the isomer structures themselves affect the resolution. The aim of this study was to explore the application of sodium dodecyl sulfate (SDS) normal micelles in aqueous solution and sodium 1,4-bis(2-ethylhexyl)sulfosuccinate (AOT) aggregates in chloroform, at a range of concentrations, to the diffusion resolution of some simple model sets of isomers such as monomethoxyphenols and short chain alcohols. It is shown that SDS micelles offer better resolution where these isomers differ in the position of a hydroxyl group, whereas AOT aggregates are more effective for isomers differing in the position of a methyl group. For both the normal SDS micelles and the less well-defined AOT aggregates, differences in the resolution of the isomers can in part be rationalised in terms of differing degrees of hydrophobicity, amphiphilicity and steric effects. Feed characteristics may influence the bacterial community composition and metabolic activities in the pig gastrointestinal tract, known to be associated with positive effects on the gut. Use of mash feed is associated with reduced Salmonella excretion, but little is known of its effect on the Escherichia coli population or of the mechanism of action. Our objectives were to assess the effect of feed texture combined with feed particle size on VFA profiles and levels, total E. coli count, and the presence of genes encoding virulence factors of pathogenic E. coli strains in the digestive tract along with their impact on pig performance of fattening pigs. Pigs (n = 840) on a commercial farm received mash or pellet diets of different particle sizes during the fattening period. Caecal and colon contents from 164 pigs were sampled at the slaughterhouse for enumeration of E. coli by quantitative PCR (qPCR) and for VFA quantification by capillary gas chromatography. The yccT gene was used to enumerate total E. coli. Improved pig performances associated with pellet texture and a 500-μm size were observed. Caecal (P = 0.02) and colon (P < 0.01) propionic acid concentrations were lower for pigs receiving pellet rather than mash feed. Similarly, caecal (P = 0.01) and colon (P < 0.001) butyric acid concentrations were also lower for pigs receiving pellet rather than mash feed, as determined by capillary gas chromatography. Moreover, caecal (P = 0.03) and colon (P < 0.001) butyric acid concentrations were higher for pigs receiving a feed with a 1,250-μm particle size rather than a 500-μm particle size. On the other hand, total caecal and colon E. coli levels were higher for pigs receiving pellet feed than for those receiving mash feed. For total E. coli enumeration, caecal (P < 0.01) and colon (P < 0.01) yccT gene copies were higher for pigs receiving pellet rather than mash feed. No effect of particle size on fatty acid concentrations or on E. coli numbers was observed. Virulence gene quantification revealed no trend. Taken together, results showed that mash feed is associated with lower growth performance but with favorable intestinal changes linked to VFA levels and E. coli reduction in the intestine.

Symbolic execution is a powerful program analysis technique, but it is very challenging to apply to programs built using event-driven frameworks, such as Android. The main reason is that the framework code itself is too complex to symbolically execute. The standard solution is to manually create a framework model that is simpler and more amenable to symbolic execution. However, developing and maintaining such a model by hand is difficult and error-prone. We claim that we can leverage program synthesis to introduce a high-degree of automation to the process of framework modeling. To support this thesis, we present three pieces of work. First, we introduced SymDroid, a symbolic executor for Android. While Android apps are written in Java, they are compiled to Dalvik bytecode format. Instead of analyzing an app’s Java source, which may not be available, or decompiling from Dalvik back to Java, which requires significant engineering effort and introduces yet another source of potential bugs in an analysis, SymDroid works directly on Dalvik bytecode. Second, we introduced Pasket, a new system that takes a first step toward automatically generating Java framework models to support symbolic execution. Pasket takes as input the framework API and tutorial programs that exercise the framework. From these artifacts and Pasket's internal knowledge of design patterns, Pasket synthesizes an executable framework model by instantiating design patterns, such that the behavior of a synthesized model on the tutorial programs matches that of the original framework. Lastly, in order to scale program synthesis to framework models, we devised adaptive concretization, a novel program synthesis algorithm that combines the best of the two major synthesis strategies: symbolic search, i.e., using SAT or SMT solvers, and explicit search, e.g., stochastic enumeration of possible solutions. Adaptive concretization parallelizes multiple sub-synthesis problems by partially concretizing highly influential unknowns in the original synthesis problem. Thanks to adaptive concretization, Pasket can generate a large-scale model, e.g., thousands lines of code. In addition, we have used an Android model synthesized by Pasket and found that the model is sufficient to allow SymDroid to execute a range of apps.

In recent years, security of industrial control systems has been the main research focus due to the potential cyber-attacks that can impact the physical operations. As a result of these risks, there has been an urgent need to establish a stronger security protection against these threats. Conventional firewalls with stateful rules can be implemented in the critical cyberinfrastructure environment which might require constant updates. Despite the ongoing effort to maintain the rules, the protection mechanism does not restrict malicious data flows and it poses the greater risk of potential intrusion occurrence. The contributions of this thesis are motivated by the aforementioned issues which include a systematic investigation of attack-related scenarios within a substation network in a reliable sense. The proposed work is two-fold: (i) system architecture evaluation and (ii) construction of attack tree for a substation network. Cyber-system reliability remains one of the important factors in determining the system bottleneck for investment planning and maintenance. It determines the longevity of the system operational period with or without any disruption. First, a complete enumeration of existing implementation is exhaustively identified with existing communication architectures (bidirectional) and new ones with strictly unidirectional. A detailed modeling of the extended 10 system architectures has been evaluated. Next, attack tree modeling for potential substation threats is formulated. This quantifies the potential risks for possible attack scenarios within a network or from the external networks. The analytical models proposed in this thesis can serve as a fundamental development that can be further researched.

The molecular profiling system was developed using directed terminal-restriction fragment length polymorphism (dT-RFLP) to characterize soil nematode assemblages by relative abundance of feeding guilds and validation by comparison to traditional morphological method. The good performance of these molecular tools applied to soil nematodes assemblages create an opportunity to develop a novel approach for rapid assessment of the biodiversity changes of benthic nematodes assemblages of marine and estuarine sediments. The main aim of this research is to combine morphological and molecular analysis of estuarine nematodes assemblages, to establish a tool for fast assessment of the biodiversity changes within habitat recovery of Zostera noltii seagrass beds; and validate the dT-RFLP as a high-throughput tool to assess the system recovery. It was also proposed to develop a database of sequences related to individuals identified at species level to develop a new taxonomic reference system. A molecular phylogenetic analysis of the estuarine nematodes has being performed. After morphological identification, barcoding of 18S rDNA are being determined for each nematode species and the results have shown a good degree of concordance between traditional morphology-based identification and DNA sequences. The digest strategy developed for soil nematodes is not suitable for marine nematodes. Then five samples were cloned and sequenced and the sequence data was used to design a new dT-RFLP strategy to adapt this tool to marine assemblages. Several solutions were presented by DRAT and tested empirically to select the solution that cuts most efficiently, separating the different clusters. The results of quantitative PCR showed differences in nematode density between two sampling stations according the abundance of the nematode density obtained by the traditional methods. These results suggest that qPCR could be a robust tool for enumeration of nematode abundance, saving time.

An absolute method is described via mass spectrometry (MS) for the structural assignment of isomers within the class of methylpiperidines. The method explores both the unimolecular and bimolecular gas phase behavior of structurally diagnostic fragment ions (SDFI). For the methylpiperidnes, the isomeric 2-methyl, 3-methyl and 4-methyl 2-azabutadienyl cations are found to function as SDFI. These fragment ions are expected to be formed from all members within the class, to be stable and to retain the structural information of the precursor molecule, and to not interconvert into one another. To characterize these SDFI, both the collision induced dissociation (CID) in argon and bimolecular ion/molecule chemistry with ethyl vinyl ether were compared.

This work aimed at determining the occurrence of heat resistant molds during the aseptic processing of tomato pulp (8° BRIX). During tomato harvest, 9 lots were sampled (3 at the beginning, 3 at the apex and 3 at the end of harvest) and other 5 lots were sampled between harvest. For each lot, the enumeration of heat resistant molds was carried out in samples collected during the aseptic process. The mean count of heat resistant molds was relatively low, ranging from <1 to 8CFU/100mL of sample. The higher counts were observed in the raw material and the pre-wash and transportation water. Fifty strains of heat resistant molds detected in the enumeration procedure were isolated, codified and stocked. One-month-old spores of each isolate were submitted to different heat shocks to select the most heat resistant mold. The most heat resistant isolated strain (survived 100° C/25 minutes) was identified as Neosartorya fischeri.

Ao mensurar-se mortalidade materna, é necessário distinguir ' mortes por causas maternas' e 'mortes maternas' Para a Organização Mundial da Saúde-OMS-,mortes maternas são as que ocorrem na gestação, no parto e até 42 dias após o parto; e mortes por causas maternas englobam as causas classificadas no Capítulo XV da Classificação Estatística Internacional de Doenças e Problemas Relacionados à Saúde, Décima Revisão (CID-10), incluindo as ocorridas quando passados 42 dias do parto. Apresentam-se resultados da investigação de mortes femininas em idade fértil-10 a 49 anos- nas capitais de Estados e no Distrito Federal do Brasil, em 2002. Adotou-se a metodologia RAMOS, comparando-se as causas básicas das declarações de óbito originais com as das declarações preenchidas após o resgate de informações, obtidas em entrevistas domiciliares e prontuários. Entre as mortes por causas maternas originais, 15,9 por cento não eram mortes maternas, de acordo com a definição da OMS. Houve, concomitantemente, subenumeração de mortes maternas. Sugestões são feitas para melhorar o preenchimento das declarações de óbito e inclusão de novas categorias na CID-10, visando melhorar a informação das causas maternas

A partir das contribui????es de Osborne e Gaebler (1992) e Abrucio (2006), buscou-se tra??ar um paralelo entre as l??gicas gerencial e fiscal e esclarecer como as mesmas refletiram nos indicadores de efic??cia, efici??ncia e efetividade do Poder Judici??rio no Brasil. Para atingir o objetivo proposto, optou-se por utilizar como m??todo a an??lise de conte??do de Bardin (2002), pela regra de enumera????o do tipo frequ??ncia, buscando, com isso, observar o papel que o Conselho Nacional de Justi??a exerceu nesse processo e a l??gica do plano de metas estabelecido pela institui????o para os anos de 2012 e 2013. Como resultado, observou-se que as metas estabelecidas para o bi??nio buscaram implantar uma gest??o mais eficaz nesse n??cleo estrat??gico do Estado, com ??nfase na celeridade processual, gest??o administrativa e informatiza????o dos procedimentos. Percebeu-se que a l??gica gerencial continua exercendo grande influ??ncia no modelo de gest??o adotado. Como novidade, verificou-se a inser????o de indicadores de efetividade, os quais podem permitir aproximar os objetivos dessa esfera de poder com os da sociedade brasileira, repercutindo na elabora????o de metas futuras.

O presente Relatório de Estágio teve como objectivo analisar e discutir as competências adquiridas e desenvolvidas, durante o estágio, no controlo da qualidade microbiológica na indústria farmacêutica O estágio, foi realizado no Laboratório de Microbiologia dos Laboratórios Atral, Grupo AtralCipan, localizado na Vala do Carregado, Castanheira do Ribatejo. Este teve a duração de aproximadamente dez meses e meio, com data de início a 21 de Setembro de 2011 e final de 10 de Agosto de 2012. Numa fase inicial, foram adquiridas as aptidões necessárias para a aplicação das metodologias realizadas no laboratório de microbiologia com o estudo das normas, procedimento e legislação aplicada, com foco na importância das farmacopeias na indústria farmacêutica e formação para a realização de ensaios em Áreas de Processamento Asséptico, conhecendo e compreendendo os procedimentos a ter nestas. Uma das principais metodologias realizadas, e desenvolvida neste relatório, compreendeu a análise de produtos farmacêuticos estéreis, com a aplicação de Testes de Esterilidade, pelo método de STERITEST e método Directo, com resultados que demonstraram a importância destas técnicas, da avaliação do ambiente em que são realizadas e do operador que as executa. Os Testes de Promoção de Crescimento, foram também explorados neste trabalho, realizados não só para avaliação dos meios de cultura, onde foi possível analisar os requisitos e resultados obtidos, como também para validação de metodologias, nomeadamente, na validação de meios e fluidos de marcas diferentes para utilização no método STERITEST e na validação do método de filtração de membrana para Enumeração Microbiana de um produto não estéril. Estas validações possibilitaram a redução de custos e melhoria das condições da análise e metodologias aplicadas. Com a realização do estágio foi possível adquirir aptidões práticas, que aliadas ao conhecimento teórico obtido no mestrado, proporcionaram um crescimento a nível pessoal, científico e profissional.

O presente relatório é a última etapa para a obtenção do Grau de Mestre em Tecnologia e Gestão das Construções leccionado no Instituto Superior de Engenharia do Porto, (ISEP). O estágio decorreu em ambiente maioritariamente de obra na empresa Monte Adriano, Engenharia e Construção, SA, e o objectivo consistiu em dar apoio e acompanhar a Obra de Ampliação do Reservatório de Custóias, inserida na Empreita de concepção, projecto e construção das infraestruturas necessárias à execução do plano de investimentos da Indáqua Matosinhos, actividades estas, decorrentes da função de adjunta de direcção de obra. Para além da breve descrição da empresa que a estagiária integrou e da empreitada geral, foi feita uma enumeração das várias actividades desempenhadas na Obra e dos vários condicionalismos encontrados que por vezes levaram a alterações do projecto. Dá-se especial ênfase à parte da execução das fundações, quer pela heterogeneidade do solo e sua relevância no geral, quer pelo facto do tempo de execução da obra ser praticamente o tempo do estágio. A obra consiste na execução de três células em painéis pré-fabricados SOPLACAS (paredes e cobertura), sendo que, as lajes de soleira de duas das células apoiam em poços e a laje de soleira da outra célula apoia directamente no terreno e em poços. Anexa às células encontra-se a câmara de manobras, executada em estrutura de betão armado.

A indústria automóvel exige, em geral, elevados índices de produtividade e qualidade. Para corresponder às exigências deste tipo de indústria, são requeridos métodos avançados de produção, tentando eliminar ao máximo operações que não gerem valor acrescentado e que possam introduzir problemas no processo de garantia da qualidade. A maquinagem por arranque de apara é um processo utilizado de forma intensiva na indústria automóvel. No entanto, enquanto em componentes críticos como o motor, os processos estão já altamente otimizados, o mesmo não se verifica na maior parte dos sistemas periféricos, normalmente realizados por empresas mais pequenas que gravitam em torno dos principais fornecedores da cadeia de produção de automóveis. Os sistemas responsáveis pela movimentação dos limpa pára-brisas e elevação dos vidros, entre outros, encontram-se neste grupo. Este trabalho visa essencialmente otimizar o processo de maquinagem de componentes periféricos de automóveis, sujeitos a diferentes operações em vários planos. No entanto, e tal como em muitas outras situações relacionadas com a variedade de versões existentes na indústria automóvel relativamente a cada sistema, pretende-se que o processo seja suficientemente versátil para poder ser aplicado em vários componentes de uma mesma família de produtos, necessitando de um número de ajustes o mais baixo possível. O estudo passou por uma análise profunda das similaridades geométricas dos diferentes componentes, análise dos planos de maquinagem de cada componente, operações envolvidas, elencagem da necessidade específica de ferramentas, elaboração de gabaritos de fabrico e apresentação da solução final, a qual passa pela introdução de um 4º eixo e do seu controlo através do sistema CNC já existente, assim como pela elaboração de novos programas.

O estudo avaliou a utilização do escore POSSUM (Physiological and Operative Severity Score for Enumeration of Mortality and Morbidity) para predizer a mortalidade na prática cirúrgica.Foram analisados 416 pacientes cirúrgicos com internação na UTI para cuidados de pós-operatório. Foram realizadas comparações entre as taxas de mortalidade predita e observada de acordo com 4 grupos de risco: 0-4%, 5-14%, 15-49%, 50% ou mais, e calculada a área sob a curva ROC do escore POSSUM e APACHE II para a mortalidade. A taxa de mortalidade foi de 22,4%. O escores POSSUM e APACHE II superestimaram o risco de morte, e a área sob a curva ROC do POSSUM foi de 0,762 e a do APACHE II de 0,737, sugerindo a utilização do POSSUM como ferramenta auxiliar na predição de risco de morte em pacientes cirúrgicos.
